AB  - The influences of the application of mono- and two-component organic modifiers on lipophilicity determination of 12 tetradentate Schiff bases by reversed-phase thin layer chromatography were investigated. The main goal is to estimate types of interaction between observed compounds and components of the applied chromatographic systems and establish some behaviour pattern in order to easier choose a combination of organic modifiers which will simulate interaction in biological systems based on the facts that the same basic intermolecular interactions are responsible for the behaviour of substances in both the biological and chromatographic system. The applied organic modifier shows the ability to modify the surface of the applied sorbent, which affects the manifestation of lipophilicity of the observed compounds. Mono-component organic modifiers from different groups of the Snyder triangle were used, as well as their two-component mixtures. In addition, we compared experimentally determined calculated parameters of lipophilicity.

Maillard reaction products formation and antioxidative power of spray dried camel milk powders increases with the inlet temperature of drying

AB  - Demand for camel milk (CM) is increasing worldwide, due to its high nutritious value and health benefits. In this study, whole CM powders were produced by spray drying (SD) at six inlet temperatures (190 °C–250 °C) and by freeze drying (FD). Physicochemical and functional properties of CM powder proteins were investigated. SD at higher inlet temperatures (230 °C–250 °C) resulted in higher extent of Maillard reaction (MR), in comparison to lower temperatures (190 °C–200 °C) and FD treatment. Both treatments had negative effect on casein solubility, while whey proteins remained soluble and slightly increased its solubility with the extent of MR. The CM powders obtained at higher inlet temperatures demonstrated improved antioxidant activity. Secondary structure of whey proteins did not differ among the samples, while surface hydrophobicity of whey proteins was higher in all SD than in FD samples, suggesting only limited denaturation of camel whey proteins at higher inlet temperatures of drying. Thus, the effects of SD under the conditions applied in our study did not decrease camel whey protein solubility, while drying procedure itself regardless of temperature decreased solubility of camel milk caseins. MR generated during CM processing could be an important means of compensating for the lack of antioxidant protection normally associated with β-lactoglobulin but happens to be absent from this milk.

AB  - Different Schiff base complexes have biological activities that make them suitable for drug design. The biological properties of tetradentate Schiff base copper(II) complexed with N,N′-bis(acetylacetone)propylenediimine have been studied. The cytotoxic activity towards Caco-2 cells were determined by MTT, Anexin V and PI apoptosis assays. N,N′-bis(acetylacetone)propylenediimine-copper(II) showed the anti-cancer and anti-proliferative properties by inducing apoptosis in Caco-2 cells. A comparison of the cytotoxic activity of this compound with cisplatin shows that it is more effective on the colorectal cancer cell line Caco-2. The binding capacity and interaction of N,N′-bis(acetylacetone)propylenediimine-copper(II) with HSA were systemically investigated by in vitro fluorescence spectroscopy, CD spectroscopy, and in silico molecular docking study. Furthermore, in vitro and in silico interaction studies indicated that the complex binds to HSA through a static quenching mechanism without changes in protein conformation. The calculated number of binding sites was in line with molecular docking studies. The obtained Ka value suggests that the compound can be released from the protein in target cells. The tetradentate Schiff base copper(II) complex exhibited in vitro biological activities against cancer epithelial cells, which depend on the molecular structure of the complex, causing apoptosis, and the complex can bind to the protein drug carrier in circulation to the target tissue.

AB  - Different Schiff base complexes have biological activities that make them suitable for drug design. The biological properties of tetradentate Schiff base copper(II) complexed with N,N′-bis(acetylacetone)propylenediimine have been studied. The cytotoxic activity towards Caco-2 cells were determined by MTT, Anexin V and PI apoptosis assays. N,N′-bis(acetylacetone)propylenediimine-copper(II) showed the anti-cancer and anti-proliferative properties by inducing apoptosis in Caco-2 cells. A comparison of the cytotoxic activity of this compound with cisplatin shows that it is more effective on the colorectal cancer cell line Caco-2. The binding capacity and interaction of N,N′-bis(acetylacetone)propylenediimine-copper(II) with HSA were systemically investigated by in vitro fluorescence spectroscopy, CD spectroscopy, and in silico molecular docking study. Furthermore, in vitro and in silico interaction studies indicated that the complex binds to HSA through a static quenching mechanism without changes in protein conformation. The calculated number of binding sites was in line with molecular docking studies. The obtained Ka value suggests that the compound can be released from the protein in target cells. The tetradentate Schiff base copper(II) complex exhibited in vitro biological activities against cancer epithelial cells, which depend on the molecular structure of the complex, causing apoptosis, and the complex can bind to the protein drug carrier in circulation to the target tissue.

AB  - Demand for camel milk (CM) is increasing worldwide, due to its high nutritious value and health benefits. In this study, whole CM powders were produced by spray drying (SD) at six inlet temperatures (190 °C–250 °C) and by freeze drying (FD). Physicochemical and functional properties of CM powder proteins were investigated. SD at higher inlet temperatures (230 °C–250 °C) resulted in higher extent of Maillard reaction (MR), in comparison to lower temperatures (190 °C–200 °C) and FD treatment. Both treatments had negative effect on casein solubility, while whey proteins remained soluble and slightly increased its solubility with the extent of MR. The CM powders obtained at higher inlet temperatures demonstrated improved antioxidant activity. Secondary structure of whey proteins did not differ among the samples, while surface hydrophobicity of whey proteins was higher in all SD than in FD samples, suggesting only limited denaturation of camel whey proteins at higher inlet temperatures of drying. Thus, the effects of SD under the conditions applied in our study did not decrease camel whey protein solubility, while drying procedure itself regardless of temperature decreased solubility of camel milk caseins. MR generated during CM processing could be an important means of compensating for the lack of antioxidant protection normally associated with β-lactoglobulin but happens to be absent from this milk.

Proteins were dissolved in sodium phosphate buffer to 50 mg/ml and defatted prior Bradford assay of protein concentration, then analyzed by Size Exclusion-Ultra High-Performance Liquid chromatography (SE-UHPLC). The CM protein aggregation and their stability were further monitored at different pH 2.0, 4.0, and 7.5 via sodium dodecyl sulphate polyacrylamide gel electrophoresis (SDS-PAGE). Freeze dried CM (FDCM) was the reference sample and our results showed that GJ50/50 and PJ40/60 with the highest fruit juice ratio had the lowest protein content in the supernatant, hence the decreased solubility. SE-UHPLC of supernatants showed a slight decrease in retention times of 11 kDa and 62 kDa proteins for GJ50/50 and PJ40/60 suggesting a possibility of adduct formation due to fortification leading to higher molecular weight. The simulated static in vitro gastrointestinal digestion of samples revealed that most soluble proteins were readily digested by pepsin, trypsin and chymotrypsin enzymes leading to small peptides. However, the SDS PAGE of pellets showed the partial resistance of casein and α-lactalbumin against peptic digestion.

AB  - Objective. Camel milk is highly nutritious food with many health benefits proposed. Demand for camel milk has increased worldwide. Production of camel milk powders facilitate its transport, prolonge shelf-life, and also offer an attractive additive for various food products. In this study we examined the effect of freeze/spray drying treatment for camel milk powder production, on physiochemical and functional properties of camel milk proteins.
Material and Methods. Whole camel milk powders were prepared by spray drying treatment at six different inlet temperatures (190°C - 250°C) or by freeze drying. The soluble protein fractions upon the treatments were analysed by combination of electrophoretic and spectroscopic techniques. Structural and functional properties of camel milk proteins such as Maillard reaction products formation, antioxidant activity and protein solubility were assessed.
Results. SDS-PAGE revealed non-uniform increase in Mw of major protein bands, while native electrophoresis revealed non-uniform decrease in pI values with increased inlet temperature of spray drying. That indicated attachement of lactose moieties to NH2-group of proteins via non-enzymatic Maillard reaction. Spectrophotometric analysis showed formation of intermediate Maillard reaction products (increased absorbance at 294 nm) and no detectable late Maillard reaction products formation. Higher inlet temperatures (230°C - 250°C) resulted in higher protein carbonyls formation and lower content of free amino groups as a result of Maillard reaction. Far-UV circular dichroism spectra showed no differences in secondary structures between freeze and spray dried samples. Antioxidant activity and protein solubility were increased with increase in inlet temperature.
Conclusions. Our results showed that spray drying treatment promotes non-enzymatic glycation of camel milk proteins and exert significant effects on the techno-functional properties of CM powder such as nutritional value and shelf life. Thus, optimization of spray drying parametars is essential for production of high quality camel milk powders.

Production of camel milk powders facilitate its transport, prolonge shelf-life, and also offer an attractive additive for various food products. In this study we examined the effect of freeze/spray drying treatment for camel milk powder production, on physiochemical and functional properties of camel milk proteins.
Material and Methods. Whole camel milk powders were prepared by spray drying treatment at six different inlet temperatures (190°C - 250°C) or by freeze drying. The soluble protein fractions upon the treatments were analysed by combination of electrophoretic and spectroscopic techniques. Structural and functional properties of camel milk proteins such as Maillard reaction products formation, antioxidant activity and protein solubility were assessed.
Results. SDS-PAGE revealed non-uniform increase in Mw of major protein bands, while native electrophoresis revealed non-uniform decrease in pI values with increased inlet temperature of spray drying. That indicated attachement of lactose moieties to NH2-group of proteins via non-enzymatic Maillard reaction. Spectrophotometric analysis showed formation of intermediate Maillard reaction products (increased absorbance at 294 nm) and no detectable late Maillard reaction products formation. Higher inlet temperatures (230°C - 250°C) resulted in higher protein carbonyls formation and lower content of free amino groups as a result of Maillard reaction. Far-UV circular dichroism spectra showed no differences in secondary structures between freeze and spray dried samples. Antioxidant activity and protein solubility were increased with increase in inlet temperature.
Conclusions. Our results showed that spray drying treatment promotes non-enzymatic glycation of camel milk proteins and exert significant effects on the techno-functional properties of CM powder such as nutritional value and shelf life. Thus, optimization of spray drying parametars is essential for production of high quality camel milk powders.

AB  - A new glucose biosensor for selective and sensitive determination of glucose in beverage samples was developed. A copper complex with a Schiff base, N,N’-bis(acetylacetonato) propylenediimine, was used as a mediator. It was mixed with glucose oxidase (GOx) and prepared as carbon paste electrode (CPE). Cyclic voltammetry was used for electrochemical measurements. The biosensor showed improved sensitivity when enzyme mediator ratio was 1 : 5, and working pH was 6.6 in 0.1 M phosphate buffer. The linear response of the biosensor for glucose concentration was in the range from 0.5 to 10 mM, with low detection limit of 0.36 mM (calculated as 3 s/n) and sensitivity of 0.16 μA/mM. The reproducibility of the biosensor, calculated as relative standard deviation, was 1.9% for 6 intra-day measurements. Biosensor was stored at +4 °C between experiments, and it was stable during 14 days after the preparation. The developed biosensor was applied on determination of glucose in commercial beverages (apple and cherry juice) and obtained results showed good agreement with the reference method.

AB  - The carbon stable isotope content of Cornelian cherry stones collected from wild tree stands in Serbia, SE Europe, was measured using elemental analyser-isotope ratio mass spectrometry, with the aim of recording natural carbon isotope composition of the fruit stones and its possible variation. The results show a significant variation in the carbon isotope values; we identified several environmental factors that, along with a number of other possible determinants, likely contributed to this variation. The obtained data are compared with the measurement of carbon isotope content of an archaeological specimen of Cornelian cherry stone discovered at the Neolithic site of Vina (ca. 5600-4500 BC) in Serbia. Notwithstanding the limitedness of the data and the complexity surrounding carbon fractionation and the isotopic variation, it is suggested that the differences/similarities in carbon isotope ratios between modern and archaeological Cornelian cherry stones, when measured for much larger assemblages, could potentially offer a glimpse into growing conditions of Cornelian cherry trees in the past.

AB  - Pyrolysis of high density polyethylene (HDPE) in the open system was studied. A plastic bag for food packing was used as a source of HDPE. Pyrolysis was performed at temperatures of 400, 450 and 500 degrees C, which were chosen based on thermogravimetric analysis. The HDPE pyrolysis yielded liquid, gaseous and solid products. Temperature rise resulted in the increase of conversion of HDPE into liquid and gaseous products. The main constituents of liquid pyrolysates are 1-n-alkenes, n-alkanes and terminal n-dienes. The composition of liquid products indicates that the performed pyrolysis of HDPE could not serve as a standalone operation for the production of gasoline or diesel, but preferably as a pre-treatment to yield a product to be blended into a refinery or petrochemical feed stream. The advantage of a liquid pyrolysate in comparison to crude oil is the extremely low content of aromatic hydrocarbons and the absence of polar compounds. The gaseous products have desirable composition and consist mainly of methane and ethene. The solid residues do not produce ash by combustion and have high calorific values. Co-pyrolysis of HDPE with mineral-rich lignite indicated positive synergetic effect at 450 and 500 degrees C, which is reflected through the increased experimental yields of liquid and gaseous products in comparison to theoretical ones.

AB  - The is work presents the development of a flow injection system for differential pulse amperometry (DPA) for diazepam determination in the presence of oxygen. The thin flow cell consisted of the bare glassy carbon electrode, reference silver/silver chloride, and stainless steel as the auxiliary electrode. Electrochemical reduction of diazepam (DZP) was characterised by cyclic voltammetry. Azomethine reduction peak was used for DZP quantification. The detector response was linear in the range 20-250 mu mol/dm(3) of diazepam, with a calculated detection limit of 3.83 mu g/cm(3). Intraday and interday precision were 1.53 and 10.8%, respectively. The method was applied on three beverage samples, energetic drink, and two different beer samples, and obtained recoveries were from 93.65 up to 104.96%. The throughoutput of the method was up to 90 analyses per hour.

Chromatographic investigation of series of 36 Schiff bases and their copper(II) and nickel(II) complexes was performed under both normaland reverse-phase conditions. Chemical structures of the compounds were characterized by molecular descriptors which are calculated from the structure and related to the chromatographic retention parameters by multiple linear regression analysis. Effects of chelation on retention parameters of investigated compounds, under normal-and reverse-phase chromatographic conditions, were analyzed by principal component analysis, quantitative structure-retention relationship and quantitative structure-activity relationship models were developed on the basis of theoretical molecular descriptors, calculated exclusively from molecular structure, and parameters of retention and lipophilicity.

AB  - Diazepam belongs to the group of benzodiazepine drugs, and its consumption with alcohol enhances its sedative effect and absorption rate. That is why it is often used as “date rape drug” [1]. In our experiments we applied the flow injection system on diazepam determination on a bare glassy carbon electrode. The electrochemical determination of diazepam is based on reduction of the azomethine group yielding a dyhidrodiazepam. The single cathodic peak is caused by the reduction of diazepam by a two electron change. The reduction potential is highly dependent on pH value of the electrolyte, shifting to more negative potentials with higher pH [2].

AB  - The lipophilic character of polydentate Schiff bases was investigated by reversed-phase thin-layer chromatography. In order to determine retention parameter as a measure of the lipophilicity of the tested compounds, three different binary solvent systems, composed of water and organic modifier (methanol, acetonitrile, and tetrahydrofuran), were used. Lipophilicity parameters together with 2D molecular descriptors were subjected to the multivariate statistical analysis. Principal component analysis was employed to reveal the most influential factors governing the lipophilicity of the investigated compounds. Finally, the influence of organic modifier on determination of lipophilicity parameters was discussed.

AB  - The retention behavior and lipophilicity parameters of some antiphychotics were determined using reversed-phase thin layer chromatography. Quantitative structure-activity relationships studies have been performed to correlate the molecular characteristics of observed compounds with their retention as well as with their chromatographically determinated lipophilicity parameters.
The effect of different organic modifiers (acetone, tetrahydrofuran, and methanol) has been studied. The retention of investigated compounds decreases linearly with increasing concentration of organic modifier. The chemical structures of the antipsychotics have been characterized by molecular descriptors which are calculated from the structure and related to chromatographically determinated lipophilicity parameters by multiple linear regression analysis. This approach gives us the possibility to gain insight into factors responsible for the retention as well as lipophilicity of the investigated set of the compounds. The most prominent factors affecting lipophilicity of the investigated substances are Solubility, Energy of the highest occupied molecular orbital, and Energy of the lowest unoccupied molecular orbital. The obtained models were used for interpretation of the lipophilicity of the investigated compounds. The prediction results are in good agreement with the experimental value. This study provides good information about pharmacologically important physico-chemical parameters of observed antipsychotics relevant to variations in molecular lipophilicity and chromatographic behavior. Established QSAR models could be helpful in design of novel multitarget antipsychotic compounds.

AB  - Schiff bases play an important role in inorganic chemistry as they easily form stable complexes
with majority of transition metal ions. Complexes of Schiff bases have been recognized as
models for biologically important species. The aim of this work is to investigate chromatographic
behaviour of series of 12 tetradentate Schiff bases and corresponding copper(II) and
nickel(II) complexes by normal- and reversed-phase thin layer chromatography, and optimization
of chromatographic conditions for their separation. Chromatographic methods have been
frequently applied for determination of retention parameter (RM), which is used for quantitative
relationships between chemical structure and retention. The chemical structures of investigated
compounds can be characterized by molecular descriptors. Quantitative structure-retention relationship
(QSRR) describes how molecular structure affects chromatographic retention. In the case
of thin layer chromatography, the QSRR studies are usually based on RM value, which is defined
by Bate-Smith and Westall eguation. In this study we used two stationary and twenty two
mobile phases. Also, in this work we used multivariant regression analysis to confirm the optimal
chromatographic conditions.

AB  - Several natural isolates of Bacillus strains namely 5B, 12B, 16B, 18 and 24B were grown at two different temperatures in submerged fermentation for the production of raw-starch-digesting alpha-amylases. All strains except Bacillus sp. 18 produced more alpha-amylase at 37 degrees C. The hydrolysis of raw cornstarch followed the same pattern. Efficient hydrolysis was obtained with alpha-amylases from Bacillus sp. 5B, 12B, 16B and 24B grown at 37 degrees C and Bacillus sp. 18 grown at 50 degrees C. Zymography after isoelectric focusing showed that alpha-amylases were produced in multiple forms, from 2 to 6, depending on the strain when they were growing at 37 degrees C, while growth at 50 degrees C induced only 1 or 2 isoforms. Thin layer chromatography (TLC) analysis of the hydrolysis products of raw corn and soluble starch by alpha-amylases revealed the production of various mixtures of oligosaccharides. In most cases, G3 was the most dominant product from soluble starch while G2, G3 and G5 were the main products of raw starch hydrolysis. This indicates that the obtained alpha-amylases could be used for starch liquidification or short-chain-oligosaccharide formation, depending on the type of starch (raw or soluble) used for the hydrolysis.
