Derivatives of L-Ascorbic Acid in Emulgel: Development and Comprehensive Evaluation of the Topical Delivery System

AB  - The dual controlled release of emulgels makes them efficient drug delivery systems of increasing interest. The framework of this study was to incorporate selected L-ascorbic acid derivatives into emulgels. From the formulated emulgels, the release profiles of actives were evaluated considering their different polarities and concentrations, and consequently their effectiveness on the skin via a long-term in vivo study that lasted for 30 days was determined. Skin effects were assessed by measuring the electrical capacitance of the stratum corneum (EC), trans-epidermal water loss (TEWL), melanin index (MI) and skin pH. In addition, the sensory and textural properties of emulgel formulations were compared with each other. The changes in the rate of the release of the L-ascorbic acid derivatives were monitored using the Franz diffusion cells. The obtained data were statistically significant, and indicated an increase in the degree of hydration of the skin and skin whitening potential, while no significant changes in TEWL and pH values were detected. The consistency, firmness and stickiness of the emulgels were estimated by volunteers applying the established sensory evaluation protocol. In addition, it was revealed that the difference in hydrophilic/lipophilic properties of L-ascorbic acid derivatives influenced their release profiles without changing their textural characteristics. Therefore, this study highlighted emulgels as L-ascorbic acid suitable carrier systems and one of the promising candidates as novel drug delivery systems.

Honey with added value – enriched with rutin and quercetin from Sophora flower

AB  - IntroductionThe aim of this study was to find the best conditions for the extraction of rutin and quercetin from Sophora flower in acacia honey as an extractant, and to formulate a new product – honey with added value.MethodsFor determination of the optimal extraction conditions of the above mentioned compounds, 10% m/m of the dried Sophora flower was added to honey and the concentration of rutin and quercetin was monitored, at 4 °C and room temperature at 1, 15, 40 and 90 day intervals. The analyses of rutin and quercetin content in honey were performed by high-performance liquid chromatography (HPLC) with ultraviolet (UV) detection. The content of rutin and quercetin in the flower was also determined to check the quality of the herbal drug used, as well as to evaluate the effectiveness of the extraction.ResultsThe extraction of rutin and quercetin, in a honey sample using 10% of the Sophora flower, stored at room temperature, for 40 days, was the most intense one (150.24 mg/kg of rutin and 1338.93 mg/kg of quercetin). Sophora flower corresponds to the pharmacopoeia requirements for the quality of an herbal drug. The concentrations of rutin and quercetin in acacia honey were 0.08 and 0.14 mg/kg, respectively. The concentration of rutin in honey from the melliferous pasture of the Sophora flower was 0.40 mg/kg, while quercetin was below the detection limit (0.1 mg/kg).ConclusionsHoney, enriched with rutin and quercetin from Sophora flower, had sensory acceptability and pleasant taste and aroma.

Stabilization of rose hip oil by pomegranate peel extract during accelerated storage

AB  - Rose hip seed oil is a rich source of polyunsaturated fatty acids, as well as tocopherols, carotenoids, sterols, phospholipids, and phenolic com­pounds. On the other hand, due to the high content of polyunsaturated fatty acids, this oil is prone to oxidation. The aim of this study was to investigate the influence of a natural antioxidant, i.e., pomegranate peel extract, and its com­bination with butylated hydroxytoluene as a commonly used synthetic anti­oxidant, on the stability of rose hip oil. The stability of samples without and with different antioxidants was monitored through analysis of the fatty acid composition and measurement of the quality and stability parameters of the oil (peroxide value, p-anisidine value, thiobarbituric acid reactive substances inhi­bition, total phenolic content and antiradical activity) during a storage period of 12 days at 65 °C. Pomegranate peel extract (0.1 %) inhibited more effectively the second stage of oxidation than butylated hydroxytoluene (0.02 %), while the first stage of oxidation was better prevented by the synthetic antioxidant. Furthermore, the addition of pomegranate peel extract increased the total phen­olic content of the rose hip oil as well as its antiradical activity. Thus, pome­granate peel extract could be used as a potent natural antioxidant for the stabil­ization of beneficial but unstable rose hip oil.
