Stable colloidal dispersions of fullerene C60, curcumin and C60-curcumin in water as potential antioxidants

AB  - Despite many advantages, the practical application of fullerene C60 and curcumin (CUR) has been limited by their hydrophobic nature, deficient solubility in aqueous media and, therefore, poor bioavailability. In this study, aqueous nanoformulations containing C60 and CUR in the absence and presence of three solubility enhancers, β-cyclodextrin (CD), Tween 80 (T80) and polyvinylpyrrolidone (PVP), were prepared and subjected to the antioxidant activity assessment. The content of C60 and CUR, stability, size and morphology of nanoparticles were determined using UV–vis, DLS and SEM techniques. Contrary to better solubility of pure C60 than the corresponding C60-CUR system, the solubilization of CUR in hybrid suspensions was improved compared to corresponding pure CUR suspensions, with the exception of the C60-CUR-PVP formulation. The DLS results revealed nearly monodisperse nature of CUR-T80 nanoformulation, while other ones ranged from widely dispersed to polydispersed. Depending on the type of solubilizer, similarity was observed in the particle size distributions of T80- and PVP-coated nanoformulations as well as water and CD nanoformulations. The presence of T80 differently and significantly reduces the particle size in all three nanoformulations, while PVP slightly reduces CUR-based nanoparticles but increases the size of C60 nanoparticles. Zeta potential values, ranging from − 9 mV to − 34 mV, show variation in the starting stability of nanoparticles, from unstable or relatively stable three PVP formulations and C60-T80 to highly stable CUR-T80 and C60-water. Compared to water nanoformulations as a control, SEM images of dispersant-coated nanoparticles lost the visible morphology of active components, showing variation in their morphology depending on the type of dispersant and active component used. Nanoformulations containing CUR in PVP and T80 expressed a very good antioxidant potential in DPPH radical scavenging assay, much stronger than ascorbic acid. At the same time, a moderate to high antioxidant activity in β-carotene bleaching assay was reached in PVP and T80 nanoformulations containing C60, CUR, and their noncovalent hybrid, as well. Our studies have shown that T80 and PVP colloidal systems containing C60-CUR hybrid, but also their individual components, can be used as stable antioxidant nanoformulations with potential application in the field of food and pharmacology.

AB  - A dysregulated and overwhelming response to an infection accompanied by the exaggerated pro-inflammatory state and metabolism disturbance leads to the fatal outcome in sepsis. Previously we showed that meldonium, an anti-ischemic drug clinically used to treat myocardial and cerebral ischemia, strongly increases mortality in faecal-induced peritonitis (FIP) in rats. We postulated that the same mechanism that is responsible for the otherwise strong anti-inflammatory effects of meldonium could be the culprit of the increased mortality. In the present study, we applied the LPS-induced model of sepsis to explore the presence of any differences from and/or similarities to the FIP model. When it comes to energy production, despite some shared similarities, it is evident that LPS and FIP models of sepsis differ greatly. A different profile of sympathoadrenal activation may account for this observation, as it was lacking in the FIP model, whereas in the LPS model it was strong enough to overcome the effects of meldonium. Therefore, choosing the appropriate model of sepsis induction is of great importance, especially if energy homeostasis is the main focus of the study. Even when differences in the experimental design of the two models are acknowledged, the role of different patterns of energy production cannot be excluded. On that account, our results draw attention to the importance of uninterrupted energy production in sepsis but also call for much-needed revisions of the current recommendations for its treatment. © 2022 by the authors. AB  - Two new Schiff base cobalt(III) ([Co(LH)Cl2], 1) and nickel(II) ([Ni(LH)ClO4], 2) complexes with a diimine-dioxime ligand, (4,9-diaza-3,10-diethyl-3,9-dodecadiene-2,11-dione bis oxime (LH2)), were synthesized and characterized. The compounds were obtained in MeOH from corresponding metal salts and LH2 in molar ratio 1:1 and further characterized by mass spectrometry, IR spectroscopy, electrochemistry, and elemental analysis. Previously reported copper(II) analog, ([Cu2(LH)2]·(ClO4)2, 3) was joined to 1 and 2, and the three metal analogs, 1-3, were further investigated in terms of their electrochemical behavior. The binding studies of the complexes with deoxyribonucleic acid (DNA) and human serum albumin (HSA) were carried out using both spectrophotometric and electrochemical methods. All three complexes exhibit binding affinity towards the DNA chain through intercalative interaction. The binding reaction with HSA showed for 1 and 3 complexes decrease in the peak current obtained in the case of complexes before the addition of HSA, while resulted compound obtained from Ni complex – HSA possesses the same electroactivity as starting complex. Furthermore, the cytotoxicity of LH2 as well as its metal complexes, and cisplatin were evaluated on CT-26 mouse colon carcinoma and human LS174T cancer cell lines employing MTT assay. The copper(II) complex exhibited very promising anticancer activity compared with cisplatin.

AB  - Different phosphates and phosphonates have shown excellent coating ability toward magnetic nanoparticles, improving their stability and biocompatibility which enables their biomedical application. The magnetic hyperthermia efficiency of phosphates (IDP and IHP) and phosphonates (MDP and HEDP) coated Fe3O4 magnetic nanoparticles (MNPs) were evaluated in an alternating magnetic field. For a deeper understanding of hyperthermia, the behavior of investigated MNPs in the non-alternating magnetic field was monitored by measuring the transparency of the sample. To investigate their theranostic potential coated Fe3O4-MNPs were radiolabeled with radionuclide 177Lu. Phosphate coated MNPs were radiolabeled in high radiolabeling yield (> 99%) while phosphonate coated MNPs reached maximum radiolabeling yield of 78%. Regardless lower radiolabeling yield both radiolabeled phosphonate MNPs may be further purified reaching radiochemical purity of more than 95%. In vitro stabile radiolabeled nanoparticles in saline and HSA were obtained. The high heating ability of phosphates and phosphonates coated MNPs as sine qua non for efficient in vivo hyperthermia treatment and satisfactory radiolabeling yield justifies their further research in order to develop new theranostic agents. © 2021, The Author(s), under exclusive licence to Springer Science+Business Media, LLC, part of Springer Nature.

AB  - With the growth of the industrial world and the rise of anthropogenic activities, environmental challenges have also increased. Wastewater, especially from industrial sources can contain a wide variety of heavy metals [1]. The presence of heavy metals in water resources is a cause for concern due to their toxicity and tendency to bioaccumulation, which leads to significant problems for the environment as well as for animals and humans [2]. There are various conventional treatment technologies that are used for the removal of heavy metals from wastewater such as reverse osmosis, electrodialysis, ultrafiltration, industrial ion exchange process, and chemical precipitation. These techniques have some drawbacks such a being expensive due to high reagent and energy requirements, incomplete metal removal at low concentrations of heavy metals and generation of sludge [1]. Therefore, biosorption has been the subject of research by scientists for almost 70 years in order to find efficient, effective, and low-cost biomaterials for wastewater treatment [3]. In this study, a sunflower husk treated with 1% hydrochloric acid was used to evaluate removal efficiency for Ni from real wastewater samples, as well as to estimate the adsorption capacity as a function of contact time between the heavy metal ions such as Cd in the aqueous solution and the sunflower husk as a biosorbent. Two techniques were used for quantitative analysis, Inductively Coupled Plasma Optical Emission Spectrometry (ICP-OES) as a standard reference method and laser-induced plasma spectroscopy (LIBS) as an innovative non-standard analytical method. What makes the LIBS superior from the other standard optical techniques is that it allows fast multi-elemental analysis without prior sample preparation, leads to minimal sample damage, and is considered almost non-destructive, this method is also by the principles of green analytical chemistry, which means that it does not use toxic reagents and therefore no chemical waste [4]. The results of these two methods were compared, to examine the possibility of using laser-induced plasma spectroscopy as an alternative green analytical technology for quantitative analysis of sunflower husk as a biosorbent and evaluation of biosorption efficiency as a function of contact time.

AB  - Objects of the present study are improved fullerene C60 drug carrier properties trough encapsulation by microbial polysaccharides, levan (LEV), pullulan (PUL), and their hydrophobized cholesterol-derivatives (CHL and CHP), that show better interaction with cancer cells. The zeta potential, polydispersity index, and the diameter of particles were determined, and their cytotoxicity against three cancer cell lines were tested. Biochemical changes in HeLa cells are analyzed by synchrotron radiation (SR) FTIR spectro-microscopy combined with the principal component analysis (PCA). The most significant changes occur in HeLa cells treated with LEV-C60 and correspond to the changes in the protein region, i.e. Amide Iband, and the changes in the structure of lipid bodies and membrane fluidity are evident. The highest cytotoxicity was also induced by LEV-C60. In HeLa cells, cytotoxicity could not be strictly associated with biochemical changes in lipids, proteins and nucleic acids, but these findings are significant contribution to the study of the mechanism of interaction of C60-based nanoparticles with cellular biomolecules. In conclusion, LEV, PUL, CHL, and CHP enhanced fullerene C60potential to be used as target drug delivery system with the ability to induce specific intracellular changes in HeLa cancer cells.
