AB - Bisbibenzyls are specialized metabolites found exclusively in liverworts, until recently; they represent chemical markers of liverworts. Their occurrence in vascular plants was noticed in 2007, when they were found in Primula veris subsp. macrocalyx from Russia. This report prompted us to chemically analyze the two most common Serbian Primula species, P. veris subsp. columnae and P. acaulis, in order to determine the presence of bisbibenzyls in them. Our study revealed nine structurally distinct bisbibenzyls (1–9), identified based on 1D and 2D NMR, IR, UV and HRESIMS data. Among them were five previously undescribed compounds (2–6). The remaining com pounds found and previously described in the literature were: the bisbibenzyls riccardin C (1), isoperrottetin A (7), isoplagiochin E (8) and 11-O-demethylmarchantin I (9), as well as 4-hydroxyphenylmethylketone (10) and 4-hydroxy-3-methoxyphenylmethylketone (11). Riccardin C was the most dominant bisbibenzyl in both species studied. Previously, it was the first bisbibenzyl found in vascular plants (P. veris subsp. macrocalyx). An assessment of the cytotoxic activity of the isolated compounds against A549 lung cancer and healthy MRC5 cell lines was also the subject of our study. Compounds 6 and 9 exhibited significant cytotoxic activity expressed by IC50 values of 12 μM, but the selectivity was not satisfactory.

AB - Phytochemistry deals with the study of secondary metabolites produced by plants that synthesize these compounds for many reasons, including their own protection against attack of herbivores and plant diseases. Secondary metabolites are believed to represent plant adaptation to various environmental factors and that they enabled the survival of the species. Secondary metabolites of plants can have curative or toxic effects in humans and animals. Herbal medicine has a long tradition in folk medicine and until the early 20th century, when synthetic organic chemistry began to develop, plants were the main source of medicines. The two basic goals of our phytochemical research are: isolation and identification of new (biologically active) compounds ? potential drugs, and chemotaxonomy (chemosystematics). In the following text through one selected example, the genus Amphoricarpos Vis., our phytochemical research is shown on both aspects.

AB - In continuation of our study on Cotinus coggygria from Serbia, 10 known flavonoids (1-10) were isolated from the methylene chloride/ methanol extract of the heartwood. They were tested for in vitro protective effect against chromosome aberrations in peripheral human lymphocytes, using the cytokinesis-block micronucleus assay. All tested compounds (in minimal doses of 1 μg/mL) exerted a beneficial effect by decreasing DNA damage of human lymphocytes in the range of 24.2% to 54.5%, better than the radio protectant control, amifostine. Functional groups, such as 3′,4′-dihydroxyphenyl (catechol), 5-OH, 3-OH, and 4-keto in flavonoids (3-keto in aurones), which play a key role in antioxidant activity, are proposed to be responsible for the DNA protective activity of the tested compounds.

AB - Metabolomics generate a profile of small molecules from plant extracts, which could be directly responsible for bioactivity effects. Using dry-column flash chromatography enabled a rapid and inexpensive method for the very efficient separation of plant extract with a high resolution. This separation method coupled to NMR and FTIR-based metabolomics is applied to identify bioactive natural products. OPLS multivariate analysis method, was used for correlation the chemical composition of the plant extracts, Amphoricarpos autariatus, with the results of cytotoxic activity against Human cervical adenocarcinoma cell line (HeLa) and epithelial lung cancer cell line (A549). In this way, the highest contribution to the cytotoxic activity was recorded for the guaianolide sesquiterpene lactones named amphoricarpolides. The compounds indicated as bioactive after metabolomics analysis were tested, and their cytotoxic activity were confirmed.

AB - The variability of n-alkanes in the needle cuticular wax of Pinus heldreichii and P. peuce in two natural populations from the Scardo-Pindic mountains was investigated for the first time. Gas chromatography (GC) and gas chromatography/mass spectrometry (GC-MS) analyses of two-year-old needles were performed us-ing an Agilent 7890A GC equipped with an inert 5975C XL EI/CI mass spectrometer detector (MSD) and a flame ionization detector (FID) connected by a capillary flow technology 2-way splitter with make-up. An HP-5MS capillary column was used. n-Alkanes ranged from C19 to C33. In P. heldreichii the most abundant were C23, C25 and C27 (16.3, 15.6 and 12.8 % on average, respectively), while in P. peuce they were C29, C25, and C27 (16.5, 15.3 and 13.5 % on average, resp.). Mid-chain and long-chain n-alkanes prevailed in both species. Principle component analysis (PCA) and Cluster analyses of five and six n-alkanes, respectively, showed divergence of the Scardo-Pindic populations from the Dinaric ones.

AB - Analysis of composition of CH2Cl2 surface extract of leaves of Amphoricarpos neumayeri Vis. revealed 16 sesquiterpene lactones with guaianolide skeleton, so called amphoricarpolides, typical for this genus. Four of them, 13–16, were new derivatives and their structures were elucidated by detailed analyses of IR, NMR and MS data. Amphoricarpolide (9), its 15-O-acetyl derivative (5), and two 9β-hydroxyamphoricarpolides, 3,15-di-O-acetyl- and 3-O-acetyl-15-O-isovaleroyl derivatives (3 and 6, respectively) were tested for in vitro protective effect on chromosome aberrations in peripheral human lymphocytes using cytochalasin-B blocked micronucleus (CBMN) assay. The tested compound exerted a beneficial effect by decreasing DNA damage of human lymphocytes.

AB - The genus Amphoricarpos Vis. is endemic of westpart of Balkan peninsula. In order to get more insight into the ambiguous taxonomy of the genus, the metabolomic study of the Amphoricarpos samples, has been undertaken. The(1)H NMR spectroscopy combined with multivariate data analysis has been applied. OPLS-DA has been shown to be the best method for clear discrimination of these samples based on the metabolites present in the extracts. The main difference between A. autariatus and A. neumayeri has been found to be the presence of characteristic sesquiterpene lactones of guaianolide type named amphoricarpolides. While the main sesquiterpene lactone in A. neumayeri was oxidized in position 2, those present in both subspecies of A. autariatus samples were oxidized in position 9. The use of sesquiterpene lactones as taxonomic characters for differentiating species of the Asteraceae family was reviewed elsewhere. For the differentiation of two subspecies, A. autariatus ssp. autariatus, and A. autariatus ssp. bertisceus, chlorogenic and malic acid have been found to be decisive, but these compounds do not have chemotaxonomic significance. Our results were in accordance with above mentioned classification of Blecic and Mayer but they differentiate from those of the recent genomic study of the genus indicating a single, genetically, morphologically and ecologically variable species, without intraspecific taxa.

AB - The detection of illegal drugs in various samples has been among the most fascinating objects for the practicing analytical chemist involved in forensic analysis. For centuries (until the end of the nineteenth century), drugs were almost entirely of natural origin, mostly herbal, and in some cases slightly chemically modified (e.g., through acetylation of morphine to heroin). By the development of synthetic organic chemistry (commencing at the beginning of the twentieth century), quite a few synthetic drugs designed to mimic the pharmacological effects of drugs originating from natural sources have been designed (e.g., cannabimimetic indazole derivatives or amphetamines, etc.). Detection of such synthetic drugs, named “designer drugs,” due to a large number of new substances continuously emerging on the market, is crucial. The efficacy of this analysis has become a tremendous challenge in the analytical forensic field. A development of fast identification and structural characterization is greatly facilitated by the advent of modern spectroscopic techniques, namely nuclear magnetic resonance spectroscopy (NMR) and modern mass spectrometry (MS) as well as separation techniques, such as gas and liquid chromatography (GC and LC). Nowadays, rapid strategies for chemical characterizations of illicit drugs are available.

AB - The n-alkane composition in the leaf cuticular waxes of natural populations of Bosnian pine (Pinus heldreichii), Austrian pine (P.nigra), and Macedonian pine (P.peuce) was compared for the first time. The range of n-alkanes was wider in P.nigra (C-16-C-33) than in P.heldreichii and P.peuce (C-18-C-33). Species also diverged in abundance and range of dominant n-alkanes (P. heldreichii: C-23, C-27, and C-25; P.nigra: C-25, C-27, C-29, and C-23; P.peuce: C-29, C-25, C-27, and C-23). Multivariate statistical analyses (PCA, DA, and CA) generally pointed out separation of populations of P.nigra from populations of P.heldreichii and P.peuce (which were, to a greater or lesser extent, separated too). However, position of these species on the basis of n-alkane composition was in accordance neither with infrageneric classification nor with recent molecular and terpene investigations.

AB - Simultaneous hydrodistillation and extraction of Pinus cembra needles from Slovakia was done (via Likens Nickerson apparatus) for the first time. In essential oil extracts 55 compounds were identified, comprising 99.6% of the extract. The most abundant were monoterpene hydrocarbons (71.0%). In the terpene profile alpha-pinene, limonene/beta-phellandrene, germacrene D, beta-pinene, and delta-cadinene dominated (53.2%, 11.4%, 9.4%, 4.6%, and 4.3%, respectively). Seven new compounds for P. cembra, such as methyl daniellate (0.5%), 1,8 cineole (0.2%) and trans-cadina-1(6),4-diene (0.2%), etc. were found. In needle cuticular wax of P. cembra the amount of nonacosan-10-ol was 75.8%. n-Alkanes ranged from C-20 to C-35 with the most dominant C-31, C-29 and C-33 (33.4%, 16.9%, and 9.6%, resp.). Differences in terpene profiles between Slovakian and Greece from one side and Romanian and Polish cembran pines on the other side could be the consequence of its disjuncted areal in Carpathian Mountains caused by glaciation and survival of species in different ecological niches. Obtained differences in n-alkane profiles among our and literature results could be the consequence of different age of trees.

AB - This is the first report of population variability of the contents of n-alkanes and nonacosan-10-ol in the needle epicuticular waxes of Serbian spruce (Picea omorika). The hexane extracts of needle samples originated from three natural populations in Serbia (Vranjak, Zmajevaki potok, and Mileevka Canyon) were investigated by GC and GC/MS analyses. The amount of nonacosan-10-ol varied individually from 50.05 to 74.42% (65.74% in average), but the differences between the three investigated populations were not statistically confirmed. The results exhibited variability of the composition of n-alkanes in the epicuticular waxes with their size ranging from C18 to C35. The most abundant n-alkanes were C29, C31, and C27 (35.22, 13.77, and 12.28% in average, resp.). The carbon preference index of all the n-alkanes (CPItotal) of the P. omorika populations (average of populations IIII) ranged from 3.3 to 11.5 (mean of 5.9), while the average chain length (ACL) ranged from 26.6 to 29.2. The principal component and cluster analyses of the contents of nine n-alkanes showed the greatest difference for the population growing in the Mileevka Canyon. The obtained results were compared with previous literature data given for other Picea species, and this comparison was briefly discussed.

AB - The essential-oil variability in seven native populations belonging to different infraspecific taxa of Pinus nigra (ssp. nigra, var. gocensis, ssp. pallasiana, and var. banatica) growing wild in Serbia was analyzed. In the needles of 195 trees from seven populations, 58 essential-oil components were identified. The major components were -pinene (43.6%) and germacrene D (29.8%), comprising together 73.4% of the total oil composition. Based on the average chemical profile of the main terpene components (with contents gt 5%), the studied populations were found to be the most similar to populations from central Italy and Greece (ssp. nigra). Cluster analysis showed the division of the populations into three principal groups: the first group consisted of PopulationsI, II, III, IV, and V (considered as ssp. nigra group), the second of PopulationVI (ssp. pallasiana group), and the third of PopulationVII, which had the most distinct oil composition (ssp. banatica group). The taxonomic implications of the essential-oil profiles of the investigated taxa of this very complex species are discussed.

AB - The composition of leaf cuticular neutral lipids of Amphoricarpos autariatus ssp. autariatus collected at canyon of river Tara (North Montenegro) was investigated by GC/MS (nonpolar fraction), LC-ESI TOF MS and H-1 NMR spectroscopy (more polar fraction). The nonpolar fraction (ca. 15% of the whole surface extract) contained C-27 (-) (33) n-alkanes, those with odd-number of carbons predominating. The LC-ESI MS and H-1 NMR of the more polar fraction revealed 13 sesquiterpene lactones, constituting ca. 97.5% of the lactone mixture, identified as the known guaianolides, so-called amphoricarpolides, found previously in the aerial parts of the genus. The lactone fraction exhibited considerable in vitro effect against eight fungi, i.e. Aspergillus ochraceus, A. niger, A. versicolor, Penicillium funiculosum, P. ochrochloron, Trichoderma viride, Fusarium verticillioides and Fulvia fulvum.

AB - The oil content and fatty acids, unsaponifiable composition and antioxidant activities of the seed oil from three Malvaceae species (Malva sylvestris L., Malva sylvestris L. var. mauritiana and Althaea officinalis L.) from Serbia were determined. The oil yields from the seeds varied from 7.18 to 9.60%. The main fatty acids of the seed oils were linoleic acid (44.14-54.49%), oleic acid (13.00-16.99%) and palmitic acid (11.45-24.29%). A small amount of cyclopropenoid acids, up to 1.85% was also established. The predominant sterol in all seed oils was beta-sitosterol (11.51-17.34 mg/g of oil). The antioxidant potential of all the investigated seed oils was evaluated by radical scavenging activity using 2,2-diphenyl-1-picrylhydrazyl (DPPH) assay.

AB - This is the first report on population variability of nonacosan-10-ol and n-alkanes in needle epicuticular waxes of Macedonian pine (Pinus peuce Griseb.) Hexane extracts of needle samples, originating from two natural populations in Montenegro (Zeletin and Sjekirica) and from one population in Serbia (Mokra Gora) were analyzed by gas chromatography (GC) and gas chromatography/mass spectrometry (GC/MS). The amount of nonacosan-10-ol varied individually from 41.3 to 72.31% (average 55.9%), with the Sjekirica population being statistically divergent (64.4% on average). The results showed n-alkanes in epicuticular waxes ranging from C18 to C33. The most abundant alkanes were C29, C25, C27, and C23 (15.5, 11.1, 10.6, and 10.5% on average, resp.). The carbon preference index of Pinus peuce ranged from 1.0 to 4.3 (1.9 on average). Average chain length ranged from 18.4 to 27.7 (average 25.7). A high level of inidividual quantitative variation in all of these hydrocarbon parameters was also detected. These results were compared with published data on other species from the Pinus genus.

abstract = "This is the first report on population variability of nonacosan-10-ol and n-alkanes in needle epicuticular waxes of Macedonian pine (Pinus peuce Griseb.) Hexane extracts of needle samples, originating from two natural populations in Montenegro (Zeletin and Sjekirica) and from one population in Serbia (Mokra Gora) were analyzed by gas chromatography (GC) and gas chromatography/mass spectrometry (GC/MS). These results were compared with published data on other species from the Pinus genus.",

AB - The troglomorphic millipede Brachydesmus troglobius Daday, 1889 (Polydesmida: Polydesmidae) secretes allomones from glands on both lateral surfaces of its body segments. The secretion was identified by gas chromatography-mass spectrometry (GC-MS) analysis with electron and chemical ionization, and was shown to be composed of a mixture of benzaldehyde, benzyl alcohol, benzoylnitrile, benzoic acid and mandelonitrile benzoate. Hydrogen cyanide was qualitatively identified by the picric acid test. This is the first identification of these compounds in a cave-dwelling polydesmid.

AB - This is the first report of individual variability and population diversity of the contents of nonacosan-10-ol and n-alkanes in the needle cuticular waxes of Bosnian pines originated from Montenegro, regarded as Pinus heldreichii var. leucodermis, and from Serbia, regarded as P. heldreichii var. pancici. The amount of nonacosan-10-ol varied individually from 27.4 to 73.2% (55.5% in average), but differences between the four investigated populations were not statistically confirmed. The size of the n-alkanes ranged from C18 to C33. The most abundant n-alkanes were C23, C27, and C25 (12.2, 11.2, and 10.8% in average, resp.). The carbon preference index (CPI) of the n-alkanes ranged from 0.8 to 3.1 (1.6 in average), while the average chain length (ACL) ranged from 20.9 to 26.5 (24.4 in average). Long-chain and mid-chain n-alkanes prevailed (49.6 and 37.9% in average, resp.). It was also found that the populations of P. heldreichii var. leucodermis had predominantly a narrower range of n-alkanes (C18?C31) than the trees of the variety pancici (C18?C33). Differences between the varieties were also significant for most of the other characteristics of the n-alkane pattern (e.g., most abundant n-alkanes, CPI, ACL, and relative proportion of short-, mid-, and long-chain n-alkanes). The principle component and cluster analyses of eleven n-alkanes confirmed the significant diversity of these two varieties.
