AB  - Herein, a screen–printed diamond electrode (SPDE) coupled with a “point-of-care” platform (30 µL-drop concepts, single-drop-detection approach) was successfully applied for the electrochemical determination of pterostilbene (PTS). Cyclic voltammetry identified irreversible oxidation of PTS, where oxidation peak was shown to be strongly dependent on the pH of the working environmental. Although the proposition of the detailed electrochemical oxidation mechanism of PTS goes out of the scope of the present research, we have determined the most probable reactive site of our analyte, by utilizing DFT-based reactivity descriptors (Fukui functions). For electrochemical quantification of PTS, oxidation peak at 0.32 V (vs. Ag/AgCl) was followed in presence of 0.5 mol L−1 of Briton–Robinson buffer solution (pH = 9). Coupled with the optimized parameters of differential pulse voltammetry (DPV), SPDE detected PTS in two linear ranges (first range was from 0.011 to 0.912 µmol L−1; second range was from 0.912 to 4.420 µmol L−1), providing the LOD and LOQ on a nanomolar level (3.1 nmol L−1 and 10.0 nmol L−1, respectively). The selectivity of the optimized DPV method was found to be excellent, with the current changes of less than 7%, in the presence of ten times higher concentrations of the certain interferences. The practical applicability of the SPDE and single-drop-detection approach in dietary supplements (with a declared PTS content of 50 mg/tablet), with the recovery values ranging from 95 to 102%, shows that the developed method has high potential for precise and accurate PTS detection, as well as exceptional miniaturization possibilities of relevant equipment for on-site sensing.

AB  - We present the green, highly atom-economical, solid-state silica gel-catalyzed synthesis of polysubstituted 1,4- and 1,2-dihydropyridines (DHPs) from commercially available materials, amines and ethyl propiolate. The DHP skeleton was assembled by heating the reactants and silica gel in a closed vessel. Aliphatic amines provided 1,4-isomers as the main or only DHP products, but the reactions of aromatic amines yielded a mixture of 1,4- and 1,2-isomers. To the best of our knowledge, this is the first example of the formation of a 1,2-DHP structure by the reaction of an amine with propiolic ester. Addition of 1 mass percent of H2SO4 to silica gel shifted the product distribution to 1,4-DHP as the main or the only isomer obtained. Experimental and theoretical analyses led to the identification of two key intermediates en route to DHPs and the explanation of the observed regioisomeric ratios. 1,2-DHPs show blue-cyan fluorescence in MeOH with the quantum yield Φ = 0.10–0.22 relative to quinine sulfate Φ = 0.58 and 1,4-DHPs show blue-violet fluorescence with Φ = 0.09–0.81.

AB  - Serotonin receptors modulate numerous behavioral and neuropsychological processes. Therefore, they are the target for the action of many drugs, such as antipsychotics, antidepressants, antiemetics, migraine remedies, and many others. The 5-HT1A receptors have been involved in the pathogenesis and treatment of anxiety and depression and represent a promising target for new drugs with reduced extrapyramidal side effects. In most antidepressants, a piperazine-based structural motif can be identified as a common moiety. Here we describe the synthesis, pharmacological, and in silico characterization of a novel arylpiperazines series with excellent 5-HT1A affinity. The final compounds, 4a, 8a, and 8b, were selected according to predictions of in silico pharmacokinetics, docking analysis, and molecular dynamics in conjunction with physical properties, and metabolic stability. The accentuated molecules could serve as a lead compound for developing 5-HT1A drug-like molecules for depression treatment.

AB  - Considering the health-benefits of berry fruits consumption and increased market demands for food authenticity
as one of the most important quality assurances, phenolic profiling by high-performance thin layer chroma-
tography and ultra-high-performance liquid chromatography hyphenated with mass spectrometry was combined
with multivariate analysis for phytochemical characterization and intercultivar discrimination of cultivated
berry seeds. The phenolic profiles of 45 berry seeds from nine genuine Serbian cultivated fruit species (straw-
berry, raspberry, blackberry, black currant, blueberry, gooseberry, cape gooseberry, chokeberry, and goji berry)
revealed a good differentiation according to botanical origin. In order to determine biomarkers responsible for
the classification, a total of 103 phenolic compounds were identified, including 53 phenolic acids and their
derivatives, 26 flavonoids and 24 glycosides. Biomarkers derived from the phenolic profile of berry seeds proved
to be a powerful tool in the authentication of botanical origin, and may be useful in detection of frauds in berry-
based seed-containing product.

AB  - Considering the health-benefits of berry fruits consumption and increased market demands for food authenticityas one of the most important quality assurances, phenolic profiling by high-performance thin layer chroma-tography and ultra-high-performance liquid chromatography hyphenated with mass spectrometry was combinedwith multivariate analysis for phytochemical characterization and intercultivar discrimination of cultivatedberry seeds. The phenolic profiles of 45 berry seeds from nine genuine Serbian cultivated fruit species (straw-berry, raspberry, blackberry, black currant, blueberry, gooseberry, cape gooseberry, chokeberry, and goji berry)revealed a good differentiation according to botanical origin. In order to determine biomarkers responsible forthe classification, a total of 103 phenolic compounds were identified, including 53 phenolic acids and theirderivatives, 26 flavonoids and 24 glycosides. Biomarkers derived from the phenolic profile of berry seeds provedto be a powerful tool in the authentication of botanical origin, and may be useful in detection of frauds in berry-based seed-containing product.

This honey, rich in phenolic content, is well recognized for its antioxidant, anti-inflammatory, and antimicrobial activities, and is used for the treatment of skin lesions as well as gastrointestinal and respiratory disorders. This study investigated whether STH alleviates genome damage in human peripheral blood lymphocytes produced by the cytotoxic drug irinotecan. The phenolic profile of STH was previously estimated by ultra-high-performance liquid chromatography coupled to a linear ion trap–Orbitrap hybrid mass spectrometer. The effects of STH were evaluated at three concentrations (1×, 5×, and 10×), based on the daily consumption of the honey by an adult person. After 2 h of in vitro exposure, standard lymphocyte cultures for the analysis of chromosome aberrations and the cytokinesis-block micronucleus cytome assay were established. Our results demonstrate that STH offered remarkable geno- and cytoprotection when administered with irinotecan. These findings are relevant for drawing preliminary conclusions regarding the in vitro safety of the tested honey. However, further studies are needed with the application of more complex experimental models.

AB  - The aim of this study is to develop a rapid, effect-directed screening method for quality assessment of bee pollen-honey mixtures. The comparative antioxidant potential and phenolic content of honey, bee pollen, and the bee pollen-honey mixtures, was performed using spectrophotometry. The total phenolic content and antioxidative activity of bee pollen-honey mixtures with 20 % bee pollen share were in the range 3.03–3.11 mg GAE/g, and 6.02–6.96 mmol TE/kg, respectively, while mixtures with 30 % bee pollen share contained 3.92–4.18 mg GAE/g, and 9.69–10.11 mmol TE/kg. Chromatographic fingerprint of bee pollen-honey mixtures was performed by high-performance thin-layer chromatography with conditions developed by authors and reported for the first time. Fingerprint analysis hyphenated with chemometrics enabled authenticity assessments of honey in mixtures. Results indicate that bee pollen-honey mixtures represent a food with highly, both, nutritious characteristics and health-promoting effect.

AB  - Propolis is a resinous natural substance collected by honeybees from different plant sources. The element content of propolis is influenced by the content of elements in the soil, climatic factors, and the degree of pollution. The aim of the study was to characterize element composition and content of toxic elements in 51 propolis collected from different locations in Serbia and to evaluate the differences among them. Determination of the 21 elements were performed using Inductively coupled plasma - optical emission spectrometry. K was the most abundant element, followed by Ca, Mg, Si, Fe, Zn, and Al. Kruskal-Wallis tests differentiate groups of samples by geographical origin. The presence of B, Fe, Sr, Ti, Zn, and As demonstrated statistical significance between six regions of Serbia.

AB  - Considering the well-being cognizance of masses, microgreens and germs have emerged as potential therapeutic functional foods for improving overall health by dietary supplementation. Microgreens and germs have an exceptional volume of various nutrients accounting for higher nutritive benefits compared to their mature counterparts [1]. These plants are difficult to store in fresh, due to their high surface area to volume ratio, high respiration rate, delicate leaves that easily wilt, rapid post-harvest decay transpiration, leakage of nutrient-rich exudates, tissue damage, and early senescence [1]. Therefore, within this study, the effect of drying on the sugar content of 12 samples of different microgreens and germs was monitored. A comparative study of the content of vitamin C and sugar between samples of microgreens and germs in both dry and fresh states was also done. The content of vitamin C was determined by the HPLC method, while the sugar profile was obtained using HPAEC-PAD. The results showed that there was a difference between samples of microgreens and germs (fig. 1), as well as the contents of vitamin C and sugar in the samples are inversely proportional (fig. 2), which is in line with the fact that part of the sugar is converted into vitamin C during ripening, which then serves as an antioxidant and protects against harmful effects [2]. Analyzes also showed that dry samples have a higher sugar content compared to fresh ones (fig.2.), which could serve as an argument to promote cost-effective production and future strategies for maintaining the shelf life and quality of these products in the sense that they do not lose their nutritional value during the drying process.

AB  - Besides being one of the most commercially grown berry fruits, strawberries (Fragaria x annanassa Duch.) are known for their nutritional value, richness in polyphenols, and antioxidant capacity. New directions of strawberry breeding and selection are set towards premium fruit quality, high sugars content, and desirable sugars/total acids ratio (sweet index, SI), which represent some of the main prerequisites for selecting new breeding materials [1]. With this intention, a set of 24 strawberry genotypes was cultivated, including 12 parental varieties and 12 of their descendants - perspective candidates obtained by crossing the parental varieties mentioned above. A total of eleven sugars were quantified using High-Performance Anion-Exchange Chromatography with Pulsed Amperometric Detection (HPAEC-PAD). Results implied that glucose, sorbitol, sucrose, and melibiose content were the most important sugars in separating the fruits of parental varieties and their progeny [2]. Results showed that old, parental varieties had a significantly higher content of glucose and sucrose, while new perspective genotypes had higher sorbitol and melibiose content (Fig.2). PCA analysis confirmed that parental (samples 1-12) and descendant genotypes (samples 13-24), could be discriminated according to sugars profile i.e. that these four sugars have an effect on their differentiation (Fig.1). This was in accordance with Mann-Whitney U test results (Fig.2).

AB  - Interest in production of protected crops has grown significantly over the past decade. Different cultivation systems (conventional, integral, and organic) affect the biological activity, which is ultimately reflected in the high quality of the fruits of different vegetable crops. In accordance with the requirements for health-safe products without residues of harmful substances in fruits, there is an increasing need for the production of tomatoes in integral and organic production systems. One of the most important features of these vegetables is their high quality and health safety, high nutritional and biological value. Agricultural production systems and growing practices are critical factors in determining the nutritional quality of tomato fruits [1]. In tomato, the sugar content is one of the important factors and one of the most significant parameters from the aspect of food quality [1]. Therefore, the aim of this work is to monitor fluctuations in sugars content, as parameters
that determine the nutritional value in tomato cultivars, induced by growing under integral and organic conditions. For this purpose, a set of sixteen samples of four types of tomatoes - beef, grapolo, mini and midi plum, and cherry - was analyzed. Each type of tomato included two varieties, grown in two agricultural systems - integral and organic. The sugars profile was obtained using High-Performance Anion Exchange Chromatography with Pulsed Amperometric Detection (HPAECPAD). The content of eleven sugar components was determined. Fructose and glucose were the major sugar compounds [2], while the sugar microcomponents were trehalose, arabinose, melibiose, sucrose, isomaltose, gentiobiose, raffinose, maltose, and panose. The results showed differences between samples produced in integral and organic growing systems, primarily in microsugar components. Higher content of trehalose and melibiose was found in samples obtained from organic production.

AB  - Besides being one of the most commercially grown berry fruits, strawberries (Fragaria x annanassa Duch.) are known for their nutritional value, richness in polyphenols, and antioxidant capacity. New directions of strawberry breeding and selection are set towards premium fruit quality, high sugars content, and desirable sugars/total acids ratio (sweet index, SI), which represent some of the main prerequisites for selecting new breeding materials [1]. With this intention, a set of 24 strawberry genotypes was cultivated, including 12 parental varieties and 12 of their descendants - perspective candidates obtained by crossing the parental varieties mentioned above. A total of eleven sugars were quantified using High-Performance Anion-Exchange Chromatography with Pulsed Amperometric Detection (HPAEC-PAD). Results implied that glucose, sorbitol, sucrose, and melibiose content were the most important sugars in separating the fruits of parental varieties and their progeny [2]. Results showed that old, parental varieties had a significantly higher content of glucose and sucrose, while new perspective genotypes had higher sorbitol and melibiose content (Fig.2). PCA analysis confirmed that parental (samples 1-12) and descendant genotypes (samples 13-24), could be discriminated according to sugars profile i.e. that these four sugars have an effect on their differentiation (Fig.1). AB  - High-Performance Thin-Layer Chromatography (HPTLC)-radical scavenging capacity (RSC) assays are stan- dard techniques for the separation and identification of antioxidants from complex mixtures. HPTLC coupled with DPPH · visualization of chromatograms allows for the detection of individual antioxidants. However, other HPTLC-RSC assays that recognize compounds exhibiting different mechanisms of radical- scavenging activity are rarely reported. In this study, we developed an integrated approach that combines five HPTLC-RSC assays, principal component analysis (PCA) and quantum chemical calculations to assess the antioxidant capacity of Sempervivum tectorum L. leaf extracts. Two HPTLC assays - potassium hexa- cyanoferrate(III) total reducing power assay (TRP) and total antioxidant capacity by phosphomolybdenum method (TAC) - were developed for the first time. The method supports a more in-depth study of the RSC of natural products, as it compares the radical scavenging fingerprints of S. tectorum leaf extracts and recognizes differences in their individual bioactive constituents. Kaempferol, kaempferol 3- O -glucoside, quercetin 3- O -glucoside, caffeic acid, and gallic acid were identified as the compounds that discriminate HPTLC-RSC assays according to their mechanism of action and capture the similarities between 20 S. tec- torum samples. Additionally, DFT calculations on M06–2X/6–31 + G (d,p) level were applied to map ther- modynamic feasibility of hydrogen atom transfer (HAT) and single electron transfer (SET) mechanisms of the identified compounds. Based on experimental and theoretical results, a combination of HPTLC-ABTS and HPTLC-TAC assays were proposed as the optimal method for mapping the antioxidants from S. tec- torum . This study represents a step forward in identifying and quantifying individual antioxidants from complex food and natural product matrices in a more rational manner.

Assessment of antibacterial activity of Sempervivum tectorum L. leaves extracts using HPTLC bioassays and chemometrics

AB  - Sempervivum tectorum L. is a perennial plant with succulent leaves arranged in a rosette. Despite its wide use in traditional medicine for the treatment of various changes on the skin, nervousness, anxiety, menstrual ailments, etc., it still does not have a biography in the European Pharmacopoeia¹. The antibacterial effect of juice squeezed from the leaves and tea made from dried leaves was tested against bacteria that cause inflammation of the ear and bladder². Direct bioautography on HPTLC plates for the investigation of antibacterial activity of the houseleek leaves extracts was done for the first time. Four HPTLC bioassays were developed for gram-positive bacteria (B. subtilis, M. lysodeikticus, MRSA, and S. aureus), and two for gramnegative bacteria (E. coli and K. pneumoniae). Antibacterial activity of grampositive bacteria comes from active compounds at RF 0.57 and 0.92. HPTLC bioassays show higher sensitivity for gram-negative bacteria, as numerous active bands were observed at RF 0.03, 0.38, 0.42, 0.56, 0.57, 0.86 and 0.92. By applying chemometrics and principal components analysis, the HPTLC bioassays of grampositive bacteria were separated into two groups on the scores plot graphic along PC1 axis. The bioassays for B. subtilis and M. lysodeikticus form a less uniform group in the upper left quadrant, while the bioassays for MRSA and S. aureus form a homogeneous group in the lower right quadrant. Presence or absence of the active bands at RF 0.57 and 0.92 affect these separations. The bioassays for the two gramnegative bacteria are separated along both axes on the scores plot graphic from each other but also from the other bioassays for the gram-positive bacteria. This separation is influenced by the presence of numerous active bands. For further investigation it is necessary to identify compounds responsible for the antibacterial activity of the houseleek leaves extracts.

To investigate their potential for extracting phenolic compounds from plant materials, we employed the COSMO-RS (COnductor like Screening MOdel for Real Solvents) computational method. Nineteen NES solvents were prepared to determine the most selective and effective solvent for extracting phenolic compounds from Teucrium chamaedrys using both theoretical and experimental evaluation. Screening of extraction efficiency was performed by quantification of individual compounds using ultra-high-performance liquid chromatography with a diode array detector and a triple-quadrupole mass spectrometer (UHPLC-DAD-MS/MS), as well as spectrophotometric assays (total phenolic content, total flavonoid content, and radical scavenging activity). In this paper, we propose a new approach to model NES properties using COSMO-RS. While the majority of previous studies have modeled these solvents as individual or pseudo-component complexes between hydrogen bond donor (HBD) and hydrogen bond acceptor (HBA), we have developed a ternary HBD:HBA:water supramolecular complex model to represent NES solution structures more accurately. This model provides insights into the intermolecular interactions driving the extraction process and predicts extraction efficiencies that agree with experimental data. Two eutectic mixtures (choline chloride:succinic acid in molar ratio 1:1 and 20% of water (w/w) and choline chloride: glycerol in molar ratio 1:1 and 20% of water (w/w)) showed strong affinity towards phenolic compounds. Overall, our findings suggest that the ternary complex model is a more appropriate approach for modeling NES properties.

AB  - Natural Deep Eutectic Solvents (NADES) are novel and promising solvents for green extraction of phytochemicals from food and agricultural products. NADES are made of natural origin compounds that are connected via hydrogen bond and have unique tuneable properties1. Numerous studies investigated the extraction of bioactive compounds using NADES resulted in better extraction efficiency compared to conventional solvents, suggesting large potential of these solvents2,3. Low-toxicity, natural origin and affordability of NADES-based extracts enables their application as a preservative in the food industry or functional ingredient of food supplements. The aim of this study was to develop an efficient eco-friendly method for the extraction of phenolic compounds from berries, blueberry, chokeberry and black goji berry, using NADES. This goal will be achieved through investigation of extraction efficiency of applied NADES based on three different approaches, a) chromatographic, through high-performance thin layer chromatography (HPTLC) fingerprint analysis, ultra-high-performance liquid chromatography with a diode array detector and a triple-quadrupole mass spectrometer (UHPLC-DAD-MS/MS) target analysis and bioautography, b) spectroscopic, through quality control parameters, total phenolic content (TPC), total flavonoid content (TFC) and radical scavenging activity (RSA), and c) microbiological, through well diffusion method and minimum inhibitory concentration (MIC).
In this study, 36 NADES mixtures, prepared from primary plant metabolites, were tested as green alternatives for extraction of phenolics from three berries. Different hydrogen-bond acceptors (HBA), such as choline chloride, L-proline, L-glycine, and L-lysine were mixed with various hydrogen-bond donors (HBD), four organic acids, two sugars, glycerol and urea, at various molar ratios for NADES preparation. Guided by the fact that extraction efficiency depends on polarity, viscosity and dissolving ability of solvents4, the influence of HBA, HBD and water content were investigated. Methanol, as conventional solvent, was used as contrastive solvent.
Choline chloride-based NADES in combination with malic acid, glycerol or urea showed as most efficient. All groups of studied NADES showed various extraction capabilities for phenolic compounds based on chromatographic evaluation and spectrophotometric tests. These results indicate great power of NADES solvents towards selective extraction of phenolics and shows that NADES are designer solvents. Appropriate selection of components for NADES preparation can result in selective extraction of specific phenolic compounds, regardless of how similar structures of targeted compounds are or how complex the matrix is, which gives a significant advantage for application of NADES.

Validation and optimization of ion chromatography based method for citric acid determination in Robinia pseudoacacia honey

AB  - Honey is very complex natural substance consisted of sugars, water, and other compounds in smaller quantities such as proteins (enzymes), organic acids, vitamins, minerals, phenolics, and a large variety of volatile compounds. Discriminating the honeys according to botanical and/or geographical origin may be possible according to their characteristic constituents including organic acids profile. These acids are related to the colour and flavour of honey and its chemical properties such as acidity, pH and electrical conductivity. Honey is deceptively acidic, as the high sugar content tends to mask the acidity in the taste. Citric acid among other acids, is present in honey in moderate amount but it's presence could be potentially used as marker for authenticity control [1].

Fraudulent addition of compounds in honey sets a necesity to establish the natural level of the specific substances, and subsequently determine when this level is exceeded, indicating adulteration. Citric acid can be added to decrease pH in adulterated honey where normal pH levels have been exceeded. Also increased citrate concentration in honeys could be a result of addition of invert syrup to honey. Therefore there is a need for a method to determine citrate content in honey quickly and efficiently1. In current work a method for determination of organic acids profile in honey using ion chromatography with conductometric detection (IC-CD) without prior separation of compounds and chemical derivatization was developed. This approach for citric acid determination is characterized by LOD and LOQ values of 0.05 and 0.20 mg/L, respectively. Low RSD Sy/x value and r2 level of 0.9999 indicates a good regression model. Trueness of method was confirmed by good recoveries (108%, 104% and 95% for spiked levels 75%, 100% and 125%, respectively), while precision was confirmed by HorRat value (0.30) less than 2 [2].

AB  - Soluble sugars and organic acids have a strong impact on the overall sensory quality of fruits. Organic acids also facilitate in stabilization of anthocyanins, and they extend the shelf-life of fresh fruits and their processed products. The production of new apple-based products with health beneficial properties is leading to agronomical and chemical re-evaluation of old apple cultivars.
In this study the mesocarp samples of five conventional, five resistant and fifteen autochthonous apple cultivars from a single growing season were deliberated. Assessment was based on contents of four sugars, one sugar alcohol and five organic acids that were determined by the usage of HPAEC-PAD and HPAEC-CD systems. Fructose was detected as dominant sugar in all cases (50.4 – 77.9 mg/kg FW) and malic acid as dominant acid (1.1 – 4.5 mg/kg FW). Regarding the average values, xylose and sorbitol were in higher amount in autochthonous cultivars compared to others. In resistant cultivars shikimate and malate were found in lower concentrations compared to other cultivar types. Among the obtained data, highest correlation values were found between citrate and isocitrate, and also between citrate and malate (0.905 and 0.839, respectively).

AB  - Strawberry (Fragaria x ananassa Duch.) is widely known for its nutritional value, richness in polyphenols and antioxidant capacity. In abundance of numerous ongoing strawberry breeding programs worldwide, some of the main parameters for breeding and selection are fruit quality, flavour and aroma which are directly affecting commercial acceptance [1]. These could be easily identified with some of objective measurements such as soluble solids concentration (°Brix), acid contents (titratable acidity, TA) and Brix/acid ratio, which could be strong predictors of quality in terms of consumer acceptability [1,2]. In this trial, we compared 24 strawberry genotypes, including 12 parental varieties and 12 perspective candidates obtained by crossing these particular parental varieties. Results of °Brix, titratable acidity and °Brix/acid ratio measurements indicated that new perspective candidates have had higher °Brix/acid ratio than the parental varieties, leading to conclusion that those genotypes had better flavour. Therefore, this ratio could potentially be used as indicator for distinguishing consumer acceptance.

abstract = "Strawberry (Fragaria x ananassa Duch.) is widely known for its nutritional value, richness in polyphenols and antioxidant capacity. In abundance of numerous ongoing strawberry breeding programs worldwide, some of the main parameters for breeding and selection are fruit quality, flavour and aroma which are directly affecting commercial acceptance [1]. These could be easily identified with some of objective measurements such as soluble solids concentration (°Brix), acid contents (titratable acidity, TA) and Brix/acid ratio, which could be strong predictors of quality in terms of consumer acceptability [1,2]. In this trial, we compared 24 strawberry genotypes, including 12 parental varieties and 12 perspective candidates obtained by crossing these particular parental varieties. Results of °Brix, titratable acidity and °Brix/acid ratio measurements indicated that new perspective candidates have had higher °Brix/acid ratio than the parental varieties, leading to conclusion that those genotypes had better flavour. Therefore, this ratio could potentially be used as indicator for distinguishing consumer acceptance.",

AB  - Bisbibenzyls are specialized metabolites found exclusively in liverworts, until recently; they represent chemical markers of liverworts. Their occurrence in vascular plants was noticed in 2007, when they were found in Primula veris subsp. macrocalyx from Russia. This report prompted us to chemically analyze the two most common Serbian Primula species, P. veris subsp. columnae and P. acaulis, in order to determine the presence of bisbibenzyls in them. Our study revealed nine structurally distinct bisbibenzyls (1–9), identified based on 1D and 2D NMR, IR, UV and HRESIMS data. Among them were five previously undescribed compounds (2–6). The remaining com­ pounds found and previously described in the literature were: the bisbibenzyls riccardin C (1), isoperrottetin A (7), isoplagiochin E (8) and 11-O-demethylmarchantin I (9), as well as 4-hydroxyphenylmethylketone (10) and 4-hydroxy-3-methoxyphenylmethylketone (11). Riccardin C was the most dominant bisbibenzyl in both species studied. Previously, it was the first bisbibenzyl found in vascular plants (P. veris subsp. macrocalyx). An assessment of the cytotoxic activity of the isolated compounds against A549 lung cancer and healthy MRC5 cell lines was also the subject of our study. Compounds 6 and 9 exhibited significant cytotoxic activity expressed by IC50 values of 12 μM, but the selectivity was not satisfactory.

AB  - The synthesis of 24 hybrid molecules, consisting of naturally occurring sclareol (SCL) and synthetic 1,2,4-triazolo[1,5-a]pyrimidines (TPs), is described. New compounds were designed with the aim of improving the cytotoxic properties, activity, and selectivity of the parent compounds. Six analogs (12a-f) contained 4-benzylpiperazine linkage, while 4-benzyldiamine linkage was present in eighteen derivatives (12g-r and 13a-f). Hybrids 13a-f consist of two TP units. After purification, all hybrids (12a-r and 13a-f), as well as their precursors (9a-e and 11a-c), were tested on human glioblastoma U87 cells. More than half of the tested synthesized molecules, 16 out of 31, caused a significant reduction of U87 cell viability (more than 75% reduction) at 30 µM. The concentration-dependent cytotoxicity of these 16 compounds was also examined on U87 cells, corresponding multidrug-resistant (MDR) U87-TxR cells with increased P-glycoprotein (P-gp) expression and activity, and normal lung fibroblasts MRC-5. Importantly, 12l and 12r were active in the nanomolar range, while seven compounds (11b, 11c, 12i, 12l, 12n, 12q, and 12r) were more selective towards glioblastoma cells than SCL. All compounds except 12r evaded MDR, showing even better cytotoxicity in U87-TxR cells. In particular, 11c, 12a, 12g, 12j, 12k, 12m, 12n, and SCL showed collateral sensitivity. Hybrid compounds 12l, 12q, and 12r decreased P-gp activity to the same extent as a well-known P-gp inhibitor - tariquidar (TQ). Hybrid compound 12l and its precursor 11c affected different cellular processes including the cell cycle, cell death, and mitochondrial membrane potential, and changed the levels of reactive oxygen and nitrogen species (ROS/RNS) in glioblastoma cells. Collateral sensitivity towards MDR glioblastoma cells was caused by the modulation of oxidative stress accompanied by inhibition of mitochondria.

AB  - Backgound/ObjectivesUrolithins (UROs) are the metabolites derived from the gut microbial action on ellagitannins and ellagic acid-rich foods. Following their absorption in the intestine, UROs are transported through the systemic circulation to various tissues where they can express their biological function as antimicrobial, anti-inflammatory, and anticancer agents. In addition to blood plasma, where they can be found as glucuronide and sulfate conjugates, they are also found in urine. Therefore, the interactions of UROs with serum proteins are of great clinical interest.MethodsA powerful technique for examining these urolithin-serum protein interactions is fluorescence spectroscopy. Bovine serum albumin (BSA) is a particularly suitable model protein because it is readily available, affordable, and similar to human serum albumin. This work aimed to study the binding of UROs (urolithin A, UROA and urolithin B, UROB) and their glucuronide conjugates (UROAG and UROBG) to BSA by quenching the intrinsic fluorescence of protein.ResultsThe spectra obtained showed that the binding process is influenced by the polyphenol's structure and the conjugation process with the glucuronide. The calculated Stern Vollmer binding constants (Ksv): UROA and UROB Ksv were 59236   ±   5706 and 69653   ±   14922, respectively, while for UROAG and UROBG, these values were 15179   ±   2770 and 9462   ±   1955, respectively, which showed that the binding affinity decreased with glucuronidation. Molecular docking studies confirmed that all of the studied molecules will bind favorably to BSA. The preferential binding site for both UROs and UROGs is Sudlow I, while UROs will also bind to Sudlow II. URO-Gs can bind to BSA in the cleft region with lower binding scores than for the Sudlow I binding site.ConclusionThe aglycone's higher hydrophobicity increases the binding affinity to BSA, thus reducing its bioavailability in the blood.

AB  - The combination of an intense absorption of visible light and p-type semiconducting nature makes spinel cobalt oxide (Co3O4) a very attractive material for various optoelectronic applications. However, the traditional methods for its synthesis have been either time- and energy-consuming or relying on toxic chemicals. To solve this issue, a simple, facile, and eco-friendly method of synthesis was successfully developed to obtain spinel Co3O4 nanoparticles. The novel method for obtaining pure and monophasic Co3O4 reported here is based on the thermal decomposition of hexaaquacobalt(II) D-camphor10-sulfonate at 900 °C. This fast solid-state synthesis route overcomes the disadvantages of many combustion methods, most critically by avoiding the use of toxic organic solvents. The synthesized material was subjected to a detailed characterization to assess its potential for use as a nanocatalyst. The band gap measurements indicated the presence of two band gaps, one at 2.10 eV and another at 1.22 eV, confirming the purity and semiconducting properties of the sample. The electrochemical studies demonstrated a significant enhancement in the electron transfer kinetics with the addition of the synthesized Co3O4 to the carbon-paste electrode, leading to an enhanced electrocatalytic performance. These prominent functional properties, suitable for a wide range of technological applications, pave way for the implementation of the reported method for the synthesis of Co3O4 on a larger industrial scale.
