AB  - “Bee pollen” is pollen collected from flowers by honey bees. It is used by the bees to nourish themselves, mainly by providing royal jelly and brood food, but it is also used for human nutrition. For the latter purpose, it is collected at the hive entrance as pellets that the bees bring to the hive. Bee pollen has diverse bioactivities, and thus has been used as a health food, and even as medication in some countries. In this paper, we provide standard methods for carrying out research on bee pollen. First, we introduce a method for the production and storage of bee pollen which assures quality of the product. Routine methods are then provided for the identification of the pollen’s floral sources, and determination of the more important quality criteria such as water content and content of proteins, carbohydrates, fatty acids, vitamins, alkaloids, phenolic and polyphenolic compounds. Finally, methods are described for the determination of some important bioactivities of bee pollen such as its antioxidant, anti-inflammatory, antimicrobial and antimutagenic properties. Métodos estándar Para la investigación del polenEl "polen de abeja" es el polen recogido de las flores por las abejas melíferas. El polen de abeja es utilizado para nutrir a las propias abejas, principalmente para proporcionar jalea real y alimento para las crías, pero también se utiliza para la nutrición humana. Para este último fin, se recoge en la entrada de la colmena en forma de gránulos que las abejas llevan a la colmena. El polen de abeja tiene diversas bioactividades, por lo que se hautilizado como alimento para la salud, e incluso como medicamento en algunos países. En este artículo, proporcionamos métodos estándar para llevar a cabo investigaciones sobre el polen de abeja. En primer lugar, presentamos un método de producción y almacenamiento de polen de abeja que garantiza la calidad del producto. A continuación, se ofrecen métodos de rutina para la identificación de las fuentes florales del polen y la determinación de los criterios de calidad más importantes, como el contenido de agua y de proteínas, carbohidratos, ácidos grasos, vitaminas, alcaloides y compuestos fenólicos y polifenólicos. Por último, se describen métodos para la determinación de algunas bioactividades importantes del polen de abeja, como sus propiedades antioxidantes, antiinflamatorias, antimicrobianas y antimutagénicas.

DFT study of the radical scavenging activity of isoxanthohumol, humulones (α-acids), and iso-α-acids from beer

AB  - Humulones and iso-humulones are potent natural antioxidants found in beer. In this study, density functional theory (DFT) method was applied for elucidating the structure-antioxidant activity relationship and molecular mechanism of antioxidant activity of eight bioactive humulones previously identified in different beer samples: isoxanthohumol, (R)- and (S)-adhumulone, cis- and trans-iso-adhumulone, cis- and trans-iso-n-humulone, and desdimethyl-octahydro-iso-cohumulone. The calculated bond dissociation enthalpies (BDEs) suggest that desdimethyl-octahydro-iso-cohumulone was the most potent compound with BDEs 5.1 and 23.9 kJ/mol lower compared to the values for resveratrol in gas phase and water, respectively. The enolic –OH is the most reactive site for hydrogen atom transfer (HAT). The presence of β-keto group with respect to enolic –OH diminishes the HAT potency via the formation of a strong intramolecular hydrogen bond. Another common antioxidant mechanism, single electron transfer followed by proton transfer (SET-PT), is only feasible for isoxanthohumol. The results of this study indicate a strong correlation between the increased antioxidant activity of beer products and the higher content of reduced iso-α-acids.

AB  - Taking into account a growing market and small number of articles related to honeydew honey, a metabolomic approach associated with multivariate analysis and modelling was proposed to discriminate five varieties of honey. Advanced analytical techniques were used for determination of 20 elements, 14 carbohydrates and stable carbon isotope ratio. No chemical marker has been found within sugar compounds, but several elements (Ba, Ca, Mg, Sr, Mn, Al, Co, Ni, Se) were marked as characteristic of honey type and allow classification of three botanical origins (Abies alba, Quercus frainetto, Quercus ilex). Sugars turanose, trehalose, arabinose and raffinose, elements Ba, Sr, P, Cd and Se, and δ13C values of honey, have different concentrations in honeys of the same botanical origin but harvested in different season. In addition to a confirmation of authenticity in terms of production, the values of δ13C of protein could be a good indicator of botanical origin.

AB  - Phytochemical analysis, the chemical investigation of plants, is a rapidly developing chemical discipline with various goals such as the determination of the substance classes and the isolation and qualitative and/or quantitative analyses of bioactive compounds or their structural analysis. The analysis of plants begins with optimizing extraction procedures because they play a key role in extraction outcomes, i.e., the isolation of bioactive compounds from natural products and the selection of the subsequent tests used in the analyses. The extraction is a critical step because it is necessary to extract the desired components of the complex natural matrix without destroying them. A wide range of techniques for the extraction of bioactive compounds from natural matrixes are currently available. This review aims to describe and compare the most commonly used methods, traditional/conventional and innovative/green. It will focus on the principles behind each method and on their strengths and limitations in order to help evaluate their suitability and economic feasibility, highlighting the advantages of new innovative techniques over conventional extraction procedures. Finally, the recent application of deep eutectic solvents as a new, eco-friendly method for the extraction of bioactive compounds from natural matrixes is also discussed.

AB  - Despite of the many beneficial health effects of strawberry tree (Arbutus unedo L.) honey due to its strong antioxidant activity derived mostly from polyphenols, a detailed phenolic profile has not been previously studied. The aims of this study were to identify the phenolic compounds, determine the total phenolic content (TPC) and evaluate the radical scavenging activity (RSA) of strawberry tree honey from south Croatia. Fifty-two polyphenolics (twenty-seven phenolic acids and twenty-five flavonoids) were identified using ultra-high-performance liquid chromatograph (UHPLC) coupled to a hybrid mass spectrometer (LTQ Orbitrap MS). Our overall results point to higher TPC (1038 mg gallic acid equivalents per kg of honey) and stronger RSA (3.32 mmol Trolox equivalents per kg of honey) compared to other monofloral honeys. Due to the presence of large quantities of polyphenolic compounds, strawberry tree honey may have great potential as a health promoting food.

AB  - Linden tea has been used in Serbian folk medicine for centuries to induce sweating for colds, relieve throat irritation and cough, reduce blood pressure, as well as a diuretic, spasmolytic, and as a sedative. The main aim of this study was to determine the comprehensive metabolite profile, free radical scavenging activity, and antimicrobial activities against 23 human and plant pathogens of both commercial and field-collected linden tea samples from Serbia. Developed high-performance thin-layer chromatography (HPTLC) method allowed identification of eight major metabolites in investigated samples, while forty-six metabolites were tentatively identified using high resolution mass spectrometry (HRMS): 14 phenolic acids and their derivatives, 15 glycosides, 9 flavonoids, and 8 procyanidins. Both commercial and field-collected linden tea samples showed similar total phenolic content and radical scavenging activity. Seven compounds such as p-hydroxybenzoic acid 4-O-hexoside, caffeic acid cinnamyl ester, pinocembrin, galangin, luteolin 7-O-glucuronide (and its isomer), isorhamnetin hexosyl hexoside were found in Tilia samples for the first time. Radical reagent 2,2-Diphenyl-1-picrylhydrazyl· (DPPH·) was used in DPPH-HPTLC assay which identified chlorogenic acid, caffeic acid, astragalin, quercitrin, tiliroside, and rutin as compounds that exhibit radical scavenging activity. Tilia samples showed inhibitory effect overwhelming only on Gram-positive bacteria, especially on Bacillus subtilis with the lowest MIC values observed, as well as towards Staphylococcus aureus and oral cavity isolates of Streptococcus pyogenes and Streptococcus mutans. Vaginal isolate of Candida glabrata showed more susceptibility than Candida albicans isolate. Generally, extract from Tilia cordata Miller (L4) showed the highest antimicrobial activity against the most of the tested pathogens, among all field-collected Tilia samples.

AB  - Erodium cicutarium is known for its total polyphenolic content, but this work reveals the first highly detailed profile of E. cicutarium, obtained with UHPLC-LTQ OrbiTrap MS4 and UHPLC-QqQ-MS/MS techniques. A total of 85 phenolic compounds were identified and 17 constituents were quantified. Overall, 25 new compounds were found, which have not yet been reported for the Erodium genera, or the family Geraniaceae. Along with methanolic extracts, the so far poorly investigated water extracts exhibited in vitro antioxidant activity according to all performed assays, including the ferric reducing/antioxidant power assay (FRAP), 2,2-diphenyl-1-picrylhydrazyl assay (DPPH), 2,2′-azinobis(3-ethylbenzthiazoline-6-sulfonic acid) assay (ABTS) and cupric ion reducing antioxidant capacity assay (CUPRAC). Elemental composition analysis performed with inductively coupled plasma atomic emission spectrometry (ICP-AES) and, additionally, hydride generation atomic absorption spectrometry (HydrEA-ETAAS) showed six most abundant elements to be decreasing as follows: Mg>Ca>K>S>P>Na, and gave first data regarding inorganic arsenic content (109.3–248.4 ng g−1). These results suggest E. cicutarium to be a valuable source of various phenolic compounds with substantial potential for further bioactivity testing.

A total of 85 phenolic compounds were identified and 17 constituents were quantified. Overall, 25 new compounds were found, which have not yet been reported for the Erodium genera, or the family Geraniaceae. Along with methanolic extracts, the so far poorly investigated water extracts exhibited in vitro antioxidant activity according to all performed assays, including the ferric reducing/antioxidant power assay (FRAP), 2,2-diphenyl-1-picrylhydrazyl assay (DPPH), 2,2′-azinobis(3-ethylbenzthiazoline-6-sulfonic acid) assay (ABTS) and cupric ion reducing antioxidant capacity assay (CUPRAC). Elemental composition analysis performed with inductively coupled plasma atomic emission spectrometry (ICP-AES) and, additionally, hydride generation atomic absorption spectrometry (HydrEA-ETAAS) showed six most abundant elements to be decreasing as follows: Mg>Ca>K>S>P>Na, and gave first data regarding inorganic arsenic content (109.3–248.4 ng g−1). These results suggest E. cicutarium to be a valuable source of various phenolic compounds with substantial potential for further bioactivity testing.",

AB  - The present study was carried out to evaluate the authenticity of local and imported brands of honey available in Tirana markets. The ranges of different parameters were: water content 11.10-18.57%, electrical conductivity 0.09-0.79 mS/cm, pH 3.37-4.58, free acidity 10.1-31.5 meq/kg and 5-hydroxymethylfurfural (HMF) 12.61-663.58 mg/kg. The most abundant sugars were fructose, glucose and sucrose, whereas two samples exhibit maltose concentration higher than 5 g/100 g. Generally the samples are within the limit values of standard of honey, except HMF values which the samples present high amounts, and specifically the samples M9 and M11 have respectively 638.7 and 663.58 mg/kg HMF, and of course in these samples the diastase activity was not detected. The high values of HMF and the absence of diastase activity, it is an indication of adulteration of honey with acid invert syrup. Also, sugar profile confirmed such behaviour, i.e. contents of sucrose and maltose was also above prescribed values. In addition the influence on the health was examined. High disaccharides concentration does not have direct negative influence, on the other hand, high HMF concentration can be very dangerous to human health. This compound is considered carcinogenic and prolonged exposure to high amounts can produce development of diseases such as liver and kidney failure, diabetes and development of various types of cancers.

AB  - Cilj ove studije je klasifikacija biološki različitih vrsta školjki na osnovu sadržaja esencijalnih elemenata primenom hemometrije. Analizitani uzorci su kupljeni u Incheonu, Koreja. Sadržaj esenicjalnih elemenata je određen primenom induktivno spregnute plazme kuplovane sa masenom spektrometrijom (ICP-MS) nakon mikrotalasne digestije. Hemometrijske tehnike pokazuju grupisanje ispitivanih uzoraka školjki prema sadržaju esencijalnih elemenata i identifikuju elemente najvažnije za klasifikaciju.
AB  - The main aim of current study was classification of four biologically different sellfish species such as bivalve molluscs Ruditapes philippinarum (Manila clam, MC), Yesso scallop (YS), Tegillarca granosa (TG) and Anadara broughtonii (AB) bought in the Incheon, South Korea. Content of essencial ellements such as Co, Cr, Cu, Mn, Ni, Se, Zn, and Fe were determined by using inductively coupled plasma mass spectrometry (ICP-MS) after closed-vessel microwave digestion. Chemometrics techniques showed classification of sellfish smaples based on biological species and identified elements most important for classification.

AB  - Bivalve molluscs, which include mussels, oysters and clams, have high nutritional value. They are regarded as a good source for proteins, lipids, carbohydrates and minerals [1]. On the other hand, seafood may also contain harmful contaminants and other undesirable substances such as mercury and persistent halogenated compounds, which has resulted in a number of risk-benefit assessments during the last decade [2].
Four species of bivalve molluscs Ruditapes philippinarum (Manila clam, MC), Yesso scallop (YS), Tegillarca granosa (TG) and Anadara broughtonii (AB) were bought in two fish markets in Incheon, Korea, in order to determine content of As, Cd, Co, Cr, Cu, Hg, Mn, Ni, Pb, Se, Zn, and Fe and consequently, to estimate the health hazards associated to dietary intake. The samples were analyzed by inductively coupled plasma mass spectrometry (ICP-MS) after closed-vessel microwave digestion. The analytical accuracy of the method was evaluated by using the SRM (TORT-2, lobster hepatopancreas). Application of principal component analysis (PCA) and hierarchical cluster analysis (HCA) showed a tendency to form three groups between samples belonging to different genus of samples.
European food safety authority (EFSA) has established recommended daily intake (RDI) values for Cu, Fe, Mn and Zn of 3, 14, 5 and 10 mg/day, respectively. We calculated the RDI for daily consumption in milligrams per 300 g of sample. Our results showed that these species could serve as a good dietary source of essential elements, especially Fe, Mn and Zn. However, all species showed As content higher than the maximum tolerable limit specified by EFSA. Seafood is the major contributor to As in the diet though As in seafood mostly occurs as organic As species [3]. In addition, content of Mn in Yesso scallop is few times higher than in other species.

An overview of recently published papers dealing with application of thin-layer chromatography in medicinal chemistry is presented.

Intercropping of soybean and proso millet for biomass production

AB  - Sustainable agriculture considers production of high quality food and feed with minimal impact on environment. Intercropping is one of the most efficient ways to produce valuable biomass for animal feed rich in nutrients. Intercrop combinations: alternating rows of soybean and proso millet (S-M), alternating strips with 2 rows of soybean and 2 rows of millet (SS-MM-SS) and alternating strips with 2 rows of soybean and 4 rows of millet (SS-MMMM-SS); single crop of soybean and single crop of proso millet were tested. The effect of bio-fertilizer Coveron was also followed. Aboveground biomass was harvested and land equivalent ratio (LER), as well as leaf area index (LAI) was determined. All intercropping combinations increased LAI of soybean when compared to monocrop, to 43% and 84% in SS-MM-SS combination with and without Coveron, respectively. Coveron slightly increased LAI. The highest values of biomass yield were obtained in S-M intercrop for soybean (39% and 42% higher in relation to monocrop, in combination with and without Coveron, respectively) and in monocrop for proso millet. Nevertheless, the highest LER value was obtained for SS-MM-SS combination without Coveron (1.12). In intercrops treated with Coveron slightly lower LER values were obtained for S-M and SS-MMMM-SS combination (0.97 and 0.98, respectively). Irrespective to sowing way, results indicate that 1:1 ratio of soybean and proso millet in intercrop (S-M and SS-MM-SS combinations) is the most promising way to achieve high biomass yield.

AB  - Honey is a valuable natural product which offers important nutritional and medicinal benefits. Consequently, it is frequently subjected to adulteration either by addition of various sugar syrups or cheaper and lower quality honey, or by mislabelling of declaration. In that sense, product authentication, as one of the most important food quality assurances, is essential in order to protect the consumers’ health and to avoid competition that can create a destabilized market. The authenticity of the honey has two aspects: the authenticity associated with the production of honey (sugar supplements, removal or addition of water, heating) and the authenticity of the honey related to its description, botanical and geographical origin.
Chemically, honey is a concentrated aqueous solution of various sugars, mainly fructose and glucose, but it also contains other mono-, oligo-, and polysaccharides. Some of the most important characteristics of honey include the sugars’ content, particularly the glucose and the fructose content and their total quantity, as well as a sucrose amount. Some authors demonstrated also that the ratio between glucose and fructose, as well as between some other characteristic sugars, is another parameter of interest in both defining botanical origin of honey and detection of its adulteration. Hence, a detailed analysis of sugars profile in honey samples is a useful tool for verifying product declaration, its botanical origin, as well as to uncover honey adulteration, and has been a long-time scientific preoccupation. At present, a variety of analytical techniques have been developed to determine the content of sugars in honey, such as spectroscopic, chromatographic, and electrochemical. The current compilation aims to provide the cross-section of the results on sugars composition, obtained by contemporary analytical methods, used in honey authentication.

AB  - Concerning the particular nutritive value of honeydew honey compared to blossom honey, and small number of studies defining botanical origin of honeydew honey, comprehensive analysis of phenolic profile of 64 honeydew honey samples of specific botanical origin was performed. Two advanced techniques of liquid chromatography hyphenated with mass spectrometry were used for identification of a total of 52 compounds and quantification of 25 of them. Pattern recognition analysis applied on data on phenolic compounds content confirmed that quercetin, naringenin, caffeoylquinic acid, hydroxyphenylacetic acid, apigenin and genistein, could be considered as potential markers of botanical origin of honeydew honey. Spectroscopic and electrochemical approaches were applied for the evaluation of the antioxidant capacity. Quercus sps. samples, Quercus frainetto and Quercus ilex, showed high biological activity and specific chemical composition. Additionally, cyclic voltammetry profiles were used for characterization and natural clustering of honeydew honey for the first time.

AB  - Concerning the particular nutritive value of honeydew honey compared to blossom honey, and small number of studies defining botanical origin of honeydew honey, comprehensive analysis of phenolic profile of 64 honeydew honey samples of specific botanical origin was performed. Two advanced techniques of liquid chromatography hyphenated with mass spectrometry were used for identification of a total of 52 compounds and quantification of 25 of them. Pattern recognition analysis applied on data on phenolic compounds content confirmed that quercetin, naringenin, caffeoylquinic acid, hydroxyphenylacetic acid, apigenin and genistein, could be considered as potential markers of botanical origin of honeydew honey. Spectroscopic and electrochemical approaches were applied for the evaluation of the antioxidant capacity. Quercus sps. samples, Quercus frainetto and Quercus ilex, showed high biological activity and specific chemical composition. Additionally, cyclic voltammetry profiles were used for characterization and natural clustering of honeydew honey for the first time.

AB  - Quality control is a crucial step in the production of effective and safe herbal remedies. The aim of this study was to develop a new, simple, and high throughput procedure for the quality assessment of herbal drugs using a high-performance thin-layer chromatography (HPTLC)/bioautography and UPLC–MS/MS approach combined with chemometrics. This was exemplarily shown for Morus alba L. root bark (sāng bái pí; SBP). Bioautography assays were developed for the identification of constituents with radical scavenging (DPPH assay) and antimicrobial activities (Bacillus subtilis, Escherichia coli) of 18 different M. alba samples, which was supported by UPLC–MS/MS analysis. Further, the combination of bioautography and chemometrics identified those samples with the most bioactive constituents. Plant materials collected from Serbia (11 samples) showed higher both radical scavenging and antimicrobial activities compared to samples provided from China (7 samples). Principal component analysis (PCA) confirmed the discrimination of geographically different samples and recognized their main markers responsible for differences between Serbian and Chinese samples. Most importantly for quality assessment, the combined HPTLC/bioautography and UPLC–MS/MS approach not only allowed for a fast chemical profiling of the investigated samples and their unambiguous identification, but also for the disclosure of major and minor bioactive constituents present in SBP.
