Production of levan by Bacillus licheniformis NS032 in sugar beet molasses-based medium

AB  - The production of levan by Bacillus licheniformis NS032 in a medium based on sugar beet molasses was studied. High polysaccharide yields were produced by using diluted molasses (100–140 g/L of total sugars) with the addition of commercial sucrose up to 200 g/L of total sugars, as well as K2HPO4. A levan yield of 53.2 g/L was obtained on a medium optimized by response surface methodology, containing 62.6% of sugar originating from molasses, and 4.66 g/L of phosphate, with initial pH value of 7.2. In comparison to the media with 200 and 400 g/L sucrose, in the molasses optimized medium, the observed bacterial growth was faster, while the maximum production of polysaccharide was achieved over a shorter time interval (48 h). The polysaccharide produced in molasses medium had a weight average molecular weight of 5.82 × 106 Da, degree of branching 12.68%, viscosity of 0.24 dL/g, and based on methylation analysis and NMR data, it did not significantly differ from levan obtained in the medium with 200 g/L sucrose. © 2018 Elsevier B.V.

AB  - Conformational analysis of 9alpha-acetoxycumambrine A 1 and 8-O-isobutiryl-9alpha-acetoxycumambrine B 2 was carried out by low-temperature NMR studies. Results suggested that lactones 1 and 2 are mixtures of two distinctive conformers, I and II. Based on low-temperature H-1 NMR spectra, in four solvents, the thermodynamic parameters of I reversible arrow II exchange process were assessed. Energy of activation of I -- gt  II reaction was obtained by dynamic NMR simulations for both compounds. Results revealed that conformational exchange of lactones 1 and 2 occurs due to "chair reversible arrow twisted chair" interconversion of a heptane ring. The same PM3 semiempirical method was applied for geometry optimization of lactones 1 and 2, as well as of 9alpha-hydroxycumambrine A 3, 9alpha-acetoxycumambrine B 4, and cumambrine B 5.

AB  - The conformations of the C-6 lactomized germacranolides 1-3 were calculated by applying the PM3 semi-empirical method, The low-temperature H-1-NMR spectra of parthenolide (1) were also measured. The relations between the calculated and the experimentally determined geometries were established using a modified Karplus equation. The applied quantum-chemical calculations proved to be an efficient and easy-to-use tool for die elucidation and prediction of die properties of germacranolide-type sesquiterpene lactones.
AB  - Konformacije germakranolida 1–3, laktonizovanih u položaju (6), izračunate su primenom PM3 semi-empirijske MO metode. Takođe su snimljeni protonski NMR spektri partenolida (1) na niskim temperaturama. Pomoću modifikovane Karplusove jednačine je uspostavljena korelacija između eksperimentalno utvrđenih i izračunatih najstabilnijih konformacija. Zaključeno je da primenjena kvantno-hemijska izračunavanja brzo i pouzdano mogu predvideti i objasniti osobine seskviterpenskih laktona tipa germakranolida.

AB  - A new germacranolide, (E)-1 alpha, 10 beta-opoxy-3 beta-acetoxy-6 alpha-hydroxygermacra-4,11(13)-dien-12,8 alpha-oxide, together with nine new highly oxygenated guaiadien-12,6 alpha-olides of anthemolide, and cumambrin type were identified in the repealed examination of the aerial parts of the flowering Anthemis carpatica. In addition, six known guaianolides belonging to the same groups. also isolated previously from A. carpatica, along with two guaianolides, 2 beta-hydroxyepiligustrin and cumambrin B, not found before in this species, were isolated this time. AB  - A new guaianolide, 3-deacetyl-9-O-acetylsalograviolide A, along with four known closely related lactones, salograviolide A, 9-O-acetylsalograviolide A, kandavanolide and salograviolide B were detected in the aerial parts of the flowering plant Centaurea nicolai. Antifungal tests performed on salograviolide A and its 9-O-acetyl and 3-O-deacetyl-9-O-acetyl derivatives revealed inhibitory activity against Aspergillus niger, A. ochraceus, Penticillium ochrochloron,Cladosporium cladosporodies, Fusharium tricinctum and Phomopsis helianthi. Neither of them was active against Trichoderma viride. (C) 1999 Elsevier Science Ltd. All rights reserved.
