Lignin and organic free radicals in maize (Zea mays L.) seeds in response to aflatoxin B1 contamination: an optical and EPR spectroscopic study

AB  - Aflatoxin B1 (AFB1), is a naturally hazardous environmental contaminant, contained mainly in various cereal seeds. The maize (Zea mays L.) seeds containing various levels of the AFB1 was investigated using optical and electron paramagnetic resonance spectroscopies. The changes of lignin content and organic free radicals as indicators of the seeds' defense response to the AFB1-stressor were estimated, in the seeds' inner and outer fractions. The lignin content was determined using the Acetyl Bromide method. No correlation was found between the lignin content and the AFB1 concentration in the inner fraction (R of 0.030, p = 0.943), while a significant positive linear correlation (R of 0.9923, p= 0.00005) between the lignin content and the AFB1 concentration in the outer fraction was obtained. Employing electron paramagnetic resonance (EPR) spectroscopy, the different response of the fractions was observed, regarding the content of free radicals with increasing AFB1 concentrations. In addition, front-face fluorescence spectroscopy in combination with the deconvolution showed a positive linear correlation between the ratio of the green and blue spectral emission components (C4/C2) area and the AFB1 concentration in the outer fraction. Thus lignin content and the (C4/C2) area ratio may be reliable indicators for the screening of lignin changes related to the level of AFB1 in the seeds.

Physicochemical parameters as indicators of the authenticity of monofloral honey from the territory of the Republic of Serbia

AB  - The aim of this research was to verify the authenticity of monofloral honeys from the territory of the Republic of Serbia on the basis of physicochemical parameters routinely measured in honey quality control using multivariate analysis. Seventeen samples of monofloral honey (11 samples of acacia honey and 6 samples of sunflower honey) from the territory of the Republic of Serbia were analyzed. Physicochemical analysis of the samples included the examination of basic quality parameters and qualitative and quantitative analyses of phenolic compounds. In the samples tested, a total of 93 phenolic compounds were tentatively identified, and 19 of them were quantified. The obtained physicochemical analysis of the data served as input for the multivariate analysis. The hеаt map, which is useful for visualizing numerical data, was used for this purpose. The obtained results showed that the applied data can serve to clearly separate acacia and sunflower honeys.

Variability Estimation of the Protein and Phenol Total Content in Honey Using Front Face Fluorescence Spectroscopy Coupled with MCR–ALS Analysis

AB  - It has been explored to what extent the ratio of the two main fluorophores in honey, originating from proteins and phenolic compounds, change between the honey extraction stage and packaging in the jars. Fluorescence spectroscopy combined with multivariate curve resolution–alternating least squares (MCR–ALS) was used to determine the ratio of the spectral components originating from phenol and proteins (Ph/Pr) fl , as a ratiometric indicator of variability in selected Lime tree (Tilia L.) honey samples. Spectrophotometric quantification of phenols and proteins in the honey samples was also performed and their ratio (Ph/Pr) sp was calculated. The values of the (Ph/Pr) fl ratio and honey protein content after packaging depended on the quality of homogenization before packaging in jars. Colorimetric and fluorometric results for phenols and proteins were in compliance. The examples are the values 3.34, 3.30, and 9, 3.14 for (Ph/Pr) fl and corresponding values 2.64, 2.18, and 12.75, 2.31 for (Ph/Pr) sp , in all pairs, the first value presenting the sample after extraction and the second value the sample after packaging in jars. Both methods show that in the former case there is no change in the phenol/protein ratio, and in the latter case the ratio decreased.

Effects of Ag+ ion doping on UV radiation absorption and luminescence profiles of fluorapatite nanomaterials obtained by neutralization method

AB  - In the present study we have analyzed effects of Ag+ ions doping on energetic profiles of nanophosphors materials based on fluorapatite crystal system. The UV radiation absorption and luminescence properties of monophase fluorapatite (FAP) and Ag+ doped fluorapatite (AgFAP) nanomaterials obtained by neutralization method were investigated using the photoluminescence spectrophotometry. The excitation-emission profiles of nanomaterials were analyzed statistically by MCR-ALS method and number of fluorophores was extracted. FAP lattice absorbed light at 350 nm in the UVA part of spectrum, and with increasing concentration of Ag+ ions new absorption maximum appeared at 270 nm in the UVC part. Fluorescence of FAP nanoparticles was in violet region of visible part of the spectrum, with a red shift to the green region when Ag+ was doped in lattice. MCR-ALS analyses of fluorescence spectra confirm formation of two maxima, at 484 and 505 nm, as a consequence of Ag+ ions doping in FAP lattice at Ca1 (4f) sites. The results of quantum chemical calculations showed that an Ag+ ion is stronger bonded to the binding site 1 (-1352:6 kcal/mol) than to the binding site 2 (-1249:0 kcal/mol). Considering that AgFAP1 nanopowder absorbs photons over all part of UV radiation spectrum, this material might be used as potential radiation protective nanomaterial.

AB  - This study examines the impact of harvest date, temperature and precipitation on an accumulation of the volatile aromatic compounds in 'Cabernet Sauvignon' wine. The research was carried out in the vineyard of King Peter I Karadjordjevic, the Royal Winery in Oplenac, Serbia, during 2010 and 2011. In each year grapes were harvested twice - when the full maturity has been reached and 15 days later. Aromatic complexes of these 4 wines were analyzed using the Gas Chromatography Mass Spectrometry (GC-MS) and the Gas Chromatography with Flame Ionization Detector (GC-FID). Higher alcohols, lactones, organic acids, esters and amides were detected in wines. Relative portions of 34 aromatic compounds were compared for the full maturity harvests in the both experimental years, as well as their change from the first to the second harvest in each year.

AB  - Steady state fluorescence spectroscopy in combination with Multivariate Curve Resolution - Alternating Least Squares (MCR-ALS) for spectral analysis has been applied to differentiate commercial samples of wines available on the market. We have chosen the wine trade marks from the two Serbian producers that contain only one wine type, since such approach is a starting step for further analyses of the more complex samples containing different wine types. We also studied changes of the emission spectra of these samples during seven days after opening of the wine bottle. The emission spectra were recorded in the wavelength range 275 - 500 nm, after excitation in the range 255 - 300 nm. The spectra of the wine samples obtained from the same producer are very similar, i.e. contain the same components in the similar ratios. The changes of spectral components at 315 nm and 430 nm were the basis for differentiation of the wine samples from the two producers, as well as for estimation of the wine stability in time after the bottle opening. The results indicate that this may be a useful approach in fingerprinting wines from various producers, as well as in screening the stability of wine.
AB  - Fluorescentna spektroskopija u kombinaciji sa metodom Multivarijaciona Rezolucija Krivih - Naizmenični Najmanji Kvadrati (Multivariate Curve Resolution - Alternating Least Squares, MCR-ALS), je primenjena za razlikovanje komercijalnih uzoraka vina dostupnih na tržištu. Odabrali smo trgovačke marke vina od dva srpska proizvođača koje sadrže samo jednu vrstu vina, jer je ovakav pristup početni korak za kasnije analize složenijih uzoraka koji sadrže različite vrste vina. Takođe smo proučavali promene emisionih spektara ovih uzoraka u toku sedam dana posle otvaranja boce. Emisioni spektri su mereni u opsegu talasnih dužina 275 - 500 nm, posle pobuđivanja u opsegu 255 - 300 nm. Spektri uzoraka vina od istog proizvođača su veoma slični, to jest sadrže iste komponente u sličnom međusobnom odnosu. Promene komponenata spektara na 315 nm i 430 nm bile su osnova za razlikovanje uzoraka vina od dva proizvođača, kao i za procenu stabilnosti vina u vremenu posle otvaranja boce. Rezultati ukazuju da ovo može biti koristan pristup u razlikovanju vina od različitih proizvođača, kao i u praćenju stabilnosti sastava vina.
