AB  - Varroa destructor is a parasitic mite responsible for the loss of honey bee (Apis mellifera) colonies. This study aimed to find a promising marker in honey for the bee colony infestation level using fluorescence spectroscopy and biochemical analyses. We examined whether the parameters of the honey samples’ fluorescence spectra and biochemical parameters, both related to proteins and phenolics, may be connected with the level of honey bee colonies’ infestation. The infestation level was highly positively correlated with the catalase activity in honey (r = 0.936). Additionally, the infestation level was positively correlated with the phenolic spectral component (r = 0.656), which was tentatively related to the phenolics in honey. No correlation was found between the diastase activity in honey and the colonies’ infestation level. The results indicate that the catalase activity in honey and the PFC1 spectral component may be reliable markers for the V. destructor infestation level of the colonies. The obtained data may be related to the honey yield obtained from the apiaries.

Intrinsic Fluorescence Markers for Food Characteristics, Shelf Life, and Safety Estimation: Advanced Analytical Approach

AB  - Food is a complex matrix of proteins, fats, minerals, vitamins, and other components. Various analytical methods are currently used for food testing. However, most of the used methods require sample preprocessing and expensive chemicals. New analytical methods are needed for quick and economic measurement of food quality and safety. Fluorescence spectroscopy is a simple and quick method to measure food quality, without sample preprocessing. This technique has been developed for food samples due to the application of a front-face measuring setup. Fluorescent compounds–fluorophores in the food samples are highly sensitive to their environment. Information about molecular structure and changes in food samples is obtained by the measurement of excitation–emission matrices of the endogenous fluorophores and by applying multivariate chemometric tools. Synchronous fluorescence spectroscopy is an advantageous screening mode used in food analysis. The fluorescent markers in food are amino acids tryptophan and tyrosine; the structural proteins collagen and elastin; the enzymes and co-enzymes NADH and FAD; vitamins; lipids; porphyrins; and mycotoxins in certain food types. The review provides information on the principles of the fluorescence measurements of food samples and the advantages of this method over the others. An analysis of the fluorescence spectroscopy applications in screening the various food types is provided.

Synthesis and characterization of luminescent Cu2+–doped fluorapatite nanocrystals as potential broad–spectrum antimicrobial agents

AB  - Nanomaterials based on metal–doped fluorapatite (FAP) have attracted considerable interest as potential next–generation antimicrobial agents. In this study, Cu2+–doped FAP nanocrystals have been successfully synthesized by a neutralization method at room temperature. Their structural, optical, antimicrobial, and hemcompatible properties have been investigated. XRD, FTIR, FESEM, and N2 adsorption–desorption studies indicate the formation of single–phase FAP mesoporous nanopowders, composed of rod–like particles. TEM images confirmed the formation of nanorodes with a length of 60 nm and a width of about 18 nm. Rietveld analysis shows that the Cu2+ ions preferentially substitute Ca2 (6 h) sites in the hexagonal fluorapatite crystal structure. Fluorescence spectroscopy accompanied by MCR–ALS method confirms substitution of Cu2+ ions in FAP crystal lattice with extracting additional d–d band transition at green color from FAP broadband self–activated luminescence in violet–blue color. Antimicrobial studies conducted on Staphylococcus aureus, Escherichia coli and Micrococcus lysodeikticus showed that FAP nanopowder with the highest Cu2+ content have strong bacteriostatic action on Staphylococcus aureus bacterial strain in mediums containing nutrition matters. In addition, this sample in comparison to pure FAP achieved a high percentage of relative reduction of bacterial population for all three species, being >90% in most cases. Fungistatic action is noticed too, throwgh the slowing down mycelium growth of fungus Aspergillus niger, Aspergillus flavus and Penicillium roqueforti and reduction of sporulation of Aspergillus niger species. Cu2+–doped FAP nanocrystals shows a synergistic antimicrobial effect with Cu2+ and F− ions. Concerning the potential biomedical applications, the hemolysis ratios of the Cu2+–doped FAP samples were below 5%. The obtained results pointed out the possible use of the synthesized nanocrystals as broad–spectrum antimicrobial agents for various biomedical and health care preparations.

Lignin and organic free radicals in maize (Zea mays L.) seeds in response to aflatoxin B1 contamination: an optical and EPR spectroscopic study

The aim was the screening of semi-volatile organic compounds from plants’ shoots using comprehensive two-dimensional gas chromatography–mass spectrometry, a powerful separation technique allowing to reach unique separation resolution, and investigation of qualitative changes after the treatment with coated cerium oxide nanoparticles. The results were analyzed by the identification of individual peaks and fingerprint analysis by image processing. Wheat samples contained a higher number of semi-volatile organic compounds (108) compared to pea (77) but were less affected by the treatments with coated nanoparticles. The highest number of compounds was detected in wheat after the treatment with levan- and pullulan-coated nanoparticles, and in pea after treatment with levan-coated nanoparticles. This article reports a successful application of a semi-volatile organic compounds profile presented only as categorical variables and unique fingerprint images for the inter-cultivar recognition. This method may be useful in screening nanoparticles’ effects on different plants.

AB  - Cerium oxide nanoparticles (nCeO2) have widespread applications, but they can be hazardous to the environment. Some reports indicate the toxic effect of nCeO2 on tested animals, but literature data are mainly contradictory. Coating of nCeO2 can improve their suspension stability and change their interaction with the environment, which can consequently decrease their toxic effects. Herein, the exopolysaccharides levan and pullulan, due to their high water solubility, biocompatibility, and ability to form film, were used to coat nCeO2. Additionally, the monosaccharide glucose was used, since it is a common material for nanoparticle coating. This is the first study investigating the impact of carbohydrate-coated nCeO2 in comparison to uncoated nCeO2 using different model organisms. The aim of this study was to test the acute toxicity of carbohydrate-coated nCeO2 on the bacterium Vibrio fischeri NRRL B-11177, the crustacean Daphnia magna, and zebrafish Danio rerio. The second aim was to investigate the effects of nCeO2 on respiration in Daphnia magna which was performed for the first time. Finally, it was important to see the relation between Ce bioaccumulation in Daphnia magna and Danio rerio and other investigated parameters. Our results revealed that the coating decreased the toxicity of nCeO2 on Vibrio fischeri. The coating of nCeO2 did not affect the nanoparticles’ accumulation/adsorption or mortality in Daphnia magna or Danio rerio. Monitoring of respiration in Daphnia magna revealed changes in CO2 production after exposure to coated nCeO2, while the crustacean's O2 consumption was not affected by any of the coated nCeO2. In summary, this study revealed that, at 200 mg L−1, uncoated and carbohydrate-coated nCeO2 are not toxic for the tested organisms, however, the CO2 production in Daphnia magna is different when they are treated with coated and uncoated nCeO2. AB  - BACKGROUND: Cereal seeds, such as maize seeds, are frequently contaminated with aflatoxin B1 (AFB1), one of the most dangerous naturally occurring carcinogens. In plants, phenolamides are involved in biotic stress response. The data on variations of phenolamides in AFB1-containing seeds are limited. RESULTS: Five polyamine conjugates, including two spermidine and three putrescine conjugates, were tentatively identified in methanolic extracts, using HPLC-DAD-MS. The ratio of putrescine to spermidine conjugates changed with increasing AFB1 concentration in a logistic dose–response manner, with a ratio of below 1 up to a concentration of 51.51 μg kg−1, and approximately 2.54 and 3 at higher concentrations of 177.4 and 308.13 μg kg−1, respectively. The observed variations of the total antioxidant activity and the total phenolic content may support this biphasic behaviour of the seeds against AFB1 stress. CONCLUSIONS: The obtained data are a contribution to the understanding of the roles of polyamine conjugates in seed defence to increasing AFB1 concentrations. According to our knowledge, this study reports for the first time the biphasic response of maize seeds to increasing AFB1 contamination level, comprising the induction of polyamine conjugate accumulation and variation in the ratio of conjugates. This dose–response relationship may provide useful information in the field of agricultural and food chemistry as an indicator of AFB1 contamination level and, hence, for selecting an appropriate seed quality.

abstract = "BACKGROUND: Cereal seeds, such as maize seeds, are frequently contaminated with aflatoxin B1 (AFB1), one of the most dangerous naturally occurring carcinogens. In plants, phenolamides are involved in biotic stress response. The data on variations of phenolamides in AFB1-containing seeds are limited. RESULTS: Five polyamine conjugates, including two spermidine and three putrescine conjugates, were tentatively identified in methanolic extracts, using HPLC-DAD-MS. The ratio of putrescine to spermidine conjugates changed with increasing AFB1 concentration in a logistic dose–response manner, with a ratio of below 1 up to a concentration of 51.51 μg kg−1, and approximately 2.54 and 3 at higher concentrations of 177.4 and 308.13 μg kg−1, respectively. The observed variations of the total antioxidant activity and the total phenolic content may support this biphasic behaviour of the seeds against AFB1 stress. CONCLUSIONS: The obtained data are a contribution to the understanding of the roles of polyamine conjugates in seed defence to increasing AFB1 concentrations. According to our knowledge, this study reports for the first time the biphasic response of maize seeds to increasing AFB1 contamination level, comprising the induction of polyamine conjugate accumulation and variation in the ratio of conjugates. This dose–response relationship may provide useful information in the field of agricultural and food chemistry as an indicator of AFB1 contamination level and, hence, for selecting an appropriate seed quality.",

AB  - Reports about the influence of cerium-oxide nanoparticles (nCeO2) on plants are contradictory due to their positive and negative effects on plants. Surface modification may affect the interaction of nCeO2 with the environment, and hence its availability to plants. The aim was to determine whether nontoxic carbohydrates alter the effect of nCeO2 on the seed germination, plant growth, and metabolism of wheat and pea. We applied 200 mgL-1 of nCeO2 on plants during germination (Ger treatment) or three week-growth (Gro treatment) in hydroponics. The plant response to nCeO2 was studied by measuring changes in Ce concentration, total antioxidative activity (TAA), total phenolic content (TPC), and phenolic profile. Our results generally revealed higher Ce concentration in plants after the treatment with coated nanoparticles compared to uncoated ones. Considering all obtained results, Ger treatment had a stronger impact on the later stages of plant development than Gro treatment. The Ger treatment had a stronger impact on TPC and plant elongation, whereas Gro treatment affected more TAA and phenolic profile. Among nanoparticles, levan-coated nCeO2 had the strongest and positive impact on tested plants. Wheat showed higher sensitivity to all treatments.

Several botanical species have been considered including mulberry (Morus alba L.), myrobalan plum (Prunus cerasifera Ehrh.), black locust (Robinia pseudoacacia L.), wild cherry (Prunus avium (L.) L.), and oak (Q. petraea (Matt.) Liebl., Q. robur L., and Q. cerris L.). A total of 37 compounds were quantified, demonstrating the presence of phenolic acids, flavonols, flavones, flavanones, flavanonol taxifolin, stilbenoids, and coumarins. Taxifolin was the most abundant in wild cherry (8455.70 mg kg−1), while ellagic acid predominated in oak wood (8872.05–10099.32 mg kg−1 in sessile oaks, and up to 15,958.80 mg kg−1 in pedunculate oak from Slavonia). The highest content of protocatechuic acid (533.39 mg kg−1) was found in myrobalan plum. Also, isoflavones were characteristic of wild cherry, while mulberry was abundant in stilbenoids. Total phenolic content, as well as antioxidant, chromatic, and fluorescence properties were studied. The spectral shapes and maxima of fluorescence emission spectra of bare wood samples were compared with those of the corresponding wood extracts. The Principal Component Analysis (PCA) was applied in order to find patterns in emission spectra for differentiation among wood samples.

abstract = "The aim of this research was to study phenolic compounds of diverse botanical species of wood commonly used in cooperage in Balkan countries. Several botanical species have been considered including mulberry (Morus alba L.), myrobalan plum (Prunus cerasifera Ehrh.), black locust (Robinia pseudoacacia L.), wild cherry (Prunus avium (L.) L.), and oak (Q. petraea (Matt.) Liebl., Q. robur L., and Q. cerris L.). A total of 37 compounds were quantified, demonstrating the presence of phenolic acids, flavonols, flavones, flavanones, flavanonol taxifolin, stilbenoids, and coumarins. Taxifolin was the most abundant in wild cherry (8455.70 mg kg−1), while ellagic acid predominated in oak wood (8872.05–10099.32 mg kg−1 in sessile oaks, and up to 15,958.80 mg kg−1 in pedunculate oak from Slavonia). The highest content of protocatechuic acid (533.39 mg kg−1) was found in myrobalan plum. Also, isoflavones were characteristic of wild cherry, while mulberry was abundant in stilbenoids. Total phenolic content, as well as antioxidant, chromatic, and fluorescence properties were studied. The spectral shapes and maxima of fluorescence emission spectra of bare wood samples were compared with those of the corresponding wood extracts. AB  - It has been explored to what extent the ratio of the two main fluorophores in honey, originating from proteins and phenolic compounds, change between the honey extraction stage and packaging in the jars. Fluorescence spectroscopy combined with multivariate curve resolution–alternating least squares (MCR–ALS) was used to determine the ratio of the spectral components originating from phenol and proteins (Ph/Pr) fl , as a ratiometric indicator of variability in selected Lime tree (Tilia L.) honey samples. Spectrophotometric quantification of phenols and proteins in the honey samples was also performed and their ratio (Ph/Pr) sp was calculated. The values of the (Ph/Pr) fl ratio and honey protein content after packaging depended on the quality of homogenization before packaging in jars. Colorimetric and fluorometric results for phenols and proteins were in compliance. The examples are the values 3.34, 3.30, and 9, 3.14 for (Ph/Pr) fl and corresponding values 2.64, 2.18, and 12.75, 2.31 for (Ph/Pr) sp , in all pairs, the first value presenting the sample after extraction and the second value the sample after packaging in jars. Both methods show that in the former case there is no change in the phenol/protein ratio, and in the latter case the ratio decreased.

Peroxidase-Sensitive Tyramine Carboxymethyl Xylan Hydrogels for Enzyme Encapsulation

AB  - Derivatives of xylans were synthesized from corncob xylan by carboxymethylation, oxidization with different molar ratios of periodate (5, 10 15 and 20 mol%) and by reductive amination with tyramine. Modifications of tyramine carboxymethyl xylans (Tyr-CMX) were confirmed by FTIR, UV and NMR spectra. Concentration of ionizable groups increased from 1.5 mmol/g for carboxymethyl xylan (CMX) to 5.4 mmol/g for Tyr-CMX oxidized with 20 mol% of periodate. All Tyr-CMXs were able to form hydrogels the cross-linking reaction with horseradish peroxidase and peroxide. Tyr-CMXs were tested for amyloglucosidase (AG) encapsulation within hydrogel microbeads obtained in a reaction of emulsion polymerization with peroxidase. Average diameter of Tyr-CMX hydrogel microbeads was 52±25 µm and after encapsulation optimization with respect to the extent of CMX modification with tyramine, the concentration of Tyr-CMX, and the amount of added AG, microbeads with AG specific activity of 2 U/mL and 20% yield of immobilization were obtained. The optimum pH of the immobilized AG was not changed compared to the soluble one, while half-life at 60 °C was increased around 10 times. The Michaelis-Menten constant for the immobilized enzyme, 1.03 mM, was significantly lower than that for the soluble one, 1.54 mM. After 5 cycles of repetitive use in batch reactor, the immobilized AG retained 68% of initial activity.

AB  - In the present study we have analyzed effects of Ag+ ions doping on energetic profiles of nanophosphors materials based on fluorapatite crystal system. The UV radiation absorption and luminescence properties of monophase fluorapatite (FAP) and Ag+ doped fluorapatite (AgFAP) nanomaterials obtained by neutralization method were investigated using the photoluminescence spectrophotometry. The excitation-emission profiles of nanomaterials were analyzed statistically by MCR-ALS method and number of fluorophores was extracted. FAP lattice absorbed light at 350 nm in the UVA part of spectrum, and with increasing concentration of Ag+ ions new absorption maximum appeared at 270 nm in the UVC part. Fluorescence of FAP nanoparticles was in violet region of visible part of the spectrum, with a red shift to the green region when Ag+ was doped in lattice. MCR-ALS analyses of fluorescence spectra confirm formation of two maxima, at 484 and 505 nm, as a consequence of Ag+ ions doping in FAP lattice at Ca1 (4f) sites. The results of quantum chemical calculations showed that an Ag+ ion is stronger bonded to the binding site 1 (-1352:6 kcal/mol) than to the binding site 2 (-1249:0 kcal/mol). Considering that AgFAP1 nanopowder absorbs photons over all part of UV radiation spectrum, this material might be used as potential radiation protective nanomaterial.

AB  - One of the most important practices in the production of some alcoholic beverages is ageing process. During that process, important changes occur, which results in improved sensory characteristics. The phenolic fraction was considered to be one of the most important parameters for evaluating quality in the choice of good alternative aging wood [3]. So far, mulberry heartwood hasn’t been investigated too much in order to characterize polyphenols. Therefore, the aim of this research was to investigate phenolic profile of industrially dried mulberry staves originating from Serbia. The results showed that mulberry was abundant in oxyresveratrol, taxifolin, (- )-catechin gallate, and phenolic acids, such as protocatechuic, p-hydroxybenzoic and ellagic acid. Characteristic emission spectrum of the wood sample showed the maximum at 420 nm, being blue shifted comparing with the maxima of most hard- and soft- wood species. This may be addressed to a relatively low content of lignin in mulberry wood. The extract showed a maximum characteristic for the major polyphenols of oxyresveratrol and coumarin type.

AB  - Prilikom starenja alkoholnih pića u prisustvu drveta dolazi do važnih promena u aromi, boji, ukusu i gorčini, usled interakcije između jedinjenja iz drveta i destilata. Iako se hrast najviše koristi za čuvanje alkoholnih pića, i druge vrste, kao što su kesten, trešnja, bagrem i dud, se mogu koristiti. Cilj rada bio je da se okarakteriše ekstrakt duga trešnjinog drveta primenom HPLC-MS i spektrofluorometrijske metode. Dobijeni rezultati ukazali su na to da je stablo trešnje bogato fenolnim jedinjenjima. Spektrofluorometrijski rezultati ukazuju na razliku trešnjinog drveta i njegovog ekstrakta u odnosu na druge vrste drveta, zbog prisustva specifičnih flavonoida u ovom drvetu. AB  - One of the most important practices in the production of some alcoholic beverages is
ageing process. During that process, important changes occur, which results in improved
sensory characteristics. 
The phenolic fraction was considered to be one of the most important parameters for
evaluating quality in the choice of good alternative aging wood [3]. So far, mulberry
heartwood hasn’t been investigated too much in order to characterize polyphenols. Therefore, the aim of this
research was to investigate phenolic profile of industrially dried mulberry staves originating
from Serbia. The results showed that mulberry was abundant in oxyresveratrol, taxifolin, (-
)-catechin gallate, and phenolic acids, such as protocatechuic, p-hydroxybenzoic and ellagic
acid.  Characteristic emission spectrum of the wood sample showed the maximum at 420
nm, being blue shifted comparing with the maxima of most hard- and soft- wood species.
This may be addressed to a relatively low content of lignin in mulberry wood. The extract
showed a maximum characteristic for the major polyphenols of oxyresveratrol and coumarin
type.

AB  - Cerium oxide (CeO2) nanoparticles (CONPs) are interesting biomaterials with various applications in biomedicine, cosmetics and the pharmaceutical industry, but with limited practical application because of their low stability in aqueous media. The aim of this study was to obtain CONPs with increased stability by coating the particles. Microbial exopolysaccharides (levan, pullulan) and glucose were used to prepare CONPs under different synthesis conditions. Coating was attempted by adding the carbohydrates during (direct coating) or after (subsequent coating) the synthesis of CONPs. The obtained nanoparticles were characterized by X-Ray diffraction analysis, Fourier transform infrared spectroscopy (FT-IR), scanning electron microscopy (SEM) and transmission electron microscopy (TEM). The suspension stability of the uncoated and coated CONPs in aqueous media was evaluated by measuring the hydrodynamic size, zeta potential and turbidity. The FT-IR spectra revealed the differences between coated CONPs and showed the success of subsequent coating with carbohydrates. Coating with carbohydrates improved the stability the CONP suspension by decreasing the size of aggregated particles. The suspensions of levan- and glucose-coated CONPs had the best stability. In this study, CONPs were prepared using non-toxic materials, which were completely environmentally friendly. The obtained results open new horizons for CONP synthesis, improving their biological applications.

Tyramine-modified pectins via periodate oxidation for soybean hull peroxidase induced hydrogel formation and immobilization

AB  - Pectin was modified by oxidation with sodium periodate at molar ratios of 2.5, 5, 10, 15 and 20 mol% and reductive amination with tyramine and sodium cyanoborohydride afterwards. Concentration of tyramine groups within modified pectin ranged from 54.5 to 538 mu mol/g of dry pectin while concentration of ionizable groups ranged from 3.0 to 4.0 mmol/g of dry polymer compared to 1.5 mmol/g before modification due to the introduction of amino group. All tyramine-pectins showed exceptional gelling properties and could form hydrogel both by cross-linking of carboxyl groups with calcium or by cross-linking phenol groups with peroxidase in the presence of hydrogen peroxide. These hydrogels were tested as carriers for soybean hull peroxidase (SHP) immobilization within microbeads formed in an emulsion based enzymatic polymerization reaction. SHP immobilized within tyramine-pectin microbeads had an increased thermal and organic solvent stability compared to the soluble enzyme. Immobilized SHP was more active in acidic pH region and had slightly decreased K (m) value of 2.61 mM compared to the soluble enzyme. After 7 cycles of repeated use in batch reactor for pyrogallol oxidation microbeads, immobilized SHP retained half of the initial activity.
