AB  - Stinging nettle (Urtica dioica L.) is one fantastic plant widely used in folk medicine, pharmacy, cosmetics, and food. This plant’s popularity may be explained by its chemical composition, containing a wide range of compounds significant for human health and diet. This study aimed to investigate extracts of exhausted stinging nettle leaves after supercritical fluid extraction obtained using ultrasound and microwave techniques. Extracts were analyzed to obtain insight into the chemical composition and biological activity. These extracts were shown to be more potent than those of previously untreated leaves. The principal component analysis was applied as a pattern recognition tool to visualize the antioxidant capacity and cytotoxic activity of extract obtained from exhausted stinging nettle leaves. An artificial neural network model is presented for the prediction of the antioxidant activity of samples according to polyphenolic profile data, showing a suitable anticipation property (the r2 value during the training cycle for output variables was 0.999).

Activity Coefficients of the System {yKCl + (1 – y)KH2PO4}(aq) at T = 298.15 K Determined by Cell Potential Measurements

AB  - Zero-current cell potential measurements were used to determine the solution activity coefficient in a ternary system {yKCl + (1 – y) KH2PO4}(aq) at temperature T = 298.15 K. The cell of the type K–ISE|KCl(mKCl), KH2PO4(mKH2PO4)|Ag|AgCl was used in the total ionic strength range, Im = 0.0886–1.0046 mol kg–1. In order to generate a set of parameters that can be applied in a wide range of mixed solution ionic strengths, the Pitzer, Scatchard, and Clegg–Pitzer–Brimblecombe models were used to fit all available experimental data, including cell potential and isopiestic measurements from the literature. The experimental and calculated values of thermodynamic properties for the studied system are in excellent agreement. Potential interactions and solution structure were discussed by means of the excess free energy of mixing via potential pairs, triplets, or quads for the investigated solution using the Scatchard model mixing parameters.

Food Waste (Beetroot and Apple Pomace) as Sorbent for Lead from Aqueous Solutions—Alternative to Landfill Disposal

AB  - This article presents studies, whose main goal was to minimize food waste. To achieve this goal, it is necessary to expand the scope of their application, for example, for the purification of polluted water from heavy metals. Millions of tons of waste from the fruit and vegetable industry, including pomace of apples and beetroots, are thrown into landfills, posing a danger to the environment. In order to solve the problems with the disposal of these wastes, the authors investigated their sorption potential for the removal of lead from wastewater. The sorbents, dried apple (AP), and beetroots (BR) pomaces were characterized by various methods (study of composition, zeta potential, FTIR-ATR, and SEM-EDX). Various models of sorption kinetics and sorption isotherms were analyzed. Kinetical studies under optimal conditions showed that the sorption process occurs through complexation and ion exchange and the determining stage limiting the rate of sorption is the diffusion of lead ions in the sorbent. The maximum sorption capacity was 31.7 and 79.8 mg/g for AP and BR, respectively. The thermodynamic data revealed the spontaneous sorption of lead ions by sorbents. The temperature rise contributes to the sorption increase by the AP sorbent, while for the BR sorbent, the opposite effect is observed. The obtained results showed that apple and beetroots pomaces can serve as effective renewable materials for the preparation of sorbents, contributing to the solution of complex environmental problems.

AB  - Morphine (MORPH) is natural alkaloid and mainly used as a pain reliever. Its monitoring in human body fluids is crucial for modern medicine. In this paper, we have developed an electrochemical sensor for submicromolar detection of MORPH. The sensor is based on modified carbon paste electrode (CPE) by investigating the FexW1-xO4 ratio in iron tungstate (FeWO4), as well as the ratio of this material in CPE. For the first time, the effect of the iron–tungsten ratio in terms of achieving the best possible electrochemical characteristics for the detection of an important molecule for humans was examined. Morphological and electrochemical characteristics of materials were studied. The best results were obtained using Fe1W3 and 7.5% of modifier in CPE. For MORPH detection, square wave voltammetry (SWV) was optimized. Under the optimized conditions, Fe1W3@CPE resulted in limit of detection (LOD) of the method of 0.58 µM and limit of quantification (LOQ) of 1.94 µM. The linear operating range between 5 and 85 µM of MORPH in the Britton–Robinson buffer solution (BRBS) at pH 8 as supporting electrolyte was obtained. The Fe1W3@CPE sensor resulted in good selectivity and excellent repeatability with relative standard deviation (RSD) and was applied in real-world samples of human urine. Application for direct MORPH detection, without tedious sample pretreatment procedures, suggests that developed electrochemical sensor has appeared to be a suitable competitor for efficient, precise, and accurate monitoring of the MORPH in biological fluids.

AB  - Antioxidants present in infant food contribute to its nutritive value and provide protection from oxidative damage. Here, a direct current polarographic method, based on a decrease of anodic current from the hydroxoperhydroxo-mercury(II) complex (HPMC) formed at a mercury dropping electrode at its potential of dissolution, was used to determine the total antioxidant capacity (TAC) of infant formulas, human milk collected from mothers of full-term and preterm infants, human milk supplemented with fortifier, and cow milk. TAC of milk samples and infant formula correlated with protein content measured by the Bradford method (Pearson correlation coefficient 0.605, p<0.001), while TAC of human milk correlated with content of free thiol groups measured by the Ellman method (0.779, p<0.001). The results obtained showed that, in contrast to the 2,2-diphenyl-1-picrylhydrazyl (DPPH) assay, HPMC enables specific insight into the quality of infant milk food which reflects the amounts of free thiol groups and protein content. The significant contribution of milk proteins and the most prevalent amino acids, particularly cysteine, arginine, and phenylalanine, to TAC was shown. Thus, the HPMC method might be useful in monitoring the quality of infant formulas and human milk for preterm infants before and after supplementation with fortifier.

AB  - In this study, a novel green walnut liqueur (GWL) based on green walnuts, as the main ingredient, with the addition of fruits, aromatic plants, chocolate and honey, was produced at the pilot-scale. Antioxidant activity (AO) of the obtained GWL was determined using multiple AO assays in parallel, standard spectrophotometric (FC, DPPH and FRAP) and recently developed electrochemical ones, HydroxoPerhydroxoMercury(II) Complex and Mercury Reduction Antioxidant Power, and compared to similar commercial alcoholic beverages. Characterization of the GWL in terms of volatile and polyphenolic components was performed using SPE-GC-MS and HPLC-DAD-MS/MS, respectively. Sensory quality assessment was performed by experts in the field of sensory analysis of alcoholic beverages, using a scoring method. According to all AO assays applied, AO activity and total phenolics content of GWL were superior in comparison to commercial spirits considered. The volatile fraction of GWL was mainly composed of eugenol,alpha-terpineol and vanillin, while the most prevalent phenolic compounds were gallic (5.054 mg/L) and chlorogenic acid (1.307 mg/L) and flavonoids such as catechine (0.882 mg/L), quercetin (0.499 mg/L) and its sugar-conjugated derivatives, quercetin 3-O-glucoside (0.774 mg/L) and quercetin-3-O-rhamnoside (0.614 mg/L). Gallic acid is the major contributor to total AO activity, especially determined by DPPH and FRAP, followed by catechine, quercetin and chlorogenic acid. Among the terpenoids, contribution of eugenol to total AO activity is estimated as the highest. Excellent sensory quality (18.52 of maximum 20 scores) was ascribed to GWL. Consequently, the presence of bioactive compounds and high AO activity of GWL, in addition to high sensory quality score, indicates a high market potential for this high-value product.

AB  - This study was focused on in vitro screening of the total antioxidant activity of the selected extracts of the mushroom Coprinus comatus and quinic acid, one of their antidiabetic ingredients, by an uncommon electrochemical assay. Indeed, direct current (DC) polarographic HydroxoPerhydroxo Mercury(II) Complex (HPMC) assay based on decrease of anodic limiting current originating from HPMC formation in alkaline solutions of hydrogen peroxide at potential of mercury dissolution, observed upon gradual addition of antioxidants, was applied herein for the estimation of the natural products' antioxidativity. Quinic acid was found to exhibit most promising antioxidant potential (4.0 ± 0.2%µL-1) being ≈ 2-fold more active than the screened C. comatus extract samples. Actually, such a finding puts some light on the antioxidativity of cyclic polyols, well understimated class of organic compounds, compared to aromatic (poly)phenolics. As a low cost, easy-to handle and accurate this polarographic assay may be thoroughly recommended for much broader use. [Formula: see text].

AB  - Isopiestic measurements have been made at 55 compositions of the {yK2HPO4 + (1 − y)KH2PO4}(aq) system at T = (298.15 ± 0.01) K, 11 for each of the limiting binary solutions and 33 for mixture compositions at K2HPO4 stoichiometric ionic strength fractions y = (0.23330, 0.47671, and 0.73177), using KCl(aq) as the reference standard. Model parameters for the binary subsystems were evaluated at this temperature for an extended form of Pitzer's ion-interaction model and also for the Clegg, Pitzer and Brimblecombe model based on the mole-fraction-composition scale, using the present isopiestic results along with critically-assessed osmotic coefficients for both of these aqueous electrolytes as extracted from the published literature. The thermodynamic models for KH2PO4(aq) extend to slightly above the saturated solution molality at T = (298.15 ± 0.01) K, whereas those for K2HPO4(aq) extend to m = 9.7429 mol·kg−1, which is the molality of the saturated solution, also at T = (298.15 ± 0.01) K. These results yield the CODATA-compatible standard Gibbs energy of formation ΔfGmo(K2HPO4·3H2O,cr,298.15K)=-2367.70±1.60kJ·mol-1. The 33 osmotic coefficients for the ternary mixtures were likewise represented with these models, using both the usual Pitzer mixing terms and also Scatchard's neutral-electrolyte model mixing terms for the extended ion-interaction model. Two mixing parameters are needed for each of the three models for {yK2HPO4 + (1 − y)KH2PO4}(aq), and both of these ion-interaction models give similar high-quality representations of the experimental results. However, the Clegg, Pitzer and Brimblecombe model had more difficulty in representing the osmotic coefficients of K2HPO4(aq), especially below 3 mol·kg−1, and consequently the corresponding mixture model with two mixing parameters is slightly less accurate for representing the osmotic coefficients. The maximum difference in calculated values of the mean molality-based activity coefficients for the two recommended extended Pitzer models with the different types of mixing terms are 0.0061 for the trace activity coefficient of K2HPO4(aq) in KH2PO4(aq) but with much better agreement at most mixture compositions.

AB  - Isopiestic measurements have been made for aqueous solutions of the common sodium cation mixtures of NaH2PO4and Na2HPO4at T = 298.15 ± 0.01 K, at NaH2PO4ionic strength fractions y = (0, 0.24851, 0.49862, 0.74544, and 1), where the ionic strength fractions were calculated by assuming complete electrolytic dissociation of NaH2PO4as 1:1 and Na2HPO4as 2:1 electrolytes; CaCl2(aq) was used as the reference standard solution. Model parameters for an extended form of Pitzer's ion-interaction model and also for the Clegg-Pitzer-Brimblecombe equations based on the mole-fraction-composition scale were evaluated at T = 298.15 K for both NaH2PO4(aq) and Na2HPO4(aq) using the isopiestic results from this study (17 values each) together with numerous critically assessed osmotic coefficients for both electrolytes taken from the published literature. The thermodynamic models for NaH2PO4(aq) extend to m = 7.5 mol·kg-1, whereas those for Na2HPO4(aq) extend to m = 2.6050 mol·kg-1, which is well above the solubility limit for the thermodynamically stable phase Na2HPO4·12H2O(cr). The 51 osmotic coefficients for the ternary mixtures were treated with these two models together with Scatchard's neutral-electrolyte model; one previous set of osmotic coefficient values for {yNaH2PO4+ (1 - y)Na2HPO4}(aq) mixtures was found in the literature [ Scharge, T.; et al. J. Chem. Thermodyn. 2015, 80, 172-183 ], and hence an analysis and comparison were made of our results with theirs.

Model parameters for an extended form of Pitzer's ion-interaction model and also for the Clegg-Pitzer-Brimblecombe equations based on the mole-fraction-composition scale were evaluated at T = 298.15 K for both NaH2PO4(aq) and Na2HPO4(aq) using the isopiestic results from this study (17 values each) together with numerous critically assessed osmotic coefficients for both electrolytes taken from the published literature. The thermodynamic models for NaH2PO4(aq) extend to m = 7.5 mol·kg-1, whereas those for Na2HPO4(aq) extend to m = 2.6050 mol·kg-1, which is well above the solubility limit for the thermodynamically stable phase Na2HPO4·12H2O(cr). The 51 osmotic coefficients for the ternary mixtures were treated with these two models together with Scatchard's neutral-electrolyte model; one previous set of osmotic coefficient values for {yNaH2PO4+ (1 - y)Na2HPO4}(aq) mixtures was found in the literature [ Scharge, T.; et al. J. Chem. Thermodyn. 2015, 80, 172-183 ], and hence an analysis and comparison were made of our results with theirs.",

An integrated approach to AO capacity determination allowed a more comprehensive comparison between samples. AB  - This study aimed to estimate antiradical, antioxidant (AO) and cytotoxic activities of the fungus Trametes versicolor ethanol fruiting body extract. The extract was found to effectively scavenge both O-2(center dot-) and NO center dot (29.62 and 52.48g/mL, respectively). It also showed a good AO activity in the polarographic HPMC assay (950%/mL). p-Hydroxybenzoic acid may be one of the responsible compounds for the afore-mentioned activities. The same extract also exhibited a concentration-dependent cytotoxicity against MCF-7 and HepG2 tumour cell lines reaching IC50 values of 123.51 and 134.29g/mL, respectively with no cytotoxic activity against normal MRC-5 cells. Gentisic, syringic and protocatechuic acids may be among the bioactive principles for the observed cytotoxicity. Taken all together, T. versicolor ethanol extract can be considered as a promising candidate for development of health promoting food supplement. [GRAPHICS] .

AB  - Locally available apricot (Prunus armeniaca L.) shells classified as a waste product from fruit processing, were alkali activated in order to develop an efficient heavy metal ions sorbent for water purification. To examine the changes occurred after alkali treatment, raw (SH) and modified apricot shells (SHM) were thoroughly characterized in terms of their chemical composition and surface properties. Chemical analysis revealed that alkaline treatment causes the disintegration of hemicellulose (its content decreased from 19.2 to 3.5%), which was in accordance with FTIR results. SEM micrographs and the mercury intrusion porosimetry revealed a larger surface area and porosity of SHM. Bohem's acid-base titration method indicated that the most of the SHM surface carboxylic groups were in sodium salt form and together with the pH of points of zero charge showed increase of surface alkalinity after modification. Treatment with NaOH enhanced the adsorption capacity by 154, 61 and 90% for Cu2+, Zn2+ and Pb2+, respectively. The amount of cations released from SHM was almost equal to the amount of adsorbed metal ions, suggesting ion exchange mechanism. The pseudo-second order kinetic indicated that the heavy metals cations were bound predominantly by complexation. In order to establish the effectiveness of the biosorbent in real wastewater sample, SHM was employed for cleaning-up of drain water emanating from atomic adsorption spectrophotometer. The SHM showed high removal efficiency towards multiple metal ions. The amounts of Fe, Pb, Cu and Cr ions were reduced by 97, 87, 81 and 80%, respectively, while Ni and Zn amounts were reduced for 33 and 14%. Used biosorbent SHM can be successfully regenerated with HCl (desorption  gt  95%) and after regeneration biosorbent can be reused or it can be safely disposed. (C) 2018 Elsevier B.V. AB  - Locally available apricot (Prunus armeniaca L.) shells classified as a waste product from fruit processing, were alkali activated in order to develop an efficient heavy metal ions sorbent for water purification. To examine the changes occurred after alkali treatment, raw (SH) and modified apricot shells (SHM) were thoroughly characterized in terms of their chemical composition and surface properties. Chemical analysis revealed that alkaline treatment causes the disintegration of hemicellulose (its content decreased from 19.2 to 3.5%), which was in accordance with FTIR results. SEM micrographs and the mercury intrusion porosimetry revealed a larger surface area and porosity of SHM. Bohem's acid-base titration method indicated that the most of the SHM surface carboxylic groups were in sodium salt form and together with the pH of points of zero charge showed increase of surface alkalinity after modification. Treatment with NaOH enhanced the adsorption capacity by 154, 61 and 90% for Cu2+, Zn2+ and Pb2+, respectively. The amount of cations released from SHM was almost equal to the amount of adsorbed metal ions, suggesting ion exchange mechanism. The pseudo-second order kinetic indicated that the heavy metals cations were bound predominantly by complexation. In order to establish the effectiveness of the biosorbent in real wastewater sample, SHM was employed for cleaning-up of drain water emanating from atomic adsorption spectrophotometer. The SHM showed high removal efficiency towards multiple metal ions. The amounts of Fe, Pb, Cu and Cr ions were reduced by 97, 87, 81 and 80%, respectively, while Ni and Zn amounts were reduced for 33 and 14%. Used biosorbent SHM can be successfully regenerated with HCl (desorption  gt  95%) and after regeneration biosorbent can be reused or it can be safely disposed. (C) 2018 Elsevier B.V. AB  - Antioxidant (AO) capacity of instant, espresso, filter and Turkish/Greek coffee brews, coffee substitutes (roasted chicory root, barley, pea, chickpea, carob and dried fig) and individual compounds (phenolic acids, flavonoids, methylxanthines, N-methyl pyridinium and HMW melanoidins) was assessed using DC polarographic assay based on decrease of anodic current originating from hydroxo-perhydroxo mercury complex formed in alkaline solutions of H2O2 at potential of mercury dissolution, as well as three spectrophotometric assays (DPPH, ABTS and FRAP). A large difference between applied assays ability to recognize various types of individual AOs was noticed. Only according to DC polarographic assay significant AO activity was ascribed to methylxanthines and N-methyl pyridinum. The total content of phenolics (TPC) present in complex samples was determined by FC assay. The highest TPC was ascribed to instant coffees and coffee substitutes while the lowest to decaffeinated filter coffee. Complex samples were grouped based on principal components analysis, phenolics AO coefficient, calculated as the ratio between AO capacity and TPC, and relative AO capacity index (RACI), calculated by assigning equal weight to all applied assays including FC. The highest values of RACI were ascribed to instant coffee brews, followed by substitutes while the lowest to the decaffeinated espresso coffee.

Determination of Copper in Wine by Anodic Stripping Voltammetry with Rotating Glassy Carbon and Microfiber Carbon Electrode

AB  - Anodic stripping voltammetry (ASV) with rotating glassy carbon electrode (RGCE) and microfiber carbon electrode (mu FCE), both filmed with mercury, were used to determine copper content in wine. Influence of phenolics on in situ mercury film formation was studied. Wine and quercetin were added gradually into solution of Hg(II) in acetic acid-sodium acetate buffer (pH = 4.60). Reduction of Hg(II) was observed in both cases. In situ filmed electrode was found unsuitable. Results obtained using ex situ filmed RGCE and mu FCE were found in good agreement with ASV using hanging mercury drop electrode. Thus, ASV with ex situ mercury filmed electrodes can be recommended for accurate quantification of copper in complex samples containing phenolics, without any special pretreatments.

AB  - The Li-ion batteries are the main power source for the high technology devices, such as mobile phones and electric vehicles. Because of that, the number of spent Li-ion batteries significantly increases. Today, the number of active mobile phones crossed 7.19 billion. It is estimated that the mass of the spent lithium ion batteries in China will exceed 500,000 t by 2020. The trouble is in the ingredients of these batteries. They contain Li, Co, Mn, Ni, Cu, Al and toxic and flammable electrolytes which have a harmful affection to the environment. Because of that, the recycling procedure attracts raising attention of researches. Several commercial spent Li-ion batteries were recycled by the relatively fast, economic and simple procedure. The three ways of separating the cathode material from Al collector were examined after the manual dismantling of the components of batteries with the Li(Co-Mn-Ni)O-2 as cathode material. These were: 1. dissolution of the Al collector in the alkali medium, 2. peeling off with N-methyl-pyrrolidone and 3. thermal decomposition of the adhesive at 700 degrees C. The procedure with the highest yield was the one with the dissolution in alkali medium. The chemical analysis of the single batteries' components (the crust, Al/Cu collector, cathode material) were done by the atomic absorption spectrometry. The components, before the analysis, were dissolved. The re-synthesis of the cathode material by the method of the citrate gel combustion was done after the separating the cathode material and dissolving it in the nitric acid. The obtained product was, after annealing, characterized by the methods of X-ray diffraction and Raman spectroscopy. The recycled product was LiCo0.59Mn0.26Ni0.15O2 stoichiometry, with the hexagonal layered structure alpha-NaFeO2 type. The functionalization of the resynthesized material was examined in the 1 M solution LiClO4 in the propylene carbonate, by galvanostatic charging, with the current density of 0.7C. The recycled material showed relatively good capacities of charging and discharging which are 94.9 i 64.8 mA h g(-1), respectively.

The interactions of vanadate monomer with the mycelium of fungus Phycomyces blakesleeanus: reduction or uptake?

AB  - The possibility of reduction of vanadate monomer in the mycelium of fungus Phycomyces blakesleeanus was investigated in this study by means of polarography. Control experiments were performed with vanadyl [V(IV)] and vanadate [V(V)] in 10 mM Hepes, pH 7.2. Addition of P. blakesleeanus mycelium resulted in disappearance of all V(IV) polarographic waves recorded in the control. This points to the uptake of all available V(IV) by the mycelium, up to 185 A mu mol/g(FW), and suggests P. blakesleeanus as a potential agent in V(IV) bioremediation. Polarographic measurements of mycelium with low concentrations (0.1-1 mM) of V(V), that only allows the presence of monomer, showed that fungal mycelia removes around 27% of V(V) from the extracellular solution. Uptake was saturated at 104 +/- 2 A mu mol/g(FW) which indicates excellent bioaccumulation capability of P. blakesleeanus. EPR, V-51 NMR and polarographic experiments showed no indications of any measurable extracellular complexation of V(V) monomer with fungal exudates, reduction by the mycelium or adsorption to the cell wall. Therefore, in contrast to vanadium oligomers, vanadate monomer interactions with the mycelium are restricted to its transport into the fungal cell, probably by a phosphate transporter.

AB  - Background: Free radicals (FR) occur in oral cavity where lot of food was transferred to through entire life under specific saliva conditions. Many enzymes, microorganism, alcohol beverages, nicotine and other harmful or indifferent substances when in contact to oral tissues might provoke oxidation process under specific condition creating FR's. The similar role might have various dental materials. Aim of the study was to record the level of antioxidant (AO) activity of several permanent (P) luting cements alone or combined with quercetin AO substance. Materials/Methods: P cements were Zn-phosphate, Zn-polycarboxilate, GIC and composite resin cement. They were prepared as original prescription and their variant by 1% weight addition of quercetin. AO activity of cements was measured by HPMC test evaluated by Student t test. Results: There were statistically significant differences among Zn-phosphate, Zn-polycarboxilate and resin dental cements (p  gt  0,05). GIC displayed significantly higher AO values (p  lt  0,01) versus other three cements. There were no difference in AO capacity between sample of original P cements and their corresponding quercetin variants (p  gt  0,05). Conclusions: Conventional GIC displayed the most powerful AO activity among P luting cements. Addition of 1% antioxidant quercetin did not improve AO capacity of investigated cements.

AB  - Antioxidant (AO) capacity of chocolates with 27, 44 and 75 % cocoa was assessed after production and during twelve months of storage by direct current (DC) polarographic assay, based on the decrease of anodic current caused by the formation of hydroxo-perhydroxyl mercury(II) complex (HPMC) in alkaline solutions of hydrogen peroxide at potentials of mercury oxidation, and two spectrophotometric assays. Relative antioxidant capacity index (RACI) was calculated by taking the average value of the AO assay (the sample mass in all assays was identical). Oxidative stability of chocolate fat was determined by differential scanning calorimetry. Measured parameters and RACI were correlated mutually and with the content of total phenols (Folin-Ciocalteu assay), flavan-3-ols (vanillin and p-dimethylaminocinnamaldehyde assay) and proanthocyanidins (modified Bate-Smith assay). During storage, the studied functional and health-related characteristics remained unchanged. Amongst applied AO assays, the DC polarographic one, whose validity was confirmed by two-way ANOVA and F-test, correlated most significantly with oxidative stability (oxidation onset temperature and induction time). In addition, principal component analysis was applied to characterise chocolate types.

AB  - Antioxidant (AO) capacity of various medicinal plants extracts and phenolic compounds was assessed by the most widely used spectrophotometric assays such as ferric reducing antioxidant power (FRAP) and scavenging of 2,2-azino-bis-3-ethylbenzothiazoline-6-sulfonic acid (ABTS) and 2,2-diphenyl-1-picrylhydrazyl (DPPH). In addition, two direct current (DC) polarographic assays, one based on a decrease of anodic current of [Hg(O2H)(OH)]-HydroxoPerhydroxoMercury(II) Complex (HPMC) formation in alkaline solution of H2O2, at the potential of mercury dissolution and another recently developed Mercury Reduction Antioxidant Power (MRAP), based on a a decrease of cathodic current of Hg(II) reduction were employed. Percentage of both currents decrease was plotted versus the volume of gradually added complex samples or the amount of individual ones and the slopes of these plots were used to express AO capacity. Total phenolic content (TPC) of extracts was determined by Folin-Ciocalteu (FC) assay. Correlations between applied assays were calculated by regression analysis. Relative Antioxidant Capacity Index (RACI), calculated by assigning equal weight to all applied assays and Phenolic Antioxidant Coefficients (PAC), calculated as a ratio between particular AO capacity and TPC, were used to achieve more comprehensive comparison between analyzed samples, as well as applied assays.

DC polarographic examination of Hg2+ reduction applicability to antioxidant activity determination

AB  - Direct current polarographic (DCP) antioxidant assay based on the decrease of anodic limiting current of HydroxoPerhydroxoMercury(II) Complex [Hg(O2H)(OH)] (HPMC) upon addition of antioxidants (AOs) was verified. Here, influence of several common AOs on DCP cathodic currents of both HPMC and Hg2+ was followed. Amperometric titrations of HPMC and Hg2+ with AOs and potentiometric titrations of Hg2+ were performed. The changes of DCP i-E curves and limiting currents decrease during amperometric titrations, as well as end points of amperometric and potentiometric titrations for each AO used as titrant proved the occurrence of Hg2+ reduction, either from complex (HPMC) or salt (HgCl2) solution. Constant H2O2 reduction current during amperometric titration of HPMC further confirmed the participation of Hg2+ as the oxidant species in reaction with AOs. The possibility of determining AO activity of flavonoids, phenolic acids, ascorbic acid and Trolox based on the decrease of Hg2+ reduction current was demonstrated. Strict correlation (R-2 = 0.96) between a new AO assay named MRAP (Mercury Reduction Antioxidant Power) and HPMC assay was found. Total AO capacity of real samples was measured by the proposed fast, simple and inexpensive MRAP assay as well. (C) 2015 Elsevier Ltd. All rights reserved.

abstract = "Direct current polarographic (DCP) antioxidant assay based on the decrease of anodic limiting current of HydroxoPerhydroxoMercury(II) Complex [Hg(O2H)(OH)] (HPMC) upon addition of antioxidants (AOs) was verified. Here, influence of several common AOs on DCP cathodic currents of both HPMC and Hg2+ was followed. Amperometric titrations of HPMC and Hg2+ with AOs and potentiometric titrations of Hg2+ were performed. The changes of DCP i-E curves and limiting currents decrease during amperometric titrations, as well as end points of amperometric and potentiometric titrations for each AO used as titrant proved the occurrence of Hg2+ reduction, either from complex (HPMC) or salt (HgCl2) solution. Constant H2O2 reduction current during amperometric titration of HPMC further confirmed the participation of Hg2+ as the oxidant species in reaction with AOs. The possibility of determining AO activity of flavonoids, phenolic acids, ascorbic acid and Trolox based on the decrease of Hg2+ reduction current was demonstrated. Strict correlation (R-2 = 0.96) between a new AO assay named MRAP (Mercury Reduction Antioxidant Power) and HPMC assay was found. MRAP was shown to be effective in the determination of AO activity of both synthetic mixtures of individual AOs and real samples containing various AOs. Agreement between the decrease of Hg2+ cathodic current and HPMC anodic current lead to the assumption that Hg2+ reduction causes a decrease in concentration of Hg2+ available for HMPC complex formation, i.e. its anodic current decrease. The validity Of MRAP assay was also confirmed through good correlation with FRAP assay.

Antioxidant Activity of Pea Protein Hydrolysates Produced by Batch Fermentation with Lactic Acid Bacteria

AB  - Nine Lactobacillus strains known for surface proteinase activity were chosen from our collection and tested for their ability to grow in pea seed protein-based medium, and to hydrolyze purified pea proteins in order to produce peptides with antioxidant (AO) activity. Two strains, Lactobacillus rhamnosus BGT10 and Lactobacillus zeae LMG17315, exhibited strong proteolytic activity against pea proteins. The AO activity of the pea hydrolysate fraction, MW  lt  10 kDa, obtained by the fermentation of purified pea proteins with Lactobacillus rhamnosus BGT10, was tested by standard spectrophotometric assays (DPPH, ABTS, Fe3+-reducing capacity) and the recently developed direct current (DC) polarographic assay. The low molecular weight fraction of the obtained hydrolysate was separated using ion exchange chromatography, while the AO activity of eluted fractions was determined by means of a sensitive DC polarographic assay without previous concentration of samples. Results revealed that the fraction present in low abundance that contained basic peptides possessed the highest antioxidant activity. Based on the obtained results, it can be concluded that Lactobacillus rhamnosus BGT10 should be further investigated as a candidate strain for large-scale production of bioactive peptides from legume proteins.
