AB  - L-DOPA (L-3,4-dihydroxyphenylalanine), the precursor of dopamine, is widely used in the treatment of Parkinson’s disease, thus determining and monitoring the concentration of L-DOPA is of utmost importance for both medical and scientific purposes. Although many analytical approaches, designed for drug detection and quantification, already exist, there is a constant need for modification of old and tailoring of new, faster, and selective methods. Redox active chemical species, such as L-DOPA, can be measured directly by electrochemical means, whereas electrochemical sensors combine sensitivity and selectivity within a small analytical device. This work demonstrates the development of such electrochemical sensor, based on carboxylated single-wall carbon nanotubes (SWCNT-COOH) decorated with SiO2 coated-Nd2O3 nanoparticles, and further application for the detection of L-DOPA. Developed SWCNT-COOH@Nd2O3-SiO2 sensor shows linear response in the range from 2 µmol L−1 to 52 µmol L−1 analyte concentration, and beside the low detection limit, it is characterized by a fast response time, as well as good life-time, reproducibility and repeatability.

AB  - Cerium oxide nanoparticles (nCeO2) have widespread applications, but they can be hazardous to the environment. Some reports indicate the toxic effect of nCeO2 on tested animals, but literature data are mainly contradictory. Coating of nCeO2 can improve their suspension stability and change their interaction with the environment, which can consequently decrease their toxic effects. Herein, the exopolysaccharides levan and pullulan, due to their high water solubility, biocompatibility, and ability to form film, were used to coat nCeO2. Additionally, the monosaccharide glucose was used, since it is a common material for nanoparticle coating. This is the first study investigating the impact of carbohydrate-coated nCeO2 in comparison to uncoated nCeO2 using different model organisms. The aim of this study was to test the acute toxicity of carbohydrate-coated nCeO2 on the bacterium Vibrio fischeri NRRL B-11177, the crustacean Daphnia magna, and zebrafish Danio rerio. The second aim was to investigate the effects of nCeO2 on respiration in Daphnia magna which was performed for the first time. Finally, it was important to see the relation between Ce bioaccumulation in Daphnia magna and Danio rerio and other investigated parameters. Our results revealed that the coating decreased the toxicity of nCeO2 on Vibrio fischeri. The coating of nCeO2 did not affect the nanoparticles’ accumulation/adsorption or mortality in Daphnia magna or Danio rerio. Monitoring of respiration in Daphnia magna revealed changes in CO2 production after exposure to coated nCeO2, while the crustacean's O2 consumption was not affected by any of the coated nCeO2. In summary, this study revealed that, at 200 mg L−1, uncoated and carbohydrate-coated nCeO2 are not toxic for the tested organisms, however, the CO2 production in Daphnia magna is different when they are treated with coated and uncoated nCeO2. AB  - BACKGROUND: Pectin is a complex polysaccharide, present in fresh and processed food as an additive, which also serves as an important source of fiber. In this work, the aim was to investigate in-vitro application of apple pectin as a possible antimicrobial, antioxidant and prebiotic agent for growth of bacteria selected from human milk and infant feces, as well as standard strains Lactobacillus rhamnosus and Bifidobacterium bifidum.
METHODS : All the bacteria isolated from the milk of mothers of preterm and term-born infants, bacteria from infant feces and standard strains were tested for fermentation of apple pectin as the main carbohydrate in selective broth media.
RESULTS: An increase in the growth of bacteria Bifidobacteria, Lactobacillus, and a consortium of bacteria selected from human milk and infant feces was noticed after pectin fermentation. It was found that pectin has a positive impact on the increase of the antioxidant capacity in all the tested samples. A high correlation was achieved, comparing electrochemical with spectrophotometric methods. Antimicrobial tests showed that pectin and fermentation products have slight or no influence at all on the growth of standard strains of Escherichia coli, Staphylococcus aureus and Candida albicans.
CONCLUSIONS: The results showed the importance of pectin as an antioxidant and its impact on the development of potential probiotic microbiota in mothers and infants. The study indicated a new potential application of pectin as a supplement in infant food.

AB  - To investigate the magnetic behavior of starch-coated cobalt ferrites, well-established synthetic methods, i.e., coprecipitation, mechanochemical, ultrasonically assisted coprecipitation, microemulsion, and microwave-assisted hydrothermal syntheses were chosen for their preparation. The obtained materials had pure single-phase spinel structures. Scanning and transmission electron microscopy analyses revealed that the morphology of the samples is not uniform, and particle aggregation is a dominant process. Fourier transform infrared spectra and thermogravimetric analysis confirmed the presence of starch in all–coated samples. The unusually higher saturation magnetization of starch-coated samples than their as-prepared analogs, obtained by coprecipitation, ultrasonically assisted coprecipitation, and microwave-assisted hydrothermal methods, might be explained by the Ostwald ripening mechanism induced by the coating process. AB  - The main biologically active components of plants belonging to the genus Allium, responsible for their biological activities, including anti-inflammatory, antioxidant and immunomodulatory, are organosulfur compounds. The aim of this study was to synthetize the mixture of dipropyl polysulfides (DPPS) and to test their biological activity in acute hepatitis. C57BL/6 mice were administered orally with DPPS 6 h before intravenous injection of Concanavalin A (ConA). Liver inflammation, necrosis and hepatocytes apoptosis were determined by histological analyses. Cytokines in liver tissue were determined by ELISA, expression of adhesive molecules and enzymes by RT PCR, while liver mononuclear cells were analyzed by flow cytometry. DPPS pretreatment significantly attenuated liver inflammation and injury, as evidenced by biochemical and histopathological observations. In DPPS-pretreated mice, messenger RNA levels of adhesion molecules and NADPH oxidase complex were significantly reduced, while the expression of SOD enzymes was enhanced. DPPS pretreatment decreased protein level of inflammatory cytokines and increased percentage of T regulatory cells in the livers of ConA mice. DPPS showed hepatoprotective effects in ConA-induced hepatitis, characterized by attenuation of inflammation and affection of Th17/Treg balance in favor of T regulatory cells and implicating potential therapeutic usage of DPPS mixture in inflammatory liver diseases.

AB  - The study investigated the influence of maturity, biomass type, and depositional settings on the distribution and relative abundance of polycyclic aromatic hydrocarbons (PAHs) for lacustrine sediments collected from depths up to 1000 m of Prebreza and Čučale stratigraphic units (the northwest part of the Toplica Basin). A recently proposed benzo[ghi]perylene/(perylene + benzo[ghi]perylene) parameter, along with commonly used Phenanthrene Alkylation Index and benzo[e]pyrene/(perylene + benzo[e]pyrene) indices, pointed out differences in maturity levels between stratigraphic units by displaying a positive linear relationship with vitrinite reflectance. However, in several immature Prebreza sediments, a substantial presence of algae and/or anoxic, mesosaline/hypersaline conditions were suitable for forming β-substituted methylphenanthrenes and 6-ring benzo[ghi]perylene. Generally, high molecular weight unsubstituted PAHs (HMWPAHs), particularly perylene, predominated Prebreza sediments. Anoxic conditions appeared to be decisive for accumulating and preserving the perylene carbon skeleton in studied stratigraphic units. Besides, more intense volcanism in the Čučale unit favored combustion processes, which prompted the accumulation of low molecular weight unsubstituted PAHs (LMWPAHs), especially phenanthrene. A general prevalence of retene over cadalene in Prebreza sediments, in which alginite and liptodetrinite predominated, implied algae as retene precursor. Selective degradation of retene or hindered demethylation of 9-methylphenanthrene under anoxic and more saline environmental settings had occurred notably in the Prebreza unit, which led to the formation of 1-methylphenanthrene and/or pimanthrene (1,7-dimethylphenanthrene). Čučale sediments with substantial amounts of vitrinite macerals or saturated diterpenoids had a predominant simonellite derived from conifers. Non-degraded and well-preserved Pinaceae conifers predominated in Prebreza sediments deposited under semi-arid climatic conditions, whereas mixed degraded/non-degraded conifers characterized Čučale sediments deposited in a wide range of climatic conditions, from semi-arid to semi-humid. © 2021, The Author(s), under exclusive licence to Springer Nature B.V.

BanLec-eGFP Chimera as a Tool for Evaluation of Lectin Binding to High-Mannose Glycans on Microorganisms

AB  - Fluorescently labeled lectins are useful tools for in vivo and in vitro studies of the structure and function of tissues and various pathogens such as viruses, bacteria, and fungi. For the evaluation of high-mannose glycans present on various glycoproteins, a three-dimensional (3D) model of the chimera was designed from the crystal structures of recombinant banana lectin (BanLec, Protein Data Bank entry (PDB): 5EXG) and an enhanced green fluorescent protein (eGFP, PDB 4EUL) by applying molecular modeling and molecular mechanics and expressed in Escherichia coli. BanLec-eGFP, produced as a soluble cytosolic protein of about 42 kDa, revealed β-sheets (41%) as the predominant secondary structures, with the emission peak maximum detected at 509 nm (excitation wavelength 488 nm). More than 65% of the primary structure was confirmed by mass spectrometry. Competitive BanLec-eGFP binding to high mannose glycans of the influenza vaccine (Vaxigrip®) was shown in a fluorescence-linked lectin sorbent assay (FLLSA) with monosaccharides (mannose and glucose) and wild type BanLec and H84T BanLec mutant. BanLec-eGFP exhibited binding to mannose residues on different strains of Salmonella in flow cytometry, with especially pronounced binding to a Salmonella Typhi clinical isolate. BanLec-eGFP can be a useful tool for screening high-mannose glycosylation sites on different microorganisms.

AB  - Based on graphene nanoplatelets capability to build block composites, as well as well-known electrochemical characteristic of the manganese oxide materials, in the present research, a nanocomposite, formed from graphene nanoplatelets (GNP) and manganese(IV)-oxide (MnO2) nanoparticles, has been proposed as a novel and convenient support for enzyme immobilization. Performance of screen printed carbon electrodes (SPCEs) was significantly improved after their modification with GNP@MnO2 (SPCE/GNP@MnO2). The polyphenolic index biosensor was prepared by applying the drop coating technique using laccase and Nafion®. Developed biosensor shows a fast and reliable amperometric response toward caffeic acid, as a model compound, at operating potential of +0.40 V (vs Ag/AgCl), with a wide linear range and detection limit of 1.9 μmol l−1. Developed procedure was successfully applied for the determination of polyphenolic indexes in wine samples. Recovery tests indicate excellent accuracy and precision of the method, concluding that the biosensor can offer a fast, accurate, reliable and precise determination of the polyphenolic index. More importantly, our results suggest a great potential for the application in real samples.

AB  - Novel polyurethane nanocomposite (PUN) materials containing different surface-functionalized mesoporous silica nanoparticles (MSNs) were prepared by in situ polymerization methodology. Polyurethane network was formed from poly(dimethylsiloxane)-based macrodiol (PDMS), 4,4′-methylenediphenyldiisocyanate (MDI), and hyperbranched polyester of the second pseudo-generation (BH-20; used as crosslinking agent). PU and PU/MSN nanocomposites contained equal ratios of soft PDMS and hard MDI-BH-20 segments. Non-functionalized and surface-functionalized (with 3-(trihydroxysilyl)propyl methylphosphonate (FOMSN) and 2-[methoxy(polyethyleneoxy)6−9propyl]trimethoxysilane (PEGMSN)) MSNs were used as the nanofillers at a concentration of 1 wt%. Prepared materials were characterized by Fourier transform infrared (FTIR) spectroscopy, atomic force microscopy (AFM), scanning electron microscopy (SEM), thermogravimetric analysis (TGA), differential scanning calorimetry (DSC), dynamic mechanical thermal analyses (DMTA), nanoindentation, equilibrium swelling and water absorption measurements. Characteristics of the prepared PUNs when in contact with a biological environment were assessed through testing their biocompatibility, protein adsorption and adhesion of endothelial cells. The favourable influence of MSNs on the physico-chemical and biological characteristics of these novel PUN materials was identified, which evidences their vast applicability potential as coatings for medical devices and implants.

AB  - Geometries of aromatic/aromatic interactions in crystal structures of small molecules from the Cambridge Structural Database (CSD) (benzene/benzene, toluene/toluene, and p-phenol/p-phenol interactions) and in protein structures from the Protein Data Bank (PDB) (Phe/Phe and Tyr/Tyr interactions) were studied and compared. The data show a larger influence of crystal packing/surrounding, more than the influence of substituents, on geometries of aromatic/aromatic interactions. While the interactions in crystal structures from the CSD show preference for parallel stacking interactions at the large offsets, in proteins from the PDB, they show preference for T-shaped geometries with small offsets.

AB  - The mutual impact of low-quality lignite and high-density polyethylene (HDPE) during open system pyrolysis was investigated, aiming to improve utilization of lignite with simultaneous treatment of HDPE waste. Pyrolysis of lignite, HDPE, and their mixture (mass ratio, 1:1) was performed at temperatures 400, 450, 500, 550, and 600 °C. Initial substrates and pyrolysis products were characterized by thermogravimetric analysis (TGA), gas chromatography–mass spectrometry (GC–MS), specific carbon isotope analysis of individual hydrocarbons (δ13C), Rock-Eval pyrolysis, and elemental analysis. The positive synergetic effect during co-pyrolysis of lignite/HDPE mixture was observed at temperatures ≥450 °C, with the greatest being at 500 °C. The highest yield of liquid co-pyrolysis products with a similar composition to that of crude oils is also noticed at 500 °C. The yields of liquid and gaseous products and quality of pyrolytic products obtained by co-pyrolysis of lignite/HDPE mixture are notably improved compared with pyrolysis of lignite alone. On the other hand, data obtained from pyrolysis of HDPE alone indicate that it cannot be concurrent to well-developed catalytic thermal processes for polymer recycling. However, concerning the huge amount of produced HDPE, at least part of this plastic material can be reused for advanced thermal treatment of lignite, particularly in countries where this low-rank coal represents the main source of energy.

AB  - Non-covalent selenium–selenium interactions between selenium-containing organic molecules were studied in crystal structures from the Cambridge Structural Database and by high-level quantum chemical calculations. Se⋯Se contacts in the crystal structures were analyzed, and the most frequent patterns were identified and used to design the model systems for quantum chemical calculations. The strongest calculated Se⋯Se interaction (ΔECCSD(T)/CBS = −2.31 kcal mol−1) was identified in the model system with a mutual parallel orientation of interacting molecules. In the crystal structures, this orientation of molecules is predominant. In the geometry with the σ-hole bonding, the interaction is somewhat weaker (ΔECCSD(T)/CBS = −2.13 kcal mol−1). NCI analysis showed that Se⋯Se interaction in the most stable geometries is further enhanced by hydrogen bonding of Se–H⋯Se or C–H⋯Se type. The results of energy decomposition analysis (SAPT) calculations revealed that the nature of the Se⋯Se interaction is predominantly dispersive with a strong electrostatic contribution. The results of the energy decomposition analysis also suggest that the electrostatic component has a crucial role in defining the geometry of selenium–selenium interactions due to their directional nature.

AB  - Solid-state reaction between BaTiO3and Fe2O3was used to produce a multiferroic heterostructure composite. Commercial BaTiO3and Fe(NO3)3•9H2O were suspended in ethanol for 30 minutes in an ultrasound bath. The prepared mixture was thermally processed at 300°C for 6 h. Sintering at 1300 °C for 1 h resulted in a mixture of different phases, BaTiO3, BaFe12O19and Ba12Ti28Fe15O84, which were confirmed by x-ray powder diffraction. A dense microstructure with a small volume fraction of closed porosity was indicated by the scanning electron microscopy, while a homogeneous distribution of Fe ions over BaTiO3phase was visible from energy dispersive spectroscopy mapping. Doping of BaTiO3with Fe2O3resulted in formation of magnetic hexaferrite phases, as confirmed by dielectric measurements that showed a broadened maximum of the permittivity measured as a function of temperature. © 2021 Authors. In our study, the main volatiles and aroma compounds of 108 non-aged plum brandies originating from three plum cultivars, and fermented using different conditions, were investigated. The chemical profiles obtained after two-step GC-FID-MS analysis were subjected to multivariate data analysis to reveal the peculiarity in different cultivars and fermentation process. Correlation of plum brandy chemical composition with its sensory characteristics obtained by expert commission was also performed. The utilization of PCA and OPLS-DA multivariate analysis methods on GC-FID-MS, enabled discrimination of brandy samples based on differences in plum varieties, pH of plum mash, and addition of selected yeast or enzymes during fermentation. The correlation of brandy GC-FID-MS profiles with their sensory properties was achieved by OPLS multivariate analysis. Proposed workflow confirmed the potential of GC-FID-MS in combination with multivariate data analysis that can be applied to assess the plum brandy quality.

AB  - Antioxidants present in infant food contribute to its nutritive value and provide protection from oxidative damage. Here, a direct current polarographic method, based on a decrease of anodic current from the hydroxoperhydroxo-mercury(II) complex (HPMC) formed at a mercury dropping electrode at its potential of dissolution, was used to determine the total antioxidant capacity (TAC) of infant formulas, human milk collected from mothers of full-term and preterm infants, human milk supplemented with fortifier, and cow milk. TAC of milk samples and infant formula correlated with protein content measured by the Bradford method (Pearson correlation coefficient 0.605, p<0.001), while TAC of human milk correlated with content of free thiol groups measured by the Ellman method (0.779, p<0.001). The results obtained showed that, in contrast to the 2,2-diphenyl-1-picrylhydrazyl (DPPH) assay, HPMC enables specific insight into the quality of infant milk food which reflects the amounts of free thiol groups and protein content. The significant contribution of milk proteins and the most prevalent amino acids, particularly cysteine, arginine, and phenylalanine, to TAC was shown. Thus, the HPMC method might be useful in monitoring the quality of infant formulas and human milk for preterm infants before and after supplementation with fortifier.

The structure of synthesized derivatives was confirmed with NMR and FT–IR spectroscopy, and with elemental analysis. For one of the compounds, single-crystal X-ray diffraction analysis was performed. Specific and non-specific molecular interactions were interpreted by LSER principles, using Catalan’s model. For additional information about the dominance and influence of the interactions presented, correlations with Hansen’s solubility parameters were calculated. Influence of the type and position of the substituent on absorption maxima was determined with LFER (linear free-energy relationship) principles, using Hammett’s equation. Acidity constants of the synthesized compounds were theoretically calculated and experimentally determined. AB  - The Pd-catalyzed N-arylation method for the synthesis of eighteen N,1-diaryl-1H-tetrazol-5-amine derivatives is reported. By running the reactions at 35 °C, compounds were isolated as single isomers since the undesired Dimroth rearrangement was completely suppressed. Furthermore, the Dimroth rearrangement of N,1-diaryl-1H-tetrazol-5-amines was rationalized by conducting comprehensive experiments and NMR analysis as well as density functional theory (DFT) calculations of thermodynamic stability of the compounds. It was established that the Dimroth rearrangement is thermodynamically controlled, and the equilibrium of the reaction is determined by the stability of the corresponding isomers. The mechanism was investigated by additional DFT calculations, and the opening of the tetrazole ring was shown to be the rate-determining step. By maneuvering Pd-catalyzed N-arylation and the subsequent Dimroth rearrangement, two more N,1-diaryl-1H-tetrazol-5-amine derivatives were acquired, which otherwise cannot be synthesized by employing the C–N cross-coupling reaction.
