AB  - This study was focused on in vitro screening of the total antioxidant activity of the selected extracts of the mushroom Coprinus comatus and quinic acid, one of their antidiabetic ingredients, by an uncommon electrochemical assay. Indeed, direct current (DC) polarographic HydroxoPerhydroxo Mercury(II) Complex (HPMC) assay based on decrease of anodic limiting current originating from HPMC formation in alkaline solutions of hydrogen peroxide at potential of mercury dissolution, observed upon gradual addition of antioxidants, was applied herein for the estimation of the natural products' antioxidativity. Quinic acid was found to exhibit most promising antioxidant potential (4.0 ± 0.2%µL-1) being ≈ 2-fold more active than the screened C. comatus extract samples. Actually, such a finding puts some light on the antioxidativity of cyclic polyols, well understimated class of organic compounds, compared to aromatic (poly)phenolics. As a low cost, easy-to handle and accurate this polarographic assay may be thoroughly recommended for much broader use. [Formula: see text].

AB  - Trehalose and its precursor, trehalose 6-phosphate (T6P), are essential regulators of plant response to abiotic and biotic stress. Here we used the specific host-insect interaction between Linaria vulgaris (Plantaginaceae) and stem-galling weevil, Rhinusa pilosa (Mecinini, Curculionidae) with the aim to distinguish carbohydrate allocation patterns in response to herbivory, gall formation (G1, 24 h after oviposition), and gall development (G2, 7 days after oviposition) under controlled conditions. The hypothesis is that herbivory and galling induce distinct responses in both leaves and stems, and that shifts in carbon allocations are regulated by signaling sugars. Systemic response to herbivory was accumulation of T6P and maltose. The main feature of G1 in the stems was accumulation of trehalose, accompanied by increased T6P, turanose and glucose content, oppositely to the leaves. In G2, galls had 3-folds higher weight than controls, with further accumulation of fructose, glucose, turanose, and total water-insoluble carbohydrates (TIC), while the sucrose/hexose ratio decreased. Analysis of fast chlorophyll fluorescence kinetic (OJIP) transients in G2 showed a slight decrease in quantum yield of electron transport flux from QA to QB, and towards photosystem I acceptor side, correlated with the decreased content of photosynthetic pigments and hexoses accumulation. Redistribution of photosynthates, and accumulation of T6P were induced in response to herbivory, indicating its signaling role. The results support the hypothesis that R. pilosa can induce plant reprogramming towards the accumulation of beneficial carbohydrates in developing gall by mechanisms which include both T6P and trehalose.

AB  - Biliverdin (BV), a product of heme catabolism, is known to interact with transition metals, but the details of such interactions under physiological conditions are scarce. Herein, we examined coordinate/redox interactions of BV with Cu2+ in phosphate buffer at pH 7.4, using spectrophotometry, HESI-MS, Raman spectroscopy, 1 H NMR, EPR, fluorimetry, and electrochemical methods. BV formed a stable coordination complex with copper in 1 : 1 stoichiometry. The structure of BV was more planar and energetically stable in the complex. The complex showed strong paramagnetic effects that were attributed to an unpaired delocalized e−. The delocalized electron may come from BV or Cu2+, so the complex is formally composed either of BV radical cation and Cu1+ or of BV radical anion and Cu3+. The complex underwent oxidation only in the presence of both O2 and an excess of Cu2+, or a strong oxidizing agent, and it was resistant to reducing agents. The biological effects of the stable BV metallocomplex containing a delocalized unpaired electron should be further examined, and may provide an answer to the long-standing question of high energy investment in the catabolism of BV, which represents a relatively harmless molecule per se.

AB  - Locally available apricot (Prunus armeniaca L.) shells classified as a waste product from fruit processing, were alkali activated in order to develop an efficient heavy metal ions sorbent for water purification. To examine the changes occurred after alkali treatment, raw (SH) and modified apricot shells (SHM) were thoroughly characterized in terms of their chemical composition and surface properties. Chemical analysis revealed that alkaline treatment causes the disintegration of hemicellulose (its content decreased from 19.2 to 3.5%), which was in accordance with FTIR results. SEM micrographs and the mercury intrusion porosimetry revealed a larger surface area and porosity of SHM. Bohem's acid-base titration method indicated that the most of the SHM surface carboxylic groups were in sodium salt form and together with the pH of points of zero charge showed increase of surface alkalinity after modification. Treatment with NaOH enhanced the adsorption capacity by 154, 61 and 90% for Cu2+, Zn2+ and Pb2+, respectively. The amount of cations released from SHM was almost equal to the amount of adsorbed metal ions, suggesting ion exchange mechanism. The pseudo-second order kinetic indicated that the heavy metals cations were bound predominantly by complexation. In order to establish the effectiveness of the biosorbent in real wastewater sample, SHM was employed for cleaning-up of drain water emanating from atomic adsorption spectrophotometer. The SHM showed high removal efficiency towards multiple metal ions. The amounts of Fe, Pb, Cu and Cr ions were reduced by 97, 87, 81 and 80%, respectively, while Ni and Zn amounts were reduced for 33 and 14%. Used biosorbent SHM can be successfully regenerated with HCl (desorption  gt  95%) and after regeneration biosorbent can be reused or it can be safely disposed. (C) 2018 Elsevier B.V. AB  - In this thesis antioxidative activities (AA) of free L-amino acids (glycine, alanine, proline, valine, isoleucine, leucine, phenylalanine, tryptophane, tyrosine, serine, threonine, asparagine, glutamine, histidine, aspartic acid, glutamic acid, lysine, arginine, cystein and methionine, homocysteine, hydroxyproline, norleucine, α-aminobutyric acid and 3,4-dihydroxyphenylalanine) against Fenton system-mediated hydroxyl radical (HO•) production in aqueous solution were studied. The rank order according to AA was: Trp > norleucine > Phe, Leu > Ile > His >3,4-dihydroxyphenylalanine, Arg > Val > Lys, Tyr, Pro > hydroxyproline> α-aminobutyric acid > Gln, Thr, Ser > Glu, Ala, Gly, Asn, Asp. AB  - Antioxidant (AO) capacity of instant, espresso, filter and Turkish/Greek coffee brews, coffee substitutes (roasted chicory root, barley, pea, chickpea, carob and dried fig) and individual compounds (phenolic acids, flavonoids, methylxanthines, N-methyl pyridinium and HMW melanoidins) was assessed using DC polarographic assay based on decrease of anodic current originating from hydroxo-perhydroxo mercury complex formed in alkaline solutions of H2O2 at potential of mercury dissolution, as well as three spectrophotometric assays (DPPH, ABTS and FRAP). A large difference between applied assays ability to recognize various types of individual AOs was noticed. Only according to DC polarographic assay significant AO activity was ascribed to methylxanthines and N-methyl pyridinum. The total content of phenolics (TPC) present in complex samples was determined by FC assay. The highest TPC was ascribed to instant coffees and coffee substitutes while the lowest to decaffeinated filter coffee. Complex samples were grouped based on principal components analysis, phenolics AO coefficient, calculated as the ratio between AO capacity and TPC, and relative AO capacity index (RACI), calculated by assigning equal weight to all applied assays including FC. The highest values of RACI were ascribed to instant coffee brews, followed by substitutes while the lowest to the decaffeinated espresso coffee.

AB  - Gljive iz roda Trichoderma spp. su kosmopolitske filamentozne gljive koje se pojavljuju kao dominantne komponente zemljišnog ekosistema. Ekološka i biološka uloga ovih gljiva je veoma raznovrsna. Kao endofitni simbionti biljaka imaju primenu u kontroli biljnih bolesti čime pospešuju biljni rast i prinos. Takođe, poznata je i njihova sposobnost da indukuju otpornost kod biljaka koje su izložene biotičkom ili abiotičkom stresu. Pored važne primene u poljoprivredi one nalaze primenu i u različitim granama industrije jer proizvode mnogobrojne enzime i metabolite. Obzirom da su mnoge vrste u okviru ovog roda ne mogu razlikovati na morfološkom nivou, neophodna je njihova identifikacja na molekularnom nivou.Cilj ove teze bio je utvrđivanje najvažnijih faktora koji utiču na varijabilnost roda Trichoderma u različitim tipovima zemljišta. Molekularna identifikacija gljiva roda Trichoderma, izolovanih iz različitih tipova zemljišta (rendzina, sirozem na lesu, ritska crnica, fluvisol, ranker, gajnjača, černozem sa različitim podtipovima i smonica) i iz rizosfere različitih biljnih kultura urađena je analizom sekvenci ITS i TEF1-α gena. Uzorci zemljišta ispitani su u funkciji fizičko-hemijskih i mikrobioloških karakteristika, kao i sadržaja ukupnih i lakopristupačnih metala (Cr, Co, Ni, Cu, Zn, Cd, Pb i Mn). U cilju ispitivanja primene ovih gljiva kao antagonista fitopatogenih mikroorganizama, testirani su odabrani izolati prema Fusarium solani species complex (FSSC), Alternaria alternata i Phoma cucurbitacearum. Enzimska karakterizacija izolata odabranih za test antagonizma, urađena je semikvantitativnim testom API-ZYM. Kako bi se ispitao efekat ovih gljiva i na abiotski stres izazvan povećanim sadržajem metala u zemljištu, između ostalih, odabrani su izolati koji su identifikovani iz zemljišta u kojima je određena povećana koncentracija metala, bakra(II) i nikla(II). Na osnovu in vitro testa, gajenjem izolata na povećanim koncentracijama bakra(II) i nikla(II), odabran je izolat, koji je pokazao najveću tolerantnost i primenjen je u ispitivanju uticaja na biljke paradajza gajene u vodnom deficitu...
AB  - Fungi of the genus Trichoderma spp. are cosmopolitan species present in different ecosystems. with important ecological and biological roles.. Beneficial effects of this fungi are responsible for their use in various fields of biotechnology and agriculture. As plant endophytes they are involved in control of plant deseases. They are known to induce plant tolerance to various biotic and abiotic stresses. Known to produce different enzymes, they can be used in preparations of commercial products, such as biopesticides. However, for their adequate use and application their identification on the molecular level is of great importance.The aim of this thesis was to investigate the most important factors that influence Trichoderma variabilty in different soil types. Molecular identification of Trichoderma spp. from different soil types (Rendzic Leptosol, Regosol (Calcaric, Arenic), Calcic Gleysol, Fluvisol, Dystric Leptosol, Eutric Cambisol, Chernozem on loess terrace Chernozem (Arenic) and Vertisol) and plant rhizosphere of different crops was done based on their ITS and TEF1-α sequences. Variability was investigated in function of physical, chemical, microbiological properties of the soil, as well as total and extractable metal (Cr, Co, Ni, Cu, Zn, Cd, Pb and Mn) content in the soil. In order to examine the application of these fungi as antagonists of phytopathogens, selected strains of Trichoderma species were tested against Fusarium solani species complex (FSSC), Alternaria alternata and Phoma cucurbitacearum. The enzyme characterization of selected strains was performed with semiquantitative test API-ZYM. Furthermore, the selected strains were tested for tolerance to various concentrations of copper and nickel. The strain T. brevicompactum SZMC 22661 which showed the best tolerance to metals was applied in the examination of drought effect on tomato plants...

Content of Total Phenolics, Flavan-3-Ols and Proanthocyanidins, Oxidative Stability and Antioxidant Capacity of Chocolate During Storage

AB  - Antioxidant (AO) capacity of chocolates with 27, 44 and 75 % cocoa was assessed after production and during twelve months of storage by direct current (DC) polarographic assay, based on the decrease of anodic current caused by the formation of hydroxo-perhydroxyl mercury(II) complex (HPMC) in alkaline solutions of hydrogen peroxide at potentials of mercury oxidation, and two spectrophotometric assays. Relative antioxidant capacity index (RACI) was calculated by taking the average value of the AO assay (the sample mass in all assays was identical). Oxidative stability of chocolate fat was determined by differential scanning calorimetry. Measured parameters and RACI were correlated mutually and with the content of total phenols (Folin-Ciocalteu assay), flavan-3-ols (vanillin and p-dimethylaminocinnamaldehyde assay) and proanthocyanidins (modified Bate-Smith assay). During storage, the studied functional and health-related characteristics remained unchanged. Amongst applied AO assays, the DC polarographic one, whose validity was confirmed by two-way ANOVA and F-test, correlated most significantly with oxidative stability (oxidation onset temperature and induction time). In addition, principal component analysis was applied to characterise chocolate types.

Measured parameters and RACI were correlated mutually and with the content of total phenols (Folin-Ciocalteu assay), flavan-3-ols (vanillin and p-dimethylaminocinnamaldehyde assay) and proanthocyanidins (modified Bate-Smith assay). During storage, the studied functional and health-related characteristics remained unchanged. Amongst applied AO assays, the DC polarographic one, whose validity was confirmed by two-way ANOVA and F-test, correlated most significantly with oxidative stability (oxidation onset temperature and induction time). In addition, principal component analysis was applied to characterise chocolate types.",

In addition, two direct current (DC) polarographic assays, one based on a decrease of anodic current of [Hg(O2H)(OH)]-HydroxoPerhydroxoMercury(II) Complex (HPMC) formation in alkaline solution of H2O2, at the potential of mercury dissolution and another recently developed Mercury Reduction Antioxidant Power (MRAP), based on a a decrease of cathodic current of Hg(II) reduction were employed. Percentage of both currents decrease was plotted versus the volume of gradually added complex samples or the amount of individual ones and the slopes of these plots were used to express AO capacity. Total phenolic content (TPC) of extracts was determined by Folin-Ciocalteu (FC) assay. Correlations between applied assays were calculated by regression analysis. Relative Antioxidant Capacity Index (RACI), calculated by assigning equal weight to all applied assays and Phenolic Antioxidant Coefficients (PAC), calculated as a ratio between particular AO capacity and TPC, were used to achieve more comprehensive comparison between analyzed samples, as well as applied assays.

AB  - Rhinusa pilosa (Gyllenhal) is a highly specific weevil that induces stem galls on the common toadflax Linaria vulgaris Mill. females oviposit the eggs near the apex of a growing shoot. The act of oviposition is accompanied by secretion of an ovipositional fluid, which is considered to be cecidogen, directly involved in gall induction. The remains of cecidogenic fluid were collected from the surface of the oviposition point on the stem. We performed a comparative analysis of the phenolics extracted from cecidogen, the stem and galls of L. vulgaris and adult and larva of R. pilosa by HPLC-DAD. One compound with A (max) at 273, 332 nm (R (t) 30.65 min) was exclusively found in the methanol extract of cecidogen. To further characterize the cecidogen and stem phenolic profiles, we used UHPLC coupled with an OrbiTrap mass analyzer. Among 49 phenolic compounds extracted from both the ovipositional fluid and the plant, protocatechuic acid and two phenolic glycosides were exclusively found in cecidogen: diosmetin-O-acetylrutinoside and an unidentified compound. The unknown compound produced an MS2 base peak at 387 and 327 and 267 m/z base peaks at MS3 and MS4 fragmentation, respectively, and had the molecular formula C32H31O18. The plausible role of phenolic compounds in the induction of gall formation on L. vulgaris is discussed.

Here, influence of several common AOs on DCP cathodic currents of both HPMC and Hg2+ was followed. Amperometric titrations of HPMC and Hg2+ with AOs and potentiometric titrations of Hg2+ were performed. The changes of DCP i-E curves and limiting currents decrease during amperometric titrations, as well as end points of amperometric and potentiometric titrations for each AO used as titrant proved the occurrence of Hg2+ reduction, either from complex (HPMC) or salt (HgCl2) solution. Constant H2O2 reduction current during amperometric titration of HPMC further confirmed the participation of Hg2+ as the oxidant species in reaction with AOs. The possibility of determining AO activity of flavonoids, phenolic acids, ascorbic acid and Trolox based on the decrease of Hg2+ reduction current was demonstrated. Strict correlation (R-2 = 0.96) between a new AO assay named MRAP (Mercury Reduction Antioxidant Power) and HPMC assay was found. AB  - We studied the specific effects of high photosynthetically active radiation (PAR, 400-700 nm) and ecologically relevant UV-B radiation (0.90 W m(-2)) on antioxidative and phenolic metabolism by exploiting the green-white leaf variegation of Pelargonium zonale plants. This is a suitable model system for examining "source-sink" interactions within the same leaf. High PAR intensity (1350 mu mol m(-2) s(-1)) and UV-B radiation induced different responses in green and white leaf sectors. AB  - Antioxidant (AO) activity of extracts of hop cones (Serbian domestic varieties) and commercial hop products (Saaz, Spalter, Spalter select, and Magnum pellets) was determined by parallel application of recently developed direct current (DC) polarographic and widely used DPPH assay. Correlations between 2,2-diphenyl-1-picrylhydrazyl radical (DPPH) scavenging and total phenolics (TPC) determined by the Folin-Ciocalteu assay (FC) (0.99), and between H2O2 scavenging,: (HPS) and humulone content (H) determined by conductometric method (0.94), total resins (TR) (0.85), and hop storage index (HIS) (-0.90), were found statistically significant at p  lt  0.05 level while complete lack of HPS correlation with TPC and DPPH was observed. To obtain an insight into differences between results of AO assays applied, activity of individual compounds, prevalent hop phenolics, and bitter acids was determined. By far superior HPS activity of humulone was followed by catechin, quercetin, xanthohumol, lupulone, and rutin. In contrast, DPPH scavenging activity of phenolics (quercetin  gt  catechin  gt  rutin  gt  xantohumol) was found substantially higher than activity of bitter acids. According to ferric reducing antioxidant power (FRAP) and scavenging of 2,2-azino-bis-3-ethylbenzothiazoline-6-sulfonic acid (ABTS), higher AO activity was ascribed to phenolics, while almost neglecting humulone. Besides reliability, low cost, and an easy-to-handle procedure, an ability to recognize humulone as the major contributor of hop AO activity could allow DC polarographic assay to be applied in analysis of various hop-derived products.

AB  - Hydrogen peroxide scavenging (HPS) activity of honey of different floral sources and its constituents such as predominant honey flavonoids, phenolic acids, amino and organic acids, and carbohydrates have been assessed by direct current (DC) polarographic assay. The assay was based on decrease of anodic current of hydrogen peroxide complex, formed in alkaline solution, at the potential of mercury dissolution. High correlations between honey HPS activity, its total phenolic content (FC-GAE), antioxidant activity measured by four standard methods (DPPH, TEAC, FRAP and ORAC), and also the relative antioxidant capacity index, were obtained. Statistical evaluation by ANOVA and F-test further confirmed the assay validity. The results for individual compounds showed that HPS activity of honey reflects an integrated action of a wide range of constituents, both phenolics and non-phenolics. The polarographic assay applied is a fast, reliable and low cost alternative to spectrophotometric antioxidant assays commonly applied in analysis of honey and can serve as an indicator of honey quality.

Development of a peroxide anodic current having form of a peak, instead of common polarographic wave, has been investigated. As a base for this investigation the interaction of H(2)O(2) with anodically dissolved mercury was followed. Formation of mercury complex [Hg(O(2)H)(OH)] has been confirmed. The relevant experimental conditions, such as temperature, concentration and pH dependence, as well as time stability of hydrogen peroxide anodic current, have been assessed. Development of an AO assay based on decrease of anodic current of hydrogen peroxide in the presence of antioxidants (AOs) has been described. Under optimized working conditions, a series of benzoic acids along with corresponding cinnamate analogues have been tested for hydrogen peroxide scavenging activity. AB  - The behavior of chlorogenic acid (CGA) and its interaction with Hg(II), Pb(II), Cu(II), Cd(II) and Zn(II) has been studied using direct current (DC) and differential-pulse polarography (DPP). Adsorptive and kinetic/catalytic anodic waves of CGA have been detected at -0.02 and -0.18 V vs. SCE, respectively; Hg(I)-CGA surface disproportionation has been assumed. Reduction of Hg(II)-ions, added in the buffered solution of CGA (pH 7.5), occurs rather than complexation, while the complex forming ability of CGA towards Pb(II) and Cu(II) has been observed. Stoichiometry 1?:?1 for Pb(II)-CGA, and 1?:?1 and 1?:?2 for Cu(II)-CGA has been established applying Jobs method of continuous variation on DPP data. UV-vis spectrophotometric measurements additionally confirmed the existence and stoichiometry of Cu(II) complexes.

AB  - This study was conducted to investigate the influence of different drying treatments on antioxidant (AO) activity and phenolic content of raspberry (Rubus idaeus), cultivar Willamette. Whole raspberry fruits were dried convectively (air-drying), osmotically, and freeze-dried. Acetone-water extracts of fresh and dried raspberries were assessed for total phenolic content by standard Folin-Ciocalteau method. Two AO assays were applied, a recently developed direct current (DC) polarographic assay based on decrease of anodic oxidation current of hydrogen peroxide and widely used radical scavenge against the 1,1-diphenyl-2-picrylhydrazyl (DPPH). Strong correlation has been obtained between both AO assays and total phenolic content. In addition, some individual phenolic compounds present in raspberry have been assessed using DPPH and DC polarographic assay. Comparison and evaluation of drying methods has been based on preservation of AO activity and total phenolic content. Obtained results confirmed superiority of freeze-drying; convective drying caused slight changes while osmotic dehydration showed a significant decrease of phenolic compounds and AO activity. Practical Application Optimization of drying process could be based on surveying of AO activity.

AB  - The possibility of determination of chloramphenicol by differential pulse voltammetry at four different carbon paste electrodes, in the full pH range (2-12) of Britton-Robinson (BR) buffer was investigated. Electrodes were prepared by mixing spectroscopic graphite powder or glassy carbon microbeads with mineral oil (Nujol) or tricresyl phosphate. Under optimal conditions (BR buffer pH 12, the electrode prepared from glassy carbon microbeads and tricresyl phosphate), linear calibration graph was obtained only in 10-5 M chloramphenicol concentration range. Determination of lower concentrations of chloramphenicol was complicated by irreproducible peak of oxygen from the carbon paste which overlapped with peak of chloramphenicol. Addition of sodium sulfite removed the oxygen peak without influence on the peak of chloramphenicol. Under optimal conditions (electrode paste made from glassy carbon microbeads, BR buffer pH 10 and 0.5 M sodium sulfite), straight calibration line was obtained in the 10(-6) and 10(-5) M chloramphenicol concentration range. Limit of determination was 5 x 10(-7) mol/l.
