Novel derivatives of aroylacrylic acid phenylamides as inducers of apoptosis through the ROS-mediated pathway in several cancer cell lines

AB  - In the present work, the α,β-double bond of the aroylacrylic acid phenylamides was suitably modified to optimize the toxicity–antiproliferative activity ratio of the resulting compounds 1–5. The phenylamides were modified by Michael's addition of suitably chosen piperidine-containing fragments: 1-amino-N-benzylpiperidine (a1), 4-benzylpiperidine (a2), and N,N-dimethyl-N-[2-(1-piperazinyl)-ethyl]amine (a3). The compounds exerted micromolar activity toward three cancer cell lines, A549, LoVo, and Skov-3, causing apoptotic cell death. It was shown that the nature of the cyclic amine moiety at position C2 of the compounds is probably the primary determinant of anticancer activity toward tested cell lines and the acute toxicity toward brine shrimp (Artemia salina). The majority of compounds revealed the ability to vigorously induce mitochondrial superoxide anion generation in all treated cell lines, which together with cell cycle arrest at the S phase and activation of intrinsic caspase cascade, indicates the possibility that apoptosis was triggered due to irreparable chromosomal damage by acute oxidative stress. Two derivatives also exerted significant antibacterial activity with one order of magnitude higher potency than chloramphenicol in most of the investigated bacterial strains. Also, the drug-like properties for all compounds were estimated by available software tools.

AB  - Benefits and risks of the fish consumption should be assessed to balance the ingestion of healthy omega-3 fatty acids and adverse chemicals. Persistent organic pollutants-POPs (organochlorine pesticides-OCPs and polychlorinated biphenyls-PCB), macro- and micro-elements and fatty acid contents were determined in six fish species from the Adriatic Sea to assess health risks for consumers (worst-case scenario, diseases development risks and benefit-risk). 16 element, 24 POPs and 14 fatty acid contents were determined in six pelagic species which can be used in human diet. Element concentrations were determined by inductively coupled plasma mass spectrometry (ICP-MS), POPs by high-resolution gas chromatography and fatty acid content by gas–liquid chromatography. Diet based on chub mackerel and round sardinella showed lower daily intake (DI) of POPs and highest DI of essential omega-3 fatty acids than other investigated species. Lower ingestion of toxic elements can be observed by consuming anchovy and round sardinella. Based on POP concentrations, there was not observed non-carcinogenic (HI) nor carcinogenic (CR) risks for consumers. Based on element concentrations, there was low HI (0.1 ≥ HI ≥ 1), while the maximum HIs and outlier values (horse mackerel and anchovy samples) implied the presence of HI (HI > 1). The most significant contributor to total non-carcinogenic and carcinogenic risks was inorganic arsenic (inorganic As). Acceptable CR for consumers was assessed, but maximum CR for consumers of horse mackerel and anchovy (CR ≥ 1 × 10–6) showed adverse effects on human health. There were low HIs for developing cardiovascular, nervous, and reproductive diseases, and maximum HIs were higher than 1. Acceptable (1 × 10−4≥CR ≥ 1 × 10−6) risks were observed for developing cancer of nervous system and reproductive organs. Among investigated fish samples, those with higher ƩBR (benefit-risks) and BR for arsenic (As) than median value have a higher risk than benefits in the human diet.

Rapid analytical approach for bioprofiling compounds with radical scavenging and antimicrobial activities from seaweeds

AB  - Brown seaweeds are traditionally used as food in Asian countries, and they are a valuable source of bioactive compounds. Herein, a novel high-throughput methodological approach was developed for the tracing of compounds with radical scavenging and antimicrobial activities in Saccharina japonica and Undaria pinnatifida methanol extracts. The seaweed metabolites were separated by a novel high-performance thin-layer chromatography method, the bioactive bands were identified by bioautography assays. The bioactive compounds were characterized with ultra-high-performance liquid chromatography coupled with linear trap quadrupole tandem mass spectrometry. Stearidonic, eicosapentaenoic, and arachidonic acids were identified as major components having radical scavenging and antimicrobial activities. The suggested method provides a fast identification and quantification of bioactive compounds in multicomponent biological samples.

Milk and dairy, although less often, also cause a significant number of allergic manifestations. The aim of this study was to identify α-Gal-containing bovine milk proteins with allergenic properties among AGS patients. Methods Thirty-eight AGS patients with IgE to milk were included in the study. Milk proteins were analyzed for the presence of α-Gal and for binding by patients’ IgE using immunoblot, ImmunoCAP, and inhibition ELISA. Allergenicity of milk and milk proteins was assessed by basophil activation test. Results More than half of the AGS patients reported allergic reactions to milk or dairy products. Bovine γ-globulin (BGG), lactoferrin (LF), and lactoperoxidase (LPO) were identified as α-Gal carrying proteins which were recognized by AGS patients’ IgE. Whey mirrored the anti-α-Gal and IgE reactivity of BGG, LF, and LPO. Eighty-nine percent of the patients displayed IgE to BGG, 91% to LF, and 57% to LPO. Inhibition of α-Gal-specific IgE binding was achieved by BGG, LF, LPO, and whey. These proteins also activated AGS patients’ basophils. Interestingly, at lower concentrations, LF was the most potent inhibitor of IgE binding, and the most potent activator of basophils. Conclusion BGG, LF, and LPO were all found to be relevant milk α-Gal-containing glycoproteins that bound AGS patients’ IgE antibodies and activated their basophils. These proteins are probably involved in the allergic reactions to milk in AGS patients. LPO was for the first time shown to be an allergen.

Impact of essential and toxic trace metals on thyroid health and cancer: A review

In this review, the impact of essential and toxic trace metals from the environment on thyroid health and TC was evaluated. Furthermore, this review clarifies which trace metals most likely contribute to the etiology of TC. Biomarkers based on trace metal ratios are proposed. AB  - Honeybee colony losses have been a focus of research in the last years, due to the importance of managed honeybee colonies for economy and ecology. Different unfavorable conditions from the outside environment have a strong impact on the hive health. The majority of losses occur mainly during winter and the exact reason is not completely understood. Only a small number of studies are dealing with content of bioelements, their function and influence on honeybee physiology. The aim of the present study was to determine seasonal and spatial variations in content of bioelements and non-essential elements, in hemolymph and whole body of honeybees originating from three regions with different degrees of urbanization and industrialization. Concentrations of 16 elements were compared: macroelements (Ca, K, Mg, Na), microelements (Cu, Fe, Mn, Zn) and non-essential elements (Al, Ba, Cd, Co, Cr, Ni, Pb, Sr) in samples collected from 3 different environments: Golija (rural region), Belgrade (urban region) and Zajača (industrial region). Content of bioelements and non-essential elements in honeybees was under noticeable influence of the surrounding environment, season and degree of honeybee activity. Hemolymph was proven to be helpful in differentiating air pollution from other sources of honeybee exposure. The results of our study demonstrated that bees can be successfully used as biomonitors since we have observed statistically significant differences among observed locations, but unless compared locations are exposed to excessively different pollution pressures, it is essential that all bees should be collected at the same season.

AB  - Proteklih nekoliko godina fermentisani beli luk i njegovi ekstrakti se sve češće koriste u kulinarstvu i u svakodnevnoj ishrani zbog svog specifičnog ukusa, nutritivnog sastava i zdravstvenih benefita. Proces fermentacije vrši se termičkom obradom belog luka uz kontrolisanu vlažnost tokom dužeg vremenskog perioda. Tokom fermentacije, hemijske reakcije i transformacije poput Majlardovih reakcija i reakcija karamelizacije uzrokuju promene ukusa, nutritivnog sastava, sadržaja makro- i mikroelemenata kao i sadržaja fenolnih jedinjenja te i antioksidativnu aktivnost. U ovom radu analizirani su uzorci belog luka i fermentisanog belog luka te je vršeno poređenje sadržaja fenolnih jedinjenja i antioksidativne aktivnosti, makro- i mikroelemenata kao i nutritivnog sastava. Sadržaj Ca, K, Mg, Na, Mn, Cu, Zn i W se povećao, dok se sadržaj Fe, Al, Cr, Ni, Mo, Hg i Pb smanjio tokom fermentacije. Sadržaj vode se smanjio tokom procesa fermentacije, dok se sadržaj šećera i ugljenih hidrata značajno povećao kao rezultat termičke razgradnje poli- i oligosaharida (fruktana i ostalih kompleksnih polisaharida). Shodno svemu navedenom, energetska vrednost fermentisanog belog luka veća je u odnosu na svež beli luk. Sadržaj ukupnih fenolnih jedinjenja veći je u fermentisanog uzorku u poređenju sa svežim, jer se u toku fermentacije događaju različite hemijske transformacije na nivou sekundarnih metabolita. Kao pokazatelji antioksidativne aktivnosti, rađena su dva eseja: DPPH i FIC i oba su pokazala veću aktivnost fermentisanog belog luka, što je u pozitivnoj korelaciji sa većim sadržajem fenolnih jedinjenja u datom uzorku.

AB  - Humulones and iso-humulones are potent natural antioxidants found in beer. In this study, density functional theory (DFT) method was applied for elucidating the structure-antioxidant activity relationship and molecular mechanism of antioxidant activity of eight bioactive humulones previously identified in different beer samples: isoxanthohumol, (R)- and (S)-adhumulone, cis- and trans-iso-adhumulone, cis- and trans-iso-n-humulone, and desdimethyl-octahydro-iso-cohumulone. The calculated bond dissociation enthalpies (BDEs) suggest that desdimethyl-octahydro-iso-cohumulone was the most potent compound with BDEs 5.1 and 23.9 kJ/mol lower compared to the values for resveratrol in gas phase and water, respectively. The enolic –OH is the most reactive site for hydrogen atom transfer (HAT). The presence of β-keto group with respect to enolic –OH diminishes the HAT potency via the formation of a strong intramolecular hydrogen bond. Another common antioxidant mechanism, single electron transfer followed by proton transfer (SET-PT), is only feasible for isoxanthohumol. The results of this study indicate a strong correlation between the increased antioxidant activity of beer products and the higher content of reduced iso-α-acids.

Sadržaj citrata u medu kao pokazatelj njegove autentičnosti

AB  - U radu je istražen uticaj dodatka natrijum-hidroksida na količinu HMF-a u medu. U svim uzorcima nakon dodatka natrijum-hidroksida je vrednost HMF-a bila niža u odnosu na koncentraciju pre dodatka hidroksida.
Pored obećavajućih rezultata, usled malog broja uzoraka ipak se ne može sa sigurnošću definisati da li je razlika u koncentraciji citrata u medu pre i nakon dodatka natrijum-hidroksida i limunske kiseline dovoljna da bi se razvila nova metoda zasnovana na određivanju granične količine citrata u medu koja bi okarakterisala med kao autentičan, usled čega su dalja opširnija istraživanja neophodna.

AB  - Predmet ove disertacije bio je proučavanje uticaja herbicida sa ili bez primene folijarnog đubriva na sadržaj fitohemikalija u listu i zrnu četiri hibrida kukuruza šećerca (ZP504su, ZP355su, ZP553su i ZP515su) tokom dve godine, u cilju definisanja njihove tolerantnosti i karakterističnog odgovora. Sadržaj fitohemikalija određen je primenom visoko-efikasne tečne hromatografije sa ultravioletnim detektorom sa više dioda i fluorescentnim detektorom i visoko-efikasne jonske hromatografije sa pulsno-amperometrijskim detektorom. Sadržaj ukupnih fenolnih jedinjenja, glutationa i antioksidativna aktivnost određeni su primenom UV/VIS spektrofotometrije. Hromatografska razdvajanja su postignuta pri nižim protocima mobilnih faza uz smanjen utrošak organskih rastvarača podržavajući na taj način trendove zelene analitičke hemije. Dobra linearnost, tačnost, niska granica detekcije, direktno HPLC određivanje ekstrakata bez derivatizacije, skraćeno vreme ekstrakcije i trajanja analize omogućili su rutinsko korišćenje primenjenih hromatografskih metoda. Rezultati su pokazali da je uticaj tretmana bio izraženiji u zrnu, dok u listu uglavnom nije bio statistički značajan. Primena oba herbicida dovela je do statistički značajnog povećanja ukupnog sadržaja svih fitohemikalija u ZP553su na osnovu čega se on može definisati kao tolerantan genotip. Primenjeni tretmani izazvali su aktiviranje neenzimskih antioksidanasa kod ZP355su, a kod ZP515su aktiviranje metabolizma ugljenih hidrata. Rezultati su pokazali da se sadržaj karotenoida, vitamina E, fenolnih kiselina i šećera može koristiti kao jedinstven pokazatelj genotipa nakon primenjenih tretmana. Pokazano je da kontrolisana upotreba herbicida može dovesti do poboljšanja fitohemijskog profila i povećanja nutritivnog statusa kukuruza šećerca.
AB  - The subject of this doctoral dissertation was the impact of herbicides with or without foliar fertilizer on phytochemicals content in leaf and grain in four hybrids of sweet maize (ZP504su, ZP355su, ZP553su, and ZP515su) during two years in order to define their tolerance and characteristic response. The content of phytochemicals was determined using high-performance liquid chromatography with an ultraviolet multi-diode detector and a fluorescent detector, and high-performance anion exchange chromatography with pulsed amperometric detection. The content of total phenolic compounds, glutathione and antioxidant activity was determined using UV / VIS spectrophotometry. Chromatographic separations were achieved at lower mobile phase flows with reduced consumption of organic solvents, thus supporting the trends of green analytical chemistry. Good linearity, accuracy, low limit of detection, direct HPLC determination of extracts without derivatization, shortened extraction time, and duration of analysis enabled the routine use of applied chromatographic methods. The results showed that the impact of the treatment was more conspicuous in the grain, while in the leaf it was generally not statistically significant. The application of both herbicides led to a statistically significant increase in the total content of all phytochemicals in ZP553su, based on which it can be defined as a tolerant genotype. The applied treatments caused the activation of non-enzymatic antioxidants in ZP355su, and in ZP515su the activation of carbohydrate metabolism. The results of this study indicated that the content of carotenoids, vitamin E, phenolic acids and sugars can be used as a characteristic phytochemical parameter of the genotype after the applied treatments. It has been shown that the controlled use of herbicides can lead to an improvement in the phytochemical profile and an increase in the nutritional status of sweet maize.

U novije vreme, dolazi do sve veće potraţnje za metodama koje bi mogle efikasno i ekonomično rešiti problem lekova koji su teško rastvorni u vodi. U ovom radu opisana je metoda povećanja rastvorljivosti i brzine rastvaranja zasnovana na kiselinsko-baznim interakcijama. Cilj ovog rada bio je da se poveća brzina rastvaranja haloperidola, fluoksetin–hidrohlorida i olanzapina – lekova koji se ponašaju kao slabe baze, korišćenjem slabe organske kiseline sa kojom izabrana jedinjenja neće nagtraditi soli.
Ispitana je brzina rastvaranja haloperidola, fluoksetin-hidrohlorida i olanzapina u kiseloj i baznoj sredini, u prisustvu slabe triprotične organske kiseline (limunska kiselina). Uzorci su pripremljeni uparavanjem vodenih rastvora u kojima je molski odnos leka i limunske kiseline različit. U toku eksperimenta praćenja vremenski zavisne rastvorljivosti, u odreĎenim vremenskim intervalima uzimani su alikvoti rastvora, u kojima je koncentracija leka odreĎena spektrofotometrijski. Konstruisani su grafici zavisnosti koncentracije odgovarajuće supstance (leka) od vremena, na osnovu kojih je se moglo zaključiti da prisustvo limunske kiseline utiče na rastvorljivost ispitivanih lekova. Brzina rastvaranja sva tri leka značajno je poboljšana u prisustvu limunske kiseline.

AB  - “Bee pollen” is pollen collected from flowers by honey bees. It is used by the bees to nourish themselves, mainly by providing royal jelly and brood food, but it is also used for human nutrition. For the latter purpose, it is collected at the hive entrance as pellets that the bees bring to the hive. Bee pollen has diverse bioactivities, and thus has been used as a health food, and even as medication in some countries. In this paper, we provide standard methods for carrying out research on bee pollen. First, we introduce a method for the production and storage of bee pollen which assures quality of the product. Routine methods are then provided for the identification of the pollen’s floral sources, and determination of the more important quality criteria such as water content and content of proteins, carbohydrates, fatty acids, vitamins, alkaloids, phenolic and polyphenolic compounds. Finally, methods are described for the determination of some important bioactivities of bee pollen such as its antioxidant, anti-inflammatory, antimicrobial and antimutagenic properties. El polen de abeja es utilizado para nutrir a las propias abejas, principalmente para proporcionar jalea real y alimento para las crías, pero también se utiliza para la nutrición humana. Para este último fin, se recoge en la entrada de la colmena en forma de gránulos que las abejas llevan a la colmena. El polen de abeja tiene diversas bioactividades, por lo que se hautilizado como alimento para la salud, e incluso como medicamento en algunos países. En este artículo, proporcionamos métodos estándar para llevar a cabo investigaciones sobre el polen de abeja. En primer lugar, presentamos un método de producción y almacenamiento de polen de abeja que garantiza la calidad del producto. A continuación, se ofrecen métodos de rutina para la identificación de las fuentes florales del polen y la determinación de los criterios de calidad más importantes, como el contenido de agua y de proteínas, carbohidratos, ácidos grasos, vitaminas, alcaloides y compuestos fenólicos y polifenólicos. Por último, se describen métodos para la determinación de algunas bioactividades importantes del polen de abeja, como sus propiedades antioxidantes, antiinflamatorias, antimicrobianas y antimutagénicas.

abstract = "“Bee pollen” is pollen collected from flowers by honey bees. It is used by the bees to nourish themselves, mainly by providing royal jelly and brood food, but it is also used for human nutrition. For the latter purpose, it is collected at the hive entrance as pellets that the bees bring to the hive. Bee pollen has diverse bioactivities, and thus has been used as a health food, and even as medication in some countries. In this paper, we provide standard methods for carrying out research on bee pollen. First, we introduce a method for the production and storage of bee pollen which assures quality of the product. Routine methods are then provided for the identification of the pollen’s floral sources, and determination of the more important quality criteria such as water content and content of proteins, carbohydrates, fatty acids, vitamins, alkaloids, phenolic and polyphenolic compounds. Finally, methods are described for the determination of some important bioactivities of bee pollen such as its antioxidant, anti-inflammatory, antimicrobial and antimutagenic properties. AB  - The identification of agricultural food production systems has gained importance in order to protect both human health and the environment. The importance of organic production system of agriculture which involves the application of natural processes and substances, and limits or completely eliminates the use of synthesized means is emphasized. Knowledge of the mineral composition in tomato samples can be used as a potent tool in the identification of chemical markers as potential indicators of the farming system. A set of tomato samples taken from two factorial randomized trials were comprehended eight different varieties, belonging to four tomato types: large—BEEF and CLUSTER, and mini and midi—CHERRY and PLUM tomatoes, cultivated under two different farming systems: integral (IPM) and organic (O) were characterized based on the composition of the minerals. A total of 44 elements were quantified. To establish criteria for the classification of the samples and confirm a unique set of parameters of variation among the types of production, sophisticated chemometric techniques were used. The results indicate that the accumulation of elements varies between 8 tomato varieties and 2 different growing systems. The contents of Al, Mn, As, Pb, and some of the rare-earth elements (REEs) are able to distinguish between production types. Examination of different hybrids, which belong to different types in two production systems: organic and integral within Zeleni hit (official Enza and Vitalis trial and breeding station), was done with the aim of reaching a methodology of diversification, ie complete traceability of organic production, and to contribute to distinguishing types of agricultural systems and enhancing the possibility of acquiring a valuable authenticity factor about the type of agricultural production system employed for the cultivation of tomatoes.

A set of 48 potato tubers of four varieties with different harvesting times, cultivated in three types of agricultural systems: conventional (C), integral (I), and organic (O) were characterized based on the composition of the elements of their bulk and peel. A total of 16 elements were quantified. In order to determine the source of variation among the types of production, parts of potato and varieties, multivariate analysis of variance (MANOVA) was conducted. The results indicate that Ca, Mg, and K may be considered as important indicators of the type of production, genotypes of potato, and harvesting time. Additionally, the analyses show that ten microelements are able to distinguish between production types and genotypes of potato.

AB  - Serological testing is important method for diagnosis of severe acute respiratory syndrome coronavirus 2 (SARS-CoV-2) infection. Nucleocapsid (N) protein is the most abundant virus derived protein and strong immunogen. We aimed to find its efficient, low-cost production.

SARS-CoV-2 recombinant fragment of nucleocapsid protein (rfNP; 58–419 aa) was expressed in E. coli in soluble form, purified and characterized biochemically and immunologically.

Purified rfNP has secondary structure of full-length recombinant N protein, with high percentage of disordered structure (34.2%) and of β-sheet (40.7%). rfNP was tested in immunoblot using sera of COVID-19 convalescent patients. ELISA was optimized with sera of RT-PCR confirmed positive symptomatic patients and healthy individuals. IgG detection sensitivity was 96% (47/50) and specificity 97% (67/68), while IgM detection was slightly lower (94% and 96.5%, respectively).

Cost-effective approach for soluble recombinant N protein fragment production was developed, with reliable IgG and IgM antibodies detection of SARS-CoV-2 infection.

SARS-CoV-2 recombinant fragment of nucleocapsid protein (rfNP; 58–419 aa) was expressed in E. coli in soluble form, purified and characterized biochemically and immunologically.

Purified rfNP has secondary structure of full-length recombinant N protein, with high percentage of disordered structure (34.2%) and of β-sheet (40.7%). rfNP was tested in immunoblot using sera of COVID-19 convalescent patients. ELISA was optimized with sera of RT-PCR confirmed positive symptomatic patients and healthy individuals. AB  - Demand for camel milk (CM) is increasing worldwide, due to its high nutritious value and health benefits. In this study, whole CM powders were produced by spray drying (SD) at six inlet temperatures (190 °C–250 °C) and by freeze drying (FD). Physicochemical and functional properties of CM powder proteins were investigated. SD at higher inlet temperatures (230 °C–250 °C) resulted in higher extent of Maillard reaction (MR), in comparison to lower temperatures (190 °C–200 °C) and FD treatment. Both treatments had negative effect on casein solubility, while whey proteins remained soluble and slightly increased its solubility with the extent of MR. The CM powders obtained at higher inlet temperatures demonstrated improved antioxidant activity. Secondary structure of whey proteins did not differ among the samples, while surface hydrophobicity of whey proteins was higher in all SD than in FD samples, suggesting only limited denaturation of camel whey proteins at higher inlet temperatures of drying. Thus, the effects of SD under the conditions applied in our study did not decrease camel whey protein solubility, while drying procedure itself regardless of temperature decreased solubility of camel milk caseins. MR generated during CM processing could be an important means of compensating for the lack of antioxidant protection normally associated with β-lactoglobulin but happens to be absent from this milk.

Interaction, binding capacity and anticancer properties of N,N′-bis(acetylacetone)-propylenediimine-copper(II) on colorectal cancer cell line Caco-2

AB  - Different Schiff base complexes have biological activities that make them suitable for drug design. The biological properties of tetradentate Schiff base copper(II) complexed with N,N′-bis(acetylacetone)propylenediimine have been studied. The cytotoxic activity towards Caco-2 cells were determined by MTT, Anexin V and PI apoptosis assays. N,N′-bis(acetylacetone)propylenediimine-copper(II) showed the anti-cancer and anti-proliferative properties by inducing apoptosis in Caco-2 cells. A comparison of the cytotoxic activity of this compound with cisplatin shows that it is more effective on the colorectal cancer cell line Caco-2. The binding capacity and interaction of N,N′-bis(acetylacetone)propylenediimine-copper(II) with HSA were systemically investigated by in vitro fluorescence spectroscopy, CD spectroscopy, and in silico molecular docking study. Furthermore, in vitro and in silico interaction studies indicated that the complex binds to HSA through a static quenching mechanism without changes in protein conformation. The calculated number of binding sites was in line with molecular docking studies. The obtained Ka value suggests that the compound can be released from the protein in target cells. The tetradentate Schiff base copper(II) complex exhibited in vitro biological activities against cancer epithelial cells, which depend on the molecular structure of the complex, causing apoptosis, and the complex can bind to the protein drug carrier in circulation to the target tissue.
