AB  - As byproducts of essential oil distillation, hydrolates are used in natural cosmetics/biomedicine due to their beneficial skin effects. However, data on their safety with relevant biological targets, such as human skin cells, are scarce. Therefore, we have tested nine hydrolates from the Lamiaceae family with skin fibroblasts that are responsible for extracellular collagenous matrix builds. Thyme, oregano, and winter savoury hydrolates showed several times higher total phenolics, which correlated strongly with their radical scavenging and antioxidative capacity; there was no correlation between their viability profiles and the reducing sugar levels. No proteins/peptides were detected. All hydrolates appeared safe for prolonged skin exposure except for 10-fold diluted lavender, which showed cytotoxicity (~20%), as well as rosemary and lavandin (~10%) using viability, DNA synthesis, and cell count testing. Clary sage, oregano, lemon balm, and thyme hydrolates (10-fold diluted) increased fibroblast viability and/or proliferation by 10–30% compared with the control, while their viability remained unaffected by Mentha and winter savoury. In line with the STITCH database, increased viability could be attributed to thymol presence in oregano and thyme hydrolates in lemon balm, which is most likely attributable to neral and geranial. The proliferative effect of clary sage could be supported by alpha-terpineol, not linalool. The major volatile organic compounds (VOCs) associated with cytotoxic effects on fibroblasts were borneol, 1,8-cineole, and terpinene-4-ol. Further research with pure compounds is warranted to confirm the roles of VOCs in the observed effects that are relevant to cosmetic and wound healing aspects.

AB  - AbstractBackground and objectiveCotinus coggygria Scop. is a valuable medicinal plant species with pronounced pharmacological potential due to its numerous biological activities. The herb is characterized by a high content of polyphenols among which is fustin. The anticancer activities of fustin, however, are extremely weakly studied. The aim of the present study was to investigate the in vitro antiproliferative potential of fustin isolated from the heartwood of C. coggygria against cell lines originating from two of the most common cancer types – breast (MDA-MB-231 and MCF7), and colon cancer (Colon 26).Materials and methodsCell growth inhibitory properties of fustin were examined by MTT assay. Subsequently, phase-contrast and fluorescence microscopy analysis as well as colonyforming assay were carried out on the most sensitive to the cytostatic action of the fustin cell line.ResultsThe obtained results showed that fustin reduced the proliferation of all studied cell lines. The highest cytostatic effect was registered towards breast cancer MDA-MB-231 cells with a half maximal inhibitory concentration (IC50) value of 56.02 μg/ml followed by colon cancer cells with an IC50 of 78.07 μg/ml. MCF7 cell proliferation was least affected with a calculated IC50 of 187.8 μg/ml. Further investigations on breast cancer MDA-MB-231 cells indicated decreased density of cell

AB  - IntroductionOregano essential oil (OEO) is one of the most widely used essential oils worldwide due to its huge therapeutic benefits. Nevertheless, data on the effects of the endemic species Origanum minutiflorum, also known as wild or Turkish oregano, is scarce. On the other hand, various chronic lung diseases, characterised by persistent inflammation, oxidative stress, and common bacterial infections, do not have effective pharmacological therapy. Hence, the aim of this study is to examine the effects of wild oregano essential oil (WOEO) on human bronchial epithelial cells and lung pathogens.MethodsWe provided a detailed chemical composition of WOEO using GC-MS and GC-FID analysis. Anti-inflammatory effects of WOEO were analysed using the qRT-PCR and ELISA methods, while antioxidative properties were examined by using the dichlorofluorescein assay in BEAS-2B cells. Antibacterial activity was tested on lung pathogens by using the agar diffusion assay.ResultsThe major constituents of WOEO, analysed in this study, were carvacrol, linalool, p-cymene, γ-terpinene, and (E)-caryophyllene. We found that treatment with WOEO attenuated LPS-induced IL8 gene expression and hydrogen peroxide-induced oxidative stress in BEAS-2B cells. Moreover, WOEO showed an inhibitory effect on pathogenic bacteria Acinetobacter baumannii and highly resistant Klebsiella pneumoniae commonly seen in healthcare-associated pneumonia.ConclusionOur work presents new insights into the anti-inflammatory, antioxidative, and antimicrobial properties of WOEO which may be used as a simple and local treatment in various chronic lung diseases.

AB  - This study aimed to show aromatic profile of wines produced from two autochthonous grape cultivars Krstač (K) and Žižak (Z). During the wine production two enzymatic preparations (EP) Lallzyme cuvee blanc (CB) and Lallzyme enzymatic preparation β (EB) and different time of skin contact (4 and 8 h) were applied. Aromatic compounds were detected by GC/FID–MS ana­lysis. Significantly higher content of total detected aromatic compounds com­pared to appropriate controls (168.54 and 161.72 mg L-1) was observed for K EB4h (176.33 mg L-1) and Z CB4h (177.29 mg L-1) wines. Skin contact and usage of EP mostly increased content of 2-phenylethyl and isoamyl alcohols. Wines from both varieties showed higher content of hexanoic and octanoic acids compared to the control. It is interesting to emphasize that content of esters that are responsible for fruity aroma of wine which is important for plea­sant taste (isoamyl acetate – banana, ethyl hexanoate – ripe banana, 2-phe­nyl­ethyl acetate – powerful fruity rose like) were increased in all samples com­pared to the controls. The highest grades, after sensory evaluation, were obtained for K EB 8h (18.0 out of 20.0) and Z CB 8h (18.2 out of 20.0).

AB  - Chemotaxonomy presents various challenges that need to be overcome in order to obtain valid and reliable results. Individual genetic and environmental variations can give a false picture and lead to wrong conclusions. Applying a holistic approach, based on multivariate data analysis, these challenges can be overcome. Thus, a metabolomics approach has to be optimized depending on the subject of research. We used 1H NMR-based metabolomics as a potential chemotaxonomic tool on the selected Euphorbia species growing wild in Serbia. Principal components analysis (PCA), soft independent modeling by class analogy (SIMCA) and Orthogonal Projections to Latent Structures Discriminant Analysis (OPLS-DA) were used to analyze obtained NMR data in order to reveal chemotaxonomic biomarkers. The standard protocol for plant metabolomics was optimized aiming to extract more specific metabolites, which are characteristic for the Euphorbia genus. The obtained models were validated, which revealed that variables unique for each species were associated with certain classes of molecules according to literature data. In E. salicifolia, acacetin-7-O-glycoside (not found before in the species) was detected, and the structure of the aglycone part was solved based on 2D NMR data. In the presented paper, we have shown that metabolomics can be successfully used in Euphorbia chemotaxonomy.

The state of the art indicates gaps in the understanding and diagnosis of this disease, but also the need for personalized and precise approaches to patients through customized medical treatment and reliable monitoring of treatment response. In order to fulfill existing gaps, the establishment of a universal set of disorder biomarkers is a necessary step. Metabolomic investigations of serum samples of Serbian patients with schizophrenia (51) and healthy controls (39), based on NMR analyses associated with chemometrics, led to the identification of 26 metabolites/biomarkers for this disorder. Principal component analysis (PCA) and orthogonal partial least squares discriminant analysis (OPLS-DA) models with prediction accuracies of 0.9718 and higher were accomplished during chemometric analysis. The established biomarker set includes aspartate/aspartic acid, lysine, 2-hydroxybutyric acid, and acylglycerols, which are identified for the first time in schizophrenia serum samples by NMR experiments. The other 22 identified metabolites in the Serbian samples are in accordance with the previously established NMR-based serum biomarker sets of Brazilian and/or Chinese patient samples. Thirteen metabolites (lactate/lactic acid, threonine, leucine, isoleucine, valine, glutamine, asparagine, alanine, gamma-aminobutyric acid, choline, glucose, glycine and tyrosine) that are common for three different ethnic and geographic origins (Serbia, Brazil and China) could be a good start point for the setup of a universal NMR serum biomarker set for schizophrenia. © 2022 by the authors.

AB  - Quince and sweet cherry are two common edible fruit-producing trees of temperate regions of Euroasia. The phenolic profiles of juices from quince and sweet cherry fruit extracted by the cold-pressing were analyzed by Ultra-High-Performance Liquid Chromatography (UHPLC). Chromatographical results identified 31 phenolic compounds while quantified thirteen for fruit juices. The quantitative data indicate that the predominant phenolic was 5-O-caffeoylquinic acid in quince (8.343 mg L-1) and cherry juice (6.407 mg L-1). Total Flavonoids (TF), Total Phenolic Content (TPC), and Total Antioxidant Capacity (TAC) were also investigated. In both fruit juices, the TF values varied between 0.30-0.97 g L-1 (Catechin Equivalent) and TPC between 0.36 to 0.94 g L-1 (Gallic Acid Equivalent). It is worth noting that the quince fruit juices possessed a higher TPC and TFC compared to sweet cherry fruit juices.

Bioactivity-guided identification and isolation of a major antimicrobial compound in Cistus creticus subsp. creticus leaves and resin “ladano”

AB  - Cistus creticus subsp. creticus is a shrubby Mediterranean plant used since ancient times in folk medicine for the treatment of different diseases. C. creticus extracts and resin contain different types of secondary metabolites, such as terpenoids (predominantly labdane type diterpenoids), and phenylpropanoids. Growth conditions seem to influence the content of labdane-type diterpenes and flavan-3-ols in leaves of C. creticus subsp. creticus. Histochemical staining of leaves’ trichomes and comprehensive phytochemical characterization of resin, leaves and their exudates, indicated that long-stalked capitate trichomes of C. creticus subsp. creticus, grown both in vitro (IV) and in greenhouse (GH), are capable of producing bioactive oleoresin-related terpenoids and phenylpropanoids compounds. Bioactivity-guided approach was implemented in search for the major antibacterial compound in C. creticus resin against two Gram-negative (Escherichia coli and Pseudomonas aeruginosa) and two Gram-positive bacteria (Bacillus cereus and Micrococcus flavus). Bioautographic assay on TLC plates with separated components of Cistus resin extract, revealed a pronounced zone of microbial growth inhibition, corresponded to a highly active compound with Rf values of 0.45, structurally characterized and identified as ent-3β-acetoxy-13-epi-manoyl oxide. This finding opens the route for focusing on isolation and functional characterization of genes involved in the biosynthesis of ent-3β-acetoxy-13-epi-manoyl oxide and its precursor ent-3β-hydroxy-13-epi-manoyl oxide, with the aim to establish sustainable in vitro biotechnological protocols for its large-scale production in homologous and heterologous plant and microbial hosts.

AB  - Lavandula sp. essential oil and hydrolate are commercially valuable in various industry branches with the potential for wide-ranging applications. This study aimed to evaluate the quality of these products obtained from L. x intermedia cv. ‘Budrovka’ for the first time cultivated on Fruška Gora Mt. (Serbia) during three successive seasons (2019, 2020, and 2021). Essential oil extraction was obtained by steam distillation, and the composition and influence of weather conditions were also assessed, using flowering tops. The obtained essential oils and hydrolates were analysed by gas chromatography with a flame ionization detector (GC-FID) and gas chromatography coupled to mass spectrometry (GC-MS). A linear regression model was developed to predict L. x intermedia cv. ‘Budrovka’ essential oil volatile compound content and hydrolate composition during three years, according to temperature and precipitation data, and the appropriate regression coefficients were calculated, while the correlation analysis was employed to analyse the correlations in hydrolate and essential oil compounds. To completely describe the structure of the research data that would present a better insight into the similarities and differences among the diverse L. x intermedia cv. ‘Budrovka’ samples, the PCA was used. The most dominant in L. intermedia cv. ‘Budrovka’ essential oil and hydrolate were oxygenated monoterpenes: linalool, 1,8-cineole, borneol, linalyl acetate, and terpinene-4-ol. It is established that the temperature was positively correlated with all essential oil and hydrolate compounds. The precipitations were positively correlated with the main compounds (linalool, 1,8-cineole, and borneol), while the other compounds’ content negatively correlated to precipitation. The results indicated that Fruška Gora Mt. has suitable agro-ecological requirements for cultivating Lavandula sp. and providing satisfactory essential oil and hydrolate. © 2022 by the authors. and corresponding hydrolate obtained from S. officinalis grown in Serbia, as well as the influence
of weather conditions (temperature and precipitations) on their chemical profiles. Furthermore,
their antimicrobial activity was investigated in vitro. The main compounds in essential oil were cisthujone, followed by camphor, trans-thujone, and 1,8-cineole, while hydrolate was slightly different
from the essential oil, with camphor, cis-thujone, and 1,8-cineole as the main compounds. Among
the eight respiratory-associated microorganisms, Klebsiella oxytoca was the most sensitive to the
tested EOs (minimum inhibitory concentration (MIC)/minimal bactericidal/fungicidal concentration
(MBC/MFC) were 14.20 and 28.4 µL mL−1
, respectively). MIC and MBC values of other tested
bacteria ranged between 28.40 and 227.25 µL mL−1 while for Candida albicans MIC/MFC ranged
from 28.40/56.81 to 56.81–113.63 µL mL−1
. Antibiotic susceptibility patterns for the analyzed eight
respiratory-associated microorganisms showed an intermediate level of resistance to commonly used
antibiotics such as ampicillin, levofloxacin, and ciprofloxacin. As a preliminary approach to the
antimicrobial profiling of the tested EO, the obtained results revealed that the tested samples possess
remarkable antibacterial activities and could be used to develop pharmaceutical formulations as an
alternative to conventional antibiotic therapy

Although it has been traditionally used in Serbia in traditional medicine for centuries as an insecticidal, antibacterial, and antifungal plant as well as a traditional culinary plant, none of the O. basilicum varieties have been commercialised until today. There are significant numbers of information across the world that oils and by-products are part of the global market, but no references to the essential oil composition of Serbian plants were found. Therefore, the objective of this work was to evaluate the antioxidant and antimicrobial potentials of essential oil and hydrolate of two different varieties: O. basilicum var. genovese and Ocimum. basilicum var. minimum originating from Serbia for further industrial production of antimicrobial- and/or antioxidant-valued products. The results of this study confirm that essential oils of O. basilicum var. genovese and var. minimum represent a significant source of bioactive compounds, especially linalool, with a high rate of biological activities. Similar behaviour is observed for hydrolates, which are the by-product of the essential oil distillation process and can be utilised as bioactive-rich waste in further investigation.

AB  - We examined essential oils (EOs) of Chamaecyparis lawsoniana and Thuja plicata (Cupressaceae), their antifeedant activity on Lymanthria dispar larvae and their antimicrobial activity. Studies of EOs showed that these two conifer species differed both in content of terpene classes and the major compounds. Total monoterpenes strongly dominated in T. plicata (96.4%), while C. lawsoniana was rich in both mono- and sesquiterpenes (40.8% and 30.3%, respectively) as well as in diterpenes (19.1%). The most dominant compounds of C. lawsoniana EO were: limonene (16.7%), oplopanonyl acetate (14.5%), beyerene (10.1%), and 13-epi-dolabradiene (6.7%). The dominant compound of T. plicata EOs was α-thujone (76.9%), followed by relatively small amounts of β- thujone (5.3%), sabinene (4.5%) and terpinene-4-ol (3.2%). The difference in EO compositions of the conifers was reflected on Lymantria dispar larvae performance. Larvae fed on the leaf discs treated by C. lawsoniana EO had a slight phagostimulatory effect at lower concentration shown by higher relative rate of food consumption and relative growth rate than the larvae in the control group. Contrastingly, leaf discs treated with EO of T. plicata EO had an antifeedant effect and lower relative consumption rate (RCR) and relative growth rate RGR than the larvae in the control group. Both tested EOs influenced substantially the colony growth of the subjected Phytophthora plurivora and P. quercina. Namely, 100% inhibitory effect was recorded at concentration of 0.1% in the case of C. lawsoniana EO, whereas of T. plicata colonies did not grow at 0.5% concentration. The implications of these findings and possibility of using the tested EOs in further experiments in vitro and in vivo are discussed.

In six Pinus taxa (P. mugo, P. nigra ssp. dalmatica, P. sylvestris, P. pinea, P. strobus and P. wallichiana) carbon preference index and average chain length values were examined for the first time. In the plane of Axis 1 (PCA), P. strobus, P. mugo and P. wallichiana were strongly separated from P. heldreichii and P. pinea. In the plane of Axis 2, P. peuce, P. strobus and P. wallichiana as well as P. heldreichii and P. pinea diverged from other examined species. In DA, P. heldreichii, P. strobus, P. peuce and P. wallichiana diversed. CA divided sections Pinus and Pinaster from the section Strobi.

AB  - Nuts and seeds are an indispensable part of the plant-based diet, which is becoming increasingly popular due to the evidence of their health benefits and contribution to sustainability and planetary health. Since the health effects of consuming nuts and seeds directly depend on their nutritional composition and consumed amount, it is essential to know the exact chemical composition of each nut and seed so that appropriate dietary interventions can be adequately planned. The present study aimed to examine the chemical composition of nuts and seeds obtained from the Serbian market and to highlight the similarities and differences in their nutritional composition. In twenty-four samples of nuts and seeds, the content of total lipids, including fatty acid profile, total proteins, including amino acid profile, total carbohydrates (sugars and fibers), phytosterols, and minerals were determined. Content of selected nutrients in grams (g) or milligrams (mg) of nuts and seeds was expressed in one portion (28 g) and as a percentage of contribution of Reference Intakes (% RI) and Dietary Reference Value (% DRV) for macronutrients and minerals, respectively. Some of the seed representatives appeared to be rich sources of essential omega-3 fatty acid, α-linolenic acid (ALA) (flax seed vs. walnut, 6.50 vs. 0.56 g per portion, respectively), dietary fibers (chia seed vs. raw almond, 10.6 vs. 3.4 g per portion, respectively), calcium (black sesame seed vs. almond roasted, 32.4% NRV vs. 8.1% NRV per portion, respectively), magnesium (hemp seed vs. Brazil nut, 38.3% NRV vs. 27.8% NRV per portion, respectively), and zinc (hemp seed vs. pine nut, 21.4% NRV vs. 17.6% NRV per portion, respectively). Our results highlighted the crucial role of seeds in the diet, especially as a better source of nutrients compared to nuts. Furthermore, it was seen that nuts and seeds are different and complementary in their composition. Thus, in order to meet the needs for certain nutrients for which nuts and seeds are used as dietary sources (essential fatty acids, minerals, dietary fibers), it would be beneficial to combine both nuts and seeds as a part of a healthy dietary pattern.

AB  - Bioactive compounds extracted from plants such as antimicrobials have attracted the attention of consumers and the food industry. This study aimed to determine the antimicrobial activity and chemical composition of Annona muricata leaf oleoresin obtained by supercritical CO2 extraction. The oleoresin was obtained by supercritical CO2 extraction and the chemical identification by gas chromatography coupled to mass spectrometry. Antimicrobial activity was evaluated by broth microdilution method against 14 foodborne fungi and bacteria. The oleoresin major chemical class was phytosterols (22.7%) and the major compounds were γ-sitosterol (15.7%), α-tocopherol (13.7%), phytol (13.1%), and hexadecanoic acid (11.5%). Minimum inhibitory concentration against bacteria ranged from 0.0025 to 0.010 mg mL−1. The oleoresin had high bactericidal activity against all bacteria, mainly Enterobacter cloacae and Pseudomonas aeruginosa with 0.005 mg mL−1 minimum bactericidal concentration. However, it had low fungicidal activity. The leaf oleoresin of A. muricata has promising applications in food, cosmetic, and pharmaceutical industries.

AB  - In continuation of our study on Cotinus coggygria from Serbia, 10 known flavonoids (1-10) were isolated from the methylene chloride/ methanol extract of the heartwood. They were tested for in vitro protective effect against chromosome aberrations in peripheral human lymphocytes, using the cytokinesis-block micronucleus assay. All tested compounds (in minimal doses of 1 μg/mL) exerted a beneficial effect by decreasing DNA damage of human lymphocytes in the range of 24.2% to 54.5%, better than the radio protectant control, amifostine. Functional groups, such as 3′,4′-dihydroxyphenyl (catechol), 5-OH, 3-OH, and 4-keto in flavonoids (3-keto in aurones), which play a key role in antioxidant activity, are proposed to be responsible for the DNA protective activity of the tested compounds.

AB  - Tumor is one of the leading diseases of today. Chemotherapy and radiation therapy healing chances are limited, so some alternative methods are resorted to. Many papers indicated that garlic and organosulphur compounds diallyldisulphide (DADS) and diallyltrisulphide, which are main components of garlic decrease the cancer risk and inhibit the cell proliferation. In this paper inhibition potency of disulphides (DADS and its synthetic analogues) and trisulphides with different alkyl and phenyl substituents on growth of various tumor (Non-Small cell Lung Cancer, Colon cancer, CNS Cancer, Melanoma, Ovarian Cancer, Renal Cancer, Prostate Cancer, Breast Cancer, Leukemia) cell lines was investigated. Concentration of each compound (DADS, diethyl disulphide, dipropyl disulphide, diphenyl disulphide, tetraethylthiuram disulphide (TETUDS), dimetil trisulphide, dipropyl trisulphide and metilpropyl trisulphide (MPTS) was 10 μmol/L. It was found that DADS has an inhibitory effect on the growth of several cancer cell lines, but synthetic analogues TETUDS and MPTS exhibit stronger effect on certain cells lines. MPTS inhibits the growth of even 12 cell lines for more than 10%, especially leukemia cell line SR and NCI-H522 cell line of Non-Small cell Lung Cancer (reduction of growth for 24% and 47%, resp.). Trisulphide analogues exhibit little higher inhibitory effects in comparison to disulphide ones.

AB  - Plum brandy (Slivovitz (en); Šljivovica(sr)) is an alcoholic beverage that is increasingly consumed all over the world. Its quality assessment has become of great importance. In our study, the main volatiles and aroma compounds of 108 non-aged plum brandies originating from three plum cultivars, and fermented using different conditions, were investigated. The chemical profiles obtained after two-step GC-FID-MS analysis were subjected to multivariate data analysis to reveal the peculiarity in different cultivars and fermentation process. Correlation of plum brandy chemical composition with its sensory characteristics obtained by expert commission was also performed. The utilization of PCA and OPLS-DA multivariate analysis methods on GC-FID-MS, enabled discrimination of brandy samples based on differences in plum varieties, pH of plum mash, and addition of selected yeast or enzymes during fermentation. The correlation of brandy GC-FID-MS profiles with their sensory properties was achieved by OPLS multivariate analysis. Proposed workflow confirmed the potential of GC-FID-MS in combination with multivariate data analysis that can be applied to assess the plum brandy quality.

AB  - Eleven 31-methylenlanostane triterpenoids, i.e., seven 21- and four 26-oic acids, as well as a lupane triterpenoid betulin, isolated from the fruiting bodies of the mushroom Fomitopsis betulina, were tested for in vitro protective effect on chromosome aberrations in peripheral human lymphocytes using cytochalasin-B blocked micronucleus (CBMN) assay. Most of the tested compounds showed a beneficial effect by reducing DNA damage of human lymphocytes more effectively than amifostine, a radioprotective agent, used as a positive control. All the tested compounds decreased MN frequency in the concentration dependent manner, with the concentration of 2.0 µg mL-1 being the most effective – with increase of the concentration the activity slightly decreases. The structure–activity relationship (SAR) studies indicated that the lanostanes containing a conjugated 7,9 (11)-diene system exhibit lower activity than Δ8-analogues. It was also demonstrated that the DNA protective activities within the Δ8-lanostane-26-oic acid group are affected by the substitution in position 3 pattern. In the Δ8 series the oxygenation at C-12 or 16 as well as 21- or 26-oic acid functionality proved beneficial for in vitro protective effect on chromosomal aberrations. Betulin exhibited the lowest protective activity, but it is still comparable to that of amifostine.

AB  - Thirteen undescribed 24-methylene lanostane triterpenoids, named polyporenic acids E-M and fomitosides L-O, as well as seventeen known analogues, were isolated from the fruiting bodies of the mushroom Fomitopsis betulina. Their structures were determined using 1D, 2D NMR, IR, and HRESIMS. Fomitoside L and fomitoside N exhibited cytotoxicity against HL60 leukemia cells (IC50 = 15.8 and 23.7 μM, respectively).
