AB - This study was focused on in vitro screening of the total antioxidant activity of the selected extracts of the mushroom Coprinus comatus and quinic acid, one of their antidiabetic ingredients, by an uncommon electrochemical assay. Indeed, direct current (DC) polarographic HydroxoPerhydroxo Mercury(II) Complex (HPMC) assay based on decrease of anodic limiting current originating from HPMC formation in alkaline solutions of hydrogen peroxide at potential of mercury dissolution, observed upon gradual addition of antioxidants, was applied herein for the estimation of the natural products' antioxidativity. Quinic acid was found to exhibit most promising antioxidant potential (4.0 ± 0.2%µL-1) being ≈ 2-fold more active than the screened C. comatus extract samples. Actually, such a finding puts some light on the antioxidativity of cyclic polyols, well understimated class of organic compounds, compared to aromatic (poly)phenolics. As a low cost, easy-to handle and accurate this polarographic assay may be thoroughly recommended for much broader use. [Formula: see text].

AB - BACKGROUND: Cereal seeds, such as maize seeds, are frequently contaminated with aflatoxin B1 (AFB1), one of the most dangerous naturally occurring carcinogens. In plants, phenolamides are involved in biotic stress response. The data on variations of phenolamides in AFB1-containing seeds are limited. RESULTS: Five polyamine conjugates, including two spermidine and three putrescine conjugates, were tentatively identified in methanolic extracts, using HPLC-DAD-MS. The ratio of putrescine to spermidine conjugates changed with increasing AFB1 concentration in a logistic dose–response manner, with a ratio of below 1 up to a concentration of 51.51 μg kg−1, and approximately 2.54 and 3 at higher concentrations of 177.4 and 308.13 μg kg−1, respectively. The observed variations of the total antioxidant activity and the total phenolic content may support this biphasic behaviour of the seeds against AFB1 stress. CONCLUSIONS: The obtained data are a contribution to the understanding of the roles of polyamine conjugates in seed defence to increasing AFB1 concentrations. According to our knowledge, this study reports for the first time the biphasic response of maize seeds to increasing AFB1 contamination level, comprising the induction of polyamine conjugate accumulation and variation in the ratio of conjugates. This dose–response relationship may provide useful information in the field of agricultural and food chemistry as an indicator of AFB1 contamination level and, hence, for selecting an appropriate seed quality.

AB - Reports about the influence of cerium-oxide nanoparticles (nCeO2) on plants are contradictory due to their positive and negative effects on plants. Surface modification may affect the interaction of nCeO2 with the environment, and hence its availability to plants. In this study, the uncoated and glucose-, levan-, and pullulan-coated nCeO2 were synthesized and characterized. The aim was to determine whether nontoxic carbohydrates alter the effect of nCeO2 on the seed germination, plant growth, and metabolism of wheat and pea. We applied 200 mgL-1 of nCeO2 on plants during germination (Ger treatment) or three week-growth (Gro treatment) in hydroponics. The plant response to nCeO2 was studied by measuring changes in Ce concentration, total antioxidative activity (TAA), total phenolic content (TPC), and phenolic profile. Our results generally revealed higher Ce concentration in plants after the treatment with coated nanoparticles compared to uncoated ones. Considering all obtained results, Ger treatment had a stronger impact on the later stages of plant development than Gro treatment. The Ger treatment had a stronger impact on TPC and plant elongation, whereas Gro treatment affected more TAA and phenolic profile. Among nanoparticles, levan-coated nCeO2 had the strongest and positive impact on tested plants. Wheat showed higher sensitivity to all treatments.

AB - The possibility of reduction of vanadate monomer in the mycelium of fungus Phycomyces blakesleeanus was investigated in this study by means of polarography. Control experiments were performed with vanadyl [V(IV)] and vanadate [V(V)] in 10 mM Hepes, pH 7.2. Addition of P. blakesleeanus mycelium resulted in disappearance of all V(IV) polarographic waves recorded in the control. This points to the uptake of all available V(IV) by the mycelium, up to 185 A mu mol/g(FW), and suggests P. blakesleeanus as a potential agent in V(IV) bioremediation. Polarographic measurements of mycelium with low concentrations (0.1-1 mM) of V(V), that only allows the presence of monomer, showed that fungal mycelia removes around 27% of V(V) from the extracellular solution. Uptake was saturated at 104 +/- 2 A mu mol/g(FW) which indicates excellent bioaccumulation capability of P. blakesleeanus. EPR, V-51 NMR and polarographic experiments showed no indications of any measurable extracellular complexation of V(V) monomer with fungal exudates, reduction by the mycelium or adsorption to the cell wall. Therefore, in contrast to vanadium oligomers, vanadate monomer interactions with the mycelium are restricted to its transport into the fungal cell, probably by a phosphate transporter.

AB - The growth inhibition of stictic acid, a secondary metabolite isolated from the lichen Lobaria pulmonaria (L.) Hoffm. (Lobariaceae), was evaluated in vitro on three human cell lines for the first time. The cell lines HT-29 and MCF-7 were utilized for measuring the activity of stictic acid against cancer cells, while the cell line MRC-5 was selected for estimation of its effect on normal cells. The results suggest a moderate anticancer activity (IC50 value for the cell line HT-29 was 29.29 mu g/ml) and a low growth inhibition on nonmalignant cells (IC50 value for the cell line MRC-5 was 2478.40 mu g/ml) of stictic acid. This natural product can be considered as a promising lead compound for the design of novel human colon adenocarcinoma drugs. (C) 2013 Production and hosting by Elsevier B.V.

AB - The fatty acid compositions of the moss species Atrichum undulatum (Hedw.) P. Beauv. (Polytrichaceae) and Hypnum andoi A.J.E. Sm. (Hypnaceae) collected in winter time were analysed by gas chromatography (GC) and gas chromatography mass spectrometry (GC-MS) as a contribution to their chemistry. Eight fatty acids were identified in the chloroform/methanol extract 1:1 of A. undulatum (linoleic acid, 26.80%, palmitic acid, 22.17%, alpha-linolenic acid, 20.50%, oleic acid, 18.49%, arachidonic acid, 6.21%, stearic acid, 3.34%, cis-5,8,11,14,17-eicosapentaenoic acid, 1.52% and behenic acid, 1.01%), while six fatty acids were found in the same type of extract of H. andoi (palmitic acid, 63.48%, erucic acid, 12.38%, stearic acid, 8.08%, behenic acid, 6.26%, lignoceric acid, 5.16% and arachidic acid, 4.64%). According to this study, the moss A. undulatum can be considered as a good source of both essential fatty acids for humans (linoleic acid and alpha-linolenic acid) during the winter.
AB - Kao doprinos izučavanju njihovog hemijskog sastava, u ovom radu GC i GC-MS analizom ispitivane su masne kiseline mahovina Atrichum undulatum (Hedw.) P. Beauv. (Polytrichaceae) i Hypnum andoi A.J.E. Sm. (Hypnaceae) sakupljene za vreme zimskog doba. U ekstraktu hloroform/metanol 1:1 vrste A. undulatum identifikovano je osam viših masnih kiselina (linolna kiselina, 26,80%, palmitinska kiselina, 22,17%, α-linolenska kiselina, 20,50%, oleinska kiselina, 18,49%, arahidonska kiselina, 6,21%, stearinska kiselina, 3,34%, cis-5,8,11,14,17-eikosapentaenska kiselina, 1,52% i behenska kiselina, 1,01%), dok je u istom ekstraktu vrste H. andoi nađeno šest viših masnih kiselina (palmitinska kiselina, 63,48%, erukinska kiselina, 12,38%, stearinska kiselina, 8,08%, behenska kiselina, 6,26%, lignocerinska kiselina, 5,16% i arahidinska kiselina, 4,64%). Dobijeni rezultati ukazuju da u ispitivanom vremenskom periodu mahovina A. undulatum predstavlja dobar izvor linolne i α-linolenske kiseline, esencijalnih masnih kiselina za ljude.

AB - The identification of nystose and 1(F)-beta-fructofuranosylnystose in the oligosaccharide-rich fraction of the moss Rhodobryum ontariense (Kindb.) Kindb. (Bryaceae) by thin layer chromatography and high pressure chromatography with anion exchange-pulsed amperometric detector is discussed. According to our knowledge, it is the first record of these saccharides in the genus Rhodobryum as well as in the group of mosses as a whole. The findings are in good agreement with the recent isolation of 1-kestose from the same plant material and confirm that short-chain fructooligosaccharides, well known as prebiotics, are present in the examined bryophyte.
