AB  - IntroductionChokeberry is a plant belonging to the family Rosaceae which has a long history of edible and medicinal use. The aims of this study are: i) to investigate of HPTLC profile of the berry samples using HPTLC technique based metabolomics approach, ii) to determine the botanical origin of investigated chokeberry adulterations iii) to identify the most important botanical markers responsible for classification.MethodsA simple, rapid, and high-throughput planar chromatography (HPTLC) method was applied in combination with multivariate techniques for differentiating chokeberry samples and identification of the main metabolites of chokeberry and four common adulterants. Images of HPTLC chromatograms obtained using two different detections i.e. visible light and derivatization with natural products reagent were used as a dataset for multivariate analysis.ResultsPrincipal component analysis and Orthogonal Partial Least Squares Discriminant Analysis confirmed the discrimination of five botanically different samples and recognized their main markers such as responsible for differences between chokeberry and four common adulterants.ConclusionThe obtained results showed that the HPTLC-based metabolomics approach can be a very reliable technique for the detection of chokeberry adulteration.

Stabilization of rose hip oil by pomegranate peel extract during accelerated storage

AB  - Rose hip seed oil is a rich source of polyunsaturated fatty acids, as well as tocopherols, carotenoids, sterols, phospholipids, and phenolic com­pounds. On the other hand, due to the high content of polyunsaturated fatty acids, this oil is prone to oxidation. The aim of this study was to investigate the influence of a natural antioxidant, i.e., pomegranate peel extract, and its com­bination with butylated hydroxytoluene as a commonly used synthetic anti­oxidant, on the stability of rose hip oil. The stability of samples without and with different antioxidants was monitored through analysis of the fatty acid composition and measurement of the quality and stability parameters of the oil (peroxide value, p-anisidine value, thiobarbituric acid reactive substances inhi­bition, total phenolic content and antiradical activity) during a storage period of 12 days at 65 °C. Pomegranate peel extract (0.1 %) inhibited more effectively the second stage of oxidation than butylated hydroxytoluene (0.02 %), while the first stage of oxidation was better prevented by the synthetic antioxidant. Furthermore, the addition of pomegranate peel extract increased the total phen­olic content of the rose hip oil as well as its antiradical activity. Thus, pome­granate peel extract could be used as a potent natural antioxidant for the stabil­ization of beneficial but unstable rose hip oil.

Evaluation of the anti-cancer potential of Mahonia aquifolium extracts via apoptosis and anti-angiogenesis

AB  - The cytotoxicity of Mahonia aquifolium ethanol and water extracts was examined using MTT test. The morphological changes were analyzed by fluorescence microscopy. Cell cycle distribution and possible activation of caspase-dependent pathway of cell death were assessed by flow cytometry. The effects of ethanol and water extracts on migration of endothelial EA. hy926 cells were analyzed by in vitro scratch assay and inhibition of angiogenesis was detected using tube formation assay. Both extracts demonstrated cytotoxic effects on cancer cell lines with very high selectivity. Morphological evaluation indicated apoptosis. These results were confirmed with cell cycle analysis, where there was accumulation of cancer cells in the subG1 phase. Ethanol and water extracts induced a caspase-dependent apoptosis in HeLa cells through activation of caspase-3 and caspase-8. Both extracts showed the ability to inbibit the migration of EA. hy926 cells and initial steps of angiogenesis. In addition, ethanol extract exerted significant anti-angiogenic effect.

Medicinal plants in Northern Montenegro: Traditional knowledge, quality, and resources

AB  - The main objectives of this study were to collect information on the use of wild medicinal plants by local people living in the high mountain region of northern Montenegro and to conduct local botanical and ecological surveys. During terrain expeditions, 412 plant species (38 endemic) were identified, and the most abundant family was Rosaceae, followed by Cupressaceae and Lamiaceae. Plant use was documented by speaking with 75 people (mean age 57, 61∈% female, 39∈% male) who were born in and residents of northern Montenegro. Information on 116 plant species belonging to 40 families for the treatment of various human ailments was gathered. Botanical families, such as Asteraceae, Rosaceae, and Lamiaceae, were represented by the highest number of species reported by informants (14, 13, and 7 species, respectively). The most frequently reported medicinal uses were ones for treating gastrointestinal and respiratory diseases. Aerial parts (33.3∈%) were found to be highly utilized plant parts followed by roots (20.8∈%) and leaves (17.3∈%). Only two endemic species (Dianthus knapii and Pinus peuce) proved useful in folk tradition. Active ingredients of plant species officinal in the European Pharmacopoeia 6.0 (Ph. Eur. 6.0) were studied, and we assessed possibilities for commercial exploitation for local economic development. This work increases the ethnobotanical data from Montenegro, safeguards local folk knowledge, and provides information on new or scarcely reported properties of medicinal plants, whose traditional use requires experimental validation. © 2014 Springer Science+Business Media New York. Active ingredients of plant species officinal in European Pharmacopoeia 6.0 (Ph. Eur. 6.0) were studied and we assessed possibilities for commercial exploitation for local economic development. Materials and methods: The 75 people that were interviewed (40-82 years old) identified 94 species for treatment of various human ailments. For each named species, the following elements are provided: botanical name, family, part(s) used, medicinal use and perceived property, listing in published pharmacopoeias, the relative abundance of each species and locality where the plant was collected. Chemical analyses were done according to prescriptions of Ph. Eur. 6.0 in order to estimate potential commercial use of native plants. Results and conclusion: The most common in traditional usage were Rosaceae (11 species) making 11.7%, Asteraceae (10 species) 10.6% and Lamiaceae (7 species) 7.4%. From 94 species reported, 35 (37.2%) are officinal in Ph. Eur. 6.0 and 12 in national pharmacopoeias (12.8%). Aerial parts were mostly used (43.6%). AB  - The chemical composition and free radical scavenging properties of the MeOH extract of Hypericum richeri and its subsequent CHCl3, n-BuOH, EtOAc and aqueous extracts were investigated. Four quercetin-3-O-glycosides, three myricetin-3-O-glycosides, 3-O- and 5-O-caffeoylquinic acid, quercetin, I3,II8-biapigenin, pseudohypericin and hypericin were identified. The constituents of the extracts were quantified using HPLC-DAD (high-performance liquid chromatography-diode array detector). Isolation of the compounds from n-BuOH and EtOAc extracts was conducted by column chromatography. Antioxidant capacity of the extracts was evaluated by superoxide radical and ABTS root+ scavenging assays and inhibition of lipid peroxidation. The EtOAc extract of H. richeri was the most potent antioxidant in all experiments. Free radical scavenging compounds were identified by HPLC-DPPH root (high-performance liquid chromatography-2,2-diphenyl-1-picrylhydrazyl) post-column derivatisation method, where all phenolic compounds except I3,II8-biapigenin exhibited radical scavenging properties. This is the first report of the presence of myricetin-3-O-rutinoside, myricetin-3-O-galactoside, myricetin-3-O-glucoside, 5-O-caffeoylquinic acid and I3,II8-biapigenin in H. richeri.

AB  - Rutin (1, main constituent) and two flavone C-glycosides, vitexin (2) and vitexin 2 ''-O-alpha-rhamnopyranoside (3) were isolated from the aerial parts of Onobrychis montana subsp. scardica. They were identified by (1)H-NMR, (13)C-NMR and UV-Vis spectroscopy (procedure with shift reagents), and high resolution ESI-MS. A relatively high content of 1 (5.27 mg/g of dry plant material), measured by HPLC, indicated O. montana subsp. scardica as a new natural source of this biologically active compound. The isolated flavonoid compounds might be of value as chemotaxonomic markers.
AB  - Iz nadzemnih delova Onobrychis montana subsp. scardica izolovani su rutin (1, glavni sastojak) i dva flavonoidna S-glikozida, viteksin (2) i njegov 2''-O-α-ramnopiranozid (3). Njihova struktura je određena primenom 1H-NMR, 13C-NMR i UV spektroskopije (procedura sa reagensima UV-pomeranja) i masene spektrometrije visokog razlaganja (HR-ESI-MS). Relativno visok sadržaj rutina (5.27 mg/g suvog materijala) određen pomoću tečne hromatografije (HPLC), ukazuje na O. montana subsp. scardica kao nov prirodni izvor ovog biološki aktivnog jedinjenja. Izolovana jedinjenja mogu biti i od vrednosti kao hemotaksonomski markeri.
