AB - Two new nitrato complexes of zinc and nickel with 1,4,8,11-tetrakis(2-pyridylmethyl)-1,4,8,11-tetraazacyclotetradecane (tpmc), have been synthesized and characterized. The IR spectral peaks showed that the coordinated and ionic nitrate ions are in agreement with the formula proposed by elemental analysis. Conductometric titrations predicted methanol to be a convenient solvent for synthesis and revealed the stoichiometry of the complexes, while molar electrical conductivities indicated a 1 : 3 complex electrolyte type for the zinc complex, and a 1 : 2 complex electrolyte type for the nickel complex. The optimized complex structure was obtained by molecular modeling and density functional theory calculations. The biological activity of the novel complexes was examined by screening eight different bacterial strains and two cancer cell lines. The zinc complex showed better antimicrobial activity against the bacterial strains, while the complexes did not show significance antiproliferative activity toward cancer cells MCF-7 and MDA-MB-231.

AB - Bronchopneumonia is the most frequent cause of calves’ ailments around theworld. The absence of adequate immune response to causative agents ofbronchopneumonia in calves remained unclear to date. The infectious agents of virusand/or bacterium origin associated with bronchopneumonia are omnipresent harmlesscommensals in the cattle population that induce illness when the host defense isdepressed by environmental stress factors and inability of calves to fully respond to anantigenic stimulus due to the fragile nature of the immune system. The disease is aleading cause of economic loss in the livestock industry worldwide which points to thenecessity of attending to this problem with great attention.Having in mind that immune response to certain pathogens responsible forbronchopneumonia in calves is IgG antibody-dependent, the presence of immunecomplexes containing these antibodies could have an important role in the pathogenesisof bronchopneumonia. The goal of this doctoral dissertation was to investigate theprotein and lipid components of circulating immune complexes (CIC) of calves threemonths old, diagnosed with bronchopneumonia caused by Pasteurella multocidabacterium, and their role as modulators of immune response.CIC were isolated by polyethylene glycol precipitation. The level ofpolyethylene glycol precipitating CIC in calves with bronchopneumonia compared tohealthy calves was higher. In calves with bronchopneumonia CIC were larger in size.CIC were composed of five groups of corpuscles; In healthy calves the most abundantcorpuscles in CIC were of 1426±291 nm in diameter; In healthy calves corpuscles withhydrodynamic radius of 225±33 nm and 734±174 nm were mostly present in CIC. Incalves with bronchopneumonia zeta potential of CIC was slightly higher than zetapotential of CIC in healthy calves. In calves with bronchopneumonia increased level of serum total proteins wasdetected. The concentration of γ and α globulins in the serum and in CIC of diseasedcalves was higher. The concentration of IgG and transferrin in the sera was higher, butremained unchanged in CIC of diseased calves...

AB - To explore the potential usage of recombinant banana lectin as an immunostimulator it was of interest to analyze the effect it exerted in vivo in mice, upon two different routes of exposure. By Western blotting analysis of the gastric and intestinal luminal content it was found that recombinant banana lectin is stable in vivo in the mouse digestive tract, where it specifically interacts with the mucosal surfaces which can be inhibited by the addition of glucose. Its attachment to mice jejunum has been confirmed by immunofluorescence. In addition, it was able to induce systemic immunity, evidenced by specific antibody production, by oral application, a property that might be exploited for the induction of systemic immune responses.
