AB  - Per- and polyfluoroalkyl substances (PFAS) are a group of synthetic chemicals that have been widely used in various industrial and consumer products due to their unique properties, such as oil and water repellency, thermal stability, and durability. However, the persistence and mobility of these chemicals in the environment have raised concerns about their potential adverse effects on human health and the environment. PFAS have been detected on a global scale in various environmental media, such as soil, water, air, and biota. As a country undergoing economic development and transition, Serbia faces the challenge of managing and addressing the PFAS contamination in its environment. This challenge requires a comprehensive and science-based strategy that can effectively reduce the exposure and risks of PFAS to human health and the environment. This document aims to provide a scientific strategy for solving the PFAS challenge in Serbia. Firstly, it is important to acknowledge that PFAS are persistent and bioaccumulative in the environment, which means that they do not break down easily and can accumulate in the food chain, posing a long-term risk to human health and the environment. Therefore, a precautionary approach is necessary to minimize the exposure and risks of PFAS. International experience and cooperation are very important for developing an effective scientifc strategy for addressing the PFAS challenge in Serbia. PFAS are a global issue, and many countries have already implemented measures to manage and reduce the exposure and risks of PFAS. Therefore, it is important to draw on international experience and best practices when developing the strategy for Serbia. International experience can provide valuable insights into the sources, pathways, and fate of PFAS, as well as the e"ectiveness of various risk management measures. For example, the United States, Canada, and some European countries have established regulatory frameworks for PFAS, which can serve as a model for Serbia. Other countries have implemented remediation measures for contaminated sites, which can provide valuable insights for selecting appropriate remediation technologies in Serbia. Moreover, international experience can provide access to the latest scientific knowledge, methods, and technologies for assessing and managing PFAS contamination. For example, international organizations such as the United Nations Environment Programme (UNEP) and the Organization for Economic Co-operation and Development (OECD) have developed guidance documents and tools for assessing and managing PFAS contamination. The scientific strategy for solving the PFAS challenge in Serbia is based on a thorough understanding of the sources, pathways, and fate of PFAS in the environment. It is also based on a comprehensive and systematic approach, including risk assessment, monitoring, regulation, remediation, and communication. This strategy is tailored to the specific context and needs of Serbia. It is based on the latest scientifc knowledge and practical experience from other countries and regions, focusing on the European Union, USA, China and Japan. The cultural, social, economic, and political factors can affect the implementation and effectiveness of the strategy, and therefore, the strategy is developed through a collaborative and participatory process involving stakeholders from different sectors and levels. The international experience provided helpful guidance and lessons learned, but ultimately, the strategy is based on local knowledge, priorities, and capacities. By implementing this strategy, we aim to contribute to that Serbia can protect its citizens and environment from the potential harm of PFAS and achieve sustainable development.

Antioxidant Capacity and Quality of Human Milk and Infant Formula Determined by Direct Current Polarography

AB  - Antioxidants present in infant food contribute to its nutritive value and provide protection from oxidative damage. Here, a direct current polarographic method, based on a decrease of anodic current from the hydroxoperhydroxo-mercury(II) complex (HPMC) formed at a mercury dropping electrode at its potential of dissolution, was used to determine the total antioxidant capacity (TAC) of infant formulas, human milk collected from mothers of full-term and preterm infants, human milk supplemented with fortifier, and cow milk. TAC of milk samples and infant formula correlated with protein content measured by the Bradford method (Pearson correlation coefficient 0.605, p<0.001), while TAC of human milk correlated with content of free thiol groups measured by the Ellman method (0.779, p<0.001). The results obtained showed that, in contrast to the 2,2-diphenyl-1-picrylhydrazyl (DPPH) assay, HPMC enables specific insight into the quality of infant milk food which reflects the amounts of free thiol groups and protein content. The significant contribution of milk proteins and the most prevalent amino acids, particularly cysteine, arginine, and phenylalanine, to TAC was shown. Thus, the HPMC method might be useful in monitoring the quality of infant formulas and human milk for preterm infants before and after supplementation with fortifier.

Investigation of pectin as a prebiotic, antioxidant and antimicrobial agent for the bacteria selected from human milk of mothers of premature infants

AB  - BACKGROUND: Pectin is a complex polysaccharide, present in fresh and processed food as an additive, which also serves as an important source of fiber. In this work, the aim was to investigate in-vitro application of apple pectin as a possible antimicrobial, antioxidant and prebiotic agent for growth of bacteria selected from human milk and infant feces, as well as standard strains Lactobacillus rhamnosus and Bifidobacterium bifidum.
METHODS : All the bacteria isolated from the milk of mothers of preterm and term-born infants, bacteria from infant feces and standard strains were tested for fermentation of apple pectin as the main carbohydrate in selective broth media.
RESULTS: An increase in the growth of bacteria Bifidobacteria, Lactobacillus, and a consortium of bacteria selected from human milk and infant feces was noticed after pectin fermentation. It was found that pectin has a positive impact on the increase of the antioxidant capacity in all the tested samples. A high correlation was achieved, comparing electrochemical with spectrophotometric methods. Antimicrobial tests showed that pectin and fermentation products have slight or no influence at all on the growth of standard strains of Escherichia coli, Staphylococcus aureus and Candida albicans.
CONCLUSIONS: The results showed the importance of pectin as an antioxidant and its impact on the development of potential probiotic microbiota in mothers and infants. The study indicated a new potential application of pectin as a supplement in infant food.

Study on the assessment of humification processes during biodegradation of heavy residual fuel oil

AB  - The aim of this study was to investigate the creation of humic substances during biodegradation of heavy residual fuel oil, because there are indications that substances similar to humic substances are generated during biodegradation of polycyclic aromatic hydrocarbons. In the study, which lasted for 110 days, biodegradation of heavy residual fuel oil was carried out in a layer of artificial soil substrate. The initial concentration of the total petroleum hydrocarbon in the prepared artificial soil substrate (biopile) was 23.1 g kg-1 dry weight (d.w.). At the end of the process, the total petroleum hydrocarbons were reduced to 8.1 g kg-1 d.w. in the inoculated biopile, while the content of humic acids increased during bioremediation from 3.15 g kg-1 d.w. to 4.95 g kg-1 d.w. The humic acids extracted from biopile during the biodegradation process were characterized by various chemical techniques (elemental analysis, spectrofluorimetric analysis, electrochemical measurements, and size exclusion chromatography). The results showed that levels of C, H and the H/C ratio decreased as the biodegradation process progressed. This indicated that humic acids aromatization process took place and this was confirmed by the spectrofluorimetric analysis. The increase of oxygen percentage and the O/C ratio in the humic acids after the biodegradation treatment indicated an increase in functional oxygen groups. Additional analyses of humic acids from the inoculated biopile showed that they were transformed during the bioremediation process. They had greater redox and buffering capacities and a larger portion of the fractions had high molecular mass. Also, the humification parameters (the CHAs/CFAs ratio and CHAs/Corg ratio) increased during the biodegradation. This is one of the few studies that describes the generation of humic substances during the biodegradation of oil compounds.

The humic acids extracted from biopile during the biodegradation process were characterized by various chemical techniques (elemental analysis, spectrofluorimetric analysis, electrochemical measurements, and size exclusion chromatography). The results showed that levels of C, H and the H/C ratio decreased as the biodegradation process progressed. This indicated that humic acids aromatization process took place and this was confirmed by the spectrofluorimetric analysis. The increase of oxygen percentage and the O/C ratio in the humic acids after the biodegradation treatment indicated an increase in functional oxygen groups. Additional analyses of humic acids from the inoculated biopile showed that they were transformed during the bioremediation process. They had greater redox and buffering capacities and a larger portion of the fractions had high molecular mass. Also, the humification parameters (the CHAs/CFAs ratio and CHAs/Corg ratio) increased during the biodegradation. This is one of the few studies that describes the generation of humic substances during the biodegradation of oil compounds.",

AB  - Pollution of the environment by petroleum products is a global problem, since its extensive exploitation in broad areas.Microorganisms can degrade or transform toxic, petroleum derivates into harmless products with high efficiency.This study aimed to investigate the biodegradation process of petroleum hydrocarbons in contaminated sediment usingindigenous and a combination of indigenous and allochthonous microorganisms.The allochthonous microorganisms that were added belonged to the genera Acinetobacter and Citrobacter. The experiment lasted two weeks, and the degradation process took place in a closed vessel on a Micro-Oxymax respirometer.Total petroleum hydrocarbons (TPH) were extracted and determined using a gas chromatograph with a flame ionization detector. The number of microorganisms at the beginning and end of the experiment was also determined.The results on the respirometer and gas chromatography indicated that the microorganisms were metabolically active.Indigenous microorganisms reduce the amount of TPH by 32.0% and the combination of indigenous-allochthonousmicroorganisms by 33.6%. The results showed that there is no significant difference between the application of autochthonous and the combination of indigenous-allochthonous microorganisms for the reduction of TPH in contaminatedsediment, suggesting a high potential of autochthonous microorganisms for bioremediation processes.

AB  - Zagađenje vode, zemljišta i sedimenta naftom i njenim derivatima, može se javiti tokom eksploatacije, transporta i skladištenja i predstavlja rizik za životnu sredinu i ljudsko zdravlje. Bioremedijacija je zelena tehnologija kojom se uklanja zagađenje iz kontaminirane životne sredine upotrebom mikroorganizama. U ovom radu su praćene promene u sadržaju mineralnih ulja i aktivnost mikroorganizama koji razgrađuju ugljovodonike tokom procesa bioremedijacije koji je trajao 210 dana. Rezultati ukazuju da mikroorganizmi koji razgrađuju ugljovodonike sa visokom efikasnošću degraduju mineralna ulja. Na osnovu količine mineralnog ulja (smanjena za 63,82 %), stepena degradacije i udela mikroorganizama naftnih degradera u ukupnom broju mikroorganizama, može se zaključiti da su faza adaptacije konzorcijuma i faza intenzivne razgradnje bile u periodu od 0. do 90. dana, praćene dalje sporijom fazom razgradnje u periodu od 90. do 210. dana.

The results indicate that hydrocarbon degrading microorganisms with high efficiency have biodegraded mineral oil. Based on the amount of mineral oil (decreased for 63.82 %), degradation rates and level of hydrocarbon degraders in the total number of microorganisms, we note that the stages of adaptation of the consortium and intensive degradation phases were in the range of 0-90 days, followed by a slower decomposition phase in the period of 90-210 days.

The results indicate that hydrocarbon degrading microorganisms with high efficiency have biodegraded mineral oil. Based on the amount of mineral oil (decreased for 63.82 %), degradation rates and level of hydrocarbon degraders in the total number of microorganisms, we note that the stages of adaptation of the consortium and intensive degradation phases were in the range of 0-90 days, followed by a slower decomposition phase in the period of 90-210 days.",

Their major advantages are low cost, high efficiency, renewability.

U radu je ispitivana mogućnost primene otpadne biomase (koštice breskve, agroindustrijskog otpada), kao jeftinog biosorbenta, za uklanjanje nafnih zagađivača iz vodenog rastvora u stacionarnim uslovima. Biosorpcioni eksperimenti su obavljeni u erlenmajerima na na orbitalnom šejkeru u kojima je konstantna količina biosorbenta od 1 g mešana sa 100 ml vode kontaminirane naftnim ugljovodonicima u koncentacijama 4 mg/L, 12 mg/L, 18 mg/L, 24 mg/L, 30 mg/L, 40 mg/L and 80 mg/L. AB  - Microbial fuel cells (MFCs) represent new potential energy sources through which electricity is generated, and among numerous advantages is that the emission of harmful gases is missing. MFC is a type of biological fuel cell, a system that converts chemical energy into electrical power by microorganisms. These systems can be constructed in many ways so that there are one-chamber, two-chamber and complex MFCs. MFCs are good alternative sources of energy offering possibility of application that can be found in the biosensor industry, hydrogen or electricity production systems and wastewater treatment plants. In this paper, characterization of the sediment used in a single chamber MFC is presented. The sediment used to form the microbial fuel cell is characterized by microbiological, chemical and analytical parameters.
AB  - Mikrobne gorivne ćelije (Microbial fuel cell-MFC) predstavljaju nove potencijalne izvore energije, preko kojih se generiše električna energija, a među brojnim prednostima je i ta što izostaje emisija štetnih gasova. MFC su vrsta bioloških gorivnih ćelija, sistema koji konvertuju hemijsku energiju u električnu pomoću mikroorganizama. Postoji više načina konstrukcije i među njima se nalaze jednokomorne, dvokomorne i složene MFC. Takođe, mikroorganizmi i supstrati koji se koriste u MFC, koje ti mikroorganizmi obrađuju u hemijskim procesima, mogu da budu različitog porekla i da imaju različite karakteristike. MFC su dobri alternativni izvori energije koji svoju potencijalnu primenu nalaze u industriji biosenzora, sistemima za proizvodnju vodonika ili električne energije i postrojenjima za prečišćavanje otpadnih voda. U ovom radu dat je opis sedimenta korišćenog u sistemu jednokomorne MFC, okarakterisan pomoću mikrobioloških, hemijskih i analitičkih parametara.

AB  - Microbial fuel cells (MFCs) represent new potential energy sources through which electricity is generated, and among numerous advantages is that the emission of harmful gases is missing. MFC is a type of biological fuel cell, a system that converts chemical energy into electrical power by microorganisms. These systems can be constructed in many ways so that there are one-chamber, two-chamber and complex MFCs. MFCs are good alternative sources of energy offering possibility of application that can be found in the biosensor industry, hydrogen or electricity production systems and wastewater treatment plants. In this paper, characterization of the sediment used in a single chamber MFC is presented. The sediment used to form the microbial fuel cell is characterized by microbiological, chemical and analytical parameters.
AB  - Mikrobne gorivne ćelije (Microbial fuel cell-MFC) predstavljaju nove potencijalne izvore energije, preko kojih se generiše električna energija, a među brojnim prednostima je i ta što izostaje emisija štetnih gasova. MFC su vrsta bioloških gorivnih ćelija, sistema koji konvertuju hemijsku energiju u električnu pomoću mikroorganizama. Postoji više načina konstrukcije i među njima se nalaze jednokomorne, dvokomorne i složene MFC. Takođe, mikroorganizmi i supstrati koji se koriste u MFC, koje ti mikroorganizmi obrađuju u hemijskim procesima, mogu da budu različitog porekla i da imaju različite karakteristike. MFC su dobri alternativni izvori energije koji svoju potencijalnu primenu nalaze u industriji biosenzora, sistemima za proizvodnju vodonika ili električne energije i postrojenjima za prečišćavanje otpadnih voda. U ovom radu dat je opis sedimenta korišćenog u sistemu jednokomorne MFC, okarakterisan pomoću mikrobioloških, hemijskih i analitičkih parametara.

AB  - The numerous naturally occurring biopolymeric materials are used as edible films and coatings (lipids, proteins, and polysaccharides). Microbial levan and pullulan have been already considered as biopolymers in food industry. The aim of this work was to obtain the levan and levan/pullulan blend films and compare them using the Atomic force microscopy (AFM) and Fourier transform infrared spectroscopy (FTIR). Two different films compositions were prepared by casting method: levan and levan/pullulan in the proportions 1:1. AFM measurements showed that levan film had a smoother surface and, consequently, a lower degree of roughness comparing to the levan/pullulan blend films. FTIR spectra showed peaks, which corresponded to all components of the obtained films.

AB  - AimThere is a discrepancy between the amount of transitional milk produced by mothers of preterm infants and the low capacity of premature infants to consume it. This milk can be used in milk banks, but previous studies found that there are large variations in the level of host-defence proteins in individual samples of milk from mothers of premature infants, which implies that large individual variations in antioxidative defence composition are also possible. MethodsMilk samples were collected from 20 healthy mothers of preterm infants. We determined the values for non-enzymatic antioxidative capacity parameters (oxygen radical absorbance capacity (ORAC)), static oxidation-reduction potential (ORP), activities of antioxidant defence enzymes and the amount of vitamin C in whole milk, skim and whey fractions of transitional milk. ResultsThe main low-molecular-weight antioxidant in transitional milk is vitamin C and most of it is contained in whey. ORAC is higher in whole transitional milk than in skim milk and whey, and ORP is lower in whole transitional milk than that in skim milk and whey. Antioxidative enzyme activities are similar in all individual samples of transitional milk from mothers of preterm infants. ConclusionsOur results indicate that transitional milk of mothers of preterm infants shows slow individual variations in antioxidative defence composition; therefore, it can be used in human milk banks.

AB  - The electrochemical techniques were used to determine the total antioxidant capacity of breast milks and the results were compared with a commonly used spectrophotometric (2,2-diphenyl-1-picrylhydrazyl, DPPH) method. Breast milk from mothers of preterm infants was monitored in three lactation phases and after storage of expressed milk by monitoring changes in the total antioxidant capacity over a two year period. Statistical analysis showed there was no significant difference between the ability of the three methods to detect changes in breast milk after storage. Either of the electrochemical techniques studied could be successfully used to replace the time-consuming spectrophotometric method and can be applied to clinical trials.

Zinc concentrations in human milk and infant serum during the first six months of lactation

AB  - Normal supply of zinc to the newborn via milk is essential for normal development. Using ICP-OES, we analyzed changes in the level of Zn in milk and infant serum in the neonatal period (Day 1 and Day 28 post partum) and at 6 months after delivery, in the cohort of 60 mothers and exclusively breastfed babies. Zn level in the serum showed increase (significant at 6 months) during this period (mg/mL): Day 1: 0.52 +/- 0.12; Day 28; 0.59 +/- 0.19; 6 months: 0.68 +/- 0.28. The concentration of Zn in the milk showed an opposite (decreasing) trend during the follow up: Day 1: 4.70 +/- 1.74 mg/L; Day 28: 2.65 +/- 1.06; 6 months: 0.46 +/- 0.36. A significant negative correlation was established between serum and milk [Zn] at day 28 (R = -0.338; p = 0.008), whereas a positive correlation was found at 6 months between these parameters (R = 0.306; p = 0.018). There was no significant correlation between [Zn] in the milk and serum and infants' body mass, mothers' age and mass at delivery. The level of Zn in the milk at 6 months of lactation is not sufficient to meet the recommended values. This implies that in Serbian population, Zn supplementation might be needed in the later phase of lactation.

AB  - Objectives: Milk banks collect, pasteurize, and freeze/store human milk. The processing may alter redox properties of milk, but the effects have not been fully examined. Methods: We collected 10 mature milk and 10 colostrum samples and applied a battery of biochemical assays and electron paramagnetic resonance spectroscopy to inspect changes that milk undergoes with pasteurization and 30 days storage at -20 degrees C. Results: Pasteurization and storage of raw milk did not affect total nonenzymatic antioxidative capacity, but specific components and features were altered. Urate radical and ascorbyl radical emerge as products of exposure of milk to hydroxyl radical-generating system. Processing shifted the load of antioxidative activity from ascorbate to urate and lowered the capacity of milk to diminish hydroxyl radical. Pasteurization caused a significant drop in the activity of 2 major antioxidative enzymes-superoxide dismutase and glutathione peroxidase, whereas freezing/storage of raw milk affected only superoxide dismutase. Colostrum showed drastically higher total nonenzymatic antioxidative capacity, hydroxyl radical scavenging ability, and glutathione reductase activity compared with mature milk. Conclusions: Pasteurization and storage affect nonenzymatic and enzymatic antioxidative agents in human milk. It appears that nonenzymatic antioxidative systems in colostrum and milk are different. The effects of processing may be partially compensated by fortification/spiking with ascorbate before use.

U mlađem gestacionom dobu koje karakteriše slabiji antioksidativni kapacitet može doći do povećanog stvaranja slobodnih radikala i reaktivnih kiseoničnih vrsta. Dosadašnjim istraživanjima je utvrđeno da majčino mleko ima veoma veliki antioksidativni kapacitet u odnosu na infant formule, koje su zamena za majčino mleko. Antioksidanti majčinog mleka još uvek nisu kompletno definisani, ali najvažniji su superoksid-dismutaza, glutation-peroksidaza, katalaza, vitamini C, A i E, koenzim Q, karotenoidi, cistein, koji sinergistički štite novorođenče od reaktivnih kiseoničnih vrsta u ranom životnom doba.Cilj ove doktorske disertacije je proučavanje ukupnog antioksidativnog potencijala infant i prelaznih formula i mleka za malu decu do 3 godine starosti i njihovo poredjenje sa ukupnim antioksidativnim svojstvima zrelog majčinog mleka i standardnog konzumnog kravljeg mleka, kao referentnog.Na osnovu dosadašnjih rezultata i aktuelnih naučnih saznanja u okviru ove oblasti istraživanja, u ovoj doktorskoj disertaciji primenjene su moderne laboratorijske tehnike i instrumentalne metode za određivanje ukupnog antioksidativnog potencijala hrane za bebe. Karakterizacija antioksidativnog potencijala hrane za bebe urađena je na osnovu: enzimskih aktivnosti uzoraka (određivanje enzimske aktivnosti superoksid-dismutaze-SOD i glutation-peroksidaze-GSH-Px), ispitivanja antioksidativnog potencijala hrane za bebe različitim elektrohemijskim metodama (potenciometrija, ciklična voltametrija, diferencijalna pulsna voltametrija, polarografija), elektron-paramagnetnom rezonancom (EPR) i spektrometrijskim metodama (UV/VIS)...
AB  - Oxidative stress occurs even during the neonatal period. At the younger gestational age, which is characterized by weaker antioxidant capacity, increased production of free radicals and reactive oxygen species is possible. Previous research has found that breast milk has very high antioxidant capacity compared to infant formula, which are a substitute for breast milk. Breast milk antioxidants are not fully defined yet, but the most important ones are superoxide dismutase, glutathione peroxidase, catalase, vitamins C, A and E, coenzyme Q, carotenoids, cysteine, which synergistically protect infants from reactive oxygen species at an early age.The objective of this dissertation is the study of total antioxidant potential of infant, follow-on and toddler formulas for children under 3 years of age and their comparison with the overall antioxidant properties of mature breast milk and standard cow's milk as a reference.Based on previous results and current scientific knowledge in this field of research, modern laboratory techniques and instrumental methods for the determination of total antioxidant potential of baby food were applied within this dissertation. AB  - The aim of this study is to investigate the significance of breast milk and infant formula in the prevention of oxidative stress, by electrochemical determination of the total antioxidant potential, demonstrating the relationship between the antioxidant capacity of milk and postnatal age. Human breast milk, commercial UHT milk, and infant formulas supplemented with prebiotics were used. Samples were diluted in phosphate buffer solution, and the total antioxidant activity was potentiometrically measured by the iodine/iodide redox couple with the Pt Fisher electrode as a working electrode and saturated calomel as a reference electrode. Cyclic voltammograms and differential pulse voltammograms were recorded with the glassy carbon electrode as the working electrode, an accessory platinum electrode, and an Ag/AgCl reference electrode. The potentiometric measurement indicates that human breast milk has the highest redox potential, while the commercial UHT milk has very low potential. Infant formulas also have high potential. The main advantage of electrochemical methods used to assess the total antioxidant activity of milk was that they directly monitored the electron-donating ability of the compounds and could be used for the quantitative analysis of the total antioxidants of different types of milk.
