AB  - Three Schiff base Cu(II) complexes, (N,N’-bis(acetylacetone)­propyl­enediimine)copper(II) complex, [Cu(acac2pn)] (1), (N,N'-bis-(ben­zoyl­acet­one)propylenediimine)copper(II) complex, [Cu(phacac2pn)] (2) and (N,N’-bis-(trifluoroacetylacetone)propylenediimine)­copper(II) complex, [Cu(tfacac2pn)] (3), were used to investigate the interactions with calf thymus DNA (ct-DNA) and bovine serum albumin (BSA) using the electronic absorption and spectro­scopic fluorescence methods. UV–Vis absorption studies showed that studied complexes interact with DNA molecule and exhibit moderate binding affinity. Fluorescence studies of complexes 1–3 also showed a possibility for DNA int­ercalation as well as a relatively high binding ability toward BSA. Among the tested complexes, the highest affinity for DNA and BSA molecules was shown by complex 1. Cytotoxic analyses, performed on human colorectal carcinoma HCT-116 and healthy lung fibroblast MRC-5 cell lines, showed that complex 2 exhibited activity on both cell lines, while complexes 1 and 3 did not show any activity.

AB  - In order to discover new anticancer drugs, novel ruthenium(III) complexes [Ru(L)Cl(H2O)], where L is tetradentate Schiff base bis(acetylacetone)ethylendiimine (acacen, 1), bis(benzoylacetone)ethylendiimine (bzacen, 2), (acetylacetone)(benzoylaceton)ethylendiimine (acacbzacen, 3), bis(acetylacetone)propylendiimine (acacpn, 4), bis(benzoylacetone)propylendiimine (bzacpn, 5) or (acetylacetone)(benzoylaceton)propylendiimine (acacbzacpn, 6), were synthesized. The complexes 1 – 6 were characterized by elemental analysis, molar conductometry, and by various spectroscopic techniques, such as UV–Vis, IR, EPR, and ESI-MS. Based on in vitro DNA/BSA experiments, complexes 2 (bzacen) and 5 (bzacpn) with two aromatic rings showed the highest DNA/BSA-activity, suggesting that the presence of the aromatic ring on the tetradentate Schiff base ligand contributes to increased activity. Moreover, these two compounds showed the highest cytotoxic effects toward human, A549 and murine LLC1 lung cancer cells. These complexes altered the ratio of anti- and pro-apoptotic molecules and induced apoptosis of A549 cells. Further, complexes 2 and 5 reduced the percentage of Mcl1 and Bcl2 expressing LLC1 cells, induced their apoptotic death and exerted an antiproliferative effect against LLC1. Finally, complex 5 reduced the volume of mouse primary heterotopic Lewis lung cancer, while complex 2 reduced the incidence and mean number of metastases per lung. Additionally, molecular docking with DNA revealed that the reduced number of aromatic rings or their absence causes lower intercalative properties of the complexes in order: 2 > 5 > 6 > 3 > 4 > 1. It was observed that conventional hydrogen bonds and hydrophobic interactions contribute to the stabilization of the structures of complex-DNA. A molecular docking study with BSA revealed a predominance of 1 – 6 in binding affinity to the active site III, a third D-shaped hydrophobic pocket within subdomain IB.

AB  - Fourteen novel quinoline-4-carboxylic acid-chalcone hybrids were obtained via Claisen–Schmidt condensation and evaluated as potential human dihydroorotate dehydrogenase (hDHODH) inhibitors. The ketone precursor 2 was synthesized by the Pfitzinger reaction and used for further derivatization at position 3 of the quinoline ring for the first time. Six compounds showed better hDHODH inhibitory activity than the reference drug leflunomide, with IC50 values ranging from 0.12 to 0.58 μM. The bioactive conformations of the compounds within hDHODH were resolved by means of molecular docking, revealing their tendency to occupy the narrow tunnel of hDHODH within the N-terminus and to prevent ubiquinone as the second cofactor from easily approaching the flavin mononucleotide as a cofactor for the redox reaction within the redox site. The results of the 3-(4,5-dimethylthiazol-2-yl)-2,5-diphenyltetrazolium bromide (MTT) assay revealed that 4d and 4h demonstrated the highest cytotoxic activity against the A375 cell line, with IC50 values of 5.0 and 6.8 µM, respectively. The lipophilicity of the synthesized hybrids was obtained experimentally and expressed as logD7.4 values at physiologicalpH while the solubility assay was conducted to define physicochemical characteristics influencing the ADMET properties.

Geological substrate-related variability of Teucrium montanum L. (Lamiaceae) essential oil

AB  - This study aimed to determine the qualitative variability of essential oils of mountain germander – Teucrium
montanum L. (Lamiaceae) at the population level, depending on the type of geological substrate. Ten samples of
essential oils originated from aerial plant parts of populations from calcareous and ten from serpentinite were
tested by GCxGC-MS method. The obtained results showed that the serpentinite populations have more than 3
times higher essential oil yield than calcareous populations. Based on the average values for the relative percentage amounts of T. montanum essential oils, it was shown that the sesquiterpenes (34.89%) are the main
compounds in analyzed essential oils. Population from calcareous habitats synthesize twice as many aliphatic
hydrocarbons than populations from serpentinite habitats, while populations from serpentinite habitats synthesize twice as much mono- and sesquiterpenes. The compounds with highest relative percentage amounts in
calcareous populations was the hydrocarbon tetracosane (9.02%), while in serpentinite populations it was
oxygenated sesquiterpene, limonen-10-ol (4.81%). Compounds which were only detected for calcareous populations are: dodecane and isophytol, while for serpentinite populations are: 3-caren-2-ol, carvone oxide and
pyronene. The obtained results indicate a significant influence of the habitat geological substrate on the qualitative characteristics of the T. montanum essential oil. Differentiation of calcareous and serpentinite populations
into two distinct groups based on the essential oil composition indicate the significant presence of specific
chemotypes developed by the influence of various edaphic characteristics.

AB  - Dithiocarbamates, with their high lipophilic character and good chelating properties, could provide stabile transition metal complexes and enable these metal-based drugs to reach their biological targets. Palladium(II) and gold(III) complexes, due to its structural similarity with platinum(II)compounds, could be competitive candidates for implementing and developing new pharmacological agents. Herein, novel gold(III) complex with triammonium N-carboxyiminodiacetate as ligand has been synthesized and characterized by elemental analysis, molar conductivity measurements, FT-IR and 1H and 13C NMR spectroscopy. The proposed structure was examined and compared with analogue palladium(II) and zinc(II) complexes by density functional theory (B3LYP/def2tzvp). Additionally, DNA-binding studies by UV/Vis-spectrometer as well as ethidium-bromide (EB) and bovine serum albumin (BSA) quenching studies by spectrofluorometer were performed for gold(III), palladium(II) and zinc(II) dithiocarbamate complexes. The investigated complexes showed a good affinity to calf thymus DNA (CT-DNA) and BSA, with a higher affinity to BSA. Palladium(II) complex exhibited 5-fold a stronger affinity to DNA binding in comparison to zinc(II), but no significantly higher than gold(III) complex. Furthermore, palladium(II) and gold(III) complexes demonstrated a similar affinity toward BSA and these interactions are stronger than showed by zinc(II) complex.

AB  - Cilj prikazanog istraživanja je utvrđivanje kvalitativne varijabilnosti i relativne zastupljenosti seskviterpenskih ugljovodonika u uzorcima vrste Teucrium montanum L. prikupljenih sa dva različita tipa podloge. Četiri uzorka etarskih ulja sa serpentinita i četiri sa krečnjaka dobijeno je metodom hidrodestilacije od nadzemnihbiljnih delova. Na osnovu rezultata, pokazano je da populacije sa serpentinitskih staništa sintetišu 2 puta više seskviterpena nego sa krečnjačkih, dok je jedinjenje 6-epi-β-kubeben detektovan samo u populacijama sa krečnjaka. Edafske karakteristikestaništa utiču na kvalitativni sastav i relativnu zastupljenost seskviterpena u etarskim uljima što ima značajnu primenu u biotehnologiji.
AB  - The aim of this study is to determine the qualitative variability and relative presence of sesquiterpene hydrocarbons in essential oils samples of Teucrium montanum L. collected from two different substrate types. Four samples of essential oils from serpentinite and four from calcareous were obtained by the method of hydrodistillation from aboveground plant parts. Based on the results, it was shown that populations from serpentinite habitats synthesize 2 times more sesquiterpenes than from calcareous ones, while the compound 6-epi-β-cubeben was detected only in populations from calcareous. Edaphic characteristics of habitats affect the qualitative composition and relative presence of sesquiterpenes in essential oils, which has a significant application in biotechnology.

Edafske karakteristikestaništa utiču na kvalitativni sastav i relativnu zastupljenost seskviterpena u etarskim uljima što ima značajnu primenu u biotehnologiji., The aim of this study is to determine the qualitative variability and relative presence of sesquiterpene hydrocarbons in essential oils samples of Teucrium montanum L. collected from two different substrate types. Four samples of essential oils from serpentinite and four from calcareous were obtained by the method of hydrodistillation from aboveground plant parts. Based on the results, it was shown that populations from serpentinite habitats synthesize 2 times more sesquiterpenes than from calcareous ones, while the compound 6-epi-β-cubeben was detected only in populations from calcareous. Edaphic characteristics of habitats affect the qualitative composition and relative presence of sesquiterpenes in essential oils, which has a significant application in biotechnology.",
