AB  - S-Adenosyl-L-Homocysteine (AdoHcy) is a crucial pathological factor in homocysteine-associated disorders. We developed an impedimetric approach for single drop S-Adenosyl-L-Homocysteine detection, based on S-Adenosyl-L-Homocysteine hydrolase (SAH), produced in the prokaryotic expression system (E. coli) by recombinant DNA technology, immobilized on citrate acid-coated gallium oxyhydroxide dispersed over single-walled carbon nanotubes (CA/GaO(OH)@SWCNT). The proposed biosensor offers detection of AdoHcy, with a limit of detection (LOD) of 0.17 μM. with operating linear range from 1 to 44 μM. The remarkable stability of gallium oxyhydroxide crystal was further improved by citric acid coating and its optical, electrical and physical anisotropy was skipped using SWCNTs.

Co(III), Ni(II), and Cu(II) complexes with tetradentate Schiff base ligand: Synthesis, Characterization, Electrochemical Behavior, Binding assessment and In vitro cytotoxicity activity

AB  - Two new Schiff base cobalt(III) ([Co(LH)Cl2], 1) and nickel(II) ([Ni(LH)ClO4], 2) complexes with a diimine-dioxime ligand, (4,9-diaza-3,10-diethyl-3,9-dodecadiene-2,11-dione bis oxime (LH2)), were synthesized and characterized. The compounds were obtained in MeOH from corresponding metal salts and LH2 in molar ratio 1:1 and further characterized by mass spectrometry, IR spectroscopy, electrochemistry, and elemental analysis. Previously reported copper(II) analog, ([Cu2(LH)2]·(ClO4)2, 3) was joined to 1 and 2, and the three metal analogs, 1-3, were further investigated in terms of their electrochemical behavior. The binding studies of the complexes with deoxyribonucleic acid (DNA) and human serum albumin (HSA) were carried out using both spectrophotometric and electrochemical methods. All three complexes exhibit binding affinity towards the DNA chain through intercalative interaction. The binding reaction with HSA showed for 1 and 3 complexes decrease in the peak current obtained in the case of complexes before the addition of HSA, while resulted compound obtained from Ni complex – HSA possesses the same electroactivity as starting complex. Furthermore, the cytotoxicity of LH2 as well as its metal complexes, and cisplatin were evaluated on CT-26 mouse colon carcinoma and human LS174T cancer cell lines employing MTT assay. The copper(II) complex exhibited very promising anticancer activity compared with cisplatin.

AB  - Type of the wood used for the aging highly influences the quality of alcoholic beverages. In this research we explored the potential of cyclic voltammetry (CV) and differential pulse voltammetry (DPV) to establish fingerprints characteristic for each wood and to enable determining the type of the wood used in the aging process. Eleven different wood samples were used to prepare three different types of spirits during 15 months. The highest extraction rate was obtained during the first month, while further aging was followed with almost constant amount of extracted polyphenols. Black locust, myrobalan plum, and mulberry extracts were discriminated from the spirits aged in oak and wild cherry wood when statistical analysis was applied. Although clear classification of all samples was not achieved, this long term study demonstrated a potential of both CV and DPV for differentiating wood species used in the aging, hence in the quality control of spirits. © 2020 Elsevier Ltd

AB  - The goal of this work was to develop green electrode material that unites all advantages of domestic made, synthesized porous carbon powder and ceria dioxide nanoparticles known due to exceptional catalytic properties. Thermal decomposition of Novolac phenol-formaldehyde resin and cerium sulfate resulting in producing a high-performance CeO2 porous carbon material highly sensitive to dopamine (DA) electrooxidation. Morphological and structural characteristics of the material were determined by SEM and XRD measurements, while electrochemical characterization was performed by EIS and CV. The sensitivity of DA determination on the proposed CeO2-doped carbon material was enhanced by adding multi-wall carbon nanotubes to finally prepare a mixture for a specific carbon paste electrode (TPCeO2&MWCNT@CPE). SWV technique was used for quantification of dopamine in Britton-Robinson buffer pH 6 in the concentration range of 0.5-100 μM of DA, with the detection limit of 0.14 μM and quantification limit of 0.44 μM. Good selectivity overstudied bioactive compounds enables the successful and efficient application of the proposed electrode and developed an analytical procedure for the determination of dopamine in spiked urine samples.

abstract = "The goal of this work was to develop green electrode material that unites all advantages of domestic made, synthesized porous carbon powder and ceria dioxide nanoparticles known due to exceptional catalytic properties. Thermal decomposition of Novolac phenol-formaldehyde resin and cerium sulfate resulting in producing a high-performance CeO2 porous carbon material highly sensitive to dopamine (DA) electrooxidation. Morphological and structural characteristics of the material were determined by SEM and XRD measurements, while electrochemical characterization was performed by EIS and CV. The sensitivity of DA determination on the proposed CeO2-doped carbon material was enhanced by adding multi-wall carbon nanotubes to finally prepare a mixture for a specific carbon paste electrode (TPCeO2&MWCNT@CPE). SWV technique was used for quantification of dopamine in Britton-Robinson buffer pH 6 in the concentration range of 0.5-100 μM of DA, with the detection limit of 0.14 μM and quantification limit of 0.44 μM. Good selectivity overstudied bioactive compounds enables the successful and efficient application of the proposed electrode and developed an analytical procedure for the determination of dopamine in spiked urine samples.",

AB  - This work presents advanced electrochemical platform based on screen printed diamond electrode (SPDE) system for the single drop “point-of-care” testing. Proposed approach was applied for the quantification of doxorubicin hydrochloride (DOX) in biological fluids and pharmaceutical product. Using a single drop (~30 μL) of the tested solution at the electrode surface, DOX showed high electroactivity over a wide range of pHs. In these conditions, single oval shaped, well-defined and pH dependent oxidation peak was observed in the potential range from 0.5 V to 1.3 V. In the reverse scan, two cathodic peaks, were noted – around 0.3 V and – 0.5 V. Similarly, first reduction peak was pH dependent, while second one was independent in the studied range. Experimental conditions for DOX quantification were optimized and natures of the electrode reactions were investigated. Working linear range obtained for DOX detection was from 0.1 to 2.5 μM. Diffusion controlled electrode reaction reveal long life time of the proposed electrode as well high potential for practical application. Developed procedure was successfully applied for the DOX analysis in biological fluids – urine and pharmaceutical formulation. Obtained results clearly indicated that given procedure can be easily implemented for pharmaceutical control and medical analysis, in both, laboratory and field conditions.

AB  - Different phosphates and phosphonates have shown excellent coating ability toward magnetic nanoparticles, improving their stability and biocompatibility which enables their biomedical application. The magnetic hyperthermia efficiency of phosphates (IDP and IHP) and phosphonates (MDP and HEDP) coated Fe3O4 magnetic nanoparticles (MNPs) were evaluated in an alternating magnetic field. For a deeper understanding of hyperthermia, the behavior of investigated MNPs in the non-alternating magnetic field was monitored by measuring the transparency of the sample. To investigate their theranostic potential coated Fe3O4-MNPs were radiolabeled with radionuclide 177Lu. Phosphate coated MNPs were radiolabeled in high radiolabeling yield (> 99%) while phosphonate coated MNPs reached maximum radiolabeling yield of 78%. Regardless lower radiolabeling yield both radiolabeled phosphonate MNPs may be further purified reaching radiochemical purity of more than 95%. In vitro stabile radiolabeled nanoparticles in saline and HSA were obtained. The high heating ability of phosphates and phosphonates coated MNPs as sine qua non for efficient in vivo hyperthermia treatment and satisfactory radiolabeling yield justifies their further research in order to develop new theranostic agents. © 2021, The Author(s), under exclusive licence to Springer Science+Business Media, LLC, part of Springer Nature.

AB  - Herein, we proposed a novel approach and state-of-the-art technology for the improvement of the materials and enzyme immobilization at the electrode surface and construction of impedimetric glucose biosensor. We silanized titanium dioxide nanoparticles using (3-aminopropyl)triethoxysilane (APTES), for the preparation of cross-linked material nanoparticles, with carboxylic graphene. The silanization of titanium dioxide nanoparticles and an increase in electron shuttle was proven feasible when this composite was able to achieve about 30% higher current than non-silanized material. The proposed approach was used for the modification of the printed three-electrode system and the development of the impedimetric glucose biosensor. The material morphology and electrochemical characteristics were confirmed by spectroscopic and electrochemical methods. The present combination effectively modified the electrode surface and serve as a promising basis for the construction of Point-of-Care devices. Developed biosensor possesses wide operating linear range toward glucose detection from 50 µmol to 1000 µmol, with the limit of detection of 24 µmol. Finally, negligible interference effect and application in the real sample indicate that the proposed mechanism can be successfully applied to the assessment of glucose level in only one drop of real sample.

AB  - In order to ensure high food quality, one of the prime importance is the detection and quantification of histamine, well known marine food poison. In this work, we constructed novel electrochemical biosensor for the detection of histamine based on gold nanoparticles decorated on manganese dioxide (Au/MnO2) and used for modification of screen-printed carbon electrode (Au/MnO2@SPCE). The constructed sensor was then used for the estimation of histamine content in a single drop. Materials used in this study were synthesized and characterized using HR-TEM, XRPD and electrochemical methods. The amperometric detection method was optimized and, under selected operating parameters (supporting electrolyte pH 6, working potential of 1 V), the proposed sensor possesses linear working range from 0.3 µM to 5.1 µM, with a detection limit of 0.08 µM. The effect of selected interferences was investigated and it was found that the developed approach offers accurate, precise, selective, fast and reproducible quantification of histamine using only one drop of the sample. In the end, this work stands as a proof-of-concept of the modified electrodes and electrochemical detection as a promising and prospective approach for the applications in real-time monitoring of the food quality.

AB  - It was reported that novel O,O′-diethyl-(S, S)-ethylenediamine-N,N′-di-2-(3-cyclohexyl) propanoate dihydrochloride (DE-EDCP) displayed in vitro antiproliferative activity on several human and mouse cancer cell lines, which was comparable to that of the prototypical anticancer drug cisplatin. In order to reveal its toxicity profile, acute and repeated-dose toxicity studies were performed in Naval Medical Research Institute (NMRI) Han mice. The intravenous LD 50 values of DE-EDCP were found to be 95.3 and 101.3 mg/kg body weight in female and male mice, respectively. In the subacute toxicity study, DE-EDCP was administered intravenously at the doses of 15, 25, and 40 mg/kg/day for a period of 28 days. There were no adverse effects on general condition, growth, feed and water consumption, and hematological parameters. There was a significant increase in urea and alanine aminotransferase in female mice and aspartate aminotransferase and alkaline phosphatase in both genders in 40 mg/kg/day dose-treated group. The histopathological changes confined to the liver and kidney, but in other organs were not found. Satellite group revealed that changes in the kidney and liver were less pronounced, suggesting their reversibility. Interactions with DNA could also be of importance for understanding DE-EDCP toxic side effects. Hyperchromic effect obtained with ultraviolet–visible, suggested electrostatic interactions between DE-EDCP and calf thymus DNA. The toxicity testing of DE-EDCP was conducted to predict human outcomes.

Furthermore, the radiotracer method using the radionuclides 99m Tc and 90 Y was applied as a reliable and powerful method for evaluating the in vivo behavior of a nanoprobe; a high radiolabeling yield (>93%), in vitro and in vivo stability of the radiolabeled nanoparticles and high heating effect were observed, thus paving the way for the possible theranostic applications of Fe 3 O 4 -DPD MNPs.

AB  - Magnetic nanoparticles (MNPs) are of immense interest for diagnostic and therapeutic applications in medicine. Design and development of new iron oxide-based MNPs for such applications is of rather limited breadth without reliable and sensitive methods to determine their levels in body tissues. Commonly used methods, such as ICP, are quite problematic, due to the inability to decipher the origin of the detected iron, i.e. whether it originates from the MNPs or endogenous from tissues and bodily fluids. One of the approaches to overcome this problem and to increase reliability of tracing MNPs is to partially substitute iron ions in the MNPs with Er. Here, we report on the development of citric add coated (Fe,Er)(3)O-4 nanopartides and characterization of their physico-chemical and biological properties by utilization of various complementary approaches. The synthesized MNPs had a narrow (6-7 nm) size distribution, as consistently seen in atomic pair distribution function, transmission electron microscopy, and DC magnetization measurements. The particles were found to be superparamagnetic, with a pronounced maximum in measured zero-field cooled magnetization at around 90 K. Reduction in saturation magnetization due to incorporation of 1.7% Er3+ into the Fe3O4 matrix was clearly observed. From the biological standpoint, citric acid coated (Fe,Er)(3)O-4 NPs were found to induce low toxicity both in human cell fibroblasts and in zebrafish (Danio rerio) embryos. Biodistribution pattern of the MNPs after intravenous administration in healthy Wistar rats was followed by the radiotracer method, revealing that Y-90-labeled MNPs were predominantly found in liver (7533% ID), followed by lungs (16.70% ID) and spleen (2.83% ID). Quantitative agreement with these observations was obtained by ICP-MS elemental analysis using Er as the detected tracer. Based on the favorable physical, chemical and biological characteristics, citric add coated (Fe,Er)(3)O-4 MNPs could be further considered for the potential application as a diagnostic and/or therapeutic agent. This work also demonstrates that combined application of these techniques is a promising tool for studies of pharmacokinetics of the new MNPs in complex biological systems.

AB  - The extensive development of radiopharmaceuticals towards early tumour detection and treatment has increased the demand for new ligands with higher tumour selectivity. Research has been done on the potential of the novel O,O'-diethylethylenediamine-N,N'-di-3-propanoate (L) ligand as a radionuclide vehicle for tumour targeting. Under alkaline conditions, L hydrolyses and produces half ester ligand (L') and diacid ligand (L"), with characteristic donor atom array N, N, O. Ligand L was successfully labelled with Tc-99m at pH=9 by coordination with the octahedral fac-[Tc-99m(CO)(3)(H2O)(3)](+) intermediate, forming the main radioproduct fac-[(TcL)-Tc-99m'(CO)(3)] (Tc1). The Tc-99m complex showed a low lipophilic character (log P = 0.48) and low binding affinity to human serum albumin (2.51 +/- 0.48%). In vitro stability studies in saline and human plasma, as well as challenge studies with cysteine and histidine, revealed high stability of the complex during 24 h. Biodistribution studies of Tc1 in female C57BL/6 mice bearing B16/F1 melanoma metastases showed significant tumour uptake: 9.81 +/- 1.19% ID g(-1) in the liver, 5.87 +/- 0.54% ID g(-1) in the lungs and 3.17 +/- 0.33% ID g(-1) in the ovary at 30 min post-injection. Favourable physicochemical properties, satisfactory in vitro/in vivo stability and biodistribution profile in the experimental metastatic melanoma model indicate the possible application of the radiolabelled ligand in tumour diagnosis.
