AB  - Lipophilicity and ionization constant of sixteen fentanyl analogues were investigated by reversed-phase thin-layer chromatography (RP-TLC). Fourteen compounds have nitrogen containing groups at C-3 position of thepiperidine ring and two compounds are 3-carbomethoxy derivatives of fentanyl. Among them, five are diaste-reomeric cis/trans couples and six novel trans diastereomers. In addition, the lipophilicity and ionization constantof fentanyl were also determined by the same method, as a reference. The physicochemical property, lip-ophilicity, expressed as retention indices RM0 , b, and C0, as well as PC1, was determined and correlated with insilico values. Ionization constants were determined on the basis of the relationships between analyte’s retentionexpressed as RF and mobile phase pH. Calculated structural descriptors together with the retention indices, weresubjected to the principal component analysis – PCA and hierarchical cluster analysis – HCA, in order to providebasic insights into the similarities among the studied compounds. The blood – brain barrier (BBB) permeationwas investigated in the function of experimentally obtained values of lipophilicity, polar surface area and mo-lecular weight. In general, results of the present research corroborate well with previously determined anti-nociceptive activities of the investigated compounds, pointing out that besides the cis/trans isomerism, anotherset of important parameters should be taken into account when designing new derivatives of C-3 substitutedfentanyl, especially lipophilicity, voluminosity and HBD/A character of the substituents. Accordingly, RP-TLCcan be considered as a valuable asset in the ligand-based drug design

AB  - Considering the health-benefits of berry fruits consumption and increased market demands for food authenticityas one of the most important quality assurances, phenolic profiling by high-performance thin layer chroma-tography and ultra-high-performance liquid chromatography hyphenated with mass spectrometry was combinedwith multivariate analysis for phytochemical characterization and intercultivar discrimination of cultivatedberry seeds. The phenolic profiles of 45 berry seeds from nine genuine Serbian cultivated fruit species (straw-berry, raspberry, blackberry, black currant, blueberry, gooseberry, cape gooseberry, chokeberry, and goji berry)revealed a good differentiation according to botanical origin. In order to determine biomarkers responsible forthe classification, a total of 103 phenolic compounds were identified, including 53 phenolic acids and theirderivatives, 26 flavonoids and 24 glycosides. Biomarkers derived from the phenolic profile of berry seeds provedto be a powerful tool in the authentication of botanical origin, and may be useful in detection of frauds in berry-based seed-containing product.

AB  - An electrochemical approach is presented based on multiwall carbon nanotubes (MWCNTs) and neodymium(III) hydroxide (Nd(OH)3) nanoflakes for detection of bovine serum albumin (BSA). The materials were characterized morphologically (XRPD, SEM, and HR-TEM) and electrochemically (DPV, EIS). The MWCNTs@Nd(OH)3 composite was used as support for bovine serum albumin polyclonal antibody (anti-BSA). After the antibody immobilization on the electrochemical platform and antigen/antibody binding time (optimum 60 min), the proposed approach shows a linear voltammetric response toward BSA concentration in the range 0.066 to 6.010 ng mL−1 at maximum peak potential of 0.13 V (vs. Ag/AgCl). Limit of detection (LOD) and limit of quantification (LOQ) were 18 pg mL−1 and 61 pg mL−1, respectively. The precision of the method calculated as relative standard deviation (RSD) of five independent measurements was better 3%. The selectivity of the optimized method regarding structurally similar proteins (human serum albumin and human hemoglobin), ions (Na+, K+, Ca2+, and NO2−), or compounds (glucose, ascorbic acid, dopamine, uric acid, paracetamol, and glycine) was found to be satisfactory, with the current changes of less than 5% in the presence of up to 1 × 105 times higher concentrations (depending on the compound) of the listed potential interfering compounds. Practical applicability of immunosensor for BSA determination in cow whey sample, with recovery values in the range 97 to 103%, shows that the developed method has high potential for precise and accurate detection of BSA, as well as exceptional miniaturization possibilities for on-site and equipment-free sensing.

AB  - An electrochemical approach is presented based on multiwall carbon nanotubes (MWCNTs) and neodymium(III) hydroxide (Nd(OH)3) nanoflakes for detection of bovine serum albumin (BSA). The materials were characterized morphologically (XRPD, SEM, and HR-TEM) and electrochemically (DPV, EIS). The MWCNTs@Nd(OH)3 composite was used as support for bovine serum albumin polyclonal antibody (anti-BSA). After the antibody immobilization on the electrochemical platform and antigen/antibody binding time (optimum 60 min), the proposed approach shows a linear voltammetric response toward BSA concentration in the range 0.066 to 6.010 ng mL−1 at maximum peak potential of 0.13 V (vs. Ag/AgCl). Limit of detection (LOD) and limit of quantification (LOQ) were 18 pg mL−1 and 61 pg mL−1, respectively. The precision of the method calculated as relative standard deviation (RSD) of five independent measurements was better 3%. The selectivity of the optimized method regarding structurally similar proteins (human serum albumin and human hemoglobin), ions (Na+, K+, Ca2+, and NO2−), or compounds (glucose, ascorbic acid, dopamine, uric acid, paracetamol, and glycine) was found to be satisfactory, with the current changes of less than 5% in the presence of up to 1 × 105 times higher concentrations (depending on the compound) of the listed potential interfering compounds. AB  - Chitosan (Ch)-derived from biowaste along with smectite, an abundant clay mineral, were used in a low-cost and eco-friendly synthesis of a new type of catalyst. Nanocomposite catalysts constituted of Co supported on smectite with chitosan-derived carbon loading were obtained using an impregnation‑carbonization procedure and denoted as Co/cCh-S-T (T stands for applied carbonization temperature). The carbonization was performed in the temperature range from 400 °C to 700 °C in the flow of N2 providing inert atmosphere. The temperature of 500 °C was found to be the most suitable for catalyst synthesis regarding catalytic performance in a peroxymonosulfate activated degradation of tartrazine. The incorporation of carbonized chitosan structure within the interlamellar space of the smectite was confirmed using X-ray powder diffraction. The high-resolution transmission electron microscopy confirmed a layered structure of nanocomposites characteristic for smectite, as well as the presence of small spherical cobalt containing nanoformations (confirmed by energy dispersive X-ray spectroscopy) well dispersed within structure. The existance of cobalt in the CoII and CoIII oxidation state was proven by X-ray photoelectron spectroscopy. The Co/cCh-S-500 catalyst was proven to be stable and efficient after 5 consecutive cycles. This work showed that nanocomposite Co-catalysts, based on smectite and biowaste-derived carbon, as peroxymonosulfate activators exhibited a very promising performance in the degradation of water pollutants.

Probing the stability of the food colourant R-phycoerythrin from dried Nori flakes

AB  - The high content of vitamins, minerals, antioxidants, and proteins makes red algae Porphyra sp. (Nori) superfood with exceptional health-promoting benefits. Its intense colour originates from R-phycoerythrin (R-PE), phycobiliprotein containing covalently attached tetrapyrrole chromophores: red phycoerythrobilin and orange phycourobilin. The present study aims to characterize the stability of R-PE, a natural colourant with a high potential for application in the food, cosmetic, and pharmaceutical industries. We purified R-PE from dried Nori flakes with a high purity ratio (A560 /A280 ≥5). Far-UV CD spectroscopic showed that α-helix is the dominant secondary structure (75%). The thermal unfolding of α-helix revealed two transitions (Tm1 and Tm2 at 56 and 72°C, respectively), ascribed to the different subunits of R-PE. Absorption measurements showed that high pressure (HP) induces dissociation of R-PE into subunits followed by subunit unfolding. Contrary to temperature, HP treatment showed a significant advantage under applied conditions: the protein unfolding is partly reversible, and the R-PE colour bleaching is minimized. Based on the fluorescence quenching approach, R-PE's binding affinities for Cu2+ and Zn2+ ions were 6.27x105 and 1.71x103 M-1, respectively. Absorption and near-UV/VIS CD spectroscopy suggested conformational changes in protein chromophores upon metal ions binding. Far-UV CD spectroscopy did not reveal that metal binding affects R-PE structure. The obtained results give new insights into the stability of R-PE with a good use-value in replacement of toxic synthetic dyes, preservation of R-PE red colour in fortified food and beverages by HP processing, and as a biosensor for Cu2+ in aquatic life systems.

The present study aims to characterize the stability of R-PE, a natural colourant with a high potential for application in the food, cosmetic, and pharmaceutical industries. We purified R-PE from dried Nori flakes with a high purity ratio (A560 /A280 ≥5). Far-UV CD spectroscopic showed that α-helix is the dominant secondary structure (75%). The thermal unfolding of α-helix revealed two transitions (Tm1 and Tm2 at 56 and 72°C, respectively), ascribed to the different subunits of R-PE. Absorption measurements showed that high pressure (HP) induces dissociation of R-PE into subunits followed by subunit unfolding. Contrary to temperature, HP treatment showed a significant advantage under applied conditions: the protein unfolding is partly reversible, and the R-PE colour bleaching is minimized. Based on the fluorescence quenching approach, R-PE's binding affinities for Cu2+ and Zn2+ ions were 6.27x105 and 1.71x103 M-1, respectively. Absorption and near-UV/VIS CD spectroscopy suggested conformational changes in protein chromophores upon metal ions binding. Far-UV CD spectroscopy did not reveal that metal binding affects R-PE structure. The obtained results give new insights into the stability of R-PE with a good use-value in replacement of toxic synthetic dyes, preservation of R-PE red colour in fortified food and beverages by HP processing, and as a biosensor for Cu2+ in aquatic life systems.

AB  - The strawberry tree (Arbutus unedo L., Ericaceae family) is an evergreen Mediterranean
shrub whose leaves and fruits are used in traditional medicine due to their antioxidant, antimicrobial,
antidiabetic, diuretic, and antiproliferative properties. The health benefits are mainly attributed to the
presence of phenolic compounds. The aim of this study was to compare the phenolic profiles, total
phenolic content (TPC), and radical scavenging activity (RSA) of A. unedo leaves and fruits collected
at two locations in Croatia. Phenolic profiles were identified using an ultra-high-performance liquid
chromatograph (UHPLC) coupled with a hybrid mass spectrometer (LTQ Orbitrap MS). TPC was
determined by Folin–Ciocalteu’s assay, while RSA was investigated using DPPH reagent. A total of
64 phenolics (60 and 42 compounds in leaves and fruits, respectively) were identified. Hyperoside
and flavan-3-ols were predominant compounds in leaves, while gallocatechin and catechin were
the major compounds found in fruits. To the authors’ knowledge, 16 and 5 phenolics in leaves and
fruits, respectively, were reported for the first time. Principal component analysis (PCA) showed
that UHPLC-LTQ Orbitrap MS could be used to identify which phenolics were able to discriminate
samples regarding plant tissue and geographical origin. TPC in leaves and fruits were in the ranges of
67.07–104.74 and 16.78–25.86 mg gallic acid equivalents (GAE)/g dried weight (dw), respectively. RSA
for leaves and fruits were in the ranges of 408.92–430.98 and 74.30–104.04 µmol Trolox equivalents
(TE)/g dw, respectively. The number of identified phenolics was lower in fruits compared to leaves.
Such a large number of bioactive phenolics identified and the strong antioxidant activity pointed to
A. unedo as a promising health-promoting plant and natural food preservative.

AB  - To replace common organic solvents that present inherent toxicity and have high volatility and to improve the extraction efficiency, a range of natural deep eutectic solvents (NADESs) were evaluated for the extraction of phenolic compounds from Agrimonia eupatoria. Screening of NADES efficiency was carried out based on the total phenolic and flavonoid content and radical-scavenging activity, determined by spectrophotometry, as well as phenolic compounds quantified, obtained using ultra-high-performance liquid chromatography with a diode array detector and a triple-quadrupole mass spectrometer. Increased extraction efficiency when compared with organic solvent was achieved using NADES mixtures choline chloride (ChCl):urea 1:2 and choline chloride:glycerol 1:1. Flavonol glycosides were the most abundant compounds in all extracts. The COSMO-RS model provided insights into the most important intermolecular interactions that drive the extraction process. Moreover, it could explain the extraction efficiency of flavonol glycosides using ChCl:glycerol NADES. The current article offers experimental evidence and mechanistic insights for the selection of optimal NADES to extract bioactive components from Agrimonia eupatoria. © 2022 by the authors.

AB  - This paper aims to develop an amperometric, non-enzymatic sensor for detecting and quantifying UA as an alert signal induced by allergens with protease activity in human cell lines (HEK293 and HeLa). Uric acid (UA) has been classified as a damage-associated molecular pattern (DAMP) molecule that serves a physiological purpose inside the cell, while outside the cell it can be an indicator of cell damage. Cell damage or stress can be caused by different health problems or by environmental irritants, such as allergens. We can act and prevent the events that generate stress by determining the extent to which cells are under stress. Amperometric calibration measurements were performed with a carbon paste electrode modified with La(OH)3@MWCNT, at the potential of 0.3 V. The calibration curve was constructed in a linear operating range from 0.67 μM to 121 μM UA. The proposed sensor displayed good reproducibility with an RSD of 3.65% calculated for five subsequent measurements, and a low detection limit of 64.28 nM, determined using the 3 S/m method. Interference studies and the real sample analysis of allergen-treated cell lines proved that the proposed sensing platform possesses excellent sensitivity, reproducibility, and stability. Therefore, it can potentially be used to evaluate stress factors in medical research and clinical practice.

Furthermore, the association between anti-SARS-CoV-2 specific IgG antibodies and ETE levels with vaccine responsiveness has not been investigated. Methods: The present study explored the status of ETEs (Mn, Cu, Zn, and Se) in sera of healthy women before and after vaccination with the anti-SARS-CoV-2 BNT162b2 mRNA vaccine in a follow-up period of six months. The main aim was to explore links between ETE levels and IgG antibodies produced against Spike glycoprotein's Receptor-Binding Domain (RBD). Results: A recombinant protein of SARS-CoV-2 comprising the receptor binding domain was successfully expressed in HEK-293 T cells. The purified protein was suitable for producing a sensitive antibody detection assay for human serum and monitored seropositivity, indicating a transient response with peak anti-SARS-CoV-2 IgG levels 2 months after vaccination. In parallel to increasing antibody titers, serum concentrations of Cu, Mn, and Se were not affected by vaccination, and concentrations remained relatively constant at the different sampling times during the 6-month observation period. Total serum Zn concentrations were slightly elevated when compared between the first and last sampling dates. Overall, no consistent effects of vaccination on any of the three trace elements analyzed in our study were observed. Conclusion: Vaccination of adult healthy female volunteers with an mRNA vaccine was not associated with consistent changes in serum trace element concentrations over a six-month observation period.

AB  - This paper aims to develop an amperometric, non-enzymatic sensor for detecting and quantifying UA as an alert signal induced by allergens with protease activity in human cell lines (HEK293 and HeLa). Uric acid (UA) has been classified as a damage-associated molecular pattern (DAMP) molecule that serves a physiological purpose inside the cell, while outside the cell it can be an indicator of cell damage. Cell damage or stress can be caused by different health problems or by environmental irritants, such as allergens. We can act and prevent the events that generate stress by determining the extent to which cells are under stress. Amperometric calibration measurements were performed with a carbon paste electrode modified with La(OH)3@MWCNT, at the potential of 0.3 V. The calibration curve was constructed in a linear operating range from 0.67 μM to 121 μM UA. The proposed sensor displayed good reproducibility with an RSD of 3.65% calculated for five subsequent measurements, and a low detection limit of 64.28 nM, determined using the 3 S/m method. Interference studies and the real sample analysis of allergen-treated cell lines proved that the proposed sensing platform possesses excellent sensitivity, reproducibility, and stability. Therefore, it can potentially be used to evaluate stress factors in medical research and clinical practice.

AB  - Background: Although essential trace elements (ETEs) play pivotal roles in life-supporting biochemical processes, their function in innate and adaptive immunity has not been fully elucidated, particularly during immunization. Furthermore, the association between anti-SARS-CoV-2 specific IgG antibodies and ETE levels with vaccine responsiveness has not been investigated. Methods: The present study explored the status of ETEs (Mn, Cu, Zn, and Se) in sera of healthy women before and after vaccination with the anti-SARS-CoV-2 BNT162b2 mRNA vaccine in a follow-up period of six months. The main aim was to explore links between ETE levels and IgG antibodies produced against Spike glycoprotein's Receptor-Binding Domain (RBD). Results: A recombinant protein of SARS-CoV-2 comprising the receptor binding domain was successfully expressed in HEK-293 T cells. The purified protein was suitable for producing a sensitive antibody detection assay for human serum and monitored seropositivity, indicating a transient response with peak anti-SARS-CoV-2 IgG levels 2 months after vaccination. In parallel to increasing antibody titers, serum concentrations of Cu, Mn, and Se were not affected by vaccination, and concentrations remained relatively constant at the different sampling times during the 6-month observation period. AB  - Compounds used in the fashion industry effect the water bodies in the vicinity of textile factories, resulting in the visible coloration of surface water. Fe-doped graphite-based in house prepared electrodes were used in the Fenton--like degradation of Reactive Blue 52 (RB52). The electrodes consisting of high-density graphite in three granulation sizes and three levels of Fe content were characterized using scanning electron microscopy (SEM). The amount of Fe in the electrodes and H2O2 concentration in syn­thetic textile wastewater were optimized. Additionally, the size of graphite grains was varied to inves­tigate whether it effects the degradation rate. Under only 10 min of electro-Fen­ton degradation, a system with 10 mmol dm-3 of H2O2 and an electrode made of 7 % of Fe and 70 µm of granulation size of graphite, degraded over 75 % of RB52, and over 99 % after 40 min of treatment. The obtained results indicate that the proposed approach could be beneficial in the field of novel materials for environmental applic­ation and that in house prepared carbon could be an excellent replacement for commercially available supports.

тилних фабрика, што резултира видљивим обојењем површинских вода. Домаће елек-
троде на бази графита допиране гвожђем биле су укључене у деградацију Reactive Blue
52 (RB52) механизма попут Фентонове реакције. Електроде су се састојале од графита
велике густине у три величине гранулације и три количине Fе и окарактерисане су
помоћу скенирајуће електронске микроскопије (SEM). Оптимизоване су количина Fe у
електродама и концентрација H2O2 у синтетичкој текстилној отпадној води. Додатно,
величина графитних зрна је варирана како би се испитало да ли утиче на брзину раз-
градње. За само 10 min електро-Фентонове деградације систем са 10 mmol dm-3 H2O2 и
електродом од 7 % Fе и 70 μm величине гранулације графита разградило се преко 75 %
RB52 и преко 99 % након 40 min третмана. Добијени резултати указују на то да пред-
ложени приступ може бити користан у области нових материјала за примену у животној
средини и да домаће припремљени угљеник може бити одлична замена за комерцијално
доступне носаче.

AB  - A series of novel polyurethane network/organoclay nanocomposites (PUN-NCs) with different soft segment contents (30–60 wt%) was prepared by in situ polymerization in solution and characterized. PU network (PUN) was made from poly(dimethylsiloxane)-based macrodiol as the soft segment and 4,4′-methylenediphenyldiisocyanate and hyperbranched polyester of the third pseudo generation as the hard segment. Nanocomposites were obtained by dispersion of organically modified montmorillonite (Cloisite 30B) nanofiller (0.5 wt%). The influence of the soft segment content on the functional properties of PUN-NCs was studied by Fourier transform infrared (FTIR), small-angle and near wide-angle X-ray scattering (SWAXS), thermogravimetric analysis (TGA), dynamic mechanical thermal analyses (DMTA), differential scanning calorimetry (DSC), nanoindentation, atomic force microscopy (AFM), scanning electron microscopy (SEM), and swelling behavior, water absorption and contact angle measurements. The biodegradation process was evaluated using mixed cultures of microorganisms that originated from soil. Mechanically strong PUN-NC materials in the form of films were obtained, pointing to good dispersion and the existence of exfoliated morphology of Cloisite 30B within the PUN matrix, and the nanocomposites with the abovementioned characteristics were obtained as a function of the soft segment content. The decrease of the soft segment content induced a higher degree of phase separated microstructure, increase of Young's modulus, hardness, plasticity, storage modulus, glass transition temperature, surface free energy and swelling ability in tetrahydrofuran, but at the same time, it is responsible for the decrease of crosslinking density and hydrophobicity of PUN-NCs. By choosing adequate soft segment content, the prepared materials can potentially be designed for coating applications, such as top coating materials in environmental conditions.

AB  - The problem of wastewater has long been ubiquitous and has great consequences for the environment and its inhabitants. Microbial fuel cells (MFCs) have enormous potential for the treatment of wastewaters polluted with azo dyes. The amount of energy that can be produced from a single-chamber MFC is sufficient to perform decolorization and degradation of such dyes, which are widely used in the textile industry. This study on the azo dye, reactive black 5 (RB5), provides an alternative method through three parallel-connected MFCs to obtain electricity that directly serves for the dye’s electrochemical degradation. We examined degradation followed by decolorization of RB5 using Fe and Pt electrodes, together with H2O2, to achieve the electro-Fenton process. The amount of voltage produced (295 mV), the current density (276 mA m−3) and the power density (50 mW m−3) were sufficient to degrade 25 mg L-1 RB5 dye with 0.5 mM H2O2 in just 2 h. The dye degradation mechanism was investigated using UV–VIS, FT-IR and HPLC-MS/MS. The ecotoxicity of the degradation products was assessed using a bacterial model, Aliivibrio fischeri. These tests showed that there was successful degradation of the dye to products whose toxicity is less than that of RB5.

These oils, originating from 13 Serbian oil fields in SE Pannonian Basin, differ according to source and depositional envi­ronment of organic matter (OM), as well as by thermal maturity and biodeg­rad­ation stage. Methylene and methyl asymmetric stretching peak absorbances were used for the branching factor calculation. CH2 peak positions exhibited3–4 cm-1 red shift with increasing the CH2/CH3 ratio, due to a greater contri­bution of trans vs. gauche rotamers in ali­pha­tic chains. Comparing IR spectra of the oils and model n-alkanes, it was established that the average (CH2)n methylene chain length per СН3 group varied from n = 3.5 to 6.5. The CH2/CH3 ratio showed significant concordance with geochemical para­meters, enabling clear distinction of the oils according to source and deposi­ti­onal environment of OM. At the same time, dependence of the CH2/CH3 ratio on oil maturity in the range from immature to mature was not observed, allowing for an accurate determination of oil genetic types irrespective of maturity. The CH2/CH3 ratio showed good accordance with oil biodegradation scale and oil viscosity.

AB  - Presented research aimed to develop an alternative approach for the estimation of biosorption capability of sunflower husk. The acid-pretreated sunflower biomass was characterized by scanning electron microscopy combined with energy dispersive X-Ray spectroscopy (SEM-EDX) analysis and Fourier transform infrared (FTIR) spectroscopy. Biosorption efficiency has been evaluated using inductively coupled plasma optical emission spectroscopy (ICP-OES) and laser-induced breakdown spectroscopy (LIBS). The adsorption capacity of the adsorbent was determined as the function of the pH of the solution, the initial concentration of heavy metal solutions, and contact time. The optimal conditions were achieved after 15 min of contact at pH 6, while the percentage of the removal was from 80.0–98.7 %, depending on the element. The results obtained from the kinetic and isotherm studies show that maximum adsorption of ions was quickly reached and followed the pseudo-second-order kinetic model. Real samples were tested and obtained Ni values by LIBS method were 3100±200 and 1240±100 mg kg-1, while estimated values by the ICP-OES were 2995±20 and 1130±10 mg kg-1, respectively. The obtained results prove that LIBS method can be used as a “green alternative“for the evaluation of biosorption efficiency.

AB  - Lipophilicity and ionization constant of sixteen fentanyl analogues were investigated by reversed-phase thin-
layer chromatography (RP-TLC). Fourteen compounds have nitrogen containing groups at C-3 position of the
piperidine ring and two compounds are 3-carbomethoxy derivatives of fentanyl. Among them, five are diaste-
reomeric cis/trans couples and six novel trans diastereomers. In addition, the lipophilicity and ionization constant
of fentanyl were also determined by the same method, as a reference. The physicochemical property, lip-
ophilicity, expressed as retention indices RM
0 , b, and C0, as well as PC1, was determined and correlated with in
silico values. Ionization constants were determined on the basis of the relationships between analyte’s retention
expressed as RF and mobile phase pH. Calculated structural descriptors together with the retention indices, were
subjected to the principal component analysis – PCA and hierarchical cluster analysis – HCA, in order to provide
basic insights into the similarities among the studied compounds. The blood – brain barrier (BBB) permeation
was investigated in the function of experimentally obtained values of lipophilicity, polar surface area and mo-
lecular weight. In general, results of the present research corroborate well with previously determined anti-
nociceptive activities of the investigated compounds, pointing out that besides the cis/trans isomerism, another
set of important parameters should be taken into account when designing new derivatives of C-3 substituted
fentanyl, especially lipophilicity, voluminosity and HBD/A character of the substituents. Accordingly, RP-TLC
can be considered as a valuable asset in the ligand-based drug design

Plant Metabolomics as a Tool for Detecting Adulterants in Edible Plant: A Case Study of Allium ursinum

AB  - Allium ursinum and poisonous adulterants Convallaria majalis and Arum maculatum were used as a model for detection of adulterants in edible plant. A. ursinum samples were spiked with C. majalis and A. maculatum to mimic adulteration. Metabolomic fingerprinting of all samples was performed using 1H NMR spectroscopy, and the resulting data sets were subjected to multivariate data analysis. As a result of this analysis, signals of adulterants were extracted from the data, and the structures of biomarkers of adulteration from partially purified samples were elucidated using 2D NMR and LC-MS techniques. Thus, isovitexin and vicenin II, azetidine-2-carboxylic acid, and trigonelline indicated adulteration of A. ursinum samples with C. majalis. Isovitexin was also recognized to be an indicator of adulteration of A. ursinum with A. maculatum. In conclusion, the case study of A. ursinum suggested that plant metabolomics approach could be utilized for identification of low molecular weight biomarkers of adulteration in edible plants. © 2022 by the authors.

AB  - Raspberries are considered valuable fruits due to their high levels of nutrients and phytochemicals, which have many beneficial effects on humans. As many external factors affect the composition of these fruits (the type of cultivation, soil characteristics, ripeness, storage time and post-harvest technologies, cultivar/genotype, and climatic conditions), the goal of this study was to analyze different raspberry cultivars grown in Norway. Considering that Norway is a country with specific climatic conditions, as well as has a limited period of fruit vegetation, another important goal of this study was also to compare raspberries from different Norwegian areas, as well as different grown cultivars. Modern analytical techniques, such as high-performance anion-exchange liquid chromatography with pulsed amperometric detection (HPEAC-PAD), ultra-high-performance liquid chromatography with diode array detector coupled to triple quadrupole mass spectrometry (UHPLC-DAD MS/MS), and inductively coupled plasma–optical emission spectrometry (ICP-OES), provided a detailed examination of the raspberry extract samples. Based on their high levels of minerals (especially N, P, and K), organic acids (predominantly citric and malic acids), sugars (glucose, fructose, sucrose, and galactose), and polyphenols (ellagic acid, syringic acid, quercetin, and rutin), Norwegian raspberries could be considered fruits with increased health-beneficial compounds. AB  - Most of the food allergens sensitized via the gastrointestinal tract resist thermal treatments and digestion, particularly digestion by pepsin. Roasted hazelnuts are more commonly consumed than raw ones. Since no studies have characterized gastric digestion protein fragments of raw and roasted hazelnuts nor their IgE binding properties, we compared these aspects of raw and roasted hazelnuts’ gastric digesta obtained by INFOGEST protocol. Their electrophoretically resolved profiles were probed with hazelnut allergic patients’ sera in 1D and 2D immunoblots. Electrophoretic profiles demonstrated pepsin digestion of all hazelnut allergens to varying extents. While 2D immunoblots indicated that roasting slightly reduced allergenicity, IgE ELISA with the pool of sera showed a slight significant (10%) increase in IgE binding in both gastric digesta. Cor a 9 isolated from the raw and roasted hazelnuts, characterized by far and near CD, remained stable after roasting, with preserved IgE reactivity. Its immunoreactivity contribution by inhibitory ELISA was noticeable in raw and roasted hazelnut digesta; its activity was slightly stronger in the roasted preparations. Roasting has a visible impact on proteins; however, it did not affect overall IgE reactivity. Gastric digestion slightly increases the overall IgE reactivity in raw and roasted hazelnuts, and may therefore impact the profiles of allergens and their fragments available to interact with the immune system in the small intestine. © 2022 by the authors.
