Novel derivatives of aroylacrylic acid phenylamides as inducers of apoptosis through the ROS-mediated pathway in several cancer cell lines

AB  - In the present work, the α,β-double bond of the aroylacrylic acid phenylamides was suitably modified to optimize the toxicity–antiproliferative activity ratio of the resulting compounds 1–5. The phenylamides were modified by Michael's addition of suitably chosen piperidine-containing fragments: 1-amino-N-benzylpiperidine (a1), 4-benzylpiperidine (a2), and N,N-dimethyl-N-[2-(1-piperazinyl)-ethyl]amine (a3). The compounds exerted micromolar activity toward three cancer cell lines, A549, LoVo, and Skov-3, causing apoptotic cell death. It was shown that the nature of the cyclic amine moiety at position C2 of the compounds is probably the primary determinant of anticancer activity toward tested cell lines and the acute toxicity toward brine shrimp (Artemia salina). The majority of compounds revealed the ability to vigorously induce mitochondrial superoxide anion generation in all treated cell lines, which together with cell cycle arrest at the S phase and activation of intrinsic caspase cascade, indicates the possibility that apoptosis was triggered due to irreparable chromosomal damage by acute oxidative stress. Two derivatives also exerted significant antibacterial activity with one order of magnitude higher potency than chloramphenicol in most of the investigated bacterial strains. Also, the drug-like properties for all compounds were estimated by available software tools.

AB  - Benefits and risks of the fish consumption should be assessed to balance the ingestion of healthy omega-3 fatty acids and adverse chemicals. Persistent organic pollutants-POPs (organochlorine pesticides-OCPs and polychlorinated biphenyls-PCB), macro- and micro-elements and fatty acid contents were determined in six fish species from the Adriatic Sea to assess health risks for consumers (worst-case scenario, diseases development risks and benefit-risk). 16 element, 24 POPs and 14 fatty acid contents were determined in six pelagic species which can be used in human diet. Element concentrations were determined by inductively coupled plasma mass spectrometry (ICP-MS), POPs by high-resolution gas chromatography and fatty acid content by gas–liquid chromatography. Diet based on chub mackerel and round sardinella showed lower daily intake (DI) of POPs and highest DI of essential omega-3 fatty acids than other investigated species. Lower ingestion of toxic elements can be observed by consuming anchovy and round sardinella. Based on POP concentrations, there was not observed non-carcinogenic (HI) nor carcinogenic (CR) risks for consumers. Based on element concentrations, there was low HI (0.1 ≥ HI ≥ 1), while the maximum HIs and outlier values (horse mackerel and anchovy samples) implied the presence of HI (HI > 1). The most significant contributor to total non-carcinogenic and carcinogenic risks was inorganic arsenic (inorganic As). Acceptable CR for consumers was assessed, but maximum CR for consumers of horse mackerel and anchovy (CR ≥ 1 × 10–6) showed adverse effects on human health. There were low HIs for developing cardiovascular, nervous, and reproductive diseases, and maximum HIs were higher than 1. Acceptable (1 × 10−4≥CR ≥ 1 × 10−6) risks were observed for developing cancer of nervous system and reproductive organs. Among investigated fish samples, those with higher ƩBR (benefit-risks) and BR for arsenic (As) than median value have a higher risk than benefits in the human diet.

The programme for professional development of chemistry teachers’ assessment competency

AB  - The aim of this paper is to investigate the effects of the programme for professional development of chemistry teachers on their competencies for conducting formative and summative assessment in chemistry teaching. The programme participants were 30 chemistry teachers from primary and secondary schools. Data were collected using a questionnaire at the beginning and at the end of the programme implementation. The programme included four workshops with the same structure: the introduction, group work and the discussion of the results obtained through group work. The workshops focused on: i) the assessment as a support for chemistry learning; ii) the harmonization of teaching and learning activities, formative and summative assessment, feedback from formative assessment and the criteria used to evaluate students in summative assessment; iii) the evaluation of the validity of tasks used for formative and summative assessment according to the curricula aims and the educational standards; iv) designing tasks for monitoring students’ progress towards certain educational standards. Teachers’ responses show the impact of the programme for the development of their competencies for assessment, particularly regarding formative and summative assessment and designing various kinds of assessment in accordance with the achievement standards.

AB  - Cadmium (Cd) ranks seventh on the list of most significant potential threats to human health based on its suspected toxicity and the possibility of exposure to it. It has been reported that some bacterial exopolysaccharides (EPSs) have the ability to bind heavy metal ions. We therefore investigated the capacity of eight EPS-producing lactobacilli to adsorb Cd in the present study, and Lactiplantibacillus plantarum BGAN8 was chosen as the best candidate. In addition, we demonstrate that an EPS derived from BGAN8 (EPS-AN8) exhibits a high Cd-binding capacity and prevents Cd-mediated toxicity in intestinal epithelial Caco-2 cells. Simultaneous use of EPS-AN8 with Cd treatment prevents inflammation, disruption of tight-junction proteins, and oxidative stress. Our results indicate that the EPS in question has a strong potential to be used as a postbiotic in combatting the adverse effects of Cd. Moreover, we show that higher concentrations of EPS-AN8 can alleviate Cd-induced cell damage.

AB  - The worldwide outbreak of COVID-19 was caused by a pathogenic virus called Severe Acute Respiratory Syndrome Coronavirus-2 (SARS-CoV-2). Therapies against SARS-CoV-2 target the virus or human cells or the immune system. However, therapies based on specific antibodies, such as vaccines and monoclonal antibodies, may become inefficient enough when the virus changes its antigenicity due to mutations. Polyphenols are the major class of bioactive compounds in nature, exerting diverse health effects based on their direct antioxidant activity and their effects in the modulation of intracellular signaling. There are currently numerous clinical trials investigating the effects of polyphenols in prophylaxis and the treatment of COVID-19, from symptomatic, via moderate and severe COVID-19 treatment, to anti-fibrotic treatment in discharged COVID-19 patients. Antiviral activities of polyphenols and their impact on immune system modulation could serve as a solid basis for developing polyphenol-based natural approaches for preventing and treating COVID-19.

AB  - In an effort to determine the thermochemistry of established organometallic transformation, the well documented reaction of alkynes with a palladacycle was investigated by isothermal titration calorimetry (ITC). Although the mechanism of the insertion of unsaturated substrates into the Pd−C bond of cyclopalladated compounds is known, no information is available so far about their thermochemistry. The enthalpies of the reactions of Ph−C≡C−Ph and MeOC(O)−C≡C(O)COMe with the bisacetonitrilo salt of the N,N-benzylamine palladacycle were determined by ITC in chlorobenzene after having optimized the conditions to ensure that only the double and a single insertion of alkynes were occurring respectively. The reaction energy profile established by DFT for the double insertion process involving Ph−C≡C−Ph confirmed earlier conclusions on the rate determining character of the first insertion. Further computations of reaction enthalpies reveal significant discrepancies between ITC and DFT-D/continuum solvation enthalpies, that are suspected to arise from an unexpected explicit noncovalent interaction of PhCl with the components of the reaction.

AB  - Dugotrajne organske zagađujuće supstance su organska jedinjenja otporna na fotolitičku, hemijsku i biološku degradaciju. Dobro se rastvaraju u lipidima, što im omogućava bioakumulaciju u u masnim tkivima živih organizama. Izlaganje ovim jedinjenjima povezano je sa širokim spektrom negativnih efekata na zdravlje ljudi, kao što su: imunotoksičnost, kancerogenost i nepovoljni uticaji na reproduktivni razvoj, nervni i endokrini sistem. Sve ove osobine dugotrajnih organski zagađujućih supstanci čine da one postanu jedna od glavnih tema u oblasti zaštite životne sredine. Kao globalno rešenje problema prisustva ovih jedinjenja doneta je Stokholmska konvencija čiji je cilj zaštita zdravlja ljudi i životne sredine.
U ovom radu prikazani su rezultati dobijeni analizom uzoraka majčinog mleka. Ispitan je sadržaj polihlorovanih bifenila, dihlorodifenil-trihloroetana i njegovih metabolita, heksahlorbenzena i grupe heksahlorocikloheksan pesticida.

Efekti primene onlajn materijala za učenje o protolitičkoj teoriji u prvom razredu gimnazije

AB  - Cilj ovog istraživanja je bilo da se ispitaju efekti primene onlajn materijala za učenje o protolitičkoj teoriji u prvom razredu gimnazije. Istraživanje je sprovedeno na 130 učenika prvog razreda dve gimnazije. Instrumenti korišćeni u istraživanju su ulazni test, kojim se proveravalo prethodno znanje učenika o kiselinama i bazama, izlazni test, kojim se proveravala uspešnost usvajanja gradiva o protolitičkoj teoroji pomoću onlajn materijala i upitnik za ispitivanje učeničkih stavova. Svim učesnicima istraživanja je bio dostavljen onlajn materijal identičnog sadržaja, ali na jeziku na kojem prate nastavu hemije (srpski i slovački), u okviru kojeg su usvajali znanja o protolitičkoj teoriji sopstvenim tempom kroz male porcije gradiva, praćene zadacima za utvrđivanje. Iz inicijalnog testa se uviđa da obe grupe učenika imaju predznanje o kiselinama i bazama. Rezultati izlaznog testa sugerišu se da je grupa Gimnazije Uroš Predić postigla bolje rezultate nakon korišćenja onlajn materijala, nego što je tu slučaj kod ispitanika iz uzorka koji pohađaju Gimnaziju Mihajlo Pupin.

Istraživanje je sprovedeno na 130 učenika prvog razreda dve gimnazije. Instrumenti korišćeni u istraživanju su ulazni test, kojim se proveravalo prethodno znanje učenika o kiselinama i bazama, izlazni test, kojim se proveravala uspešnost usvajanja gradiva o protolitičkoj teoroji pomoću onlajn materijala i upitnik za ispitivanje učeničkih stavova. Svim učesnicima istraživanja je bio dostavljen onlajn materijal identičnog sadržaja, ali na jeziku na kojem prate nastavu hemije (srpski i slovački), u okviru kojeg su usvajali znanja o protolitičkoj teoriji sopstvenim tempom kroz male porcije gradiva, praćene zadacima za utvrđivanje. Iz inicijalnog testa se uviđa da obe grupe učenika imaju predznanje o kiselinama i bazama. Rezultati izlaznog testa sugerišu se da je grupa Gimnazije Uroš Predić postigla bolje rezultate nakon korišćenja onlajn materijala, nego što je tu slučaj kod ispitanika iz uzorka koji pohađaju Gimnaziju Mihajlo Pupin.",

U odnosu na to, procenat tačnih odgovora je značajno niži na zahtevima da se odredi broj protona, elektrona i neutrona u atomima elemenata na osnovu položaja elementa u Periodnom sistemu, a niži je i za prepoznavanje trendova u Periodnom sistemu. Učenici svih razreda u najvećoj meri iz Periodnog sistema elemenata koriste podatke o atomskom i masenom broju, simbolu i nazivu elementa, kao i položaju elementa u PSE.

Synthesis and Characterization of Azido Esters as Green Energetic Plasticizers

AB  - Three azido-esters based green energetic plasticizers were synthesized from their chlorides including DEGBAA (diethyleneglycol bis(azidoacetate)), DPGBAA (dipropyleneglycol bis(azidoacetate)) and HETTAA (hexanetriol tris(azidoacetate)). The syntheses were carried out in a two-step process: the first step was esterification of glycol or triol using chloroacetyl chloride, and the second step was substitution of chloracetate with sodium azide that yields corresponding azido derivatives. The parameters of synthesis such as molar ratio of hydroxyl and acyl groups, and amount and type of solvent (dimethyl sulfoxide and dimethyl formamide) were optimized to achieve maximal conversion and purity of the products. The obtained products were characterized by elemental analysis, nuclear magnetic resonance (NMR), and infrared vibrational spectroscopy (IR). Thermal and rheological properties were determined using DSC and Modular Compact Rheometer. Condensed phase heat of formation and several properties important for high-energy materials were predicted from quantum chemical calculations using CBS-4M method. Detonation and combustion performance of energetic compounds were calculated with the thermochemical computer code EXPLO5V06.05. using the predicted heats of formation and experimentally determined densities as input. The energetic and physical properties of the synthesized compounds were compared to the literature data for common plasticizers.

AB  - Pheochromocytoma (PCC) is an unusual benign adrenal tumor with an unexamined status of the elements. This study delivers the first insight into the levels of microelements (Mn, Co, Cu, Zn, As, Se, Th), toxic elements (Cd, Pb, U), and macroelements (Na, K, Mg, Ca) in both adrenal tissue and whole blood samples collected from PCC patients. The results were strengthened by comparing recorded findings with the patients’ healthy adrenal tissue (HAT) and with whole blood samples from the healthy individuals. PCCs had significantly higher levels of Zn, Se, Na, K, and Mg and lower levels of Mn, Co, Pb, and As than that of HATs. Compared to healthy blood samples, the patients’ blood exhibited considerably higher levels of Na, K, and Ca but significantly lower levels of Mn, Cu, Zn, Se, and Mg. Females had significantly higher levels of essential Se and toxic Pb and Cd in their PCC tissue samples compared to males. PCC tissue levels of Mn and Cu were significantly elevated in smokers over levels in nonsmokers and in PCC patients with tumor sizes below 5 cm compared to PCC diameters above 5 cm. The data presented in this study provide a new insight into the pathophysiology of PCC. Thus, recorded elements should be considered as initiators/modifiers of PCC and potential inductors of malignant transformation. The findings of this research deepen scientific understanding of this rare adrenal disease, which, in turn, could highlight the pathogenesis of PCC.

AB  - Zeolitic imidazolate framework-8 (ZIF-8) is widely used as a protective coating to encapsulate proteins via biomimetic mineralization. The formation of nucleation centers and further biocomposite crystal growth is entirely governed by the pure electrostatic interactions between the protein’s surface and the positively charged Zn(II) metal ions. It was previously shown that enhancing these electrostatic interactions by a chemical modification of surface amino acid residues can lead to a rapid biocomposite crystal formation. However, a chemical modification of carbohydrate components by periodate oxidation for glycoproteins can serve as an alternative strategy. In the present study, an industrially important enzyme glucose oxidase (GOx) was selected as a model system. Periodate oxidation of GOx by 2.5 mM sodium periodate increased negative charge on the enzyme molecule, from −10.2 to −36.9 mV, as shown by zeta potential measurements and native PAGE electrophoresis. Biomineralization experiments with oxidized GOx resulted in higher specific activity, effectiveness factor, and higher thermostability of the ZIF-8 biocomposites. Periodate oxidation of carbohydrate components for glycoproteins can serve as a facile and general method for facilitating the biomimetic mineralization of other industrially relevant glycoproteins.

AB  - Aromaticity–antiaromaticity switch upon singlet–triplet transition of some biologically and synthetically important monoheterocycles (heteroatom = N, O, Si, P, and S) derived from benzene, naphthalene and anthracene was studied by employing energetic, magnetic and structural aromaticity criteria, at the density functional theory (DFT) level. The relationship between spin density distribution, (anti)aromaticity and singlet–triplet energy gaps, in the studied molecules, was found. In general, spin delocalization results in antiaromaticity, spin density localization to one ring in bi- and tricycles localizes antiaromaticity and spin localization on a heteroatom reduces global and local antiaromaticity. The latter reaches nonaromaticity in the case of silicon atoms which have larger orbitals and show more tendency to accept unpaired electrons. Spin density localization in bi- and tricycles allows benzene subunit(s) to develop local aromaticity, which, when combined with nonaromatic silacycle and weak global antiaromaticity, results in overall triplet state weak aromaticity. The singlet–triplet energy gaps decrease with a decrease in the triplet state antiaromaticity and are the lowest for silicon-containing compounds.

AB  - BACKGROUND Movement of the red flour beetle, Tribolium castaneum (Herbst) (Coleoptera: Tenebrionidae), in stored products is mediated by food volatiles and other semiochemicals. RESULTS In two-way olfactometer assays, T. castaneum was more attracted to wheat bran previously infested with conspecifics than to uninfested bran. Chemical analysis showed that 1-pentadecene was present in the headspace of T. castaneum-infested wheat bran, but not detectable in the headspace of uninfested bran. An olfactometer was used to test the effect on T. castaneum behaviour of 1-pentadecene, and of volatiles from wheat bran with and without 1-pentadecene. The lowest concentration of 1-pentadecene exhibited an attractive effect, compared to the control (n-hexane). Slightly higher concentrations showed a neutral effect, while the highest concentrations repelled T. castaneum. Wheat bran with a low 1-pentadecene concentration was more attractive than wheat bran alone, whereas higher concentrations of 1-pentadecene were repellent. CONCLUSION The results provide important information on intraspecific, semiochemical-mediated behaviour in T. castaneum, which could potentially be used to develop new methods to monitor the flour beetles in stored products.

Zn(II) complex possesses a distorted trigonal bipyramidal geometry while surroundings around Bi(III) ion are extended pentagonal bipyramidal. Antimicrobial activity, brine shrimp assay and DPPH radical scavenging activity of both complexes, including previously synthesized complexes with HL2Cl ligand (Zn(II) and Ni(II)) and complexes with (E)-N,N,N-trimethyl-2-oxo-2-(2-(1-(pyridin-2-yl)ethylidene)hydrazinyl)ethan-1-aminium chloride (HL3Cl) (Zn(II), Cu(II), Cd(II), Co(II), Fe(III), Ni(II)), were evaluated. For the most active complexes, cytotoxic activity against five malignant cancer cell lines (HeLa, A375, MCF7, PC-3 and A549) and normal cell line HaCaT, as well as generation of reactive oxygen species (ROS), was tested.

The aim was the screening of semi-volatile organic compounds from plants’ shoots using comprehensive two-dimensional gas chromatography–mass spectrometry, a powerful separation technique allowing to reach unique separation resolution, and investigation of qualitative changes after the treatment with coated cerium oxide nanoparticles. The results were analyzed by the identification of individual peaks and fingerprint analysis by image processing. Wheat samples contained a higher number of semi-volatile organic compounds (108) compared to pea (77) but were less affected by the treatments with coated nanoparticles. The highest number of compounds was detected in wheat after the treatment with levan- and pullulan-coated nanoparticles, and in pea after treatment with levan-coated nanoparticles. This article reports a successful application of a semi-volatile organic compounds profile presented only as categorical variables and unique fingerprint images for the inter-cultivar recognition. This method may be useful in screening nanoparticles’ effects on different plants.

AB  - S-Adenosyl-L-Homocysteine (AdoHcy) is a crucial pathological factor in homocysteine-associated disorders. We developed an impedimetric approach for single drop S-Adenosyl-L-Homocysteine detection, based on S-Adenosyl-L-Homocysteine hydrolase (SAH), produced in the prokaryotic expression system (E. coli) by recombinant DNA technology, immobilized on citrate acid-coated gallium oxyhydroxide dispersed over single-walled carbon nanotubes (CA/GaO(OH)@SWCNT). The proposed biosensor offers detection of AdoHcy, with a limit of detection (LOD) of 0.17 μM. with operating linear range from 1 to 44 μM. The remarkable stability of gallium oxyhydroxide crystal was further improved by citric acid coating and its optical, electrical and physical anisotropy was skipped using SWCNTs.
