Copper(II) complexes with 4-(diethylamino)salicylaldehyde and α-diimines: Anticancer, antioxidant, antigenotoxic effects and interaction with DNA and albumins

AB  - In this article, cytotoxicity, the mechanisms of cytotoxic activity, genotoxicity, and interaction with DNA and proteins, of two Cu(II) complexes with a salicylaldehyde derivative (4-(diethylamino)salicylaldehyde) and α-diimine (2,2′-bipyridine (bipy) and 1,10-phenanthroline (phen)) are reported. Both Cu(II) complexes performed cytotoxic effects against all tested malignant cell lines. Complexes exerted highest cytotoxicity against HeLa and A375 malignant cell lines. The cytotoxic activity of Cu(II) complex with phen as a α-diimine co-ligand was significantly higher in comparison with cytotoxic activity of Cu(II) complex with bipy. Pretreatment with specific inhibitors of caspase-3, caspase-8 or caspase-9, in order to clear up the mode of cell death triggered by two Cu(II) complexes in HeLa cells, indicated the ability of these complexes to induce apoptosis through activation of target caspases. Cu(II)-phen complex exhibited significant antioxidant activity compared with Cu(II)-bipy complex, and showed a better effect on reducing intracellular ROS levels in HeLa cells. Tested complexes did not display genotoxic potential in human peripheral blood leucocytes, but exhibited an antigenotoxic effect in post-treatment, after H2O2 exposure. The study of the in vitro biological properties regarding their affinity towards CT (calf-thymus) DNA and serum albumins showed that the compounds can intercalate to CT DNA, and bind reversibly and tightly to the albumins. Molecular docking studies of the ability of compounds to bind to biomacromolecules are consistent with in vitro studies.

AB  - Two non-steroidal antiinflammatory drugs indomethacin and mefenamic acid were coordinated to Ru(II)-arenes to afford four new complexes. The cytotoxic activities of the ligands and ruthenium complexes were tested in three human cancer cell lines (K562, A549, MDA-MB-231) and non-tumour human fetal lung fibroblast cells (MRC-5) by MTT assay. Cytotoxicity studies revealed that indomethacin Ru(II)-arene complexes 1 and 3 displayed good cytotoxicity and apparent cytoselective profiles. The IC50 values obtained in leukemia K562 cells were comparable to those of cisplatin (10.3 mu M (CDDP), 11.9 mu M (1) and 13.2 mu M (3)). Flow cytometric analysis of 1 and 3 in triple-negative breast cancer MDA-MB-231 cells revealed an interesting mechanism of action. At IC50 concentrations, 1 and 3 arrested cell cycle progression in S phase and caused rapid accumulation of cells in sub-G1 phase (up to 48%), while Annexin V-FITC/PI staining showed simultaneous occurrence of apoptotic and necrotic cell populations at approximately similar levels of 20%. Measurement of reactive oxygen species (ROS) production by DCFH-DA staining confirmed the potential of 1 and 3 to increase ROS even more than cisplatin. The interaction of the complexes with serum albumins showed their potential ability to bind tightly and reversibly to albumins. The affinity of the complexes to calf-thymus DNA was investigated by UV-vis spectroscopy, viscosity measurements and fluorescence emission spectroscopy for competitive studies of the complexes with ethidium bromide, revealing that their interaction probably occurs via intercalation. Taken together, the results strongly suggest the potential of complexes 1 and 3 to alter cell cycle progression and cause DNA-damage by means of direct DNA-binding or indirectly by ROS production.

The complexes were characterized by spectroscopy (IR, UV-vis, H-1 NMR), elemental analysis and molar conductivity measurements. The structures for three complexes (2, 3 and 5) were determined by X-ray crystallography providing different coordination modes of the 5-Br-salicylaldehydo ligand. The complexes bind to DNA mainly by intercalation, as concluded by the viscosity measurements and the EB-displacement studies from the EB-DNA complex and present high DNA-binding constants (K-b), as calculated by the Wolfe-Shimer equation and plots. The interaction of the complexes with serum albumins was studied by fluorescence emission spectroscopy and the determined binding constants exhibit relative high values. (C) 2016 Elsevier Ltd. Based on spectroscopic results ( IR, UV, NMR), elemental analysis and conductivity measurements an octahedral geometry around cadmium metal ion is suggested, with the 5- NO2- salicylaldehyde ligand having different coordination modes. The structures determined by X-ray crystallography verified neutral mononuclear 1-3 and trinuclear 4. Simultaneous TG/DTG-DTA technique was used to analyze the thermal behavior of 1, 2, and 3. The complexes bind tightly to calf-thymus DNA mainly by intercalation, as concluded by DNA viscosity measurements and exhibit significant displacement of EB from the EB- DNA complex.

abstract = "Five Cd( II) complexes with the anion of 5- NO2- salicylaldehydeH ( 5- NO2- saloH) in the absence or presence of the alpha-diimine 1,10- phenanthroline ( phen), 2,2'- dipyridylamine ( dpamH), 2,2'- dipyridine ( bipy), or 2,9- dimethyl- 1,10- phenanthroline ( neoc) were synthesized and characterized as [Cd(5- NO2- salo)(2)(CH3OH)(2)] (1), [Cd(5-NO2- salo)(2)( phen)] center dot 2CH3OH center dot H2O (2), [Cd(5- NO2- salo) 2( dpam.)] (3), [Cd-3(5-NO2- salo)(6)(bipy)(2)] (4), and [ Cd(5-NO2-salo)(neoc)(NO3)](2) (5). Based on spectroscopic results ( IR, UV, NMR), elemental analysis and conductivity measurements an octahedral geometry around cadmium metal ion is suggested, with the 5- NO2- salicylaldehyde ligand having different coordination modes. The structures determined by X-ray crystallography verified neutral mononuclear 1-3 and trinuclear 4. Simultaneous TG/DTG-DTA technique was used to analyze the thermal behavior of 1, 2, and 3. The complexes bind tightly to calf-thymus DNA mainly by intercalation, as concluded by DNA viscosity measurements and exhibit significant displacement of EB from the EB- DNA complex.",
