AB  - Vegetables play an important role in human nutrition and their importance in diet is determined by its chemical composition. Numerous studies indicate a high degree of contamination of soil and plants produced in certain agro-ecological conditions, especially near urban and industrial areas. Accordingly, toxic and potentially toxic elements (aluminum, chromium, copper, manganese, nickel, lead and zinc) were quantified in edible parts of four vegetable species (potato, cabbage, carrot and broccoli) by means of inductively coupled plasma–optical emission spectrometry (ICP–OES). In addition, health risk assessment was done and expressed through estimated weekly intake of elements (EWI) and hazard quotient (HQ). Samples of each species originating from different localities were collected from three green markets in the city of Belgrade (Serbia). The content of studied elements ranged from 1.32 to 4.00, 0.31 to 5.77, 3.50 to 23.11, 1.82 to 11.17 and 2.11 to 10.62 mg/kg for aluminum, cooper, manganese, nickel and zinc, respectively. Chromium was detected in carrot sample (1.76 mg/kg), whereas concentration of lead in two samples of broccoli exceeded the maximum allowed concentration (>0.3 mg/kg) set by the national regulations. The average estimated weekly intakes for Al, Cu, and Zn were within the safe limits set by FAO/WHO, while for Ni and Pb in some vegetables were considerably higher than proposed values. In respect to the obtained results, a great weekly health risk was indicated for chromium, nickel and lead with the highest HQ values: 2.44, 10.32 and 2.02, respectively. The results of the study impose the necessity for continuous monitoring of harmful elements content in individual vegetable crops as well as strict regulative guide-lines in order to diminish possibility of contamination.

AB  - Nanotechnology is an emerging field of science, and nanotechnological concepts have been intensively studied for potential applications in the food industry. Nanoparticles (with dimensions ranging from one to several hundred nanometers) have specific characteristics and better functionality, thanks to their size and other physicochemical properties. Polyphenols are recognized as active compounds that have several putative beneficial properties, including antioxidant, antimicrobial, and anticancer activity. However, the use of polyphenols as functional food ingredients faces numerous challenges, such as their poor stability, solubility, and bioavailability. These difficulties could be solved relatively easily by the application of encapsulation. The objective of this review is to present the most recent accomplishments in the usage of polyphenol-loaded nanoparticles in food science. Nanoparticles loaded with polyphenols and their applications as active ingredients for improving physicochemical and functional properties of food, or as components of active packaging materials, were critically reviewed. Potential adverse effects of polyphenol-loaded nanomaterials are also discussed.

AB  - The preliminary assessment of the properties of alginate immobilized aquatic weed Myriophyllum spicatum beads-MsAlg in a multi-element system of nine Serbian lakes water samples was done. Herein, the results obtained in the biosorption experiment with MsAlg contents of twenty-two elements analysed by inductively coupled plasma-optical emission spectrometry, biosorption capacity, element removal efficiency, total hardness (TH) and quality index of water (WQI) are presented. Scanning electron microscopy with energy dispersive X-ray spectroscopy was used for the characterization of M. spicatum and its beads. The study showed that aluminium, magnesium and strontium were adsorbed by MsAlg in the water samples from all examined lakes; barium and iron in the water samples from six lakes. The overall average efficiency of MsAlg in biosorption of elements was in the following order: Al  gt  Ba  gt  Sr  gt  Fe  gt  Mg (58.6, 51.7, 48.2, 23.9 and 17.7%, respectively). The increase of TH and WQI values after the biosorption was noticed in all studied lake water samples. The most significant correlations for pH were regarding the contents of B, Mg and Ca, whereas WQI was highly correlated to the contents of B and Mg, and pH. The complexity of the obtained data was explained by Cluster Analysis and Principal Component Analysis, which showed good discrimination capabilities between the water samples taken from different locations. Considering that the invasive M. spicatum is natural, widespread and that its immobilization is cheap and eco-friendly, presented findings could be helpful in further assessment of MsAlg beads for its potential use as biofilter. Levels of toxic elements in canned fish from the Serbian markets and their health risks assessment

AB  - A total of 138 samples of canned tuna, sardines and mackerel from the Serbian market were analyzed for toxic (As, Cd, Pb, Hg) element levels using ICPMS. The highest average contents of toxic elements were found to be 1.70 mg kg(-1) for arsenic and 0.026 mg kg(-1) for cadmium in sardine and 0.068 mg kg(-1) for mercury in tuna. The lead content was very similar (similar to 0.048 mg kg(-1)) of the three types of canned fish. None of the fish analyzed contained toxic elements in levels exceeding the maximum allowable levels currently in force. Human health risk assessment of As, Cd, Pb and Hg in canned fish were investigated. A risk assessment based on the measured levels of examined elements indicated that consumption of canned fish according to current eating habits in Serbia presents little risk to human health since the carcinogenic risk is within the acceptable range, 10(-4)-10(-6).

The highest average contents of toxic elements were found to be 1.70 mg kg(-1) for arsenic and 0.026 mg kg(-1) for cadmium in sardine and 0.068 mg kg(-1) for mercury in tuna. AB  - The preliminary assessment of the properties of alginate immobilized aquatic weed Myriophyllum spicatum beads-MsAlg in a multi-element system of nine Serbian lakes water samples was done. Herein, the results obtained in the biosorption experiment with MsAlg contents of twenty-two elements analysed by inductively coupled plasma-optical emission spectrometry, biosorption capacity, element removal efficiency, total hardness (TH) and quality index of water (WQI) are presented. Scanning electron microscopy with energy dispersive X-ray spectroscopy was used for the characterization of M. spicatum and its beads. The study showed that aluminium, magnesium and strontium were adsorbed by MsAlg in the water samples from all examined lakes; barium and iron in the water samples from six lakes. The overall average efficiency of MsAlg in biosorption of elements was in the following order: Al  gt  Ba  gt  Sr  gt  Fe  gt  Mg (58.6, 51.7, 48.2, 23.9 and 17.7%, respectively). The increase of TH and WQI values after the biosorption was noticed in all studied lake water samples. The most significant correlations for pH were regarding the contents of B, Mg and Ca, whereas WQI was highly correlated to the contents of B and Mg, and pH. The complexity of the obtained data was explained by Cluster Analysis and Principal Component Analysis, which showed good discrimination capabilities between the water samples taken from different locations. Considering that the invasive M. spicatum is natural, widespread and that its immobilization is cheap and eco-friendly, presented findings could be helpful in further assessment of MsAlg beads for its potential use as biofilter. AB  - Three Cabernet Franc (CF) clones (Nos. 02,010 and 012; not yet commercial) obtained in the last phase of clonal selection were examined within this study. Indeed, the content of 27 chemical elements in the vineyard soil along with CF grape and wine samples were determined by inductively coupled plasma - optical emission spectroscopy. The relative dependence of the selected elements in all samples were estimated using correlation matrices. While elemental profiles of both CF grape and wine samples were highly variable among the analysed clones, bioaccumulation factors (grape/soil) of Mg, Na and Sr were found to be specific for each single clone. Applying principal component analysis, the grape clones were differentiated among each other according to the content of 10 elements (Al, B, Ca, Cu, Fe, K, Mg, Mn, Na and Sr). Finally, hierarchical cluster analysis of CF grape and wine samples pointed out the similarity of the clones Nos. 010 and 012, since they were grouped within the same subcluster.

AB  - The water samples from Rasina District (Serbia) were evaluated for principal physical and chemical parameters, as well as for microbiological contaminants. Results were compared to National and World Health Organization (WHO) water quality standards. Several samples contained total organic matter, ammonia, residual chlorine, nitrite, nitrate, iron and manganese above proposed legislation limits. For samples contaminated with faecal bacteria, Streptococcus faecalis, aerobic mesophilic bacteria, coliform bacteria and sulfite-reducing clostridia special attention should be payed to drinking water disinfecting methods. The potential health risks of waterborne diseases due to consumption of water from contaminated sources could be implied.

The contents of twelve elements were determined in selected vegetable species taken at the city green market using ICP-OES analysis. To assess the safety of dietary intake of these metals through the consumption of vegetables, the estimated weekly intake (EWI) was calculated and compared with the provisional tolerable weekly intake (PTWI). The most abundant element was Al, whereas As, Cd, Co, Sb and Mo were not found in studied vegetables. Lead exceeded the maximum allowed concentration (MAC) set by national regulations. The average estimated weekly intake for aluminum, chromium, copper, manganese, nickel, lead, and zinc were 0.392, 0.010, 0.015, 0.096, 0.019, 0.038 and 0.124 mg kg(-1), respectively. Cr, Ni and Pb in some vegetables were considerably higher than the safe limits set by FAO/WHO.

Wine Chemical Composition and Radical Scavenging Activity of Some Cabernet Franc Clones

AB  - Background: Three clones of Cabernet Franc (Nos. 02, 010 and 012) were selected in the last phase of clonal selection in Serbia. Wines made from each clone were assessed for quality parameters and taste during five consecutive vintages (2008-2012) and compared to the standard. The wine quality was determined based on the following parameters: alcohol, total extract, anthocyanins, tannins, pH, titratable acidity, volatile acidity, aldehydes, esters and reducing sugars, relative density, ash, colour, tonality, and tasting score. In the last year of the study, grapes and wines of Cabernet Franc clones and a standard were subjected to a chemical analysis of their phenolic composition, resveratrol and radical scavenging activity. In the last year of the study, grapes and wines of Cabernet Franc clones and a standard were subjected to a chemical analysis of their phenolic composition, resveratrol and radical scavenging activity. Methods: Chemical analyses of grapes and wines along with sensory and radical scavenging activity evaluations were done according to the standard procedures. Results: The wines of the clone No. 010 showed some superior properties compared to the other two clones and the standard; in five-year period the average concentration of anthocyanins (179 +/- 3.8 mg/L) and polyphenolics (1.85 +/- 0.02 g/L) was significantly higher than in wines of clones and the standard, (168-173 mg/L and 1.63-1.74 g/L for anthocyanins and phenolics, respectively). Furthermore, the same clone had a higher alcohol content (13.97 +/- 0.03%) in each year of the study, which indicated that it ripened faster than other clones (13.06-13.08 %) and compared to the standard (13.04 +/- 0.07%). This finding suggested that the clone No. 010 could possibly have a significant economic impact and further increase popularity of Cabernet Franc in a cooler climate viticultural region. It was also found to have the highest contents of aldehydes (488 +/- 1.54 mg/L) and esters (322 +/- 0.71 mg/L) compared to aldehydes (452-467 mg/L) and esters (290-310 mg/L) measured in other wines. Finally, the highest amount of phenolic compounds (1220 +/- 40 mg/kg) and resveratrol (70 +/- 3.3 mg/kg) were found in the grapes of the clone No. 010. The present study revealed a strong correlation of total phenolic contents and anthocyanins with radical scavenging activities (0.936 and 0.929, P lt 0.001, respectively) indicating that this activity of wines was derived mainly from their phenolic compounds. Conclusion: The wine of the clone No. 010 contained the highest concentration of aldehydes, esters, anthocyanins, polyphenolics and resveratrol and consequently achieved the best tasting score. AB  - In this study, grapes and corresponding wines of Merlot /No 022, 025 and 029/ and Cabernet Franc /No 02, 010 and 012/ clones (recently developed) were evaluated regarding the total phenolic content, total anthocyanin content, and radical scavenging activity, aiming to better understand their quality and market potential. The nineteen individual polyphenols were quantified in studied grape samples using UHPLC coupled to a triple-quadrupole mass spectrometer. The mother grapes and wines were used as the relevant standards. In the grape, studied characteristics were monitored at three stages of berry development: green berry, veraison and mature berry. The mature grape of clones presented high values of total phenolics (3.81-10.89 g gallic acid equivalent kg(-1) frozen weight), anthocyanin content (359.00-1668.18 mg malvidin-3-O-glucoside kg(-1) frozen weight) and the radical scavenging activity (41.37-80.48 mmol trolox equivalent kg(-1) frozen weight) depending on the clone. Grapes and wines of Merlot No 025 and Cabernet Franc No 010 stood out with the highest values of all three parameters. Generally, the high correlation was observed between TPC and RSA values for green berries and mature grapes, as well as for wine samples. The most abundant phenolics in both Merlot and Cabernet Franc grapes were gallic acid, p-hydroxybenzoic acid, ferulic acid, catechin, epicatechin, gallocatechin gallate, catechin gallate, and rutin. Catechin, epicatechin, epigallocatechin, and catechin gallate, typical for the ripe grape of Merlot and Cabernet Franc clones, showed significant correlation with RSA values. Concentrations of individual polyphenols varied depending on the sample being studied and berry development stages. Merlot No 025 and Cabernet Franc No 010 grapes showed particularly interesting attributes for the production of high quality wines.

AB  - Development, characterization and evaluation of the efficiency of cost-effective medium for the removal of Pb2+, Cd2+ and Ni2+ from aqueous systems, as a novel, eco-friendly solution for wastewater remediation were done. The precursors for low-cost adsorbent were lignoceilulosic raw materials (sweet/sour cherry kernels), as industrial byproducts and components of organic solid waste. Activated carbon synthesis was carried out by thermochemical conversion (H3PO4, 500 degrees C) in the complete absence of inert atmosphere. Characterization of the activated carbon was performed by elemental analysis, FTIR, SEM, EDX and BET. BET surface area corresponds to 657.1 m(2) g(-1). The evaluation also included the influence of pH, contact time, solute concentration and adsorbent dose on the separation efficiency in the batch operational mode. The equilibrium and kinetic studies of adsorption were done. The maximum adsorption capacity of the activated carbon for Cd2+ ions was calculated from the Langmuir isotherm and found to be 198.7 mg g(-1). Adsorption of Pb2+ and Ni2+ were better suitable to Freundlich model with the maximum adsorption capacity of 180.3 mg g(-1) and 76.27 mg g(-1), respectively. The results indicate that the pseudo-second-order model best describes adsorption kinetic data. Based on desorption study results, activated carbon was successfully regenerated with HNO3 for 3 cycles. In order to provide the results for basic cost-effective analysis, competing ion-effects in a real sample have been evaluated. (C) 2016 Elsevier Ltd. All rights reserved.

AB  - Polycyclic aromatic hydrocarbons (PAHs) are products of the incomplete combustion or pyrolysis of organic material and they are among the most harmful compounds. During process of meat smoking, wood combustion is one of the most important sources of PAH compounds, which can be adsorbed by the surface of meat. The EFSA (European Food Safety Authority) Panel on Contaminants in the Food Chain (CONTAM Panel) recommended to the member states of European Union to use the sum of benzo[a] pyrene (BaP), benz[a] anthracene (BaA), benzo[b] fluoranthene (BbF) and chrysene (CHR), (PAH4 compounds), as a marker for the occurrence and impact of carcinogenic PAHs in food, instead of benzo[a]pyrene. The maximum content of BaP and sum of all four compounds (PAH4) has been established by European Commission Regulation No. 835/2011. For smoked foods, from 1st September 2014, the maximum BaP content was lowered to 2 mu g/kg, while the content of PAH4 is allowed to 12 mu g/kg. The new maximum residue limits (MRL) both for BaP and sum of PAH4 compounds in smoked meat and meat products were defined by the legislation of Serbia, as well, and it is in accordance with EU regulation. The aim of this paper was the validation of the method for identification and determination of benzo[a] pyrene, chrysene, benz[a] anthracene and benzo[b] fluoranthene in smoked meat and smoked meat products. Accelerated solvent extraction (ASE) was used for extraction of lipids and lipophilic compounds. Solid Phase Extraction (SPE) was used in order to remove lipids from analysed samples. High-performance liquid chromatographic with fluorescence detection (HPLC-FL) was applied for identification and quantification of PAH4 compounds. Fluorescence detector operated at excitation/emission wavelength 275/385 nm for BaA and CHR, 256/446 nm for BbF and 260/410 nm for BaP, respectively. On the base of experimental results, it is possible to conclude as follows: The in-house validation procedure of the method meets all criteria (applicability, specificity, repeatability, reproducibility, recovery, LOD and LOQ) set out by EU Regulation No. 836/2011. The method is suitable for operative control of PAH4 content in smoked meat products.

Agrobiological and Wine Quality Traits of Vitis Vinifera Cv. Merlot Clones Selected in Serbia

AB  - Aims: The selection of cv. Merlot clones performed in the Grocka winegrowing region, Republic of Serbia, and the study of agrobiological and technological traits of three selected clones and Merlot standard. Methods and results: The selection of clones was done using the method of individual clone selection in four phases over 15-20 years (Cindric, 2003). O.I.V. methods were used in determining the most important agrobiological and economic-technological traits of grapes. The results of studied traits of grape and wine suggest that the quality of the clones surpassed the quality of the standard. Although the highest values for most of the studied traits were obtained for clone 025, there was no statistically significant difference between the clones. Indeed, the significant distinction between the clones was obtained for total polyphenol content (TPC) and total anthocyanin content (TAC) in wines : the wine of clone 025 had the highest TPC and TAC values. According to the tasting scores, wines of the clones may be classified as quality wines with geographical indications. Conclusion: The selected clones were characterised by good quality grapes and wines. Improved viticultural parameters, versus the mother vine, indicate the need for further research work on the chemistry of both grapes and wines, aiming to better understand the characteristics of the selected clones and their market potential. Significance and impact of the study: The development of new clones (namely 022, 025 and 029) of better quality than the mother vine will, in the long term, lead to the introduction of the best clone(s) in viticultural practices and production. Until today, the clonal selection of Merlot has not been done in the Republic of Serbia and there are no recognised clones.

Influence of frost damage on the sugars and sugar alcohol composition in quince (Cydonia oblonga Mill.) floral nectar

AB  - Cold stress adversely affects growth and productivity, and triggers a series of morphological, physiological, biochemical and molecular changes in plants. Since sugars are present in all floral nectars in greater amounts than any other constituent, the aim of this study was to examine how frost exposure changes sugar metabolism and how it affects on the content of sugar components in the nectar of quince. Three quince cultivars ('Vranjska', 'Triumph' and 'Leskovac. ka') were investigated in this study. The contents of sugars (glucose, fructose, sucrose, trehalose, maltose, isomaltose, rhamnose, arabinose, ribose, melezitose, raffinose, and panose) and sugar alcohols (sorbitol, erythritol, mannitol and galactitol) were analyzed by high performance anion exchange chromatography (HPAEC) with amperometric detection. The results showed that after late spring frosts and irreversible damage of flower parts, the nectar of the three quince cultivars contained elevated levels of fructose, trehalose, arabinose, ribose, rhamnose, raffinose, galactitol and mannitol, indicating an impairment of central carbohydrate metabolism. The ratios between individual sugars, such as the glucose/fructose ratio, were changed in the nectar of damaged flowers in all three quince cultivars. The examined cultivars showed similar sugar response to cold stress. The only exception was 'Leskovac. ka' for the glucose and melezitose pathway, which means that composition of those two sugars changed significantly according to the genotype. The larger are the carbohydrates reserves in different parts of a fruit tree, the higher is the tolerance to any form of frost damage, the results of this study could help in the understanding of how different quince cultivars react to this kind of stress and how they modulate their sugar metabolism.

AB  - Smoked meat and smoked meat products represent a significant part of the human diet in Serbia. The process of smoking, i.e. wood combustion, is one of the most important sources of polycyclic aromatic hydrocarcarbons (PAHs). Considering the carcinogenic and mutagenic properties of some PAHs, these compounds have been in the spotlight of scientific interest. The EFSA (European Food Safety Authority) Panel on Contaminants in the Food Chain (CONTAM Panel) reviewed the available data on occurrence and toxicity of PAHs in food. In 2008, the CONTAM Panel concluded that benzo[a]pyrene is not a suitable indicator for the occurrence of PAHs in food. Based on the currently available data relating to occurrence and toxicity, the CONTAM Panel concluded that the sum of benzo[a]pyrene, chrysene, benz[a]anthracene and benzo[b]fluoranthene (PAH4) are the most suitable indicators of PAHs in food. These proposals have become part of the legislation both of EU and Serbia. From 1st September 2014, maximum residue limits (MRL) both for benzo[a]pyrene (2 μg/kg) and sum of PAH4 compounds (12 μg/kg), in smoked meat and meat products, were defined by the legislation of Serbia, which is in accordance with EU regulation. In this paper, the method has been developed for the determination of benzo[a]pyrene, chrysene, benz[a]anthracene and benzo[b]fluoranthene (PAH4 compounds) in smoked meat and smoked meat products. Accelerated solvent extraction (ASE) was used for extraction of lipids and lipophilic compounds. Solid Phase Extraction (SPE) was used in order to remove lipids from analysed samples. High-performance liquid chromatographic with fluorescence detection (HPLC-FL) was applied for identification and quantification of benzo[a]pyrene, chrysene, benz[a]anthracene and benzo[b]fluoranthene. Different conditions of HPLC-FL analysed were applied (mobile phase, HPLC column, oven temperature, flow) in order to achieve optimal conditions for qualitative and quantitative analysis of PAH4 compounds.
AB  - Dimljeno meso i proizvodi od mesa zauzimaju važno mesto u ishrani stanovništva u Srbiji. S obzirom, da tokom dimljenja tj. procesom sagorevanja drveta nastaju policiklični aromatični ugljovodonici (Polycyclic Aromatic Hydrocarbons, PAH), koji su klasifikovani kao karcinogena i mutagena jedinjenja, proučavanje ovih jedinjenja uvek zaokuplja pažnju javnosti. Na osnovu podataka, koji su rezultat novih bioloških i toksikoloških naučnih istraživanja, evropska naučna komisija o zagađivačima u lancu ishrane je u junu 2008. godine predložila da se suma sadržaja benzo[a]pirena, benzo[a]antracena, benzo[b]fluorantena i hrizena, tj. PAH4 jedinjenja koristi kao marker prisustva drugih PAH jedinjenja u različitoj hrani. Ovi predlozi postali su deo zakonske regulative, kako Evropske unije, tako i Srbije. Zakonska regulativa Srbije, koja je u saglasnosti sa propisima EU, od 1. septembra 2014. godine definiše maksimalno dozvoljenu količinu (MDK) za sumu sadržaja PAH4 jedinjenja (12 μg/kg), kao i za sadržaj benzo[a]pirena (2 μg/ kg) u dimljenom mesu i proizvodima od mesa. U ovom radu razvijena je metoda za određivanje benzo[a]pirena, benzo[a]antracena, benzo[b]fluorantena i hrizena tj. PAH4 jedinjenja u dimljenom mesu i proizvodima od mesa. Za ekstrakciju lipida i lipofilnih jedinjenja iz uzoraka korišćena je ubrzana ekstrakcija pomoću rastvarača. Ekstrakcija na čvrstoj fazi je korišćena kao postupak za uklanjanje molekula lipida iz ispitanih uzoraka. Identifikacija i kvantifikacija benzo[a]pirena, benzo[a]antracena, benzo[b]fluorantena i hrizena rađena je korišćenjem visokoefikasne tečne hromatografije sa fluorescentnim detektorom (HPLC-FL). Primenjeni su različiti uslovi HPLC analize (mobilna faza, HPLC kolona, temperatura peći, protok mobilne faze) u cilju postizanja optimalnih uslova za kvalitativnu i kvantitativnu analizu PAH4 jedinjenja.

AB  - This study reports results on 16 EU priority polycyclic aromatic hydrocarbons in smoke produced by beech wood combustion from Zlatibor region, Serbia. Smoke samples were collected with two different types of tubes (PUF and XAD-2), at a height of 2 and 5 m away from the firebox; 16 EU priority polycyclic aromatic hydrocarbons [European Commission (2005), Commission Recommendation 2005/108/EC, Official Journal of European Union L34:4345; JECFA (2005), Summary and Conclusion. Paper No. JECFA/64/SC, Joint FAO/WHO Expert Committee on Food Additives, Sixty-Fourth Meeting, Rome, Italy, February 817] were identified and quantified by Fast-GC/HRMS. The analyzed PAHs were: benzo[c]fluorene (BcL), benzo[a]anthracene (BaA), cyclopenta[c,d]pyrene (CPP), chrysene (CHR), 5-methylchrysene (5MC), benzo[b]fluoranthene (BbF), benzo[j]fluoranthene (BjF), benzo[k]fluoranthene (BkF), benzo[a]pyrene (BaP), benzo[g,h,i]perylene (BgP), dibenzo[a,h]anthracene (DhA), indeno[1,2,3-cd]pyrene (IcP), dibenzo[a,e]pyrene (DeP), dibenzo[a,h]pyrene (DhP), dibenzo[a,i]pyrene (DiP) and dibenzo[a,l]pyrene (DlP). Results show that total emission of polycyclic aromatic hydrocarbons at a height of 2 m was 6.7 and 7.4 mg m3 , in PUF and XAD-2 tubes, respectively. At height of 5 m polycyclic aromatic hydrocarbons emission was 2.4 and 4.3 mg m3 , in PUF and XAD-2 tubes, respectively. AB  - Serum amyloid A proteins (SAA) are very sensitive acute phase proteins, displaying multiple isoforms in plasma and different body fluids. They are currently under investigation as biomarkers of diseases. The aim of the present study was to compare the concentration and isoform expression of SAA in serum and milk of cows with bacteriologically negative milk (control group) and naturally occurring Staphylococcus aureus (S. aureus) subclinical mastitis (subclinical mastitis group). Somatic cell count (SCC) and bacteriological analyses were performed to establish the control and subclinical mastitis group. SAA concentration was evaluated using a commercial ELISA kit, while expression of different isoforms (serum A-SAA and milk M-SAA3 isoforms) was visualized by denaturing isoelectrical focusing and immunoblotting. The SAA concentrations in sera and milk of cows in the subclinical mastitis group were three and 100 times higher than in those from the control group of cows, respectively. Cows in the subclinical mastitis group had more acidic SAA isoforms in serum with the most prominent one at pI 5.5. This isoform was not detected in sera from the control group. Milk samples in the subclinical mastitis group contained abundant highly alkaline M-SAA3 isoforms and most of the serum isoforms, except for that at pI 5.5. In the subclinical mastitis group SAA isoforms with equivalent pI as serum isoforms accounted for 20% of the total SAA concentration in milk. There were significant differences in the concentrations and isoform patterns of SAA in serum and milk between the control and subclinical mastitis groups of cows. Also, we demonstrated that serum SAA isoforms were not transferred to milk proportion to their plasma content. (C) 2011 Elsevier B.V. All rights reserved.
