AB  - Considering the health-benefits of berry fruits consumption and increased market demands for food authenticity
as one of the most important quality assurances, phenolic profiling by high-performance thin layer chroma-
tography and ultra-high-performance liquid chromatography hyphenated with mass spectrometry was combined
with multivariate analysis for phytochemical characterization and intercultivar discrimination of cultivated
berry seeds. The phenolic profiles of 45 berry seeds from nine genuine Serbian cultivated fruit species (straw-
berry, raspberry, blackberry, black currant, blueberry, gooseberry, cape gooseberry, chokeberry, and goji berry)
revealed a good differentiation according to botanical origin. In order to determine biomarkers responsible for
the classification, a total of 103 phenolic compounds were identified, including 53 phenolic acids and their
derivatives, 26 flavonoids and 24 glycosides. Biomarkers derived from the phenolic profile of berry seeds proved
to be a powerful tool in the authentication of botanical origin, and may be useful in detection of frauds in berry-
based seed-containing product.

AB  - Lipophilicity and ionization constant of sixteen fentanyl analogues were investigated by reversed-phase thin-
layer chromatography (RP-TLC). Fourteen compounds have nitrogen containing groups at C-3 position of the
piperidine ring and two compounds are 3-carbomethoxy derivatives of fentanyl. Among them, five are diaste-
reomeric cis/trans couples and six novel trans diastereomers. In addition, the lipophilicity and ionization constant
of fentanyl were also determined by the same method, as a reference. The physicochemical property, lip-
ophilicity, expressed as retention indices RM
0 , b, and C0, as well as PC1, was determined and correlated with in
silico values. Ionization constants were determined on the basis of the relationships between analyte’s retention
expressed as RF and mobile phase pH. Calculated structural descriptors together with the retention indices, were
subjected to the principal component analysis – PCA and hierarchical cluster analysis – HCA, in order to provide
basic insights into the similarities among the studied compounds. The blood – brain barrier (BBB) permeation
was investigated in the function of experimentally obtained values of lipophilicity, polar surface area and mo-
lecular weight. In general, results of the present research corroborate well with previously determined anti-
nociceptive activities of the investigated compounds, pointing out that besides the cis/trans isomerism, another
set of important parameters should be taken into account when designing new derivatives of C-3 substituted
fentanyl, especially lipophilicity, voluminosity and HBD/A character of the substituents. Accordingly, RP-TLC
can be considered as a valuable asset in the ligand-based drug design

The physicochemical property, lip-ophilicity, expressed as retention indices RM0 , b, and C0, as well as PC1, was determined and correlated with insilico values. Ionization constants were determined on the basis of the relationships between analyte’s retentionexpressed as RF and mobile phase pH. Calculated structural descriptors together with the retention indices, weresubjected to the principal component analysis – PCA and hierarchical cluster analysis – HCA, in order to providebasic insights into the similarities among the studied compounds. The blood – brain barrier (BBB) permeationwas investigated in the function of experimentally obtained values of lipophilicity, polar surface area and mo-lecular weight. In general, results of the present research corroborate well with previously determined anti-nociceptive activities of the investigated compounds, pointing out that besides the cis/trans isomerism, anotherset of important parameters should be taken into account when designing new derivatives of C-3 substitutedfentanyl, especially lipophilicity, voluminosity and HBD/A character of the substituents. Additionally, they show a higher phenolic content and a stronger antioxidant activity, in comparison to commercial cultivars. Therefore, they are more likely to be used as a functional food. The subjects of this study were seventeen samples of fruits and leaves from autochthonous apple cultivars, five international standard cultivars, and six resistant apple cultivars. The phenolic profile was determined using ultra-high performance liquid chromatography (UHPLC), coupled with a diode array detector and a TSQ Quantum Access Max triple-quadrupole mass spectrometer. A total of twenty compounds were quantified in the samples. Most of the analyzed phenolics were detected in higher amounts in the peel, compared to the mesocarp. The results of the multivariate analysis of variance (MANOVA) indicate that 5-O-caffeoylquinic acid is present in the highest amount in the mesocarp, while in the peel and leaves, quercetin-glycosides were detected in the highest amount. According to the MANOVA: phloretin, phlorizin, 5-O-caffeoylquinic acid, kaempferol, and p-coumaric acid are present in significantly higher levels in the autochthonous cultivars, compared to the standard and resistant ones (in both fruits and leaves). Therefore, these compounds can be used as chemical tracers of the apple varietal origin.

AB  - Product authentication is one of the most important food quality assurances. Considering the importance of
consumption of berry fruits with proven health-beneficial properties, high sensory values and rich composition in
bioactive substances, the aim of this study was to evaluate a straightforward and simple procedure for the protein
fingerprinting of berry seeds. For this purpose, protein profiles of 45 samples of genuine berry fruit cultivars
(strawberry, raspberry, blackberry, black currant, blueberry, gooseberry, chokeberry, cape gooseberry, and
gojiberry) were analyzed by SDS-PAGE electrophoresis in combination with advanced chemometric tools. The
most important parameters for discrimination among berry seeds were polypeptides at 12.8; 15.1; 25.0; 26.4;
30.0; 41.8; 44.4; 46.0; 48.5; 52.3 and 56.4 kDa. Biomarkers obtained from the protein profile of berry seeds
proved to be a powerful tool in the authentication of their botanical origin, as well as for potential detection of
berry-based products adulteration

This work confirmed that unsaturated fatty acids (oleic and linoleic fatty acids) were the most represented in peach kernel oil and their content depended of peach genotype. Additionally, the fatty acids in combination with length, weight, and moisture of peach kernels could be used as a parameter of authenticity assessment. This research may contribute for the peach cultivar discrimination and recommendation of cultivars/genotypes with high kernel quality which could be used for the extractions of oil rich in unsaturated fatty acids and further use in food, pharmaceutical and cosmetic industry. AB  - Research background. Considering the importance of consumption of berry fruits with
proven health-beneficial properties and difficulties in quality control of products of specific botanical and geographic origin, a fingerprint method was developed, based on advanced data analysis (pattern recognition, classification), in order to relate the variability
of nutrients in the selected cultivars to primary metabolite profile.
Experimental approach. Forty-five samples of genuine berry fruit cultivars (strawberry,
raspberry, blackberry, black currant, blueberry, gooseberry, chokeberry, cape gooseberry
and goji berry) were characterized according to chromatographic profiles of primary metabolites (sugars, lipids and fatty acids) obtained by three chromatographic techniques
(high-performance thin-layer chromatography, gas chromatography coupled to mass
spectrometry, and high-performance anion-exchange chromatography with pulsed amperometric detection).
Results and conclusions. Comprehensive analysis allowed monitoring and identification
of metabolites belonging to polar lipids, mono-, di- and triacylglycerols, free fatty acids,
free sterols, sterol esters, mono- to heptasaccharides and sugar alcohols. Chemical fingerprint of berry seeds showed the uniformity of primary metabolites within each fruit species, but revealed differences depending on the botanical origin. All three chromatographic methods provided a discriminative, informative and predictive metabolomics
methodology, which proved to be useful for chemotaxonomic classification.
Novelty and scientific contribution. A novel methodology for the identification of bioactive compounds from primary metabolites of natural products was described. The proposed untargeted metabolite profiling approach could be used in the future as a routine
method for tracing of novel bioactive compounds. The knowledge of metabolite composition obtained in this study can provide a better assessment of genotypic and phenotypic differences between berry fruit species and varieties, and could contribute to the development of new breeding programs.

AB  - The strawberry tree (Arbutus unedo L., Ericaceae family) is an evergreen Mediterranean
shrub whose leaves and fruits are used in traditional medicine due to their antioxidant, antimicrobial,
antidiabetic, diuretic, and antiproliferative properties. The health benefits are mainly attributed to the
presence of phenolic compounds. The aim of this study was to compare the phenolic profiles, total
phenolic content (TPC), and radical scavenging activity (RSA) of A. unedo leaves and fruits collected
at two locations in Croatia. Phenolic profiles were identified using an ultra-high-performance liquid
chromatograph (UHPLC) coupled with a hybrid mass spectrometer (LTQ Orbitrap MS). TPC was
determined by Folin–Ciocalteu’s assay, while RSA was investigated using DPPH reagent. A total of
64 phenolics (60 and 42 compounds in leaves and fruits, respectively) were identified. Hyperoside
and flavan-3-ols were predominant compounds in leaves, while gallocatechin and catechin were
the major compounds found in fruits. To the authors’ knowledge, 16 and 5 phenolics in leaves and
fruits, respectively, were reported for the first time. Principal component analysis (PCA) showed
that UHPLC-LTQ Orbitrap MS could be used to identify which phenolics were able to discriminate
samples regarding plant tissue and geographical origin. TPC in leaves and fruits were in the ranges of
67.07–104.74 and 16.78–25.86 mg gallic acid equivalents (GAE)/g dried weight (dw), respectively. RSA
for leaves and fruits were in the ranges of 408.92–430.98 and 74.30–104.04 µmol Trolox equivalents
(TE)/g dw, respectively. The number of identified phenolics was lower in fruits compared to leaves.
Such a large number of bioactive phenolics identified and the strong antioxidant activity pointed to
A. unedo as a promising health-promoting plant and natural food preservative.

The analyses of rutin and quercetin content in honey were performed by high-performance liquid chromatography (HPLC) with ultraviolet (UV) detection. The content of rutin and quercetin in the flower was also determined to check the quality of the herbal drug used, as well as to evaluate the effectiveness of the extraction.ResultsThe extraction of rutin and quercetin, in a honey sample using 10% of the Sophora flower, stored at room temperature, for 40 days, was the most intense one (150.24 mg/kg of rutin and 1338.93 mg/kg of quercetin). Sophora flower corresponds to the pharmacopoeia requirements for the quality of an herbal drug. The concentrations of rutin and quercetin in acacia honey were 0.08 and 0.14 mg/kg, respectively. The concentration of rutin in honey from the melliferous pasture of the Sophora flower was 0.40 mg/kg, while quercetin was below the detection limit (0.1 mg/kg).ConclusionsHoney, enriched with rutin and quercetin from Sophora flower, had sensory acceptability and pleasant taste and aroma.

to antimicrobial investigation guided with its ethnobotanical use. The results of performed in vitro
screening on Staphylococcus aureus, Pseudomonas aeruginosa, and Candida albicans strains, show that
E. cicutarium has antimicrobial activity, with a particular emphasis on clinical S. aureus strains—
both the methicillin sensitive (MSSA) and the methicillin resistant (MRSA) S. aureus. Experimental
design consisted of general methods (the serial microdilution broth assay and the agar well diffusion
assay), as well as observing bactericidal/bacteriostatic activity through time (the “time‐kill”
assay), investigating the effect on cell wall integrity and biofilm formation, and modulation of bacterial
hemolysis. Observed antibacterial activity from above‐described methods led to further activity‐
guided fractionation of water and methanol extracts using bioautography coupled with UHPLCLTQ
OrbiTrap MS4. It was determined that active fractions are predominantly formed by gallic acid
derivatives and flavonol glycosides. Among the most active phytochemicals, galloyl‐shikimic acid
was identified as the most abundant compound. These results point to a direct connection between
galloyl‐shikimic acid and the observed E. cicutarium antibacterial activity, and open several new
research approaches for future investigation

AB  - Strawberry tree (Arbutus unedo L.) honey (STH) is widely used as part of traditional medicine
in the Mediterranean area, especially in the treatment of wounds, burns and infections. In
this study, we investigated the cyto-/genoprotective properties of STH at cell level following
short-term exposure to UVB radiation at 2 kJ/m2. Isolated human peripheral blood lymphocytes
(PBL) were selected as model cells. Phenolics in STH were determined using an ultrahigh-
performance liquid chromatograph (UHPLC) coupled to a linear ion trap-Orbitrap hybrid
mass spectrometer (LTQ Orbitrap MS), while homogentisic acid (HGA) was quantified using a
gas chromatograph-mass spectrometer (GC-MS). Primary DNA damage in PBLs treated with
STH before and after irradiation was assessed by alkaline comet assay, while their viability
was determined using the apoptosis/necrosis assay. The levels of thiobarbituric acid reactive
substances (TBARS) and glutathione (GSH) were determined in plasma samples. STH applied
as pre-treatment at a concentration equivalent to its average daily portion efficiently counteracted
the cytotoxic and genotoxic effects and diminished UVB-induced oxidative stress.
Post-radiation treatment with STH was not as beneficial. Protective effects could be associated
to the complex phytochemical profile of STH and the high content of HGA (306.8mg/
kg). It was confirmed that an intake of a complex mixture of bioactive constituents as STH
prior to potential cyto/genotoxic insults stimulated a broad-spectrum of protective mechanisms,
including intracellular antioxidative defence and DNA repair systems that helped pretreated
cells to counteract harmful exposures more efficiently.

AB  - Strawberries are an important fruit in the European diet because of their unique taste and high content of essential nutrients and bioactive compounds. The anthocyanins are known to be colorful phenolics in strawberries. In 17 samples of six strawberry cultivars produced in Serbia, i.e., the common varieties Alba, Asia, and Clery as well as promising breeding materials (11.29.11, 11.34.6, and 11.39.3), the anthocyanin profile as well as antimicrobial and antioxidative activity profiles were determined. All investigated extracts showed antioxidative and antibacterial activities against Gram-negative Aliivibrio fischeri. The responses were quite similar in number and intensity. The HPTLC-DPPH• scavenging assay and HPTLC-Aliivibrio fischeri bioassay coupled with high-resolution mass spectrometry identified pelargonidin-3-O-glucoside (Pg-3-glc) as the main anthocyanin and prominent antioxidative and antimicrobial compound in strawberries. The density functional theory calculations at the M06-2X/6-31+G(d,p) level showed that Pg-3-glc quenches free radicals via sequential proton loss electron transfer mechanism in water and in pentyl ethanoate, where the 5-OH group is the most reactive site for proton and hydrogen atom transfer. The results were confirmed via spectrophotometry. The highest total phenolic content was found in Clery and 11.39.3, while statistically significant differences between the genotypes regarding the antioxidant activity were not confirmed. Although very similar in the anthocyanin, antioxidative, and antimicrobial profile patterns, the strawberry genotypes were successfully classified using principal component analysis.

AB  - To replace common organic solvents that present inherent toxicity and have high volatility and to improve the extraction efficiency, a range of natural deep eutectic solvents (NADESs) were evaluated for the extraction of phenolic compounds from Agrimonia eupatoria. Screening of NADES efficiency was carried out based on the total phenolic and flavonoid content and radical-scavenging activity, determined by spectrophotometry, as well as phenolic compounds quantified, obtained using ultra-high-performance liquid chromatography with a diode array detector and a triple-quadrupole mass spectrometer. Increased extraction efficiency when compared with organic solvent was achieved using NADES mixtures choline chloride (ChCl):urea 1:2 and choline chloride:glycerol 1:1. Flavonol glycosides were the most abundant compounds in all extracts. The COSMO-RS model provided insights into the most important intermolecular interactions that drive the extraction process. Moreover, it could explain the extraction efficiency of flavonol glycosides using ChCl:glycerol NADES. The current article offers experimental evidence and mechanistic insights for the selection of optimal NADES to extract bioactive components from Agrimonia eupatoria. © 2022 by the authors.

The density functional theory calculations at the M06-2X/6-31+G(d,p) level showed that Pg-3-glc quenches free radicals via sequential proton loss electron transfer mechanism in water and in pentyl ethanoate, where the 5-OH group is the most reactive site for proton and hydrogen atom transfer. The results were confirmed via spectrophotometry. The highest total phenolic content was found in Clery and 11.39.3, while statistically significant differences between the genotypes regarding the antioxidant activity were not confirmed. Although very similar in the anthocyanin, antioxidative, and antimicrobial profile patterns, the strawberry genotypes were successfully classified using principal component analysis.

The anthocyanins are known to be colorful phenolics in strawberries. bioactive compounds are becoming increasingly popular. This review aims to provide an overview of thin-layer
chromatography methods used in quality control, authenticity testing and chemical profiling of bee-products in
order to help scientists engaged in the field of bee-products chemistry to utilize the advantages of this technique
in the detection and elimination of fraudulent practices in bee-product manufacturing. Recently, hyphenation of
thin-layer chromatography, image analysis and chemometrics support bee-products analysis by simultaneous determination of analytes with different detection principles, identification of individual
bioactive compounds as well as structure elucidation of compounds. Highlighted opportunities of thin-layer

AB  - The European Commission requires that fruit products distributed on the market meet standards of high quality and authenticity. For the quality assessment of apricot products susceptible to food fraud, an environmentally friendly, simple and cost-effective analytical profiling was developed by high-performance thin-layer chromatography multi-imaging (HPTLC-FLD/Vis). The new phytochemical profiling was applied for analysis of authentic samples (7 apricot and 5 pumpkin samples) and simulated adulterated products (11 mixture samples prepared by addition of 2.5–53% pooled pumpkin to pooled apricot). Based on the analytical eco-scale assessment, the HPTLC-FLD/Vis method was proven as an excellent green analytical method with low energy and solvent consumption. Chemometric data analysis confirmed the difference between apricot and apricot-pumpkin mixtures based on the phytochemical profile. Chemical markers responsible for their differentiation were identified. The results indicated that frauds by adding pumpkin to apricot products can be detected at added contents as low as 2.5%.

AB  - Sweet maize has worldwide importance due to its high nutritional value and health benefits. In order to provide valuable health-related information to consumers, the phytochemicals profile of raw sweet maize kernels and those exposed to different treatments (cooking, frozen storage before cooking, sucrose addition during cooking, and blanching) was examined herein. Thus, carotenoids, tocopherols and free phenolic acids content was determined by liquid chromatography, while the content of water soluble sugars was analyzed by anion exchange chromatography. In response to immediate cooking, as well as, to blanching prior to shorter frozen storage and cooking afterwards, all hybrids exhibited a significant decrease in carotenoids, and a significant increase in concentration of tocopherols, fructose, glucose, protocatechuic, ferulic and p-coumaric acids, in comparison to the fresh samples. However, treatments with blanching prior to longer frozen storage and cooking afterwards, contributed only to increased tocopherols content. Distinct grouping of processed samples in comparison to their corresponding control samples, revealed the existence of unique linkage between changes in phytochemicals content upon treatment in a hybrid-dependent manner. Combined blanching prior to shorter frozen storage with sucrose addition during cooking is highly recommended as a potent tool for phytochemicals enhancement.

AB  - Over the past few years, fermented garlic and its extracts have been increasingly used in cooking and in the daily diet due to their specific taste, nutritional composition and health benefits. The fermentation process is performed by heat treatment of garlic with controlled humidity for a longer period of time. During fermentation, chemical reactions and transformations such as Maillard reactions and caramelization reactions cause the changes in taste, nutritional composition, content of macro- and microelements as well as the content of phenolic compounds and antioxidant activity of garlic. In this paper, samples of garlic and fermented garlic were analyzed and the content of phenolic compounds, antioxidant activity, macro- and microelements as well as nutritional composition were compared. The content of Ca, K, Mg, Na, Mn, Cu, Zn, and W increased, while the content of Fe, Al, Cr, Ni, Mo, Hg and Pb decreased during fermentation. The water content decreased during the fermentation process, while the sugar and carbohydrate content increased significantly as a result of thermal decomposition of the poly- and oligosaccharides (fructan and other complex polysaccharides). Accordingly, the energy value of fermented garlic is higher than that of fresh garlic. The content of total phenolic compounds is higher in the fermented sample compared to fresh one, indicating  different chemical transformations of secondary metabolites during the fermentation process. As a measure of antioxidant activity, two assays were performed: DPPH and FIC and both showed higher activity of fermented garlic, which is positively correlated with the higher content of phenolic compounds in the sample.
