AB - Due to the promising applications of propolis as a natural food preservative, functional ingredient or supplement, on one side, and the inherent toxicity and high volatility of organic solvents used for extraction of phenolics from propolis, a range of natural eutectic solvents (NESs) was evaluated for the extraction of phenolic compounds from poplar type propolis to create “green-labelled” food products. The phenolic profile of novel green propolis extracts was evaluated using high-performance thin-layer chromatography, while 16 phenolic compounds were quantified by ultra-high-performance liquid chromatography hyphenated with mass spectrometry. Pinocembrin and chrysin were the most dominant phenolics with an average concentration of 3111 and 3039 mg/kg in lactic acid-based NES, respectively. Biological activity, such as antioxidative and antimicrobial activities, were determined using spectrophotometric and well diffusion methods. Considering all spectrophotometric assays lactic acid-based presented as the best extraction medium for phenolic extraction, with TPC, TFC and RSA values 150.0 mg GAE/g, 190.0 mg RUE/g, and 132.8 mg TE/g on average, respectively. From 38 studied NESs, choline-chloride:tartaric acid (1:1 n/n), glycerol:lactic acid (1:1 n/n) and glycine: lactic acid (1:3 n/n) were the most promising extraction medium for obtaining the propolis extracts enriched with bioactive compounds. Additionally, proposed green solvents were proved to be the designer solvents and showed potential to be tuned to target specific phenolic compounds. Improved, or at least maintained, biological activity of NES propolis extracts support its application in agriculture industry.

AB - Considering the health-benefits of berry fruits consumption and increased market demands for food authenticity
as one of the most important quality assurances, phenolic profiling by high-performance thin layer chroma-
tography and ultra-high-performance liquid chromatography hyphenated with mass spectrometry was combined
with multivariate analysis for phytochemical characterization and intercultivar discrimination of cultivated
berry seeds. The phenolic profiles of 45 berry seeds from nine genuine Serbian cultivated fruit species (straw-
berry, raspberry, blackberry, black currant, blueberry, gooseberry, cape gooseberry, chokeberry, and goji berry)
revealed a good differentiation according to botanical origin. In order to determine biomarkers responsible for
the classification, a total of 103 phenolic compounds were identified, including 53 phenolic acids and their
derivatives, 26 flavonoids and 24 glycosides. Biomarkers derived from the phenolic profile of berry seeds proved
to be a powerful tool in the authentication of botanical origin, and may be useful in detection of frauds in berry-
based seed-containing product.

AB - DES are mixtures of a hydrogen bond donor (HBD) and a hydrogen bond acceptor (HBA) that, when combined in a specific molar ratio, exhibit a significant decrease in melting point and become liquid at room temperature. DES can be tailored to meet different physicochemical requirements by altering the HBA:HBD ratio or the nature of the compounds. In this study, we utilized the COSMO-RS method to predict the activity coefficients of 12 phenolic compounds in 105 NaDES. These NaDES contained equimolar mixtures of betaine, carnitine, creatine, and ornithine as HBAs and various sugars and amino acids as HBDs. The NaDES were compared to methanol, and several mixtures exhibited higher affinities to all phenolics. The most promising NaDES were betaine-glycerol, betaine-xylitol, and carnitine-urea. We are currently conducting experimental validation of the extraction efficiency of these cosmetically acceptable solvents. In summary, we have identified promising NaDES mixtures that could replace conventional solvents and contribute to the development of sustainable, bio-based extracts.

AB - The latest studies show that UV and blue light, emitted by electronic devices with screens, slows the proliferation of the keratinocytes and generates free radicals leading to skin ageing1. Polyphenols, a compounds found in plants, can prevent light-induced damage to the skin. A constant screening of plant extracts is requisite in order to discern light-protective formulations to constitute cosmetic products. Using mass spectrometry, phenolic compounds were identified and quantified in plant extracts from 18 plants grown in Eastern Serbia. The cytotoxicity of the obtained extracts was tested on HaCaT cells (immortalized human keratinocytes) and IC50 values were determined. The obtained extracts are rich mixtures of phenolic compounds that must be dosed appropriately since they also damage skin cells at larger concentrations.

abstract = "The latest studies show that UV and blue light, emitted by electronic devices with screens, slows the proliferation of the keratinocytes and generates free radicals leading to skin ageing1.

AB - Due to their strong antioxidative potential, flower petals are considered important starting materials for the development of cosmetic products1. Present study explored phenolic profile and antioxidative potential of 17 flower petals and their potential application in the cosmetics industry. The phenolic profile of investigated extracts was determined using High-Performance Thin Layer Chromatography (HPTLC). The antioxidative potential was assessed using both ABTS and DPPH spectrophotometric tests, and total phenol content. DPPH-HPTLC assay was applied to identify compounds with the highest antioxidative activity. Five phenols were identified in the investigated extracts. Based on radical antioxidative tests, Paeonia lactifora, Paeonia decora, Rosa istriaca and Pelargonium peltatum were identified as species with the highest antioxidative potential, while gallic acid was recognized as the strongest antioxidants.

AB - Sorghum grain (Sorghum bicolor L. Moench) is a gluten-free cereal with excellent nutritional value and is a good source of antioxidants, including polyphenols, as well as minerals with proven health benefits. Herein, the phenolic composition, elemental profile, and antioxidant activity of sixteen food-grade sorghum grains (S1–S16) grown under agroecological conditions in Serbia were determined. Nine phenolic compounds characteristic of sorghum grains, such as luteolinidin, 5-methoxyluteolinidin, luteolidin derivative, luteolidin glucoside, apigeninidin, 7-methoxyapigeninidin, apigeninidin glucoside, and cyanidin derivative, were quantified. The antioxidant potential of the analyzed sorghum grains was evaluated by UV/Vis (DPPH, ABTS, and FRAP) and Electron Paramagnetic Resonance spectroscopy (hydroxyl and ascorbyl radical scavenging assays). The content of macro- and microelements was determined by Inductively Coupled Plasma Optical Emission spectroscopy. Theoretical daily intakes of selected major and trace elements were assessed and compared with the Recommended Daily Allowance or Adequate Intake. Sample S8 had the highest amount of phenolic compounds, while S4, S6, and S8 exhibited the strongest antioxidative potential. The sorghum studied could completely satisfy the daily needs of macro- (K, Mg, and P) and microelements (Se, Zn, Fe). Pattern recognition techniques confirmed the discrimination of samples based on phenolic profile and elemental analysis and recognized the main markers responsible for differences between the investigated samples. The reaction between hydroxyl radicals and luteolinidin/apigeninidin was investigated by Density Functional Theory and thermodynamically preferred mechanism was determined.

AB - Natural products and their analogues have contributed significantly to treatment options, especially for anti-inflammatory and infectious diseases. Thus, the primary objective of this work was to compare the bioactivity profiles of selected medicinal plants that are historically used in folk medicine to treat inflammation and infections in the body. Chemical HPTLC fingerprinting was used to assess antioxidant, phenolic and flavonoid content, while bioassay-guided HPTLC was used to detect compounds with the highest antibacterial and anti-inflammatory activities. The results of this study showed that green tea leaf, walnut leaf, St. John’s wort herb, wild thyme herb, European goldenrod herb, chamomile flower, and immortelle flower extracts were strong radical scavengers. Green tea and nettle extracts were the most active extracts against E. coli, while calendula flower extract showed significant potency against S. aureus. Furthermore, green tea, greater celandine, and fumitory extracts exhibited pronounced potential in suppressing COX-1 activity. The bioactive compounds from the green tea extract, as the most bioactive, were isolated by preparative thin-layer chromatography and characterized with their FTIR spectra. Although earlier studies have related green tea’s anti-inflammatory properties to the presence of catechins, particularly epigallocatechin-3-gallate, the FTIR spectrum of the compound from the most intense bioactive zone showed the strongest anti-inflammatory activity can be attributed to amino acids and heterocyclic compounds. As expected, antibacterial activity in extracts was related to fatty acids and monoglycerides.

AB - The aim of this study is to develop a rapid, effect-directed screening method for quality assessment of bee pollen-honey mixtures. The comparative antioxidant potential and phenolic content of honey, bee pollen, and the bee pollen-honey mixtures, was performed using spectrophotometry. The total phenolic content and antioxidative activity of bee pollen-honey mixtures with 20 % bee pollen share were in the range 3.03–3.11 mg GAE/g, and 6.02–6.96 mmol TE/kg, respectively, while mixtures with 30 % bee pollen share contained 3.92–4.18 mg GAE/g, and 9.69–10.11 mmol TE/kg. Chromatographic fingerprint of bee pollen-honey mixtures was performed by high-performance thin-layer chromatography with conditions developed by authors and reported for the first time. Fingerprint analysis hyphenated with chemometrics enabled authenticity assessments of honey in mixtures. Results indicate that bee pollen-honey mixtures represent a food with highly, both, nutritious characteristics and health-promoting effect.

AB - Propolis is a resinous natural substance collected by honeybees from different plant sources. The element content of propolis is influenced by the content of elements in the soil, climatic factors, and the degree of pollution. The aim of the study was to characterize element composition and content of toxic elements in 51 propolis collected from different locations in Serbia and to evaluate the differences among them. Determination of the 21 elements were performed using Inductively coupled plasma - optical emission spectrometry. K was the most abundant element, followed by Ca, Mg, Si, Fe, Zn, and Al. The microelements B, Ba, Mn, Na, Pb, and Ti were also present in propolis, while Co, Cr, Cu, Mo, Ni, Sr, and V were found in trace. Toxic elements such as Pb, As, and Cd were found in the trace. Kruskal-Wallis tests differentiate groups of samples by geographical origin. The presence of B, Fe, Sr, Ti, Zn, and As demonstrated statistical significance between six regions of Serbia.

AB - High-Performance Thin-Layer Chromatography (HPTLC)-radical scavenging capacity (RSC) assays are stan- dard techniques for the separation and identification of antioxidants from complex mixtures. HPTLC coupled with DPPH · visualization of chromatograms allows for the detection of individual antioxidants. However, other HPTLC-RSC assays that recognize compounds exhibiting different mechanisms of radical- scavenging activity are rarely reported. In this study, we developed an integrated approach that combines five HPTLC-RSC assays, principal component analysis (PCA) and quantum chemical calculations to assess the antioxidant capacity of Sempervivum tectorum L. leaf extracts. Two HPTLC assays - potassium hexa- cyanoferrate(III) total reducing power assay (TRP) and total antioxidant capacity by phosphomolybdenum method (TAC) - were developed for the first time. The method supports a more in-depth study of the RSC of natural products, as it compares the radical scavenging fingerprints of S. tectorum leaf extracts and recognizes differences in their individual bioactive constituents. Kaempferol, kaempferol 3- O -glucoside, quercetin 3- O -glucoside, caffeic acid, and gallic acid were identified as the compounds that discriminate HPTLC-RSC assays according to their mechanism of action and capture the similarities between 20 S. tec- torum samples. Additionally, DFT calculations on M06–2X/6–31 + G (d,p) level were applied to map ther- modynamic feasibility of hydrogen atom transfer (HAT) and single electron transfer (SET) mechanisms of the identified compounds. Based on experimental and theoretical results, a combination of HPTLC-ABTS and HPTLC-TAC assays were proposed as the optimal method for mapping the antioxidants from S. tec- torum . This study represents a step forward in identifying and quantifying individual antioxidants from complex food and natural product matrices in a more rational manner.

AB - Sempervivum tectorum L. is a perennial plant with succulent leaves arranged in a rosette. Despite its wide use in traditional medicine for the treatment of various changes on the skin, nervousness, anxiety, menstrual ailments, etc., it still does not have a biography in the European Pharmacopoeia¹. The antibacterial effect of juice squeezed from the leaves and tea made from dried leaves was tested against bacteria that cause inflammation of the ear and bladder². Direct bioautography on HPTLC plates for the investigation of antibacterial activity of the houseleek leaves extracts was done for the first time. Four HPTLC bioassays were developed for gram-positive bacteria (B. subtilis, M. lysodeikticus, MRSA, and S. aureus), and two for gramnegative bacteria (E. coli and K. pneumoniae). Antibacterial activity of grampositive bacteria comes from active compounds at RF 0.57 and 0.92. HPTLC bioassays show higher sensitivity for gram-negative bacteria, as numerous active bands were observed at RF 0.03, 0.38, 0.42, 0.56, 0.57, 0.86 and 0.92. By applying chemometrics and principal components analysis, the HPTLC bioassays of grampositive bacteria were separated into two groups on the scores plot graphic along PC1 axis. The bioassays for B. subtilis and M. lysodeikticus form a less uniform group in the upper left quadrant, while the bioassays for MRSA and S. aureus form a homogeneous group in the lower right quadrant. Presence or absence of the active bands at RF 0.57 and 0.92 affect these separations. The bioassays for the two gramnegative bacteria are separated along both axes on the scores plot graphic from each other but also from the other bioassays for the gram-positive bacteria. This separation is influenced by the presence of numerous active bands. For further investigation it is necessary to identify compounds responsible for the antibacterial activity of the houseleek leaves extracts.

AB - Natural Eutectic Solvents (NES) are a promising class of environmentally friendly liquids that offer an alternative to conventional organic solvents. To investigate their potential for extracting phenolic compounds from plant materials, we employed the COSMO-RS (COnductor like Screening MOdel for Real Solvents) computational method. Nineteen NES solvents were prepared to determine the most selective and effective solvent for extracting phenolic compounds from Teucrium chamaedrys using both theoretical and experimental evaluation. Screening of extraction efficiency was performed by quantification of individual compounds using ultra-high-performance liquid chromatography with a diode array detector and a triple-quadrupole mass spectrometer (UHPLC-DAD-MS/MS), as well as spectrophotometric assays (total phenolic content, total flavonoid content, and radical scavenging activity). In this paper, we propose a new approach to model NES properties using COSMO-RS. While the majority of previous studies have modeled these solvents as individual or pseudo-component complexes between hydrogen bond donor (HBD) and hydrogen bond acceptor (HBA), we have developed a ternary HBD:HBA:water supramolecular complex model to represent NES solution structures more accurately. This model provides insights into the intermolecular interactions driving the extraction process and predicts extraction efficiencies that agree with experimental data. Two eutectic mixtures (choline chloride:succinic acid in molar ratio 1:1 and 20% of water (w/w) and choline chloride: glycerol in molar ratio 1:1 and 20% of water (w/w)) showed strong affinity towards phenolic compounds. Overall, our findings suggest that the ternary complex model is a more appropriate approach for modeling NES properties.

AB - Owing to the use of large volumes of toxic solvents and reagents, green analytical methods have gained increasing importance in natural product research. Herein, a green approach is proposed using the natural deep eutectic solvents (NADES) for the extraction and separation of phenols from Teucrium chamaedrys by use of an ecofriendly high-performance thin-layer chromatography (HPTLC) method. Principal component analysis was used to classify NADES extracts based on HPTLC data. The method’s greenness was assessed using two different tools: the National Environmental Methods Index (NEMI) and the analytical eco-scale. This method can also find application in routine quality control analysis of the investigated medicinal herb with minimal harm to the environment or human health.

AB - Natural Deep Eutectic Solvents (NADES) are novel and promising solvents for green extraction of phytochemicals from food and agricultural products. NADES are made of natural origin compounds that are connected via hydrogen bond and have unique tuneable properties1. Numerous studies investigated the extraction of bioactive compounds using NADES resulted in better extraction efficiency compared to conventional solvents, suggesting large potential of these solvents2,3. Low-toxicity, natural origin and affordability of NADES-based extracts enables their application as a preservative in the food industry or functional ingredient of food supplements. The aim of this study was to develop an efficient eco-friendly method for the extraction of phenolic compounds from berries, blueberry, chokeberry and black goji berry, using NADES. This goal will be achieved through investigation of extraction efficiency of applied NADES based on three different approaches, a) chromatographic, through high-performance thin layer chromatography (HPTLC) fingerprint analysis, ultra-high-performance liquid chromatography with a diode array detector and a triple-quadrupole mass spectrometer (UHPLC-DAD-MS/MS) target analysis and bioautography, b) spectroscopic, through quality control parameters, total phenolic content (TPC), total flavonoid content (TFC) and radical scavenging activity (RSA), and c) microbiological, through well diffusion method and minimum inhibitory concentration (MIC).
In this study, 36 NADES mixtures, prepared from primary plant metabolites, were tested as green alternatives for extraction of phenolics from three berries. Different hydrogen-bond acceptors (HBA), such as choline chloride, L-proline, L-glycine, and L-lysine were mixed with various hydrogen-bond donors (HBD), four organic acids, two sugars, glycerol and urea, at various molar ratios for NADES preparation. Guided by the fact that extraction efficiency depends on polarity, viscosity and dissolving ability of solvents4, the influence of HBA, HBD and water content were investigated. Methanol, as conventional solvent, was used as contrastive solvent.
Choline chloride-based NADES in combination with malic acid, glycerol or urea showed as most efficient. All groups of studied NADES showed various extraction capabilities for phenolic compounds based on chromatographic evaluation and spectrophotometric tests. These results indicate great power of NADES solvents towards selective extraction of phenolics and shows that NADES are designer solvents. Appropriate selection of components for NADES preparation can result in selective extraction of specific phenolic compounds, regardless of how similar structures of targeted compounds are or how complex the matrix is, which gives a significant advantage for application of NADES.

abstract = "Natural Deep Eutectic Solvents (NADES) are novel and promising solvents for green extraction of phytochemicals from food and agricultural products.

AB - The cosmetic industry is currently experiencing substantial growth, accompanied by a surge in the number of consumers. Due to natural ingredients' safety and efficiency, an increasingly prominent trend in this industry is the design of herbal-based cosmetic products and formulations. To meet demand, integrating innovative natural resources like plant-based formulations is essential [1]. Skin ageing-related enzymes, elastase and tyrosinase, contribute to the emergence of aesthetic issues such as wrinkles, freckles, and loss of elasticity, as well as hyperpigmentation and melasma [2]. Hence, there is an increasing focus on finding inhibitors for enzymes associated with skin ageing. The fact is that plant metabolites, particularly phenolic acids and flavonoids, are frequently concentrated in plant leaves and flowers. Accordingly, we assessed the inhibitory effects on elastase and tyrosinase in seventeen particularly selected methanolic extracts obtained from various flower petal varieties cultivated in Serbia. Our analysis revealed that petals exhibit significantly more pronounced inhibitory effects, expressed as IC50 values, on elastase compared to tyrosinase. White peony (Paeonia lactifora) (230 ± 1 μg/mL), pelargonium (Pelargonium peltatum) (206 ± 3 μg/mL) and purple rose (Rosa centifolia, purple) (222 ± 8 μg/mL) emerged as the most potent elastase inhibitors, surpassing the reference compound epigallocatechin gallate (348 ± 9 μg/mL). Additionally, the lowest IC50 value for tyrosinase inhibition was attributed to lilac (Syringa vulgaris) petal extract (272 ± 28 μg/mL), which was compared to the standard compound kojic acid (50 ± 14 μg/mL). In conclusion, white peony, pelargonium, purple rose, and lilac represent promising candidates for further investigation and incorporation into cosmetic products, showcasing their potential to effectively address skin-related concerns.

AB - To replace common organic solvents that present inherent toxicity and have high volatility and to improve the extraction efficiency, a range of natural deep eutectic solvents (NADESs) were evaluated for the extraction of phenolic compounds from Agrimonia eupatoria. Screening of NADES efficiency was carried out based on the total phenolic and flavonoid content and radical-scavenging activity, determined by spectrophotometry, as well as phenolic compounds quantified, obtained using ultra-high-performance liquid chromatography with a diode array detector and a triple-quadrupole mass spectrometer. Increased extraction efficiency when compared with organic solvent was achieved using NADES mixtures choline chloride (ChCl):urea 1:2 and choline chloride:glycerol 1:1. Flavonol glycosides were the most abundant compounds in all extracts. The COSMO-RS model provided insights into the most important intermolecular interactions that drive the extraction process. Moreover, it could explain the extraction efficiency of flavonol glycosides using ChCl:glycerol NADES. The current article offers experimental evidence and mechanistic insights for the selection of optimal NADES to extract bioactive components from Agrimonia eupatoria. © 2022 by the authors.

AB - Strawberries are an important fruit in the European diet because of their unique taste and high content of essential nutrients and bioactive compounds. The anthocyanins are known to be colorful phenolics in strawberries. In 17 samples of six strawberry cultivars produced in Serbia, i.e., the common varieties Alba, Asia, and Clery as well as promising breeding materials (11.29.11, 11.34.6, and 11.39.3), the anthocyanin profile as well as antimicrobial and antioxidative activity profiles were determined. All investigated extracts showed antioxidative and antibacterial activities against Gram-negative Aliivibrio fischeri. The responses were quite similar in number and intensity. The HPTLC-DPPH• scavenging assay and HPTLC-Aliivibrio fischeri bioassay coupled with high-resolution mass spectrometry identified pelargonidin-3-O-glucoside (Pg-3-glc) as the main anthocyanin and prominent antioxidative and antimicrobial compound in strawberries. The density functional theory calculations at the M06-2X/6-31+G(d,p) level showed that Pg-3-glc quenches free radicals via sequential proton loss electron transfer mechanism in water and in pentyl ethanoate, where the 5-OH group is the most reactive site for proton and hydrogen atom transfer. The results were confirmed via spectrophotometry. The highest total phenolic content was found in Clery and 11.39.3, while statistically significant differences between the genotypes regarding the antioxidant activity were not confirmed. Although very similar in the anthocyanin, antioxidative, and antimicrobial profile patterns, the strawberry genotypes were successfully classified using principal component analysis.

AB - The houseleek (Sempervivum tectorum L.) has a wide application in traditional medicine for skin changes treatment, ear, throat and bladder infections, stomach ulcers. Recently, several studies confirmed antioxidative, antimicrobial and antiinflammatory effects of houseleek [1,2]. Several spectroscopic techniques have been proposed to measure the free radical scavenging capacity (RSC), regardless of the individual compounds which contribute to the total free RSC. On the other hand, high-performance thin-layer chromatography (HPTLC) hyphenated with biochemical detection allows identification of single compounds responsible for radical scavenging activity (RSA). The main aim of current study is to identify single radical scavengers using four HPTLC-based assays such as 2,2-Diphenyl-1picrylhydrazyl assay (DPPH), 2,2’-Azino-bis(3-ethylbenzothiazoline-6-sulfonic acid) assay (ABTS), potassium hexacyanoferrate(III) total reducing power assay (TRP) and total antioxidant capacity by phosphomolybdenum assay (TAC) in combination with principal component analysis (PCA). The best of our knowledge, HPTLC-TRP and HPTLC-TAC were developed for the first time. PCA was used for the classification of samples with similar RSA and the identification of radical scavengers responsible for classification. HPTLC-DPPH, HPTLC-ABTS and HPTLC-TRP chromatograms showed similar bioactive profiles. Further, HPTLCTAC fingerprint differs from the others with observed numerous new zones with hRF at 73, 83, 89, and 98 values. Based on PCA, HPTLC-ABTS and HPTLC-TRP assays showed similar grouping of samples. Phenolic compounds such as gallic acid (hRF = 48), kaempferol (hRF = 69) and unknown compound at hRF = 18 are recognized as markers responsible for the separation between samples.

AB - Excess of free radicals in skin causes oxidative stress, which results to skin aging and
appearance of age spots. The enzyme tyrosinase is responsible for mentioned process.
Slowing down of this unwanted process is one of the challenges of modern science. Thus,
the aim of this study is to examine the potential of selected plant extracts to reduce
oxidation stress and the appearance of hyperpigmentation. The extracts were characterized
by the use of high-performance thin-layer chromatography and high-performance liquid
chromatography coupled with mass spectrometry. The antioxidant activity of the extracts
and potential for the inhibition of tyrosinase enzyme were determined. Nineteen phenolic
compounds were quantificated in the tested extracts. The obtained results demonstrate that
the extracts of Morus alba (92%), Rubus (86%) and Agrimona eupatoria (84%) are the
most effective inhibitors. The Teucrium chamaedrys and Agrimona eupatoria showed high
values of antioxidant activity. The listed samples are commercial.
