AB  - Phenothiazines are a large group of heterocyclic, aromatic molecules with nitrogen and sulphur between two benzene rings. Their derivatives, N-alkylphenothiazines have substituent on heterocyclic nitrogen atom which gives them different properties. Also, a series of these molecules have substitution on carbon atom at place 2 of phenothiazine benzene ring. Alkylphenothiazines contain aminoalkyl substituent and their alkyl, acyl and sulphonil derivatives, as well as monocyclic and bicyclic heterocycles attached at thiazine nitrogen atom or directly linked to benzene ring. The N-alkylphenothiazines have been known as antipsychotic drugs, but they also possess antibacterial, antifungal, anticancer activity, and ability to react with macromolecules and to coordinate to the metals. Metal complexes with N-alkylphenothiazines are biological active compounds with different antimicrobial activities and cytotoxic effect against tumor cell lines. The large field of application of N-alkylphenothiazines is very attractive in terms of synthesis of new related derivatives, metal complexes, studying their properties and applications. This article presents a review of the literature and a contemporary view at N-alkylphenothiazines - their synthesis and application, as well as their metal complexes which have promising biological effects.

In Vivo Enzymes Activities of Some Ru(Ii) Compounds with N-Alkylphenothiazines

AB  - The purpose of the present study was to investigate and compare the effects of two ruthenium complexes with trifl uoperazine on acethylcholinesterase enzyme activity and lactate dehydrogenase levels in vivo under physiological conditions in rats blood. Complexes 1 and 2 showed positive effects on acethylcholinesterase at all doses and did not disturb its normal activity. Total LDH activity was inhibited in the presence of both complexes, but Ru(II) complexes showed different effects on the activity of LDH isoenzymes. The activities of LDH1 and LDH2 isoenzymes were decreased in all applied doses of the complex 2, while the activity of LDH2 reduced using complex 1 in the same doses. Results of the present study suggest the neuro-and cardio protective potential of oral administration of complexes 1 and 2, as non-toxic compounds under physiological conditions. These protective effects are the result of their potent antioxidant activity.
AB  - Cilj ovog rada je da se ispitaju i uporede efekti dva kompleksa rutenijuma sa trifluoperazinom na aktivnost enzima acetilholinesteraze i laktat-dehidrogenase in vivo pod fiziološkim uslovima u krvi pacova. Kompleksi 1 i 2 pokazali su pozitivan efekat na aktivnost acetiholinesteraze u svim primenjenim dozama. Ukupna aktivnost LDH je inhibirana u prisustvu oba kompleksa, ali kompleksi Ru(II) pokazuju različite rezultate na izoenzimske oblike ovog enzima. Aktivnosti izoenzima LDH1 i LDH2 su smanjene u svim primenjenim dozama kompleksa 2, dok kompleks 1 smanjuje aktivnost samo izoenzima LDH2 u tim istim koncentracijama. Rezultati prikazanog istraživanja ukazuju na neuro - i kardio zaštitni potencijal oralne primene kompleksa 1 i 2, kao netoksičnih jedinjenja pod fiziološkim uslovima, indukovano preko njihovog snažnog antioksidativnog efekta.

AB  - A novel efficient differential pulse voltammetric (DPV) method for determination gallic acid (GA) was developed by using an electrochemical sensor based on [Cu(2)tpmc](ClO4)(4) immobilized in PVC matrix and coated on graphite (CGE) or classy carbon rod (CGCE). The proposed method is based on the gallic acid oxidation process at formed [Cu(2)tpmcGA](3+) complex at the electrode surface. The complexation was explored by molecular modeling and DFT calculations. Voltammograms for both sensors, recorded in a HNO3 as a supporting electrolyte at pH 2 and measured in 2.5 x 10(-7) to 1.0 x 10(-4) M of GA, resulted with two linear calibration curves (for higher and lower GA concentration range). The detection limit at CGE was 1.48 x 10(-7) M, while at CGCE was 4.6 x 10(-6) M. CGE was successfully applied for the determination of the antioxidant capacity based on GA equivalents for white, rose and red wine samples.

The complexes were characterized by elemental analysis, molar conductivity measurement, and spectral (IR, H-1, C-13, 2D H-1-C-13 heteronuclear correlation spectra, Pt-195 NMR, and MS) methods. Outer-coordination sphere was proposed for 1; while in 2, the ligand was coordinated to the metal. The complexes exhibit antibacterial effect on strains of Bacillus subtilis, Bacillus cereus, Bacillus pumilus, and methicillin-resistant Staphylococci as Gram-positive bacteria and an Escherichia coli as Gram-negative bacteria, as well as the reference strains.

AB  - The complexation reactions between octaazamacrocyclic ligand N,N',N '',N'"-tetrakis(2-pyridylmethyl)-1,4,8,11-tetraazacyclotetradecane (tpmc) and Co2+, Ni2+, Zn2+ and Cu2+ metal cations have been studied conductometrically in acetonitrile and aqueous solutions. The conductance data exhibit different stoichiometry of the complexes in two studied solutions. In most cases 1:1 (ML) complexes were formed while in some cases 2:1 (M2L) complexes are evident. The stability constants of the resulting 1:1 complexes were determined by computer fitting of the conductance/mole ratio data. Theoretical DFT calculations were used to explain metal binding capability of tpmc ligand and the binding sequences were found to follow the order: Cu2+  gt  gt  Ni2+  gt  Co2+  gt  Zn2+. The combination of results obtained by this theoretical approach and conductometric study successfully explains sensitivity and selectivity of recently developed tpmc based ISE and could play a guiding role in planning experiments with large macrocyclic ligands.

AB  - Five new dioxomolybdenum(VI) complexes of the general formula [MoO2(Rdtc)(2)], 1-5, where Rdtc(-) refer to piperidine (Pipdtc), 4-morpholine (Morphdtc), 4-thiomorpholine (Timdtc), piperazine (Pzdtc) or N-methylpiperazine (N-Mepzdtc) dithiocarbamates, respectively, have been prepared. The complexes were characterized by elemental analysis, conductometric measurements, electronic, IR and NMR spectroscopy. The complexes 1-5 contain a cis-MoO2 group and have an octahedral geometry. Two dithiocarbamato ions join as bidentates with both the sulfur atoms to the molybdenum atom. The presence of different heteroatoms in the piperidino moiety influences the v(C=N) and v(C=S) vibrations, which wavelengths decrease in the order: Pipdtc  gt  N-Mepzdtc  gt  Morphdtc  gt  Pzdtc  gt  Timdtc ligands. Based on their spectral data, the molecular structures of complexes 1-5 were optimized at the semi-empirical molecular-orbital level, and the geometries, as obtained from calculations, are described. The antimicrobial activities of the complexes were tested against nine different laboratory control strains of bacteria and two strains of the yeast Candida albicans. All the tested strains were sensitive. Complexes bearing heteroatom in position 4 of piperidine moiety were significantly more potent against the tested bacteria compared to the corresponding ligands.
AB  - Sintetisano je pet novih dioksomolibden(VI) kompleksa, opšte formule [MoO2(Rdtc-)2], sa Rdtc- ligandima: piperidin- (Pipdtc), 4-morfolin- (Morphdtc), 4-tiomorfolin- (Timdtc), piperazin- (Pzdtc) i N-metilpiperazin- (N-Mepzdtc) ditiokarbamatima. Kompleksi su okarakterisani elementalnom analizom, IR i NMR spektroskopijom kao i merenjem molarne provodljivosti. Pretpostavljena geometrija svih kompleksa je oktaedarska. Rdtc- ligandi su bidentatno koordinovani preko oba atoma sumpora za atom molibdena. Prisustvo različitih heteroatoma utiče na promenu položaja v(C=N) i v(C=S) vibracija, čiji opada sledećim redom liganada: Pipdtc  gt  N-Mepzdtc  gt  Morphdt  gt  Pzdtc  gt  Timdtc. Na osnovu spektralnih podataka, strukture svih kompleksa su optimizovane na semiempirijskom molekulsko-orbitalnom nivou upotrebom PM6 metoda. Antimikrobna aktivnost ispitivana je na jedanaest različitih patogena. Uočeno je da kompleksi koji imaju heteroatom u položaju 4 piperidinskog prstena ispoljavaju značajno veću jačinu antimikrobnog dejstva prema bakterijama, u poređenju sa odgovarajućim ligandima.

Complexes bearing heteroatom in position 4 of piperidine moiety were significantly more potent against the tested bacteria compared to the corresponding ligands., Sintetisano je pet novih dioksomolibden(VI) kompleksa, opšte formule [MoO2(Rdtc-)2], sa Rdtc- ligandima: piperidin- (Pipdtc), 4-morfolin- (Morphdtc), 4-tiomorfolin- (Timdtc), piperazin- (Pzdtc) i N-metilpiperazin- (N-Mepzdtc) ditiokarbamatima. Kompleksi su okarakterisani elementalnom analizom, IR i NMR spektroskopijom kao i merenjem molarne provodljivosti. Pretpostavljena geometrija svih kompleksa je oktaedarska. Rdtc- ligandi su bidentatno koordinovani preko oba atoma sumpora za atom molibdena. Prisustvo različitih heteroatoma utiče na promenu položaja v(C=N) i v(C=S) vibracija, čiji opada sledećim redom liganada: Pipdtc  gt  N-Mepzdtc  gt  Morphdt  gt  Pzdtc  gt  Timdtc. Na osnovu spektralnih podataka, strukture svih kompleksa su optimizovane na semiempirijskom molekulsko-orbitalnom nivou upotrebom PM6 metoda. Antimikrobna aktivnost ispitivana je na jedanaest različitih patogena. Uočeno je da kompleksi koji imaju heteroatom u položaju 4 piperidinskog prstena ispoljavaju značajno veću jačinu antimikrobnog dejstva prema bakterijama, u poređenju sa odgovarajućim ligandima.",

AB  - Three new ruthenium(II) complexes 1-3 containing N-alkylphenothiazine molecules were synthesized by reaction of [RuCl2(eta(6)-P-cymene)](2) with chlorpromazine hydrochloride (1), trifluoperazine dihydrochloride (2) or thioridazine hydrochloride (3). The compounds of the general formula L[RuCl3(eta(6)-p-cymene)] were characterized by elemental analysis and spectroscopic methods (FT-IR, UV-Vis, H-1 and C-13 NMR). Complex 2 was structurally characterized by single crystal X-ray diffraction. In vitro cytotoxic activity of complexes 1-3 were assayed in four human carcinoma cell lines MCF-7, MDA-MB-453 (breast carcinoma), SW-480 (colon carcinoma) and IM9 (myeloma multiple cells). The highest cytotoxicity (12.1  lt = IC50  lt = 17.3 mu M) and induced a total (SW-480) or almost total cell death (MCF-7. MDA-MB-453) at 25 mu M in 48 h of treatment were observed for complex 2. The influence of three different doses (0.4, 4.5 and 90.4 mu M/kg bw) of complex 2 on activities of antioxidants enzymes (superoxide dismutase (SOD) and catalase (CAT)) and lactate dehydrogenase (LDH) were investigated under physiological conditions. AB  - Three new complexes of the general formula L[RuCl(3)(DMSO)(3)] (1-3), where L = chlorpromazine hydrochloride, trifluoroperazine dihydrochloride or thioridazine hydrochloride, were prepared and characterized by elemental analysis and spectroscopic methods (FT-IR, UV-Vis, (1)H NMR and (13)C NMR). In addition, the crystal structure of the complex 2 containing trifluoroperazine dihydrochloride was solved by single crystal X-ray diffraction. The complex crystallizes in the monoclinic system, space group P2(1)/n, with a = 10.4935(7) angstrom, b = 18.6836(12) angstrom, c = 19.9250(13) angstrom, beta = 98.448(2)degrees, V = 3864.0(4) angstrom(3). The structure was refined to the agreement factors of R = 4.79%, R(w) = 11.23%. The effect of three different doses (0.4, 4.5 and 90.4 mu M/kg bw) of complex 2 on superoxide dismutase (SOD) and catalase (CAT) activity was investigated under physiological conditions. Influence on nitrite production (NO(2)(-)) and the level of erythrocytes malondialdehyde (MDA) in rats blood was also evaluated. Complex 2 did not affect the CAT enzyme activity in vivo and did not cause the hydroxyl radicals production. In the 0.4 and 4.5 mu M/kg bw doses it showed almost the same or lower SOD activity and nitrite levels, while the dose of 90.4 mu M/kg bw significantly increased these parameters. Finally, the cytotoxicity of complexes were assayed in four human carcinoma cell lines MCF-7, MDA-MB-453 (breast carcinoma), SW-480 (colon adenocarcinoma) and IM9 (myeloma multiple cells). AB  - The syntheses of a new ligand 2-(diphenylphosphino)benzaldehyde selenosemicarbazone (HL) and the corresponding complex with Ni(II) of the formula [Ni(L)(NCS)] are reported. The complex was characterized by single crystal X-ray analysis. The compound complex is diamagnetic and has a markedly deformed square-planar structure with PNSe set of donor atoms of the ligand, the Schiff base monoanion, and the N atom of the coordinated NCS group. The phosphorus atom deviates even by 0.494(3) Angstrom from the mean plane defined by the other coordination atoms (NSeN) and the metal atom. (C) 2003 Published by Elsevier B.V.

AB  - Cytotoxicity and cell growth inhibition studies were\performed for five distinct cobalt(II) [CO2(acac)tpmc](CIO4)(3), [Co-2(dibzac)tpmc](CIO4)(3). [Co-2(hfac)tpmc](CIO4)(3). [Co-2(tmhd)tpmc](CIO4)(3), and [Co-2(ox)tpmc](CIO4)(2).3H(2)O and five molybdenum(VI) complexes, [MoO2(pipdtc)(2)]. [MoO2(morphdtc)(2)]. [MoO2(timdtc)(2)], [MoO2(pzdtc)(2)] and [MoO2(N-Mepzdtc)(2)]. They showed to have relatively high toxicity in K562 cells and a relatively low cytotoxicity in U937 cells. as assessed by both MTT and Trypan Blue assays. The five molybdenum complexes were tested in human promyelotic U937 cell line and they showed to have high toxicity.

AB  - Four new Cu(II) and Co(II) complexes having an octaazamacrocyclic ligand derived from cyclam, N,N'N,",N'''-tetrakis(2-pyridylmethyl)-1,4,8,11-tetraazacyclotetradecane, with either an mu-carbonato, oxalato or malonato as the additional ligand were isolated and investigated by elemental analysis, electronic and IR spectroscopy, as well as molar conductivity, magnetic and cyclic voltammetry (CV) measurements. Their microbiological activity was also tested. Binuclear or tetranuclear structures, depending on the bridging ligand and metal ion, was proposed.

AB  - 4-Morpholine-, piperidine-, 4-piperazine- and N-methyl- piperazine-dithiocarbamate complexes of Cobalt (III) with 1, 4, 8, 11-tetraazacyclotetradecane, of general formula [Co(Rdtc)cyclam] (ClO4)(2), have been prepared and have been characterized. The complexes adopt cis-octahedral geometry with folded macrocyclic ligand and with the dithiocarbamate bound as a bidentate. trans-influence of the dithiocarbamate ligands was studied by NMR spectroscopy and established the order piperidine-dtc  gt 4-morpholine-dtc gt N-methyl-piperazine-dtc.
