AB  - Serotonin receptors modulate numerous behavioral and neuropsychological processes. Therefore, they are the target for the action of many drugs, such as antipsychotics, antidepressants, antiemetics, migraine remedies, and many others. The 5-HT1A receptors have been involved in the pathogenesis and treatment of anxiety and depression and represent a promising target for new drugs with reduced extrapyramidal side effects. In most antidepressants, a piperazine-based structural motif can be identified as a common moiety. Here we describe the synthesis, pharmacological, and in silico characterization of a novel arylpiperazines series with excellent 5-HT1A affinity. The final compounds, 4a, 8a, and 8b, were selected according to predictions of in silico pharmacokinetics, docking analysis, and molecular dynamics in conjunction with physical properties, and metabolic stability. The accentuated molecules could serve as a lead compound for developing 5-HT1A drug-like molecules for depression treatment.

AB  - We present the green, highly atom-economical, solid-state silica gel-catalyzed synthesis of polysubstituted 1,4- and 1,2-dihydropyridines (DHPs) from commercially available materials, amines and ethyl propiolate. The DHP skeleton was assembled by heating the reactants and silica gel in a closed vessel. Aliphatic amines provided 1,4-isomers as the main or only DHP products, but the reactions of aromatic amines yielded a mixture of 1,4- and 1,2-isomers. To the best of our knowledge, this is the first example of the formation of a 1,2-DHP structure by the reaction of an amine with propiolic ester. Addition of 1 mass percent of H2SO4 to silica gel shifted the product distribution to 1,4-DHP as the main or the only isomer obtained. Experimental and theoretical analyses led to the identification of two key intermediates en route to DHPs and the explanation of the observed regioisomeric ratios. 1,2-DHPs show blue-cyan fluorescence in MeOH with the quantum yield Φ = 0.10–0.22 relative to quinine sulfate Φ = 0.58 and 1,4-DHPs show blue-violet fluorescence with Φ = 0.09–0.81.

AB  - In Sečovlje Salina Nature Park, the therapeutic mud matured in the natural sedimentary environmental site. This work aimed to determine the influence of the peloid maturation process on the hydrocarbon and elemental distributions, as well as changes in morphology. For this purpose, the sample before and after maturation was examined using various methods. n-Alkanes were the most abundant among saturated hydrocarbons in both immature and mature peloid samples. The results showed that the maturation mainly influenced the change in distribution and concentration (from 378 to 1958 ppm) of n-alkanes. The organic matter (OM) of the immature peloid sample was characterized by a slight prevalence of long-chain and odd carbon-numbered n-alkanes, maximizing at n-C27. However, mature peloid's OM showed a similar share of short-, mid- and long-chain n-alkanes with a slight dominance of short-chain members, maximizing at n-C16. The origin of short-chain and even carbon-numbered n-alkanes was attributed to microbial precursors (e.g., Leptolyngbyaceae). Hopanes were considerably more dominant compared to steranes in both peloids. The hopane series of immature peloid was characterized by the dominance of 22,29,30-trinor-hop-5(6)-ene (C27 hopene), as well as the presence of C30-hop-22(29)-ene (diploptene), which are widespread in cyanobacterial species. The aromatic fraction of immature peloid pointed to the predominance of polycyclic aromatic hydrocarbons (PAHs). As peloid aging progressed, the sample was richer in methyl-branched alkanes, carboxylic acids, their methyl esters, and thermodynamically more stable hopanes and steranes. The presence of elements with toxicological relevance during maturation was reduced below the limits prescribed in most of the directives for cosmetic products. It specifically refers to: As, Ni and Se. A higher concentration of total sulfur in the mature peloid can be related to gypsum precipitation in the summer and/or more intensive microbial activity.

AB  - Triclosan (TCS) is a polychlorinated phenoxy phenol (PCPPs) used as a disinfectant and a broad-spectrum antibacterial and antifungal agent in personal hygiene products. TCS easily forms diphenyl ethers and dioxins, which are persistent organic pollutants.

This work used a double approach for the TSC sensing: a) screen-printed (SPE) electrochemical platform for on-site application, modified with lanthanum iron oxide and graphitic carbon nitride composite (LaFeO3/Fe2O3@g-C3N4/SPE); and b) carbon paste electrode (CPE), modified with the same material and used in laboratory conditions. Linear range from 0.1 μM to 10 μM, the limit of detection (LOD) of 29 nM and the limit of quantification (LOQ) of 91 nM were obtained for CP electrode in BRBS pH 8. SPE showed the best analytical parameters in BRBS at pH 3, with a linear range from 0.3 μM to 7 μM, LOD of 0.09 μM and LOQ of 0.28 μM. Furthermore, the influence of potential interferents was investigated and proven to be negligible. Determination of TSC was performed to estimate the environmental impact of this compound as well as the practical usefulness of the proposed sensor in the real sample analysis, confirmed with a HPLC analysis.

Microspherical Titanium-Phosphorus Double Oxide: Hierarchical Structure Development for Sensing Applications

AB  - Stable, water-soluble titanium complexed with mandelic acid was used as a precursor for titanium phosphorus double oxide obtained in hydrothermal conditions in the presence of phosphoric acid. Surprisingly, hydrolysis of organic complexes provided a microstructured sphere with narrow size distribution, low aggregation and a small fraction of morphological irregularities. Obtained microspheres had a complex structure comprised of flakes, whose size could be manipulated with temperature conditions. Samples were found to be electrochemically active against sulcotrione, a well-recognized herbicide. Electrochemical sensors based on the synthesized microspheres were successfully adapted for natural water reservoir analysis and exhibited low levels of detection of 0.61 µM, limit of quantification of 1.86 µM, wide dynamic linear range from 2 to 200 µM, good selectivity, excellent reproducibility and in-time stability.

AB  - Triclosan (TCS) is a polychlorinated phenoxy phenol (PCPPs) used as a disinfectant and a broad-spectrum antibacterial and antifungal agent in personal hygiene products. TCS easily forms diphenyl ethers and dioxins, which are persistent organic pollutants.This work used a double approach for the TSC sensing: a) screen-printed (SPE) electrochemical platform for on-site application, modified with lanthanum iron oxide and graphitic carbon nitride composite (LaFeO3/Fe2O3@g-C3N4/SPE); and b) carbon paste electrode (CPE), modified with the same material and used in laboratory conditions. Linear range from 0.1 μM to 10 μM, the limit of detection (LOD) of 29 nM and the limit of quantification (LOQ) of 91 nM were obtained for CP electrode in BRBS pH 8. SPE showed the best analytical parameters in BRBS at pH 3, with a linear range from 0.3 μM to 7 μM, LOD of 0.09 μM and LOQ of 0.28 μM. Furthermore, the influence of potential interferents was investigated and proven to be negligible. Determination of TSC was performed to estimate the environmental impact of this compound as well as the practical usefulness of the proposed sensor in the real sample analysis, confirmed with a HPLC analysis.

Differently prepared PbO2/graphitic carbon nitride composites for efficient electrochemical removal of Reactive Black 5 dye

AB  - In this paper, electrochemical degradation of Reactive Black 5 (RB5) textile azo dye was examined in regard to different synthesis procedures for making PbO2–graphitic carbon nitride (g-C3N4) electrode. The reaction of with ClO− in the presence of different surfactants, i.e., cetyltrimethylammonium bromide (CTAB) and tetrabutylammonium phosphate (TBAP), under conventional conditions, resulted in the formation of PbO2 with varying morphology. The obtained materials were combined with g-C3N4 for the preparation of the final composite materials, which were then characterized morphologically and electrochemically. After optimizing the degradation method, it was shown that an anode comprising a steel electrode coated with the composite of PbO2 synthesized using CTAB as template and g-C3N4, and using 0.15 M Na2SO4 as the supporting electrolyte, gave the best performance for RB5 dye removal from a 35 mg/L solution. The treatment duration was 60 min, applying a current of 0.17 A (electrode surface 4 cm2, current density of 42.5 mA/cm2), while the initial pH of the testing solution was 2. The reusability and longevity of the electrode surface (which showed no significant change in activity throughout the study) may suggest that this approach is a promising candidate for wastewater treatment and pollutant removal.

Although the proposition of the detailed electrochemical oxidation mechanism of PTS goes out of the scope of the present research, we have determined the most probable reactive site of our analyte, by utilizing DFT-based reactivity descriptors (Fukui functions). For electrochemical quantification of PTS, oxidation peak at 0.32 V (vs. Ag/AgCl) was followed in presence of 0.5 mol L−1 of Briton–Robinson buffer solution (pH = 9). Coupled with the optimized parameters of differential pulse voltammetry (DPV), SPDE detected PTS in two linear ranges (first range was from 0.011 to 0.912 µmol L−1; second range was from 0.912 to 4.420 µmol L−1), providing the LOD and LOQ on a nanomolar level (3.1 nmol L−1 and 10.0 nmol L−1, respectively). The selectivity of the optimized DPV method was found to be excellent, with the current changes of less than 7%, in the presence of ten times higher concentrations of the certain interferences. The practical applicability of the SPDE and single-drop-detection approach in dietary supplements (with a declared PTS content of 50 mg/tablet), with the recovery values ranging from 95 to 102%, shows that the developed method has high potential for precise and accurate PTS detection, as well as exceptional miniaturization possibilities of relevant equipment for on-site sensing.

Cyclic voltammetry identified irreversible oxidation of PTS, where oxidation peak was shown to be strongly dependent on the pH of the working environmental. Although the proposition of the detailed electrochemical oxidation mechanism of PTS goes out of the scope of the present research, we have determined the most probable reactive site of our analyte, by utilizing DFT-based reactivity descriptors (Fukui functions). For electrochemical quantification of PTS, oxidation peak at 0.32 V (vs. Ag/AgCl) was followed in presence of 0.5 mol L−1 of Briton–Robinson buffer solution (pH = 9). Coupled with the optimized parameters of differential pulse voltammetry (DPV), SPDE detected PTS in two linear ranges (first range was from 0.011 to 0.912 µmol L−1; second range was from 0.912 to 4.420 µmol L−1), providing the LOD and LOQ on a nanomolar level (3.1 nmol L−1 and 10.0 nmol L−1, respectively). The selectivity of the optimized DPV method was found to be excellent, with the current changes of less than 7%, in the presence of ten times higher concentrations of the certain interferences. The practical applicability of the SPDE and single-drop-detection approach in dietary supplements (with a declared PTS content of 50 mg/tablet), with the recovery values ranging from 95 to 102%, shows that the developed method has high potential for precise and accurate PTS detection, as well as exceptional miniaturization possibilities of relevant equipment for on-site sensing.",

leads to the appearance of side effects, as a result of which the control and monitoring of its concentrations are of great importance. In this work, we have designed a new electrochemical sensor for
detecting L-DOPA using a carbon paste electrode (CPE) modified with Eu2O3@Cr2O3 composite
nanoparticles. Rare earth elements, including Eu, are increasingly used to design new electrode
nanocomposites with enhanced electrocatalytic properties. Europium has been considered a significant lanthanide element with greater redox reaction behavior. We conducted a hydrothermal
synthesis of Eu2O3@Cr2O3 and, for the first time, the acquired nanoparticles were used to modify
CPE. The proposed Eu2O3@Cr2O3/CPE electrode was investigated in terms of its electrocatalytic
properties and then used to develop an analytical method for detecting and quantifying L-DOPA.
The proposed sensor offers a wide linear range (1–100 µM), high sensitivity (1.38 µA µM−1
cm−2
) and
a low detection limit (0.72 µM). The practical application of the proposed sensor was investigated by
analyzing commercially available pharmaceutical tablets of L-DOPA. The corresponding results indicate
the excellent potential of the Eu2O3@Cr2O3/CPE sensor for application in real-time L-DOPA detection

AB  - Efficient cathodes for the hydrogen evolution reaction (HER) in acidic water electrolysis rely on the use of expensive platinum group metals (PGMs). However, to achieve economically viable operation, both the content of PGMs must be reduced and their intrinsically strong H adsorption mitigated. Herein, we show that the surface effects of hydrogenated TiO2 nanotube (TNT) arrays can make osmium, a so far less-explored PGM, a highly active HER electrocatalyst. These defect-rich TiO2 nanostructures provide an interactive scaffold for the galvanic deposition of Os particles with modulated adsorption properties. Through systematic investigations, we identify the synthesis conditions (OsCl3 concentration/temperature/reaction time) that yield a progressive improvement in Os deposition rate and mass loading, thereby decreasing the HER overpotential. At the same time, the Os particles deposited by this procedure remain mainly sub-nanometric and entirely cover the inner tube walls. An optimally balanced Os@TNT composite prepared at 3 mM/55 °C/30 min exhibits a record low overpotential (η) of 61 mV at a current density of 100 mA cm–2, a high mass activity of 20.8 A mgOs–1 at 80 mV, and a stable performance in an acidic medium. Density functional theory calculations indicate the existence of strong interactions between the hydrogenated TiO2 surface and small Os clusters, which may weaken the Os–H* binding strength and thus boost the intrinsic HER activity of Os centers. The results presented in this study offer new directions for the fabrication of cost-effective PGM-based catalysts and a better understanding of the synergistic electronic interactions at the PGM|TiO2 interface.

Enhancement of the hydrogen evolution reaction is an indication of electrocatalytic properties of the material, whereby developed method emerges as a candidate for straightforward application in electrochemical advanced oxidation processes (EAOPs). We have enriched experimental data with theoretical insights, provided by Density Functional Theory (DFT), and proposed oxidation mechanism of 1. Based on obtained results, we propose the new nanomaterial as a potent electrochemical modifier, suitable for catalytic treatment and process monitoring of the 1-polluted waters.

AB  - In this work, an advanced electrochemical platform was developed for the simple, rapid and sensitive determination of the polyphenolic compound tannic acid in various beverages using the combination of batch injection analysis with amperometric detection on a screen-printed carbon electrode. Several experimental parameters (pH of supporting electrolyte, detection potential, dispensing rate, injected volume, stirring) were consistently evaluated. The most favorable analytical performance in terms of sensitivity, selectivity, repeatability and sampling frequency was obtained in Britton-Robinson buffer pH 5.0 at a detection potential of +0.6 V vs. Ag/AgCl, a dispensing rate of 204 µL/s and an injected volume of 80 µL under stirring condition (1500 r.p.m.). The presented platform has advantages for routine analysis including portable and small-scale experimental setup, low sample consumption (∼100 µL), simple sample preparation (dilution in supporting electrolyte), high sampling frequency (180 injections per hour), low limit of detection (80 nM) and suitable precision (RSD = 4.2% for 10 μM tannic acid, n = 20). The applicability of the method was verified by analyzing several beverage samples (tea, wine) in spike-recovery assay with the recovery values for tannic acid ranging from 94 to 101 %.

The purpose of our study is to better understand the chemistry of the pygidial gland secretions of subterranean ground beetles, as well as the function and structure of the glands. We studied both the chemical composition of the pygidial gland secretion and morphology of the glands in adults of the troglophilic ground beetle species Laemostenus (Antisphodrus) cavicola (Schaum, 1858). The chemical composition of its defensive secretion was revealed using gas chromatography-mass spectrometry (GC-MS), while pygidial gland morphology of the beetle was investigated using bright-field microcopy. In total, seven chemical compounds were detected in the secretion mixture. Formic acid was the most dominant compound, followed by dodecyl acetate and undecane. Other chemicals were present in minor amounts. The morphological structure of the pygidial glands of L. (A.) cavicola was compared with the structure of the glands of the related congeneric troglophilic species Laemostenus (Pristonychus) punctatus (Dejean, 1828). Summary data on the semiochemicals that have been recorded so far in subterranean ground beetle species are presented, and the differences in the chemical composition of the secretions between and among troglobitic and troglophilic species are discussed. So far, forty-four compounds have been detected in four subterranean ground beetle species (two troglobites belonging to the tribe Trechini and two troglophiles belonging to the tribe Sphodrini). The results of this study indicate the great diversity of chemicals in the pygidial gland secretions of subterranean ground beetles. © 2023 by the authors.

AB  - The pygidial glands of carabids produce strong-smelling vapours. In this study, we examined the chemical composition of the gland secretions and the structure of the glands in five species of Carabini ground beetles (one species from the subtribe Calosomatina and four species from the subtribe Carabina): Calosoma (Calosoma) maderae (Fabricius, 1775), Carabus (Carabus) granulatus Linnaeus, 1758, C. (Limnocarabus) clathratus Linnaeus, 1760, C. (Carabus) ulrichii Germar, 1823, and C. (Procerus) gigas Creutzer, 1799. Additionally, we tested the antibacterial potential of the pygidial gland secretions of the two latter species against 11 bacterial strains. In order to detect the chemical content of the secretions, we used gas chromatography–mass spectrometry (GC–MS). The secretion extracts were applied against selected strains of medically important bacteria. We used bright-field microscopy to examine the morphology of the glands. We discovered a total of 11 chemical compounds in the pygidial gland extracts of the ground beetles we analysed. Ten of these compounds were identified as seven carboxylic acids, two hydrocarbons, and one aromatic aldehyde, while one chemical remained unidentified. Most of the components were isolated from the secretion of C. (L.) clathratus (nine), while the lowest number of compounds was found in C. (P.) gigas (two). Methacrylic acid was the most dominant compound by percentage in all five species, while angelic acid was also detected in all samples. As expected, salicylaldehyde was exclusively found in the species of the genus Calosoma Weber, 1801. The secretion of C. (P.) gigas was shown to achieve the highest level of antibacterial activity against Pseudomonas aeruginosa, Salmonella enterica, and S. typhimurium (even the same level as the positive control streptomycin), while the secretion of C. (C.) ulrichii achieved the highest antimicrobial potential against Staphylococcus epidermidis, S. aureus, Listeria monocytogenes, and Bacillus cereus. The most noticeable difference in the structure of the glands between the two genera is that the reservoir in Calosoma is more significantly narrowed as it leads into the efferent duct, compared to that of Carabus.

AB  - Bipolar disorder (BD) is a brain disorder that causes changes in a person’s mood, energy, and ability to function. It has a prevalence of 60 million people worldwide, and it is among the top 20 diseases with the highest global burden. The complexity of this disease, including diverse genetic, environmental, and biochemical factors, and diagnoses based on the subjective recognition of symptoms without any clinical test of biomarker identification create significant difficulties in understanding and diagnosing BD. A 1H-NMR-based metabolomic study applying chemometrics of serum samples of Serbian patients with BD (33) and healthy controls (39) was explored, providing the identification of 22 metabolites for this disease. A biomarker set including threonine, aspartate, gamma-aminobutyric acid, 2-hydroxybutyric acid, serine, and mannose was established for the first time in BD serum samples by an NMR-based metabolomics study. Six identified metabolites (3-hydroxybutyric acid, arginine, lysine, tyrosine, phenylalanine, and glycerol) are in agreement with the previously determined NMR-based sets of serum biomarkers in Brazilian and/or Chinese patient samples. The same established metabolites (lactate, alanine, valine, leucine, isoleucine, glutamine, glutamate, glucose, and choline) in three different ethnic and geographic origins (Serbia, Brazil, and China) might have a crucial role in the realization of a universal set of NMR biomarkers for BD.

AB  - In the present study, we investigated in vitro the antimicrobial activity of the pygidial gland secretion of the guanophilic ground beetle Laemostenus (Pristonychus) punctatus (Dejean, 1828) and some of its chemicals against resistant and non-resistant bacteria and Candida species, the synergistic and additive potential of combinations of selected chemicals and antimicrobial drugs against resistant bacterial and fungal strains, anti-adhesive and anti-invasive potential of the secretion and formic acid alone and in selected combinations with antimicrobial drugs against methicillin-resistant Staphylococcus aureus (MRSA) toward spontaneously immortalized human keratinocyte cell line (HaCaT cells). In addition, we examined the antiproliferative activity of the secretion and formic acid in vitro. The tested secretion and the standards of formic and oleic acids possessed a significant level of antimicrobial potential against all tested strains (P < 0.05). The isolate from guano Pseudomonas monteilii showed the highest resistance to the secretion and formic acid, while MRSA achieved a significantly high level of susceptibility to all agents tested, particularly to the combinations of formic acid and antibiotics, but at the same time showed a certain level of resistance to the antibiotics tested individually. Candida albicans and C. tropicalis were found to be the most sensitive fungal strains to the secretion. Formic acid (MIC 0.0005 mg/mL) and gentamicin (MIC 0.0010 mg/mL) in the mixture achieved synergistic antibacterial activity against MRSA (FICI = 0.5, P < 0.05). The combination of formic acid, gentamicin and ampicillin accomplished an additive effect against this resistant bacterial strain (FICI = 1.5, P < 0.05). The secretion achieved a better inhibitory effect on the adhesion ability of MRSA toward HaCaT cells compared to formic acid alone, while formic acid showed better results regarding the invasion (P < 0.001). The combinations of gentamicin and ampicillin, as well as of formic acid and gentamicin and ampicillin achieved similar anti-adhesive and anti-invasive effects, with a slight advantage of formic acid and antibiotics in combination (P < 0.001). The secretion and formic acid were found to be non-toxic to HaCaT cells in vitro (IC50 ≥ 401 μg/mL).

Exploring and strengthening the potential of R-phycocyanin from Nori flakes as a food colourant

AB  - This study aimed to purify, characterise and stabilise the natural food colourant, R-phycocyanin (R-PC), from the red algae Porphyra spp. (Nori). We purified R-PC from dried Nori flakes with a high purity ratio (A618/A280 ≥ 3.4) in native form (α-helix content 53%). SAXS measurements revealed that R-PC is trimeric ((αβ)3) in solution. The thermal denaturation of α-helix revealed one transition (Tm at 52 °C), while the pH stability study showed R-PC is stable in the pH range 4–8. The thermal treatment of R-PC at 60 °C has detrimental and irreversible effects on R-PC colour and antioxidant capacity (22 % of residual capacity). However, immobilisation of R-PC within calcium alginate beads completely preserves R-PC colour and mainly retains its antioxidant ability (78 % of residual capacity). Results give new insights into the stability of R-PC and preservation of its purple colour and bioactivity by encapsulation in calcium alginate beads.

AB  - This study investigates the impact of Porphyra sp. extracts on HT29 cell line growth and viability at reduced serum conditions. The concentration-dependent effects of phycobiliproteins (PBPs) on cell proliferation were examined over various time intervals. Lower concentrations of PBPs (20 μg/mL) demonstrated an increase in HT29 cell viability after 48 hours and 5 days of cultivation at reduced serum concentration (final serum concentration was in the range from 5 to 8%). This suggests a potential positive influence on cell proliferation, likely due to their antioxidant properties. Conversely, higher concentrations of PBPs exhibited inhibitory effects on cell growth, possibly due to cytotoxicity at elevated levels. Remarkably, when HT29 cells were cultured solely in algal extract without fetal calf serum (FCS), complete growth inhibition was observed after 72
hours. This finding underscores the insufficient nutrient and growth factor provision of PBPs alone for sustaining cell viability. Morphological differences observed in cells cultured with 70 μg/mL of PBPs indicated potential alterations in cellular morphology. Notably, 70 μg/mL of PBPs in RPMI medium with 5% FCS displayed growth inhibition compared to the control (5% FCS). Furthermore, we assessed HT29 cell adaptability to changes in FCS concentration and PBP supplementation. Cells incubated under varying FCS and PBP conditions were subcultured into RPMI medium with lower FCS concentration and PBPs from Porphyra. The viability of cells following subculturing indicated sustained adaptability to reduced FCS levels. Overall, this study provides valuable insights into the concentration-dependent effects of PBPs from Porphyra extracts
on HT29 cell growth and viability. The findings underscore the potential benefits of PBPs at lower concentrations for cell proliferation at reduced serum conditions and reveal the adaptability of HT29 cells to changing culture conditions.

AB  - Common to all biological systems and living organisms are molecular interactions, which
may lead to specific physiological events. Most often, a cascade of events occurs, establishing an equilibrium between possibly competing and/or synergistic processes. Biochemical pathways that sustain life depend on multiple intrinsic and extrinsic factors contributing to aging and/or diseases. This article deals with food antioxidants and human proteins from the circulation, their interaction, their effect on the structure, properties, and function of antioxidant-bound proteins, and the possible impact of complex formation on antioxidants. An overview of studies examining interactions between individual antioxidant compounds and major blood proteins is presented with findings. Investigating antioxidant/protein interactions at the level of the human organism and determining antioxidant distribution between proteins and involvement in the particular physiological role is a very complex and challenging task. However, by knowing the role of a particular protein in certain pathology or aging, and the effect exerted by a particular antioxidant bound to it, it is possible to recommend specific food intake or resistance to it to improve the condition or slow down the process.

Ultrasensitive Quantification of Crustacean Tropomyosin by Immuno-PCR

AB  - Tropomyosin is the major and predominant allergen among shellfish. This study developed
an ultrasensitive immuno-PCR method for the quantification of crustacean tropomyosin in foods.
The method couples sandwich ELISA with the real-time PCR (rtPCR) amplification of marker DNAs. Monoclonal anti-TPM antibody was the capture antibody, polyclonal rabbit anti-shrimp tropomyosin antibody was the detection antibody, while natural shrimp tropomyosin served as the standard. A double-stranded amino-DNA was covalently conjugated to a secondary anti-rabbit antibody and subsequently amplified and quantified via rtPCR. The quantification sensitivity of immuno-PCR was 20-fold higher than analogous ELISA, with LOQ 19.8 pg/mL. The developed immuno-PCR method is highly specific for the detection of crustacean tropomyosin and is highly precise in a broad concentration range. Tropomyosin recovery in the spiked vegetable soup was 87.7–115.6%. Crustacean tropomyosin was also quantified in commercial food products. The reported immuno-PCR assay is the most sensitive method for the quantification of crustacean tropomyosin and is the first immuno-PCR-based assay for the quantification of food allergen and food protein in general. The described method could be easily adapted for the specific and ultrasensitive immuno-PCR-based detection of traces of any food allergen that is currently being quantified with ELISA, which is of critical importance for people with food allergies.

abstract = "Tropomyosin is the major and predominant allergen among shellfish. This study developed
an ultrasensitive immuno-PCR method for the quantification of crustacean tropomyosin in foods.
The method couples sandwich ELISA with the real-time PCR (rtPCR) amplification of marker DNAs. Monoclonal anti-TPM antibody was the capture antibody, polyclonal rabbit anti-shrimp tropomyosin antibody was the detection antibody, while natural shrimp tropomyosin served as the standard. A double-stranded amino-DNA was covalently conjugated to a secondary anti-rabbit antibody and subsequently amplified and quantified via rtPCR. The quantification sensitivity of immuno-PCR was 20-fold higher than analogous ELISA, with LOQ 19.8 pg/mL. The developed immuno-PCR method is highly specific for the detection of crustacean tropomyosin and is highly precise in a broad concentration range. Tropomyosin recovery in the spiked vegetable soup was 87.7–115.6%. Crustacean tropomyosin was also quantified in commercial food products. The reported immuno-PCR assay is the most sensitive method for the quantification of crustacean tropomyosin and is the first immuno-PCR-based assay for the quantification of food allergen and food protein in general. The described method could be easily adapted for the specific and ultrasensitive immuno-PCR-based detection of traces of any food allergen that is currently being quantified with ELISA, which is of critical importance for people with food allergies.",

Sensitive and specific quantification of traces of allergens present in food samples is of critical importance for people with food allergies. This study thus aimed to develop a highly sensitive immuno-PCR method for detecting crustacean tropomyosin in foods. The sensitivity of immuno-PCR for quantification of tropomyosin was increased by up to 20-fold compared to ELISA, demonstrating accuracy as low as 19.8 pg/mL. Recovery of tropomyosin in vegetable soup as a food matrix was in the 87.7–115.6% range, with relative standard deviations in the 5–24.5% range. Tropomyosin was also quantified in the commercially available food products. Developed immuno-PCR technique thus shows the potential to be a method of choice for specific and ultrasensitive detection of tropomyosin in food samples, with the final aim of reducing risks of accidental food contamination.

AB  - Viral exonucleases play role in many processes essential for genome ma intenance,including DNA repair and recombination. Lambda exonuclease (λ -exo), isolated fromlambda bacteriophage, hydrolases double-stranded DNA (dsDNA) in the highly processivemanner in 5’→3’ direction, yielding mononucleotides and single -stranded DNA (ssDNA).This unique enzymatic properties offer several promising biotechnological applications,such as highly sensitive quantification of DNA modifications and single -moleculesequencing. Hence, optimization of the expression conditions is a prerequisite to achievehigh-level production of λ-exo. Here we have tested λ -exo expression in five different E.coli strains under various temperature regimes in order to establish the optimal conditionsfor efficient production of recombinant λ -exo. The N-terminally His -tagged λ-exo wassuccessfully expressed in E.coli BL21(AI), SHuffle T7, C41(DE3) and C43(DE3) strains inLB broth. Collected aliquots were analysed by SDS-PAGE, followed by CBB staining.Relative yield of target protein bands was determined by densitometry in total cell lysate, aswell as in soluble and insoluble cytoplasmatic fractions. We identified E.coli BL21(AI),SHuffle T7 and C41(DE3) as good producers of recombinant λ -exo, and upon scaling up, λ-exo was purified from crude cell lysates by metal affinity chromatography in satisfactoryyield. Our data suggest that densitometric analysis could serve as a powerful low-costscreening platform for improving recombinant protein expression strategies.

AB  - Introduction: Lambda exonuclease (λ-exo), isolated from lambda bacteriophage, plays a crucial role inDNA replication, repair and recombination. The enzyme specifically hydrolases double-stranded DNA(dsDNA) in a highly processive manner in 5’�����3’ direction, yielding mononucleotides and single-strandedDNA (ssDNA). This efficient unidirectional degradation makes it an invaluable tool in various molecularbiology techniques, including novel sequencing technologies. Hence, optimization of the expressionconditions is a prerequisite to achieving high-level production of λ-exo.Methods: The N-terminally His-tagged λ-exo fusion construct with thrombin cleavage site (AddGene#104531) was successfully transformed into five different E. coli strains (BL21(AI), Shuffle T7, C41(DE3),C43(DE3), and BL21(DE3)). Expression was tested under three temperature regimes (20 ˚C, 30 ˚C, and 37˚C) over time for enhanced soluble production. Crude extracts were analysed by SDS-PAGE for total proteinexpression, soluble and insoluble cytoplasmatic fractions. The exonuclease activity of the extractswas monitored via in-house developed fluorescence-based screening assay. Optimal conditions for highyieldproduction were determined by densitometric analysis using NIH ImageJ software. The soluble andactive enzyme was produced on the large scale in a shaking flask culture under optimal conditions, andpurified to homogeneity from the soluble lysate via metal affinity chromatography.Results:We identified E. coli BL21(AI), SHuffle T7, and C41(DE3) as good producers of recombinant λ-exoand determined optimal conditions (30 ˚C, 6 h post-induction) for high-yield expression. The enzyme waseluted from Ni2+-IDA-Sepharose 6B column in 300 mM imidazole and maintained its activity upon purificationassessed by an in-house developed fluorescence-based screening assay.Conclusion: This study provides a scalable cost-effective approach for soluble λ-exo production in selectedE. coli strains. This expression system would be a helpful platform for development of high-yieldproduction of λ-exo, easing its exploitation in biotechnology and other scientific frontiers. Additionally,we provided a valuable low-cost screening assay for monitoring exonuclease activity during each purificationstep.
