AB  - It was reported that novel O,O′-diethyl-(S, S)-ethylenediamine-N,N′-di-2-(3-cyclohexyl) propanoate dihydrochloride (DE-EDCP) displayed in vitro antiproliferative activity on several human and mouse cancer cell lines, which was comparable to that of the prototypical anticancer drug cisplatin. In order to reveal its toxicity profile, acute and repeated-dose toxicity studies were performed in Naval Medical Research Institute (NMRI) Han mice. The intravenous LD 50 values of DE-EDCP were found to be 95.3 and 101.3 mg/kg body weight in female and male mice, respectively. In the subacute toxicity study, DE-EDCP was administered intravenously at the doses of 15, 25, and 40 mg/kg/day for a period of 28 days. There were no adverse effects on general condition, growth, feed and water consumption, and hematological parameters. There was a significant increase in urea and alanine aminotransferase in female mice and aspartate aminotransferase and alkaline phosphatase in both genders in 40 mg/kg/day dose-treated group. The histopathological changes confined to the liver and kidney, but in other organs were not found. Satellite group revealed that changes in the kidney and liver were less pronounced, suggesting their reversibility. Interactions with DNA could also be of importance for understanding DE-EDCP toxic side effects. Hyperchromic effect obtained with ultraviolet–visible, suggested electrostatic interactions between DE-EDCP and calf thymus DNA. The toxicity testing of DE-EDCP was conducted to predict human outcomes.

AB  - The advances in nanotechnology are directed towards the development of new theranostic agents based on magnetic nanoparticles that can be used for both cancer detection and treatment. In this study, 2,3-dicarboxypropane-1,1-diphosphonic acid-coated magnetite nanoparticles (Fe 3 O 4 -DPD MNPs) were evaluated for their theranostic application using different methods. The magnetic hyperthermia efficiency of the Fe 3 O 4 -DPD MNPs was investigated in saline solution with ionic strengths between 0.05 and 1.0 mol dm −3 . For a better understanding of hyperthermia, the behavior of Fe 3 O 4 -DPD MNPs under a non-alternating magnetic field was studied, and the transparency of the sample was measured. Furthermore, the radiotracer method using the radionuclides 99m Tc and 90 Y was applied as a reliable and powerful method for evaluating the in vivo behavior of a nanoprobe; a high radiolabeling yield (>93%), in vitro and in vivo stability of the radiolabeled nanoparticles and high heating effect were observed, thus paving the way for the possible theranostic applications of Fe 3 O 4 -DPD MNPs.

AB  - Magnetic nanoparticles (MNPs) are of immense interest for diagnostic and therapeutic applications in medicine. Design and development of new iron oxide-based MNPs for such applications is of rather limited breadth without reliable and sensitive methods to determine their levels in body tissues. Commonly used methods, such as ICP, are quite problematic, due to the inability to decipher the origin of the detected iron, i.e. whether it originates from the MNPs or endogenous from tissues and bodily fluids. One of the approaches to overcome this problem and to increase reliability of tracing MNPs is to partially substitute iron ions in the MNPs with Er. Here, we report on the development of citric add coated (Fe,Er)(3)O-4 nanopartides and characterization of their physico-chemical and biological properties by utilization of various complementary approaches. The synthesized MNPs had a narrow (6-7 nm) size distribution, as consistently seen in atomic pair distribution function, transmission electron microscopy, and DC magnetization measurements. The particles were found to be superparamagnetic, with a pronounced maximum in measured zero-field cooled magnetization at around 90 K. Reduction in saturation magnetization due to incorporation of 1.7% Er3+ into the Fe3O4 matrix was clearly observed. From the biological standpoint, citric acid coated (Fe,Er)(3)O-4 NPs were found to induce low toxicity both in human cell fibroblasts and in zebrafish (Danio rerio) embryos. Biodistribution pattern of the MNPs after intravenous administration in healthy Wistar rats was followed by the radiotracer method, revealing that Y-90-labeled MNPs were predominantly found in liver (7533% ID), followed by lungs (16.70% ID) and spleen (2.83% ID). Quantitative agreement with these observations was obtained by ICP-MS elemental analysis using Er as the detected tracer. Based on the favorable physical, chemical and biological characteristics, citric add coated (Fe,Er)(3)O-4 MNPs could be further considered for the potential application as a diagnostic and/or therapeutic agent. This work also demonstrates that combined application of these techniques is a promising tool for studies of pharmacokinetics of the new MNPs in complex biological systems.

Complementary approaches for the evaluation of biocompatibility of 90Y-labeled superparamagnetic citric acid (Fe,Er)3O4 coated nanoparticles

AB  - Magnetic nanoparticles (MNPs) are of immense interest for diagnostic and therapeutic applications in medicine. Design and development of new iron oxide-based MNPs for such applications is of rather limited breadth without reliable and sensitive methods to determine their levels in body tissues. Commonly used methods, such as ICP, are quite problematic, due to the inability to decipher the origin of the detected iron, i.e. whether it originates from the MNPs or endogenous from tissues and bodily fluids. One of the approaches to overcome this problem and to increase reliability of tracing MNPs is to partially substitute iron ions in the MNPs with Er. Here, we report on the development of citric add coated (Fe,Er)(3)O-4 nanopartides and characterization of their physico-chemical and biological properties by utilization of various complementary approaches. The synthesized MNPs had a narrow (6-7 nm) size distribution, as consistently seen in atomic pair distribution function, transmission electron microscopy, and DC magnetization measurements. The particles were found to be superparamagnetic, with a pronounced maximum in measured zero-field cooled magnetization at around 90 K. Reduction in saturation magnetization due to incorporation of 1.7% Er3+ into the Fe3O4 matrix was clearly observed. From the biological standpoint, citric acid coated (Fe,Er)(3)O-4 NPs were found to induce low toxicity both in human cell fibroblasts and in zebrafish (Danio rerio) embryos. Biodistribution pattern of the MNPs after intravenous administration in healthy Wistar rats was followed by the radiotracer method, revealing that Y-90-labeled MNPs were predominantly found in liver (7533% ID), followed by lungs (16.70% ID) and spleen (2.83% ID). Quantitative agreement with these observations was obtained by ICP-MS elemental analysis using Er as the detected tracer. Based on the favorable physical, chemical and biological characteristics, citric add coated (Fe,Er)(3)O-4 MNPs could be further considered for the potential application as a diagnostic and/or therapeutic agent. This work also demonstrates that combined application of these techniques is a promising tool for studies of pharmacokinetics of the new MNPs in complex biological systems.

AB  - Hematite core-shell nanoparticles with plate-like morphology were synthesized using a one-step hydrothermal synthesis. An XRPD analysis indicates that the sample consist of single-phase alpha-Fe2O3 nanoparticles. SEM and TEM measurements show that the hematite sample is composed of uniform core-shell nanoplates with 10-20 nm thickness, 80-100 nm landscape dimensions (aspect ratio 5) and 3-4 nm thickness of the surface shells. We used computational methods for the quantitative analysis of the core-shell particle structure and circularity shape descriptor for the quantitative shape analysis of the nanoparticles from TEM micrographs. The calculated results indicated that a percentage of the shell area in the nanoparticle area (share [%]) is significant. The determined values of circularity in the perpendicular and oblique perspective clearly show shape anisotropy of the nanoplates. The magnetic properties revealed the ferromagnetic-like properties at room temperature with high coercivity H-C = 2340 Oe, pointing to the shape and surface effects. These results signify core-shell hematite nanoparticles' for practical applications in magnetic devices. The synthesized hematite plate-like nanoparticles exhibit low cytotoxicity levels on the human lung fibroblasts (MRC5) cell line demonstrating the safe use of these nanoparticles for biomedical applications.

AB  - The extensive development of radiopharmaceuticals towards early tumour detection and treatment has increased the demand for new ligands with higher tumour selectivity. Research has been done on the potential of the novel O,O'-diethylethylenediamine-N,N'-di-3-propanoate (L) ligand as a radionuclide vehicle for tumour targeting. Under alkaline conditions, L hydrolyses and produces half ester ligand (L') and diacid ligand (L"), with characteristic donor atom array N, N, O. Ligand L was successfully labelled with Tc-99m at pH=9 by coordination with the octahedral fac-[Tc-99m(CO)(3)(H2O)(3)](+) intermediate, forming the main radioproduct fac-[(TcL)-Tc-99m'(CO)(3)] (Tc1). The Tc-99m complex showed a low lipophilic character (log P = 0.48) and low binding affinity to human serum albumin (2.51 +/- 0.48%). In vitro stability studies in saline and human plasma, as well as challenge studies with cysteine and histidine, revealed high stability of the complex during 24 h. Biodistribution studies of Tc1 in female C57BL/6 mice bearing B16/F1 melanoma metastases showed significant tumour uptake: 9.81 +/- 1.19% ID g(-1) in the liver, 5.87 +/- 0.54% ID g(-1) in the lungs and 3.17 +/- 0.33% ID g(-1) in the ovary at 30 min post-injection. Favourable physicochemical properties, satisfactory in vitro/in vivo stability and biodistribution profile in the experimental metastatic melanoma model indicate the possible application of the radiolabelled ligand in tumour diagnosis. AB  - Magnetic nanocomposites can show enhanced properties compared to their single phase counterparts. Novel soft ferrite Fe3O4/soft ferrite MnFe2O4 nanocomposite, as well as pure MnFe2O4 and Fe3O4 for the sake of comparison, were synthesized by co-precipitation method. The spinel type of structure (space group Fd (3) over barm) of the samples was confirmed by XRPD and SAED. Samples are composed of spherical particles with size in the range 5-15 nm. Nanocomposite (92 wt%) MnFe2O4/(8 wt%) Fe3O4 exert single phase like magnetic behavior. Saturation magnetization (M-S) of Fe3O4/MnFe2O4 is 26% greater than M-S of MnFe2O4 and even 50% than M-S of Fe3O4. At the same time coercivity field of the nanocomposite is twice less value (H-C=180 Oe) than in binary ferrites (H-C=360 Oe). Magnetization enhancement and softening in Magnetite/Mn-ferrite is discussed related to particle interactions among two different ferrites and intrinsic properties of nanoparticles. Tailoring phase ratio and synthesis conditions can be a tool for tuning magnetic properties of nanocomposites. (C) 2014 Elsevier B.V. All rights reserved.

AB  - Different synthesis methods for the preparation of nanocrystalline nickel ferrites are reported: the thermal decomposition of precursors, made of: (i) metal-nitrate salts with carboxylic acids (citric, malonic and tartaric), and (ii) metal-nitrate salts and polyethylene glycol (PEG), in the presence of potassium chloride as a capping agent. The as-prepared gel precursors were characterized by TGA/DTA, while the samples obtained after annealing at 450 degrees C were investigated by FTIR, FESEM, XRD and Mossbauer spectroscopy. Regardless of the type of carboxylic acid used, nanocrystallites prepared by (i) method are similar in size (11-16 nm), while the method (ii) gives crystallites similar to 33 nm in size with negligible microstrain. The differences in the lattice parameter, ranging from 8.3369(2) to 8.3574(2) angstrom, result from cation distribution, nonstoichiometry and structural imperfections in the nickel ferrite nanoparticles. The Mossbauer spectra analysis indicates existence of large distortions of tetrahedral and octahedral sites in these spinel compounds. (C) 2013 Elsevier Ltd and Techna Group S.r.l. AB  - Herein a comparative study of five nanocrystalline cerium oxides (CeO2-delta) synthesised by different methods and calcined at 500 degrees C is reported. XRPD analysis showed that stoichiometry parameter delta, crystallite size/strain and lattice constant were only slightly affected by the method utilized. All ceria nanoparticles are nearly spherical in shape with faceted morphology, free of defects and with a relatively uniform size distribution. The average microstrain was found to be approximately 10 times higher than that of bulk counterpart. The absorption edge of nanocrystalline materials was shifted towards a higher wavelengths (red shift) in comparison with bulk counterpart, and band gap values were in the range 2.7-3.24 eV (3.33 eV for bulk counterpart).

The resulting nanoparticles with a composition of the (Mn,Fe)(3-delta)O-4 type are found to have a core/shell structure with different Mn/Fe ratios in the core and at the surface. XPS analysis points to valences of +2, +3, and +4 for Mn and +3 for Fe at the particle surface. Combined results of XRPD, Mossbauer spectroscopy, and EDX analysis suggest that there is a deviation from stoichiometry in the nanoparticle core compared to the shell, accompanied by creation of cation polyvalence and vacancies. The value of saturation magnetization, M-S, of 73.5 emu/g at room temperature, is among the highest reported so far among nanocrystalline ferrite systems of similar composition. X-ray line broadening analysis of mixed samples Ce1-XEuXO2-delta (0.05  lt = x  lt = 0.30) showed that the crystallite size was lower and root mean square strain higher than in pure ceria. However, within mixed samples Ce1-XEuXO2-delta (0.05  lt = x  lt = 0.30) crystallite size and root mean square strain were independent of Eu3+ concentration. Raman spectroscopy results indicated that europium ions yield disorder by breaking the phonon propagation and therefore making the non-centre Brillouin zone modes Raman active. The absorption bands in the spectra of mixed oxides were blue-shifted in comparison to pure CeO2-delta nanopowder. The samples show red emission typical for Eu ions. The biggest photoluminescent intensity was observed for the highest Eu3+ concentration (x = 0.30) and further enhanced with the increase in crystallinity.
