AB  - Stinging nettle (Urtica dioica L.) is one fantastic plant widely used in folk medicine, pharmacy, cosmetics, and food. This plant’s popularity may be explained by its chemical composition, containing a wide range of compounds significant for human health and diet. This study aimed to investigate extracts of exhausted stinging nettle leaves after supercritical fluid extraction obtained using ultrasound and microwave techniques. Extracts were analyzed to obtain insight into the chemical composition and biological activity. These extracts were shown to be more potent than those of previously untreated leaves. The principal component analysis was applied as a pattern recognition tool to visualize the antioxidant capacity and cytotoxic activity of extract obtained from exhausted stinging nettle leaves. An artificial neural network model is presented for the prediction of the antioxidant activity of samples according to polyphenolic profile data, showing a suitable anticipation property (the r2 value during the training cycle for output variables was 0.999).

AB  - Stinging nettle (SN) is an extraordinary plant from the Urticaceae botanical family. It is well-known and widely used in food and folk medicine to treat different disorders and diseases. This article aimed to study the chemical composition of SN leaves extracts, i.e., polyphenolic compounds and vitamins B and C, because many studies ascribed high biological potency to these compounds and their significance in the human diet. Besides the chemical profile, the thermal properties of the extracts were studied. The results confirmed presence of many polyphenolic compounds and vitamins B and C. It also showed that the chemical profile closely correlated with the applied extraction technique. The thermal analysis showed that analyzed samples were thermally stable up to about 160 °C. Thermal degradation of samples UAE, MAE, and MAC took place in four steps, and sample SE in three steps. Altogether, results confirmed the presence of health-beneficial compounds in stinging nettle leaves and indicated the possible application of its extract in pharmaceutical and food industries as both a medicinal and food additive.

Процена степена оптерећења површинских вода органским загађујућим супстанцама на подручју Ковина, Србија

AB  - У индустријској зони града Ковина налазe се фабрике које се баве производњом алкохола и сточног квасца, хране за кућне љубимце, сечењем и завршном обрадом челика, производњом шприцева и амбалаже за вакцине и лекове, као и највећа на Балкану фарма печурака. Поред тога, на 11 km од Ковина је рудник који угаљ вади са дна језера које је повезано са Дунавом, и представља јединствену подводну експлоатацију угља на свету. Упркос наведеним бројним делатностима које могу имати утицаја на квалитет површинских вода, нема лабораторије која се бави њиховим мониторингом. Стога је циљ овог рада прелиминарна карактеризација степена оптерећења површинских вода укупно присутним и специфичним, најзаступљенијим угљоводоничним загађујућим супстанцама. Узорци коришћени у раду узети су током новембра 2022. године, са пет репрезентативних локација на подручју Ковина (слика 1). Након одређивања основних параметара (табела 1), воде су анализиране применом стандардне методе ASTM D1252-06 [1], како би се утврдиле вредности хемијске потрошње кисеоника (ХПК). У циљу специфицирања најзаступљенијих угљоводоничних загађујућих супстанци, примењена је екстракција узорака вода помоћу трихлорметана. Добијени екстракти су након сушења и концентровања хроматографски пречишћени и анализирани на гасном хроматографу Agilent 7890A GC куплованом са квадруполним масеним детектором Agilent 5975C. Четири од пет анализираних узорака показују вредности ХПК карактеристичне за класу III површинских вода (табела 1; [2]). Најзаступљeнија специфична угљоводонична једињења у свим узорцима су n-алкани (слика 1). Нижи n-алкани са мање од 20 угљеникових атома су присутни само у траговима, што указује да ове воде углавном нису изложене загађењу сировом нафтом, дизелом или керозином. У три узорка (Дунавац, Шљункара и Црна Бара), CPI вредности за цео опсег n-алкана (С16- С34), као и за и више (С25-С34) и ниже (С15-С20, са изузетком Шљункаре) n-алканске хомологе су ниже или блиске 1, што уз n-алканске максимуме у опсегу С24-С26 указује на допринос органске супстанце акватичних макрофита, бактерија, васкуларних биљака и маховина рода Sphagnum [3]. Присуство нижих n-алкана (С15-С20) карактерише алге и бактерије, док се доминација непарних нижих хомолога у узорку Шљункара може повезати за цијанобактеријама. У узорицима Канал и Поњавица CPI вредности за цео опсег n-алкана (С16-С34), као и за више хомологе (С25-С34) веће су од 1, што уз n-алкански максимум у узорку Канал на С27 указује на допринос виших сувоземних биљака. Међутим поред наведених природних извора идентификованих n-алкана, треба истаћи да веома сличне n-алканске расподеле имају моторна уља, теже фракције уља за ложење и течни производи пиролизе лигнита. Од посебног значаја је и резултат да је у екстракту узорка воде са локације Поњавица доминантно једињење елементарни сумпор (слика 1), што се може повезати са великом количином муља у којем преовлађују аноксични услови, услед чега су фаворизовани редукциони хемијски/биохемијски процеси. Резултати прелиминарног истраживања показују да је неопходно наставити континуално и знатно детаљније истраживање предметних вода, како би се предупредило њихово даље загађење и негативан утицај на живи свет у њима, као и на подземне воде и земљиште у непосредној околини.

Preliminarno ispitivanje sastava aromatične frakcije ekstrakta prašine iz biblioteke Hemijskog fakulteta u Beogradu

AB  - Usled modernog načina života ljudi provode oko 90 % vremena u zatvorenim prostorijama,
kao što su kuće/stanovi, kancelarije, automobili itd. Prašina je sve prisutna u svim
pomenutim sredinama i nalazi se kao čestice suspendovane u vazduhu ili istaložena na
površinama. Čestice prašine se mogu lako udahnuti i mogu da oštete respiratorni sistem i
pluća. Pored toga, čestice prašine akumuliraju hemikalije iz zatvorenih prostorija i
predstavljaju važan put izlaganja ljudi tim jedinjenjima. U kućnoj prašini se mogu naći
ftalati, policiklični aromatični ugljovodonici i ostala poluisparljiva jedinjenja.
Ftalati se često koriste kao plastifikator u proizvodnji polivinil hlorida (PVC-a) i predmeta
opšte upotrebe kao što su građevinski materijal, dečije igračke i nameštaj, ali i u proizvodnji
podnih obloga, veštačke kože, lepkova, tepiha, maziva, cevi, žica, sredstava za čišćenje
kuće, proizvoda za negu tela (fiksatori mirisa, proizvodi za negu kože, pene i šamponi) i
ambalaže za namirnice. Primena ftalata zavisi od njihove molekulske mase. Ftalati male
molekulske mase, kao što su di-metil-ftalat (DMP), di-etil-ftalat (DEP), di-propil-ftalat
(DPP), di-izobutil-ftalat (DiBP), di-n-butil ftalat (DBP), di-n-pentil-ftalat (DNPP) i benzil-
butil-ftalat (BBP), se koriste u rastvaračima, lepkovima, voskovima, mastilu, lekovima,
kozmetici i insekticidima.Visokomolekulski ftalati, kao di-2-etilheksil-ftalat (DEHP), di-
ciklo-heksil-ftalat (DCHP) i di-izo-nonil-ftalat (DiNP), su plastifikatori u PVC proizvodima
i predmetima od opšte upotrebe.
Estri ftalata su povezani sa velikim brojem zdravstvenih problema kod ljudi. Pre svega,
ftalati deluju kao endokrini disruptori i doprinose stvaranju varijacija u reprodukcionom
sistemu kao što su opadanje broja spermatozoida, oštećenje DNK u spermatozoidima,
prevremeni razvoj dojki i promena u reproduktivnim hormonima. Ftalati utiču na pojavu
astme, alergija i poremećaja funkcije nervnog sistema. Pored toga, DEHP je okarakterisan
kao 2b kancerogena supstanca, tj. kao potencijalno kancerogen za ljude, od strane IARC-a
(Međunarodna agencija za istraživanje raka, eng. International Agency for Research on
Cancer). Treba napomenuti da su pojedini metaboliti ftalata često toksičniji od samih
nepromenjenih ftalata. Ljudi mogu biti izloženi ftalatima inhalacijom, ingestijom i
apsorpcijom preko kože.
Oko 1 g istaložene prašine je sakupljen iz biblioteke Hemijskog fakulteta u Beogradu, sa
visine iznad 1m od poda. Uzorci su odmereni, ekstrahovani, prečišćeni i aromatična frakcija je snimljena metodom gasne hromatografije, povezanom sa masenim spektrometrom.

Dobijeni rezultati ukazuju na to da, pored prirodnih proizvoda kao što su metil estri masnih
kiselina (koje su najverovatnije znatno obilnije u alifatičnoj frakciji), uzorak sadrži 3,5-di-
terc-butil-4-hidroksibenzaldehid, metabolit 2,6-di-terc-butil-4-methilfenola, koji se koristi
kao antioksidans u namirnicama i kozmetičkim proizvodima [3]. Najzastupljenija jedinjenja
u aromatičnoj frakciji su DiBP), DBP i DEHP. Interesantno je primetiti da se detektovani
ftalati koriste u dve različite svrhe, kao aditivi u plastici i drugim proizvodima (DiBP i DBP)
i kao plastifikatori u polimerima (DEHP). Kao što je prethodno pomenuto, DEHP je
klasifikovan kao potencijalno kancerogena supstanca za ljude. Pored toga, primena DiBP,
DBP i DEHP je ograničena od strane Evropske komisije. Zbog prisustva ftalata koji
imaju veliki uticaj na ljudsko zdravlje u ekstraktu prašine, potrebno je izvršiti dodatna
ispitivanja na većem broju uzoraka i uraditi detaljnu kvantifikaciju i procenu rizika po
ljudsko zdravlje.

abstract = "Usled modernog načina života ljudi provode oko 90 % vremena u zatvorenim prostorijama,
kao što su kuće/stanovi, kancelarije, automobili itd. Prašina je sve prisutna u svim
pomenutim sredinama i nalazi se kao čestice suspendovane u vazduhu ili istaložena na
površinama. Čestice prašine se mogu lako udahnuti i mogu da oštete respiratorni sistem i
pluća. Pored toga, čestice prašine akumuliraju hemikalije iz zatvorenih prostorija i
predstavljaju važan put izlaganja ljudi tim jedinjenjima. U kućnoj prašini se mogu naći
ftalati, policiklični aromatični ugljovodonici i ostala poluisparljiva jedinjenja.
Ftalati se često koriste kao plastifikator u proizvodnji polivinil hlorida (PVC-a) i predmeta
opšte upotrebe kao što su građevinski materijal, dečije igračke i nameštaj, ali i u proizvodnji
podnih obloga, veštačke kože, lepkova, tepiha, maziva, cevi, žica, sredstava za čišćenje
kuće, proizvoda za negu tela (fiksatori mirisa, proizvodi za negu kože, pene i šamponi) i
ambalaže za namirnice. Primena ftalata zavisi od njihove molekulske mase. Ftalati male
molekulske mase, kao što su di-metil-ftalat (DMP), di-etil-ftalat (DEP), di-propil-ftalat
(DPP), di-izobutil-ftalat (DiBP), di-n-butil ftalat (DBP), di-n-pentil-ftalat (DNPP) i benzil-
butil-ftalat (BBP), se koriste u rastvaračima, lepkovima, voskovima, mastilu, lekovima,
kozmetici i insekticidima.Visokomolekulski ftalati, kao di-2-etilheksil-ftalat (DEHP), di-
ciklo-heksil-ftalat (DCHP) i di-izo-nonil-ftalat (DiNP), su plastifikatori u PVC proizvodima
i predmetima od opšte upotrebe.
Estri ftalata su povezani sa velikim brojem zdravstvenih problema kod ljudi. Pre svega,
ftalati deluju kao endokrini disruptori i doprinose stvaranju varijacija u reprodukcionom
sistemu kao što su opadanje broja spermatozoida, oštećenje DNK u spermatozoidima,
prevremeni razvoj dojki i promena u reproduktivnim hormonima. Ftalati utiču na pojavu
astme, alergija i poremećaja funkcije nervnog sistema. Pored toga, DEHP je okarakterisan
kao 2b kancerogena supstanca, tj. kao potencijalno kancerogen za ljude, od strane IARC-a
(Međunarodna agencija za istraživanje raka, eng. International Agency for Research on
Cancer). Treba napomenuti da su pojedini metaboliti ftalata često toksičniji od samih
nepromenjenih ftalata. Ljudi mogu biti izloženi ftalatima inhalacijom, ingestijom i
apsorpcijom preko kože.
Oko 1 g istaložene prašine je sakupljen iz biblioteke Hemijskog fakulteta u Beogradu, sa
visine iznad 1m od poda. Uzorci su odmereni, ekstrahovani, prečišćeni i aromatična frakcija je snimljena metodom gasne hromatografije, povezanom sa masenim spektrometrom.
Lokacija uzorkovanja i hromatogram aromatične frakcije je prikazan na slici 1.

Испитивање ефикасности екстракције виших n-алкана из ваздуха

AB  - Испитивање угљоводоника у ваздуху претежно обухвата лакоиспарљива једињења и полицикличне ароматичне угљоводонике, док се мање пажње посвећује n-алканима дугог низа. Антропогено порекло виших n-алкана углавном се приписује угљу, биомаси, моторним уљима и уљима за ложење, док је њихово биогено порекло резултат физиолошких процеса виших сувоземних биљака, као и појединих микроорганизама. Виши n-алкани (>С20) су стабилна, неиспарљива, у води нерастворна једињења, те се у ваздуху претежно налазе адсоpбовани на честицама, и то углавном PM2.5. Стога је значајно да се ова једињења у ваздуху анализирају и да се детаљније расветли њихова хемодинамика. Циљ овога рада је испитивање брзе и јефтине методе за екстракцију виших n-алкана из ваздуха, при чему су као растварачи коришћени n-хексан и дихлорметан. Наведени растварачи су одабрани због способности да добро растварају угљоводонике и економске приступачности. Узорци ваздуха узети су са висине од 2 метра, мерено од асфалта, на локацији Капетан Мишине улице у Београду. Ова улица се налази у центру града, умерено је оптерећена само аутомобилским саобраћајем, али је транспорт знатно мање интензиван него на аутопуту и прометним саобраћајницама кроз које се крећу возила градског превоза. Ваздух је помоћу аутоматског узоркивача са пумпом на електрични погон, уз константан проток, увођен у стаклену испиралицу напуњену прописаним количинама n-хексана, односно дихлорметана. Након дефинисаног времена узорковања, из добијених екстраката је најпре уклоњена влага, а потом су концентровани на ротационом вакуум упаривачу. Добијени концентрати су пречишћени на хроматографској колони са активираним магнезијум-силикатом и анализирани на гасном хроматографу Agilent 7890A GC куплованом са квадруполним масеним детектором Agilent 5975C. На слици 1 приказан је хроматограм виших n-алкана у узорку екстрахованом дихлорметаном, који је показао већи апсорпциони потенцијал у односу на n-хексан, при идентичним експерименталним условима. У екстрактима ваздуха идентификовани су виши n-алкани у опсегу од C21 до C32. CPI (Carbon Preference Index) вредности за цео опсег n-алкана (С21-С31) износиле су 0,99. n-Алканске расподеле се карактеришу највећом заступљеношћу хомолога С23-С26, са максимумима на С24 и С25. Веома сличне n-алканске расподеле са уједначеном заступљеношћу парних и непарних хомолога имају моторна уља, теже фракције уља за ложење и течни производи пиролизе лигнита. Природни извори С23-С26 алкана су акватичне макрофите, бактерије, васкуларне биљке и маховине рода Sphagnum. Непарни n-алкани С27-С31 карактеристични су за више сувоземне биљке, при чему се доминација С27 и С29 углавном везује за дрвеће и жбуње, а С31 за траве и маховине. Добијени резултати показују да је коришћени експериментални поступак екстракције дихлорметаном ефикасан за анализу виших n-алкана у ваздуху. Поред тога, у питању је брза, јефтина метода, која је креирана тако да захтева минималан број аналитичких фаза, чиме се губици аналита своде на најмању меру, што је важно узимајући у обзир њихову ниску концентрацију у ваздуху. Резултати испитивања су основа за наставак истраживања, у циљу оптимизације методе за екстракцију и квантификацију виших n-алкана у узорцима ваздуха.

AB  - Research background. Considering the importance of consumption of berry fruits with
proven health-beneficial properties and difficulties in quality control of products of specific botanical and geographic origin, a fingerprint method was developed, based on advanced data analysis (pattern recognition, classification), in order to relate the variability
of nutrients in the selected cultivars to primary metabolite profile.
Experimental approach. Forty-five samples of genuine berry fruit cultivars (strawberry,
raspberry, blackberry, black currant, blueberry, gooseberry, chokeberry, cape gooseberry
and goji berry) were characterized according to chromatographic profiles of primary metabolites (sugars, lipids and fatty acids) obtained by three chromatographic techniques
(high-performance thin-layer chromatography, gas chromatography coupled to mass
spectrometry, and high-performance anion-exchange chromatography with pulsed amperometric detection).
Results and conclusions. Comprehensive analysis allowed monitoring and identification
of metabolites belonging to polar lipids, mono-, di- and triacylglycerols, free fatty acids,
free sterols, sterol esters, mono- to heptasaccharides and sugar alcohols. Chemical fingerprint of berry seeds showed the uniformity of primary metabolites within each fruit species, but revealed differences depending on the botanical origin. All three chromatographic methods provided a discriminative, informative and predictive metabolomics
methodology, which proved to be useful for chemotaxonomic classification.
Novelty and scientific contribution. A novel methodology for the identification of bioactive compounds from primary metabolites of natural products was described. The proposed untargeted metabolite profiling approach could be used in the future as a routine
method for tracing of novel bioactive compounds. The knowledge of metabolite composition obtained in this study can provide a better assessment of genotypic and phenotypic differences between berry fruit species and varieties, and could contribute to the development of new breeding programs.

AB  - Acute ischemia/reperfusion (I/R) liver injury is a clinical condition challenging to treat. Meldonium is an anti-ischemic agent that shifts energy production from fatty acid oxidation to less oxygen-consuming glycolysis. Thus, we investigated the effects of a 4-week meldonium pre-treatment (300 mg/kg b.m./day) on the acute I/R liver injury in Wistar strain male rats. Our results showed that meldonium ameliorates I/R-induced liver inflammation and injury, as confirmed by liver histology, and by attenuation of serum alanine- and aspartate aminotransferase activity, serum and liver high mobility group box 1 protein expression, and liver expression of Bax/Bcl2, haptoglobin, and the phosphorylated nuclear factor kappa-light-chain-enhancer of activated B cells. Through the increased hepatic activation of the nuclear factor erythroid 2-related factor 2, meldonium improves the antioxidative defence in the liver of animals subjected to I/R, as proved by an increase in serum and liver ascorbic/dehydroascorbic acid ratio, hepatic haem oxygenase 1 expression, glutathione and free thiol groups content, and hepatic copper-zinc superoxide dismutase, manganese superoxide dismutase, catalase, glutathione peroxidase, and glutathione reductase activity. Based on our results, it can be concluded that meldonium represent a protective agent against I/R-induced liver injury, with a clinical significance in surgical procedures.

AB  - Sepsis is a life-threatening condition caused by the dysregulated and overwhelming response to infection, accompanied by an exaggerated pro-inflammatory state and lipid metabolism disturbance leading to sequential organ failure. Meldonium is an anti-ischemic and anti-inflammatory agent which negatively interferes with lipid metabolism by shifting energy production from fatty acid oxidation to glycolysis, as a less oxygen-demanding pathway. Thus, we investigated the effects of a four-week meldonium pre-treatment on faecal-induced sepsis in Sprague-Dawley male rats. Surprisingly, under septic conditions, meldonium increased animal mortality rate compared with the meldonium non-treated group. However, analysis of the tissue oxidative status did not provide support for the detrimental effects of meldonium, nor did the analysis of the tissue inflammatory status showing anti-inflammatory, anti-apoptotic, and anti-necrotic effects of meldonium. After performing tissue lipidomic analysis, we concluded that the potential cause of the meldonium harmful effect is to be found in the overall decreased lipid metabolism. The present study underlines the importance of uninterrupted energy production in sepsis, closely drawing attention to the possible harmful effects of lipid-mobilization impairment caused by certain therapeutics. This could lead to the much-needed revision of the existing guidelines in the clinical treatment of sepsis while paving the way for discovering new therapeutic approaches.

AB  - The main goal of this study was to establish the chemical profile of Osage orange (Maclura pomifera) leaf extracts, obtained by conventional maceration technique, and to examine its antimicrobial activity. The identification and quantification of the extract compounds were done using ultra‐high‐performance liquid chromatography, with a diode array detector coupled with triple‐quadrupole mass spectrometer and gas chromatography–mass spectrometry techniques. Thirty‐one polyphenolic compounds were detected and identified in the ethanolic extracts, whereby 5‐O‐caffeoylquinic acid was found to be the dominant compound. Among other compounds, pentacosane and palmitic acid were the most abundant compounds in the dichloromethane extract. The preliminary antimicrobial activity screening shows that Gram‐positive bacteria tend to be more sensitive to the investigated extracts. The highest antimicrobial activity was determined against Enterococcus faecalis ATCC 19433 and Listeria monocytogenes ATCC 35152. From these results, Osage orange leaves can be considered as plant material with significant antimicrobial properties.

Evaluation of novel green walnut liqueur as a source of antioxidants: Multi-method approach

AB  - In this study, a novel green walnut liqueur (GWL) based on green walnuts, as the main ingredient, with the addition of fruits, aromatic plants, chocolate and honey, was produced at the pilot-scale. Antioxidant activity (AO) of the obtained GWL was determined using multiple AO assays in parallel, standard spectrophotometric (FC, DPPH and FRAP) and recently developed electrochemical ones, HydroxoPerhydroxoMercury(II) Complex and Mercury Reduction Antioxidant Power, and compared to similar commercial alcoholic beverages. Characterization of the GWL in terms of volatile and polyphenolic components was performed using SPE-GC-MS and HPLC-DAD-MS/MS, respectively. Sensory quality assessment was performed by experts in the field of sensory analysis of alcoholic beverages, using a scoring method. According to all AO assays applied, AO activity and total phenolics content of GWL were superior in comparison to commercial spirits considered. The volatile fraction of GWL was mainly composed of eugenol,alpha-terpineol and vanillin, while the most prevalent phenolic compounds were gallic (5.054 mg/L) and chlorogenic acid (1.307 mg/L) and flavonoids such as catechine (0.882 mg/L), quercetin (0.499 mg/L) and its sugar-conjugated derivatives, quercetin 3-O-glucoside (0.774 mg/L) and quercetin-3-O-rhamnoside (0.614 mg/L). Gallic acid is the major contributor to total AO activity, especially determined by DPPH and FRAP, followed by catechine, quercetin and chlorogenic acid. Among the terpenoids, contribution of eugenol to total AO activity is estimated as the highest. Excellent sensory quality (18.52 of maximum 20 scores) was ascribed to GWL. Consequently, the presence of bioactive compounds and high AO activity of GWL, in addition to high sensory quality score, indicates a high market potential for this high-value product.

Characterization of the GWL in terms of volatile and polyphenolic components was performed using SPE-GC-MS and HPLC-DAD-MS/MS, respectively. Sensory quality assessment was performed by experts in the field of sensory analysis of alcoholic beverages, using a scoring method. According to all AO assays applied, AO activity and total phenolics content of GWL were superior in comparison to commercial spirits considered. The volatile fraction of GWL was mainly composed of eugenol,alpha-terpineol and vanillin, while the most prevalent phenolic compounds were gallic (5.054 mg/L) and chlorogenic acid (1.307 mg/L) and flavonoids such as catechine (0.882 mg/L), quercetin (0.499 mg/L) and its sugar-conjugated derivatives, quercetin 3-O-glucoside (0.774 mg/L) and quercetin-3-O-rhamnoside (0.614 mg/L). Gallic acid is the major contributor to total AO activity, especially determined by DPPH and FRAP, followed by catechine, quercetin and chlorogenic acid. Among the terpenoids, contribution of eugenol to total AO activity is estimated as the highest. Excellent sensory quality (18.52 of maximum 20 scores) was ascribed to GWL. Consequently, the presence of bioactive compounds and high AO activity of GWL, in addition to high sensory quality score, indicates a high market potential for this high-value product.",

AB  - Laurus nobilis (bay laurel) form Serbia (SRBL) and Russia (RFBL) were analyzed in order to establish the chemical profile, thermal properties and biological activity. The GC/MS analysis showed that α-pinene (31.97%) is principal compound in the RFBL followed by eucalyptol (27.95%) and α-terpineol (10.28%). In the case of the SRBL, eucalyptol (40.51%) is the most abundant compound followed by α-terpineol (15.46%), while α-pinene was found in much lower amount (4.45%). Thermal analysis showed that the evaporation process of both oils was complex process and took place in two phases. Moreover, results indicated that the SRBL evaporated slower at the higher temperatures than the RFBL. In order to assess biological activity, the antioxidant and antimicrobial activities were investigated. The SRBL was more potent antioxidant agent in the case of both assays (DPPH and total reduction capacity). In the case of the antimicrobial activity, the SRBL showed greater potential of antimicrobial activities toward all tested microorganisms, while RFBL was found to be effective against all the bacteria and yeast, but did not possess any inhibitory action against fungi. Results indicate that both oils may be used in the food and pharmaceutical industries as antioxidant and antimicrobial agent.

AB  - Acute renal ischemia/reperfusion (I/R) injury is a clinical condition that is challenging to treat. Meldonium is an anti-ischemic agent that shifts energy production from fatty acid oxidation to less oxygen-consuming glycolysis. Thus, in this study we investigated the effects of a four-week meldonium pre-treatment (300 mg/kg b.m./day) on acute renal I/R in male rats (Wistar strain). Our results showed that meldonium decreased animal body mass gain, food and water intake, and carnitine, glucose, and lactic acid kidney content. In kidneys of animals subjected to I/R, meldonium increased phosphorylation of mitogen-activated protein kinase p38 and protein kinase B, and increased the expression of nuclear factor erythroid 2-related factor 2 and haeme oxygenase 1, causing manganese superoxide dismutase expression and activity to increase, as well as lipid peroxidation, cooper-zinc superoxide dismutase, glutathione peroxidase, and glutathione reductase activities to decrease. By decreasing the kidney Bax/Bcl2 expression ratio and kidney and serum high mobility group box 1 protein content, meldonium reduced apoptotic and necrotic events in I/R, as confirmed by kidney histology. Meldonium increased adrenal noradrenaline content and serum, adrenal, hepatic, and renal ascorbic/dehydroascorbic acid ratio, which caused complex changes in renal lipidomics. Taken together, our results have confirmed that meldonium pretreatment protects against I/R-induced oxidative stress and apoptosis/necrosis.

AB  - The main goal of this research was to determine the relationship among chemical structure, bioactivity and temperature of chamomile during subcritical water extraction in isobaric conditions (45 bar) at seven different temperatures (65-210 degrees C). The influence of temperature on phenolic profiles was defined by UHPLC-HESI-MS/MS. The overall results indicate that the presence of conjugated double bonds, side chains, glucose moiety or ether moiety in molecules influence the efficiency of polyphenols' extraction in subcritical water. In terms of antioxidant activity, the extracts were the most active towards ABTS radicals (IC50 = 7.3-16.8 mu g/mL), whereby temperature of 150 degrees C was optimal. On the other hand, the extracts obtained at 115 degrees C showed highest cytotoxicity. Inhibition of alpha-amylase and alpha-glucosidase was the highest at 65 and 85 degrees C, i.e. 0.51 and 4.13 mmolAE/g, respectively. Activity against tyrosinase was the highest at 210 degrees C (17.92 mgKAE/g). The data showed that different non-phenolic compounds may also participate in bio-activities of the extracts.

AB  - The study was designed to determine the relationship between chemical structure, bioactivity and pressure during the subcritical water extraction (SCW) of chamomile. Extraction was carried out at isothermal conditions (100 degrees C) at five different pressures (10, 30, 45, 60 and 90 bar). Twenty three polyphenolic compounds were identified in the extracts, whereby apigenin was found to be the dominant compound (61.53-1344.99 mg/kg). Results suggest that the lowest applied pressure has negligible effect on phenolic recovery from chamomile, but also the use of pressures above than 45 bar was proven as needlessly. By using in vitro assays, influence of pressure on antioxidant, cytotoxic and enzyme-inhibitory activities of the extracts was evaluated. Extracts obtained at 30, 45 and 60 bar exhibited stronger bioactivities than at 10 and 90 bar. It was concluded that pressure exert a significant influence on chemical composition of extracts, and thus on biological activity of chamomile extracts.

The purity of isolated compound was confirmed by spectroscopic analysis. Biological activity of extracts obtained under optimal conditions were determined as well. In order to identify polyphenolic compounds UHPLC-HESI-MS/MS analysis was performed.
