AB  - The Pd-catalyzed N-arylation method for the synthesis of eighteen N,1-diaryl-1H-tetrazol-5-amine derivatives is reported. By running the reactions at 35 °C, compounds were isolated as single isomers since the undesired Dimroth rearrangement was completely suppressed. Furthermore, the Dimroth rearrangement of N,1-diaryl-1H-tetrazol-5-amines was rationalized by conducting comprehensive experiments and NMR analysis as well as density functional theory (DFT) calculations of thermodynamic stability of the compounds. It was established that the Dimroth rearrangement is thermodynamically controlled, and the equilibrium of the reaction is determined by the stability of the corresponding isomers. The mechanism was investigated by additional DFT calculations, and the opening of the tetrazole ring was shown to be the rate-determining step. By maneuvering Pd-catalyzed N-arylation and the subsequent Dimroth rearrangement, two more N,1-diaryl-1H-tetrazol-5-amine derivatives were acquired, which otherwise cannot be synthesized by employing the C–N cross-coupling reaction.

Triplet state (anti)aromaticity of some monoheterocyclic analogues of benzene, naphthalene and anthracene

AB  - Aromaticity–antiaromaticity switch upon singlet–triplet transition of some biologically and synthetically important monoheterocycles (heteroatom = N, O, Si, P, and S) derived from benzene, naphthalene and anthracene was studied by employing energetic, magnetic and structural aromaticity criteria, at the density functional theory (DFT) level. The relationship between spin density distribution, (anti)aromaticity and singlet–triplet energy gaps, in the studied molecules, was found. In general, spin delocalization results in antiaromaticity, spin density localization to one ring in bi- and tricycles localizes antiaromaticity and spin localization on a heteroatom reduces global and local antiaromaticity. The latter reaches nonaromaticity in the case of silicon atoms which have larger orbitals and show more tendency to accept unpaired electrons. Spin density localization in bi- and tricycles allows benzene subunit(s) to develop local aromaticity, which, when combined with nonaromatic silacycle and weak global antiaromaticity, results in overall triplet state weak aromaticity. The singlet–triplet energy gaps decrease with a decrease in the triplet state antiaromaticity and are the lowest for silicon-containing compounds.

AB  - Serological testing is important method for diagnosis of severe acute respiratory syndrome coronavirus 2 (SARS-CoV-2) infection. Nucleocapsid (N) protein is the most abundant virus derived protein and strong immunogen. We aimed to find its efficient, low-cost production.

SARS-CoV-2 recombinant fragment of nucleocapsid protein (rfNP; 58–419 aa) was expressed in E. coli in soluble form, purified and characterized biochemically and immunologically.

Purified rfNP has secondary structure of full-length recombinant N protein, with high percentage of disordered structure (34.2%) and of β-sheet (40.7%). rfNP was tested in immunoblot using sera of COVID-19 convalescent patients. ELISA was optimized with sera of RT-PCR confirmed positive symptomatic patients and healthy individuals. IgG detection sensitivity was 96% (47/50) and specificity 97% (67/68), while IgM detection was slightly lower (94% and 96.5%, respectively).

Cost-effective approach for soluble recombinant N protein fragment production was developed, with reliable IgG and IgM antibodies detection of SARS-CoV-2 infection.

AB  - The main goal of this study was to establish the chemical profile of Osage orange (Maclura pomifera) leaf extracts, obtained by conventional maceration technique, and to examine its antimicrobial activity. The identification and quantification of the extract compounds were done using ultra‐high‐performance liquid chromatography, with a diode array detector coupled with triple‐quadrupole mass spectrometer and gas chromatography–mass spectrometry techniques. Thirty‐one polyphenolic compounds were detected and identified in the ethanolic extracts, whereby 5‐O‐caffeoylquinic acid was found to be the dominant compound. Among other compounds, pentacosane and palmitic acid were the most abundant compounds in the dichloromethane extract. The preliminary antimicrobial activity screening shows that Gram‐positive bacteria tend to be more sensitive to the investigated extracts. The highest antimicrobial activity was determined against Enterococcus faecalis ATCC 19433 and Listeria monocytogenes ATCC 35152. From these results, Osage orange leaves can be considered as plant material with significant antimicrobial properties.

AB  - Demand for camel milk (CM) is increasing worldwide, due to its high nutritious value and health benefits. In this study, whole CM powders were produced by spray drying (SD) at six inlet temperatures (190 °C–250 °C) and by freeze drying (FD). Physicochemical and functional properties of CM powder proteins were investigated. SD at higher inlet temperatures (230 °C–250 °C) resulted in higher extent of Maillard reaction (MR), in comparison to lower temperatures (190 °C–200 °C) and FD treatment. Both treatments had negative effect on casein solubility, while whey proteins remained soluble and slightly increased its solubility with the extent of MR. The CM powders obtained at higher inlet temperatures demonstrated improved antioxidant activity. Secondary structure of whey proteins did not differ among the samples, while surface hydrophobicity of whey proteins was higher in all SD than in FD samples, suggesting only limited denaturation of camel whey proteins at higher inlet temperatures of drying. Thus, the effects of SD under the conditions applied in our study did not decrease camel whey protein solubility, while drying procedure itself regardless of temperature decreased solubility of camel milk caseins. MR generated during CM processing could be an important means of compensating for the lack of antioxidant protection normally associated with β-lactoglobulin but happens to be absent from this milk.

SD at higher inlet temperatures (230 °C–250 °C) resulted in higher extent of Maillard reaction (MR), in comparison to lower temperatures (190 °C–200 °C) and FD treatment. Both treatments had negative effect on casein solubility, while whey proteins remained soluble and slightly increased its solubility with the extent of MR. The CM powders obtained at higher inlet temperatures demonstrated improved antioxidant activity. Secondary structure of whey proteins did not differ among the samples, while surface hydrophobicity of whey proteins was higher in all SD than in FD samples, suggesting only limited denaturation of camel whey proteins at higher inlet temperatures of drying. Thus, the effects of SD under the conditions applied in our study did not decrease camel whey protein solubility, while drying procedure itself regardless of temperature decreased solubility of camel milk caseins. MR generated during CM processing could be an important means of compensating for the lack of antioxidant protection normally associated with β-lactoglobulin but happens to be absent from this milk.

AB  - Serological testing is important method for diagnosis of severe acute respiratory syndrome coronavirus 2 (SARS-CoV-2) infection. Nucleocapsid (N) protein is the most abundant virus derived protein and strong immunogen. We aimed to find its efficient, low-cost production.SARS-CoV-2 recombinant fragment of nucleocapsid protein (rfNP; 58–419 aa) was expressed in E. coli in soluble form, purified and characterized biochemically and immunologically.Purified rfNP has secondary structure of full-length recombinant N protein, with high percentage of disordered structure (34.2%) and of β-sheet (40.7%). rfNP was tested in immunoblot using sera of COVID-19 convalescent patients. ELISA was optimized with sera of RT-PCR confirmed positive symptomatic patients and healthy individuals. IgG detection sensitivity was 96% (47/50) and specificity 97% (67/68), while IgM detection was slightly lower (94% and 96.5%, respectively).Cost-effective approach for soluble recombinant N protein fragment production was developed, with reliable IgG and IgM antibodies detection of SARS-CoV-2 infection.

AB  - Twenty-six major and trace elements, in the seed and the mesocarp of wild and cultivated rosehips from different locations in Serbia, were quantified by means of inductively coupled plasma–atomic emission spectroscopy (ICP–AES). The cultivated rosehip was investigated for the first time. In both wild and cultivated rosehips, the most abundant elements were K and Ca. Among trace elements, Mn (in both seed and mesocarp); Fe (in seeds); and B, Ba, and Sr (in mesocarp) were quantified in the highest concentrations. The higher content of Cu, K, Mn, P, and S in the seed of cultivated rosehip, as well as Ca, Mg, and Sr in the mesocarp of wild rosehip, was observed, both significant at p < 0.05. Additionally, differences between the seed and the mesocarp of studied rosehip were noticed in the content of B, Cu, Fe, K, Ni, P, S, Sr, and Ti (p < 0.05). Nutritional assessment revealed that both wild and cultivated rosehips are a valuable source of essential elements (Ca, Cu, K, Mg, Mn, and P). The absence of toxic and potentially toxic elements additionally contributes to the quality of studied Serbian rosehip. A high impact of factors such as variety, location, as well as their interaction on the content of elements in cultivated rosehip mesocarp and seed was observed. Pattern recognition techniques, principal component analysis (PCA), and hierarchical cluster analysis (HCA) were applied in order to provide insight into similarities among the analyzed samples.

abstract = "Zemljište predstavlja značajan izvor kako nutrijenata, tako i potencijalno toksičnih supstanci za biljke. Putem lanca ishrane toksična jedninjenja vrlo lako mogu dospeti do čoveka. Neki od policikličnih aromatičnih ugljovodonika (eng., polycyclic aromatic hydrocarbons, PAH) zvanično pripadaju grupi dokazanih kancerogena za ljude, dok je većina potencijalno kancerogena. Upravo je zbog toksičnog, kancerogenog, mutagenog i teratogenog dejstva njihova analiza u uzorcima iz životne sredine od velikog značaja. Američka agencija za zaštitu životne sredine i Evropska unija odredile su 16 PAH jedninjenja čije se koncentracije u životnoj sredini nužno prate. Njihova analiza predstavlja veliki izazov zbog male koncentracije ovih jedinjenja u uzorcima, slične strukture pojedinih jedinjenja i nedovoljne selektivnosti organskih rastvarača. 
U ovom radu je urađena analiza PAH jedinjenja i procena porekla u uzorcima koji potiču iz Slovačke Republike. Analizirani uzorci su zemljište i materijal biljnog porekla, pri čemu u okviru materijala biljnog porekla bilo je dva seta poduzoraka: biljni materijal i listići.",

Na taj način efikasnost reakcije višestruko je povećana, što se vidi u smanjenju vremena reakcije (na 3-10 min u odnosu na 24 h) i količine reagenasa u poređenju sa našim prethodnim istraživanjem.
Sintetisano je ukupno 12 jedinjenja. Osim metil karbamata, sintetisani su i benzil karbamati, koji su kao što je poznato u literaturi, veoma značajni kao zaštitne grupe za amine. -Amino karboksamidi dali su ciklične uree, u skladu sa prethodno dobijenim rezultatima.
Reakcija je, u načelu, pokazala veoma dobre rezultate sa sterno nezaštićenim alifatičnim i aktiviranim aromatičnim karboksamidima, a pod ovim reakcionim uslovima nisu primećeni sporedni proizvodi oksidativnog raspada ili aromatičnog bromovanja.
U svim slučajevima očekivani metil ili benzil karbamati izolovani su u vrlo dobrim prinosima, najčešće bez potrebe za hromatografskim prečišćavanjem. Konverzija reaktanata je bila potpuna, a prisustvo sporednih proizvoda minimalno (0-5%). Stoga se može tvrditi da je metod premeštanja po Hofmann-u, primenom kombinacije NBA/LiOH.H2O, MeOH ili BnOH, uz kratkotrajno zagrevanje značajno bolji od poznatih literaturnih metoda.",

abstract = "Kancer je maligno oboljenje koje je, pored kardiovaskularnih oboljenja, vodeći uzrok smrtnosti u svetu (Khan, Reale, et al. 2019). Mutacija unutar gena TP53, koji kodira protein p53 je najĉešći uzrok nastanka kancera (Mantovani, Collavin et al. 2019).
Protein p53 je tumorski supresor koji se veoma ĉesto naziva i “ĉuvarom genoma” kako bi se naglasila njegova izuzetno važna uloga u spreĉavanju nastanka kancera. Svoju ulogu ostvaruje kroz mnogobrojne procese unutar ćelije kao što su: blokiranje ćelijskog ciklusa, apoptoza, proces popravke DNK molekula i drugo. Gubitak ovih funkcija za posledicu ima nastanak kancerskih ćelija. Najĉešće mutacije dešavaju se u okviru DNK – vezujućeg domena proteina p53 (De Oliveira, Petronilhno et al., 2020). Poslednjih godina se izuĉavaju pristupi u terapiji kancera koji se zasnivaju na stabilizaciji p53 razliĉitim molekulima koji bi mu vratili prvobitnu funkciju i poboljšali stabilnost.
U ovom radu je izolovan i preĉišćen rekombinantni protein p53 iz E. coli i ispitivani su uslovi u kojima je protein stabilan i funkcionalan. Ispitivane su interakcije proteina p53 i benzoil - akrilne kisleline, kao i interakcije p53 i 1-4-(4-hlorofenil)-4-oksobut-2-enoinske kiseline, nazivane hloro – derivatom dalje u radu.",

AB  - Novel polyurethane nanocomposite (PUN) materials containing different surface-functionalized mesoporous silica nanoparticles (MSNs) were prepared by in situ polymerization methodology. Polyurethane network was formed from poly(dimethylsiloxane)-based macrodiol (PDMS), 4,4′-methylenediphenyldiisocyanate (MDI), and hyperbranched polyester of the second pseudo-generation (BH-20; used as crosslinking agent). PU and PU/MSN nanocomposites contained equal ratios of soft PDMS and hard MDI-BH-20 segments. Non-functionalized and surface-functionalized (with 3-(trihydroxysilyl)propyl methylphosphonate (FOMSN) and 2-[methoxy(polyethyleneoxy)6−9propyl]trimethoxysilane (PEGMSN)) MSNs were used as the nanofillers at a concentration of 1 wt%. Prepared materials were characterized by Fourier transform infrared (FTIR) spectroscopy, atomic force microscopy (AFM), scanning electron microscopy (SEM), thermogravimetric analysis (TGA), differential scanning calorimetry (DSC), dynamic mechanical thermal analyses (DMTA), nanoindentation, equilibrium swelling and water absorption measurements. Characteristics of the prepared PUNs when in contact with a biological environment were assessed through testing their biocompatibility, protein adsorption and adhesion of endothelial cells. The favourable influence of MSNs on the physico-chemical and biological characteristics of these novel PUN materials was identified, which evidences their vast applicability potential as coatings for medical devices and implants.

abstract = "Cilj ovog rada je bio ekspresija lakaze na površini ćelija Saccharomyces cerevisiae liziranje
dobijenih ćelija organskim rastvaračem sa ciljem povećanja specifične en zimske aktivnosti i njihova
imobilizacija . Imobilizacija je vršena u kuglicama L dopa alginata, sintetisanog u reakcijama
perjodatne okcidacije, a zatim i reduktivne aminacije sa L hidroksifenilalaninom (L dopom). Dobijeni
derivat alginata je okarakterisan UV Vis spektroskopijom i korišćen za imobilizaciju enzima. Dobijene
kuglice sa lakazom na ćelijskim zidovima su pored jonskog umrežavanja kalcijumom dodatno
fotoumrežavane kovalentno sa eozinom Y, u prisustvu led zelenog svetla. Upoređena je specifična
ak tivnost enzima u kuglicama bez dodatnog fotoumreženja i sa fotoumreženjem, kao i temperaturna
stabilnost ovih imobilizata.",
