AB  - In vivo measurements of the optical properties of plant leaves, by spectrophotometric methods, could potentially provide very useful information for the assessment of the plant’s health. This paper describes a novel experimental set-up that enables continuous measurements of the optical reflection and transmission coefficients of broad-leaved plants. The optical spectral characteristics of leaves, in the broad wavelength range, are often a discussed topic in the scientific literature. Nevertheless, to the best of our knowledge, the real-time spectral analyses of the leaf haven’t been reported so far. Monitoring the evolution of the plant activity in real-time has resulted in the graph of the spectral Circadian rhythm as a function of time. The set-up was tested on Phaseolus vulgaris and the results of the measurements are presented and discussed.

AB  - The plants from genus Salvia, as one of the largest genus in Lamiaceae family, are frequently in use for various purposes, as foods, in cosmetic industry, or in traditional and official medicine. Salvia verticillata L. (liliac sage) is one of sidelined sage species with potential bioactivity, reported in traditional medicine. The aim of this study was to acquire a phytochemical profile of the methanol extract obtained from S. verticillata aerial parts and to evaluate its antioxidant, antimicrobial, and biocompatibility potential. Characteristic compounds of the genus Salvia, such as rosmarinic and caffeic acids, along with their derivatives (e.g. salvianolic and yunnaneic acids isomers) and flavonoids, have been identified by ultrahigh-performance Orbitrap metabolomic fingerprinting as the main phenolic metabolites in S. verticillata. The extract displayed moderate antimicrobial properties and significant antioxidant potential, with the half maximal inhibitory concentration values (IC50) ranging from 33 to 73 μg/mL. Importantly, full biocompatibility of the extract with eukaryotic cell lines was observed up to 72 h. The obtained results revealed the presence of polyphenolic bioactive compounds in S. verticillata extract with promising antioxidant potential and significant biocompatibility. In this regard, S. verticillata can find new perspectives of application as a food ingredient, in cosmetic and pharmaceutical industries, as it represents a valuable source of compounds with prominent health properties, with a special focus on rosmarinic acid.

AB  - Vranac, an old autochthonous red grapevine variety of Montenegro, was first mentioned in a historical document published in the 15th century. As currently the study of indigenous varieties is of particular importance, the subject of this work was detailed characterization of phenolic compounds in the autochthonous grapevine variety Vranac, from the Montenegrin Podgorica subregion. Phenolic profiles of leaves, berries (skin, seeds, and pulp were examined separately) and young monovarietal wine were determined using ultra-high performance liquid chromatography (UHPLC) with linear trap quadrupole (LTQ)-Orbitrap XL mass spectrometry (MS). Total phenolic content (TPC) and radical scavenging activity (RSA) were higher for the grape seeds extracts, followed by extracts of grape skins and pulps. As expected, the total anthocyanin content (TAC) was higher in grape skin than in wine. A total of one hundred twenty nine compounds (forty two phenolic acids and their derivatives, twenty three flavan-3-ols, twenty one flavanols, five stilbenes and thirty eight anthocyanins) were identified in the investigated extracts. To our best knowledge, this is the first report of tentative identification of (epi)catechin 3-O-coumarate in grape seed and chalcan-flavan 3-ol dimers in wine and grape seed.

AB  - Background and objectives: Quinoa is considered to be a “natural functional food” due to a lot of bioactive compounds that are beneficial for human health by helping prevent the risks of different diseases. The aim of this study was to test the health-promoting characteristics of Puno and Titicaca quinoa seeds grown in Serbia by assessing their phenolic and flavonoid content, antioxidant activities, and potential anticancer effect on human colorectal cancer cells. Findings: Thirteen phenolic compounds were identified and quantified in the seeds of both cultivars, but their profile and concentration of individual phenolic compounds differed. Cytotoxic activities were present in both cultivars, whereas the pronounced concentration and time-dependent effects were more expressed in Puno extracts. Conclusions: The extracts of the seeds of both cultivars are a rich source of phenolic and flavonoid compounds and with high antioxidant activities. Potent anticancer activity against the human colorectal cancer was expressed in both investigated cultivars. Significance and novelty: The study confirmed the health benefit potential of Puno and Titicaca quinoa cultivars, grown in Serbia. These results are the first to demonstrate a potent anticancer activity of quinoa seed extract against the human colorectal cancer cell line HCT-116, as well as the presence of 7 new phenolic and flavonoid compounds.

AB  - The present study was principally aimed at ascertaining the differences in metabolomics profiles and biological activities between non-hydrolyzed (ME) and hydrolyzed methanol extract (HME) of Centaurium erythraea Rafn. UHPLC-MS/MS Orbitrap analysis showed that the enzymatic hydrolysis of the extract caused changes in beta-Dglycoside/aglycone ratio of both flavonoid and secoiridoid compounds. UHPLC/DAD/ + HESI - qqqMS characterization and/or quantification of secoiridoid glucosides (SGs) and their aglycones in both ME and HME revealed gentiopicral and erythrocentaurin as the major aglycones, the same metabolic products which appear after the hydrolysis of pure swertiamarin, the dominant secoiridoid glucoside of C. erythraea. SGs played an antioxidant role only in ABTS assay, whilst the remarkable antioxidant potential of C. erythraea methanol extract is ascribed chiefly to phenolics detected in it. Interestingly, antioxidant activities of swertiamarin and sweroside recorded in ABTS assay increased after the compounds have been hydrolyzed, which highlighted their possible antioxidant role during ingestion. Strong antimicrobial activities of ME and HME against a vast array of pathogens, which exceed the effects of the reference antibiotics and antimycotics, largely depended on the amount of secoiridoids in either of the glycosylation forms. Extracts and pure secoiridoids were especially effective against most of the tested Penicillium species. On the other hand, P. funiculosum has evolved an efficient mechanism of detoxification of sub-lethal concentrations of secoiridoid glucosides, involving their biotransformation and complete digestion. The presented findings will contribute to clarify the fate and role of the SGs after C. erythraea ingestion within in vivo systems, and to further promote this remarkable plant as a food preservation additive with significant health benefits.

UHPLC-MS/MS Orbitrap analysis showed that the enzymatic hydrolysis of the extract caused changes in beta-Dglycoside/aglycone ratio of both flavonoid and secoiridoid compounds. UHPLC/DAD/ + HESI - qqqMS characterization and/or quantification of secoiridoid glucosides (SGs) and their aglycones in both ME and HME revealed gentiopicral and erythrocentaurin as the major aglycones, the same metabolic products which appear after the hydrolysis of pure swertiamarin, the dominant secoiridoid glucoside of C. erythraea. SGs played an antioxidant role only in ABTS assay, whilst the remarkable antioxidant potential of C. erythraea methanol extract is ascribed chiefly to phenolics detected in it. Interestingly, antioxidant activities of swertiamarin and sweroside recorded in ABTS assay increased after the compounds have been hydrolyzed, which highlighted their possible antioxidant role during ingestion. Strong antimicrobial activities of ME and HME against a vast array of pathogens, which exceed the effects of the reference antibiotics and antimycotics, largely depended on the amount of secoiridoids in either of the glycosylation forms. Extracts and pure secoiridoids were especially effective against most of the tested Penicillium species. On the other hand, P. funiculosum has evolved an efficient mechanism of detoxification of sub-lethal concentrations of secoiridoid glucosides, involving their biotransformation and complete digestion. The presented findings will contribute to clarify the fate and role of the SGs after C. erythraea ingestion within in vivo systems, and to further promote this remarkable plant as a food preservation additive with significant health benefits.",

AB  - Species within the genus Centaurium readily hybridize and polyploid complexes are often seen in natural populations. We describe phytochemical profiles of newly discovered allohexaploid hybrid, here named Centaurium pannonicum, and its parental tetraploid species C. erythraea and rare C. littorale ssp. compressum. Our aim was to examine chemodiversity of these taxa in the area of Vojvodina (North Serbia) and to perform metabolomics search for chemical classifiers which would provide high resolution discrimination of parental and hybrid individuals. In sum, UHPLC-MS/MS Orbitrap metabolomics fingerprinting revealed seventy compounds in methanol extracts. Despite the lack of qualitative chemical novelty in hybrid plants, UHPLC-qqqMS targeted metabolomics approach, aimed at three secoiridoid compounds and seventeen phenolics, pointed to considerable differences in quantitative composition of these dominant compounds among the plant taxa studied. In addition to the difference in the ploidy levels, the hybrid taxon was well distinguished from both parental species based on metabolite profiles, and, for most individuals, positioned intermediately to the parental taxa in both PCA and hierarchical clustering. After optimizing and comparing several statistical learning methods, it was possible to narrow the number of taxonomic classifiers to five (three xanthones, one secoiridoid glycoside, and one phenolic acid), while increasing the differentiation resolution. The presented metabolomics approach will certainly, along with morphometrics and molecular genetics studies, have high impact on further elucidation of complex relationships among taxa within the genus Centaurium.

AB  - Cucumber (Cucumis sativus) fruit has been used in cuisine worldwide, while its leaves are rich in immunomodulatory compounds. Cucumber leaf extract (CLE) was characterized by the predominance of triterpenoids cucurbitacins and significant levels of phenolics. Effects of CLE on CD4(+) T helper (Th) cells and macrophages, as the major encephalitogenic cells in the autoimmunity of the central nervous system were investigated in our study. CLE potently inhibited production of major pathogenic Th cytokines: interferon-gamma and interleukin-17, as well as of nitric oxide and reactive oxygen species in macrophages. Antigen-presenting activity of macrophages and dendritic cells was also affected by CLE. The effects of CLE were co-incident with modulation of NFKB and p38 MAPK signaling. Concentrations of CLE used in vitro did not show toxic effects on zebrafish embryos. Moreover, CLE inhibited generation of encephalitogenic cells in vivo. These results demonstrate that CLE deserve further investigation on its anti-encephalitogenic therapeutic properties. (C) 2017 Elsevier Ltd. All rights reserved.

AB  - The polyphenolic profiles of leaves from three raspberry (Rubus idaeus L.) cultivars ('Willamette', 'Tulameen', 'Meeker') and one blackberry (Rubus fruticosus L) cultivar, ('Cganska Bestrna') grown in West Serbia were evaluated. To characterise the extracts, two hyphenated techniques were employed. Ultra-high performance liquid chromatography (UHPLC) coupled with an LTQOrbiTrap mass spectrometer was used to identify polyphenolics. A rapid and accurate procedure in negative ion mode using a gradient elution within 15 min was established. This allowed us to identify 83 compounds, mostly hydroxycinnamic acid esters, flavonol glycosides, flavan-3-ols and ellagic acid derivatives. Hydroxybenzoic acid derivatives were identified in all investigated samples. Quantification was achieved with a time selected reaction monitoring (tSRM) method using UHPLC coupled with a diode array detector (DAD) and connected to a triple-quadrupole mass spectrometer. Among 23 compounds that were quantitated using available standards, ellagic acid was found to be the main phenolic acid in the Rubus leaves [ranging from 1574 to 2875 mg kg(-1) dry weight (DW)]. The raspberry leaves had higher contents of catechin, epigallocatechin gallate, rutin, ferulic acid, chlorogenic acid and p-hydroxybenzoic acid than the blackberry leaf sample. On the other hand, there was more caffeic acid and aesculin in the blackberry leaf sample than in raspberry leaves. All leaves were characterized by a high total phenolic content (84.64-144.20 mg GAEg(-1) DW), favourable radical scavenging capacity (38.05-83.77%), adequate ferric ion reducing power (59.74-92.80 mu mol L-1 of an ascorbic acid solution), good superoxide anion radical scavenging activity (28.43-39.87%), but low metal chelating ability (1.55-8.86%). AB  - Introduction - Nepeta species contain a variety of secondary metabolites, including iridoid monoterpenes - nepetalactones and phenolic acids - that are considered the main bioactive constituents. This work represents the first attempt to comparatively explore variations in these two major groups of secondary metabolites within the genus. Objective - To develop an efficient analytical methodology for simultaneous analysis of nepetalactones and phenolic acids in methanol extracts of selected Nepeta species, and to evaluate its potential application in chemotaxonomic studies. Material and methods - A UHPLC combined with linear-trap quadrupole (LTQ) orbitrap MS method was used to characterise chemical diversity and complexity of phenolics among 12 selected Nepeta species. A targeted metabolomic approach using UHPLC coupled to a diode array detector (DAD) and combined with (+/-) heated electrospray ionisation (HESI) MS/MS was developed and validated for quantitative analysis of six hydroxycinnamic acid derivatives and four nepetalactones. Results - Phenolic profiling provided a valuable database of bioactive compounds in the plant group studied, including phenolic acids (hydroxybenzoic and hydroxycinnamic acids) and flavonoids (flavones, flavonols and flavanones). Principal component analysis and cluster analysis suggested the applicability of 10 targeted compounds as chemomarkers for chemotaxonomic studies. Pearson's correlation analysis revealed significant positive correlations between metabolites involved in different biosynthetic pathways (phenylpropanoid or monoterpenoid). Conclusion - The described targeted metabolomic approach proved to be highly beneficial in designing a phytochemical overview of the genus Nepeta, and might have applications in further clarification of phylogenetic relations. Furthermore, it has the potential to be implemented in a routine quality control of plant material and herbal preparations.

The quantitative data on the targeted compounds (25 phenolic compounds, 14 carbohydrates and 25 minerals) together with the TPC and RSA data served as a pool of variables for multivariate analysis, which provided useful information for the accurate authentication of unifloral sage honey and its discrimination from other unifloral types of honey. The proposed markers, together with chemometrics, could further contribute, as a powerful tool, to the quality control of Croatian unifloral S. officinalis honey and thus, possibly certify its commercial value.
