AB - We present the green, highly atom-economical, solid-state silica gel-catalyzed synthesis of polysubstituted 1,4- and 1,2-dihydropyridines (DHPs) from commercially available materials, amines and ethyl propiolate. The DHP skeleton was assembled by heating the reactants and silica gel in a closed vessel. Aliphatic amines provided 1,4-isomers as the main or only DHP products, but the reactions of aromatic amines yielded a mixture of 1,4- and 1,2-isomers. To the best of our knowledge, this is the first example of the formation of a 1,2-DHP structure by the reaction of an amine with propiolic ester. Addition of 1 mass percent of H2SO4 to silica gel shifted the product distribution to 1,4-DHP as the main or the only isomer obtained. Experimental and theoretical analyses led to the identification of two key intermediates en route to DHPs and the explanation of the observed regioisomeric ratios. 1,2-DHPs show blue-cyan fluorescence in MeOH with the quantum yield Φ = 0.10–0.22 relative to quinine sulfate Φ = 0.58 and 1,4-DHPs show blue-violet fluorescence with Φ = 0.09–0.81.

AB - Herein, a screen–printed diamond electrode (SPDE) coupled with a “point-of-care” platform (30 µL-drop concepts, single-drop-detection approach) was successfully applied for the electrochemical determination of pterostilbene (PTS). Cyclic voltammetry identified irreversible oxidation of PTS, where oxidation peak was shown to be strongly dependent on the pH of the working environmental. Although the proposition of the detailed electrochemical oxidation mechanism of PTS goes out of the scope of the present research, we have determined the most probable reactive site of our analyte, by utilizing DFT-based reactivity descriptors (Fukui functions). For electrochemical quantification of PTS, oxidation peak at 0.32 V (vs. Ag/AgCl) was followed in presence of 0.5 mol L−1 of Briton–Robinson buffer solution (pH = 9). Coupled with the optimized parameters of differential pulse voltammetry (DPV), SPDE detected PTS in two linear ranges (first range was from 0.011 to 0.912 µmol L−1; second range was from 0.912 to 4.420 µmol L−1), providing the LOD and LOQ on a nanomolar level (3.1 nmol L−1 and 10.0 nmol L−1, respectively). The selectivity of the optimized DPV method was found to be excellent, with the current changes of less than 7%, in the presence of ten times higher concentrations of the certain interferences. The practical applicability of the SPDE and single-drop-detection approach in dietary supplements (with a declared PTS content of 50 mg/tablet), with the recovery values ranging from 95 to 102%, shows that the developed method has high potential for precise and accurate PTS detection, as well as exceptional miniaturization possibilities of relevant equipment for on-site sensing.

AB - Serotonin receptors modulate numerous behavioral and neuropsychological processes. Therefore, they are the target for the action of many drugs, such as antipsychotics, antidepressants, antiemetics, migraine remedies, and many others. The 5-HT1A receptors have been involved in the pathogenesis and treatment of anxiety and depression and represent a promising target for new drugs with reduced extrapyramidal side effects. In most antidepressants, a piperazine-based structural motif can be identified as a common moiety. Here we describe the synthesis, pharmacological, and in silico characterization of a novel arylpiperazines series with excellent 5-HT1A affinity. The final compounds, 4a, 8a, and 8b, were selected according to predictions of in silico pharmacokinetics, docking analysis, and molecular dynamics in conjunction with physical properties, and metabolic stability. The accentuated molecules could serve as a lead compound for developing 5-HT1A drug-like molecules for depression treatment.

AB - Natural Deep Eutectic Solvents (NADES) are novel and promising solvents for green extraction of phytochemicals from food and agricultural products. NADES are made of natural origin compounds that are connected via hydrogen bond and have unique tuneable properties1. Numerous studies investigated the extraction of bioactive compounds using NADES resulted in better extraction efficiency compared to conventional solvents, suggesting large potential of these solvents2,3. Low-toxicity, natural origin and affordability of NADES-based extracts enables their application as a preservative in the food industry or functional ingredient of food supplements. The aim of this study was to develop an efficient eco-friendly method for the extraction of phenolic compounds from berries, blueberry, chokeberry and black goji berry, using NADES. This goal will be achieved through investigation of extraction efficiency of applied NADES based on three different approaches, a) chromatographic, through high-performance thin layer chromatography (HPTLC) fingerprint analysis, ultra-high-performance liquid chromatography with a diode array detector and a triple-quadrupole mass spectrometer (UHPLC-DAD-MS/MS) target analysis and bioautography, b) spectroscopic, through quality control parameters, total phenolic content (TPC), total flavonoid content (TFC) and radical scavenging activity (RSA), and c) microbiological, through well diffusion method and minimum inhibitory concentration (MIC).
In this study, 36 NADES mixtures, prepared from primary plant metabolites, were tested as green alternatives for extraction of phenolics from three berries. Different hydrogen-bond acceptors (HBA), such as choline chloride, L-proline, L-glycine, and L-lysine were mixed with various hydrogen-bond donors (HBD), four organic acids, two sugars, glycerol and urea, at various molar ratios for NADES preparation. Guided by the fact that extraction efficiency depends on polarity, viscosity and dissolving ability of solvents4, the influence of HBA, HBD and water content were investigated. Methanol, as conventional solvent, was used as contrastive solvent.
Choline chloride-based NADES in combination with malic acid, glycerol or urea showed as most efficient. All groups of studied NADES showed various extraction capabilities for phenolic compounds based on chromatographic evaluation and spectrophotometric tests. These results indicate great power of NADES solvents towards selective extraction of phenolics and shows that NADES are designer solvents. Appropriate selection of components for NADES preparation can result in selective extraction of specific phenolic compounds, regardless of how similar structures of targeted compounds are or how complex the matrix is, which gives a significant advantage for application of NADES.

AB - Owing to the use of large volumes of toxic solvents and reagents, green analytical methods have gained increasing importance in natural product research. Herein, a green approach is proposed using the natural deep eutectic solvents (NADES) for the extraction and separation of phenols from Teucrium chamaedrys by use of an ecofriendly high-performance thin-layer chromatography (HPTLC) method. Principal component analysis was used to classify NADES extracts based on HPTLC data. The method’s greenness was assessed using two different tools: the National Environmental Methods Index (NEMI) and the analytical eco-scale. This method can also find application in routine quality control analysis of the investigated medicinal herb with minimal harm to the environment or human health.

AB - Strawberry (Fragaria x ananassa Duch.) is widely known for its nutritional value, richness in polyphenols and antioxidant capacity. In abundance of numerous ongoing strawberry breeding programs worldwide, some of the main parameters for breeding and selection are fruit quality, flavour and aroma which are directly affecting commercial acceptance [1]. These could be easily identified with some of objective measurements such as soluble solids concentration (°Brix), acid contents (titratable acidity, TA) and Brix/acid ratio, which could be strong predictors of quality in terms of consumer acceptability [1,2]. In this trial, we compared 24 strawberry genotypes, including 12 parental varieties and 12 perspective candidates obtained by crossing these particular parental varieties. Results of °Brix, titratable acidity and °Brix/acid ratio measurements indicated that new perspective candidates have had higher °Brix/acid ratio than the parental varieties, leading to conclusion that those genotypes had better flavour. Therefore, this ratio could potentially be used as indicator for distinguishing consumer acceptance.

AB - The aim of this study was to comparatively analyze chemical composition and biological activity of wild- and in vitro grown Rindera umbellata. Explants were cultivated on 0.003–0.3 M sucrose, fructose, or glucose. HPLC-DAD for quantifying rosmarinic (RA) and lithospermic B (LAB) acids and GC-MS/FID for qualitative pyrrolizidine alkaloids (PAs) detection were used. Antioxidant activity (DPPH and ABTS assays) and cytotoxicity (MTT test) were monitored. Identified PAs were 7-angeloyl heliotridane, lindelofine, 7-angeloyl heliotridine, 7-angeloyl-9-(+)-trachelanthylheliotridine, punctanecine, and heliosupine, with higher variability reported in wild-growing samples. Total phenolic contents (TPCs) were comparable in wild-growing and in vitro samples, but total flavonoid (TFC) and RA levels were multifold higher in in vitro samples. Notably, high concentration of LAB was detected in wild-growing roots. Amounts of 0.3 M and 0.1 M of sucrose were optimal for TFC and RA production, while maximal antioxidant activity was monitored in plants grown on 0.3 M sucrose. The MTT test indicated colorectal HT-29 as more sensitive than A549 lung adenocarcinoma and normal MRC-5 cells, showing selective sensitivity to wild-growing and 0.3 M sucrose samples. In conclusion, PAs in vitro, as well as TPC, TFC, RA, and LAB in both growing conditions were detected for the first time in R. umbellata.

AB - Honey is very complex natural substance consisted of sugars, water, and other compounds in smaller quantities such as proteins (enzymes), organic acids, vitamins, minerals, phenolics, and a large variety of volatile compounds. Discriminating the honeys according to botanical and/or geographical origin may be possible according to their characteristic constituents including organic acids profile. These acids are related to the colour and flavour of honey and its chemical properties such as acidity, pH and electrical conductivity. Honey is deceptively acidic, as the high sugar content tends to mask the acidity in the taste. Citric acid among other acids, is present in honey in moderate amount but it's presence could be potentially used as marker for authenticity control [1].

Fraudulent addition of compounds in honey sets a necesity to establish the natural level of the specific substances, and subsequently determine when this level is exceeded, indicating adulteration. Citric acid can be added to decrease pH in adulterated honey where normal pH levels have been exceeded. Also increased citrate concentration in honeys could be a result of addition of invert syrup to honey. Therefore there is a need for a method to determine citrate content in honey quickly and efficiently1. In current work a method for determination of organic acids profile in honey using ion chromatography with conductometric detection (IC-CD) without prior separation of compounds and chemical derivatization was developed. This approach for citric acid determination is characterized by LOD and LOQ values of 0.05 and 0.20 mg/L, respectively. Low RSD Sy/x value and r2 level of 0.9999 indicates a good regression model. Trueness of method was confirmed by good recoveries (108%, 104% and 95% for spiked levels 75%, 100% and 125%, respectively), while precision was confirmed by HorRat value (0.30) less than 2 [2].

AB - Natural Eutectic Solvents (NES) are a promising class of environmentally friendly liquids that offer an alternative to conventional organic solvents. To investigate their potential for extracting phenolic compounds from plant materials, we employed the COSMO-RS (COnductor like Screening MOdel for Real Solvents) computational method. Nineteen NES solvents were prepared to determine the most selective and effective solvent for extracting phenolic compounds from Teucrium chamaedrys using both theoretical and experimental evaluation. Screening of extraction efficiency was performed by quantification of individual compounds using ultra-high-performance liquid chromatography with a diode array detector and a triple-quadrupole mass spectrometer (UHPLC-DAD-MS/MS), as well as spectrophotometric assays (total phenolic content, total flavonoid content, and radical scavenging activity). In this paper, we propose a new approach to model NES properties using COSMO-RS. While the majority of previous studies have modeled these solvents as individual or pseudo-component complexes between hydrogen bond donor (HBD) and hydrogen bond acceptor (HBA), we have developed a ternary HBD:HBA:water supramolecular complex model to represent NES solution structures more accurately. This model provides insights into the intermolecular interactions driving the extraction process and predicts extraction efficiencies that agree with experimental data. Two eutectic mixtures (choline chloride:succinic acid in molar ratio 1:1 and 20% of water (w/w) and choline chloride: glycerol in molar ratio 1:1 and 20% of water (w/w)) showed strong affinity towards phenolic compounds. Overall, our findings suggest that the ternary complex model is a more appropriate approach for modeling NES properties.

AB - AbstractBackground and objectiveCotinus coggygria Scop. is a valuable medicinal plant species with pronounced pharmacological potential due to its numerous biological activities. The herb is characterized by a high content of polyphenols among which is fustin. The anticancer activities of fustin, however, are extremely weakly studied. The aim of the present study was to investigate the in vitro antiproliferative potential of fustin isolated from the heartwood of C. coggygria against cell lines originating from two of the most common cancer types – breast (MDA-MB-231 and MCF7), and colon cancer (Colon 26).Materials and methodsCell growth inhibitory properties of fustin were examined by MTT assay. Subsequently, phase-contrast and fluorescence microscopy analysis as well as colonyforming assay were carried out on the most sensitive to the cytostatic action of the fustin cell line.ResultsThe obtained results showed that fustin reduced the proliferation of all studied cell lines. The highest cytostatic effect was registered towards breast cancer MDA-MB-231 cells with a half maximal inhibitory concentration (IC50) value of 56.02 μg/ml followed by colon cancer cells with an IC50 of 78.07 μg/ml. MCF7 cell proliferation was least affected with a calculated IC50 of 187.8 μg/ml. Further investigations on breast cancer MDA-MB-231 cells indicated decreased density of cell

Further investigations on breast cancer MDA-MB-231 cells indicated decreased density of cell",

AB - Soluble sugars and organic acids have a strong impact on the overall sensory quality of fruits. Organic acids also facilitate in stabilization of anthocyanins, and they extend the shelf-life of fresh fruits and their processed products. The production of new apple-based products with health beneficial properties is leading to agronomical and chemical re-evaluation of old apple cultivars.
In this study the mesocarp samples of five conventional, five resistant and fifteen autochthonous apple cultivars from a single growing season were deliberated. Assessment was based on contents of four sugars, one sugar alcohol and five organic acids that were determined by the usage of HPAEC-PAD and HPAEC-CD systems. Fructose was detected as dominant sugar in all cases (50.4 – 77.9 mg/kg FW) and malic acid as dominant acid (1.1 – 4.5 mg/kg FW). Regarding the average values, xylose and sorbitol were in higher amount in autochthonous cultivars compared to others. In resistant cultivars shikimate and malate were found in lower concentrations compared to other cultivar types. Among the obtained data, highest correlation values were found between citrate and isocitrate, and also between citrate and malate (0.905 and 0.839, respectively).

AB - Blackberry wine is produced by a yeast fermentation of natural saccharides present in blackberry juice. This wine is traditionally consumed in Balkans, mostly as a dessert wine, but in recent years it is gaining more attention due to unique flavour and potential health benefits. Blackberries are a natural source of many minerals and bioactive phytochemicals such as vitamins, folic acid, anthocyanins, and phenolics1,2. Considering biological potential of blackberries and their products aim of this study was to evaluate quality of Serbian blackberry wines. This quality assessment was achieved through determination of: (a) the phenolic profile by high-performance thin layer chromatography (HPTLC); (b) total phenolic (TPC) and anthocyanin content (TAC); (c) in vitro antioxidant activity by DPPH and ABTS radical scavenging assays and total antioxidant capacity (d) in vitro antimicrobial activity by the agar diffusion test; and (e) the content of macro, micro and toxic elements by inductively coupled plasma mass spectrometry (ICP-MS). Twelve blackberry wines from different geographical regions were tested indicating various phenolic profiles and great antioxidant and antimicrobial activity. Strong positive correlation was confirmed between TPC and antioxidant activity determined by DPPH and ABTS assays. Additionally, high levels of antibacterial activity were shown against all six tested strains, but primarily against Staphylococcus aureus and Micrococcus lysodeikticus. Samples were rich in macroelements, such as Ca, K, Mg, Na, P and Zn, and microelements Cu and Fe, while Mn was present in higher concentration only in one sample. Toxic metals Cd and Pb were present in low concertation in all samples. These results indicate great quality of Serbian blackberry wines concerning minerals, phenolics, antioxidant and antimicrobial activity, which support its health potential.

AB - High-Performance Thin-Layer Chromatography (HPTLC)-radical scavenging capacity (RSC) assays are stan- dard techniques for the separation and identification of antioxidants from complex mixtures. HPTLC coupled with DPPH · visualization of chromatograms allows for the detection of individual antioxidants. However, other HPTLC-RSC assays that recognize compounds exhibiting different mechanisms of radical- scavenging activity are rarely reported. In this study, we developed an integrated approach that combines five HPTLC-RSC assays, principal component analysis (PCA) and quantum chemical calculations to assess the antioxidant capacity of Sempervivum tectorum L. leaf extracts. Two HPTLC assays - potassium hexa- cyanoferrate(III) total reducing power assay (TRP) and total antioxidant capacity by phosphomolybdenum method (TAC) - were developed for the first time. The method supports a more in-depth study of the RSC of natural products, as it compares the radical scavenging fingerprints of S. tectorum leaf extracts and recognizes differences in their individual bioactive constituents. Kaempferol, kaempferol 3- O -glucoside, quercetin 3- O -glucoside, caffeic acid, and gallic acid were identified as the compounds that discriminate HPTLC-RSC assays according to their mechanism of action and capture the similarities between 20 S. tec- torum samples. Additionally, DFT calculations on M06–2X/6–31 + G (d,p) level were applied to map ther- modynamic feasibility of hydrogen atom transfer (HAT) and single electron transfer (SET) mechanisms of the identified compounds. Based on experimental and theoretical results, a combination of HPTLC-ABTS and HPTLC-TAC assays were proposed as the optimal method for mapping the antioxidants from S. tec- torum . This study represents a step forward in identifying and quantifying individual antioxidants from complex food and natural product matrices in a more rational manner.

AB - The optimization of expression conditions is a tedious, but necessary procedure for thesuccessful efficient production of recombinant proteins. Design of experiment (DoE) savestime and resources by using statistics to carefully select only a few experimental points(expression conditions) to obtain information about the whole tested range of variedconditions, unlike the most commonly used one-factor-at-a-time (OFAT) method, whichrequires full imput space screening for optimization. Here, we have optimized theexpression conditions of His 6 and His8-mCerulean3-TEV-α-synuclein production from thethe pDUET vector, as well as the His 6-tagged protein from the pET-20b vector andexpressed into the periplasm. The proteins for which optimization was conducted wereexpressed in Escherichia coli BL21(DE3) and BL21(DE3)pLysS. We have used DoE toplan adequate experimental points for optimization according to a Box-Behnken design, with IPTG concentration, temperature and time variation. After collecting the cultures atgiven experimental points, they were analyzed by SDS-PAGE and densitometry, thenmodelled and statistically analysed. The importance of correct sample preparation and gelloads for densitometric analysis is evident according to the obtained results. The largestexpression level was obtained from the His 8-tagged protein coded by the pDUET vector inE. coli BL21(DE3). We have also used densitometry to analyze expression levels indifferent culture media.

AB - In this work, we investigated the morphological and electrochemical properties of gallium/bismuth mixed oxide. The bismuth concentration was varied from 0 to 100%. The correct ratio was determined with inductively coupled plasma-optical emission spectroscopy (ICP-OES), while surface characteristics were determined using scanning electron microscopy (SEM) and X-ray diffraction (XRD) measurement. Electrochemical characteristics were studied using electrochemical impedance spectroscopy (EIS) in the Fe2+/3+ couple. The obtained materials were tested for adrenaline detection. After square wave voltammetry (SWV) optimization, the best electrode showed a wide linear working range from 7 to 100 µM at pH 6 of the Britton–Robinson buffer solution (BRBS) supporting electrolyte. The limit of detection (LOD) for the proposed method was calculated as 1.9 µM, with a limit of quantification (LOQ) of 5.8 µM. The excellent selectivity of the proposed method, with good repeatability and reproducibility, strongly suggests the possible application of the procedure for the determination of adrenaline in artificially prepared real samples. The practical applicability with good recovery values indicates that the morphology of the materials is closely connected with other parameters, which further suggests that the developed approach can offer a low-cost, rapid, selective, and sensitive method for adrenaline monitoring.

AB - Triclosan (TCS) is a polychlorinated phenoxy phenol (PCPPs) used as a disinfectant and a broad-spectrum antibacterial and antifungal agent in personal hygiene products. TCS easily forms diphenyl ethers and dioxins, which are persistent organic pollutants.

This work used a double approach for the TSC sensing: a) screen-printed (SPE) electrochemical platform for on-site application, modified with lanthanum iron oxide and graphitic carbon nitride composite (LaFeO3/Fe2O3@g-C3N4/SPE); and b) carbon paste electrode (CPE), modified with the same material and used in laboratory conditions. Linear range from 0.1 μM to 10 μM, the limit of detection (LOD) of 29 nM and the limit of quantification (LOQ) of 91 nM were obtained for CP electrode in BRBS pH 8. SPE showed the best analytical parameters in BRBS at pH 3, with a linear range from 0.3 μM to 7 μM, LOD of 0.09 μM and LOQ of 0.28 μM. Furthermore, the influence of potential interferents was investigated and proven to be negligible. Determination of TSC was performed to estimate the environmental impact of this compound as well as the practical usefulness of the proposed sensor in the real sample analysis, confirmed with a HPLC analysis.

AB - Bisphenol A (BPA) is an organic pollutant that is widely spread in waterbody with effluents as a result of its extensive use in the plastics industry. It posses to the class of compounds that are endocrine disruptors and has a great negative impact on living organisms. Therefore, the progress in development of green technology for BPA removal from the aquatic media is crucial for environmental protection. This paper presents BPA removal by electrochemical oxidation on SnO2-MWCNT (tin dioxide-multi walled carbon nanotube) nanocomposite anode. The nanocomposite was structural and morphological characterized by X-ray powder diffraction and transmission electron microscopy. Electrochemical properties of the SnO2-MWCNT anode were investigated using cyclic voltammetry, linear sweep voltammetry and electrical impedance spectroscopy. The electrochemical behavior of anodes toward BPA in sodium sulfate solutions has shown that BPA is oxidized by indirectly mechanism via hydroxyl radicals (verified by quenching tests). Electrolysis of BPA has been done in galvanostatic regime at current densities 2.5, 5.0, 10.0, 15.0 and 20.0 mA cm−2 in 0.1 M sodium sulfate supported electrolyte pH 4.0. Complete degradation of BPA was accomplished after 5 h of electrolysis at a current density of 20 mA cm−2. The proposed electrochemical technology using a low-cost SnO2-MWCNT electrode is promising for wastewater BPA removal.

AB - In this paper, electrochemical degradation of Reactive Black 5 (RB5) textile azo dye was examined in regard to different synthesis procedures for making PbO2–graphitic carbon nitride (g-C3N4) electrode. The reaction of with ClO− in the presence of different surfactants, i.e., cetyltrimethylammonium bromide (CTAB) and tetrabutylammonium phosphate (TBAP), under conventional conditions, resulted in the formation of PbO2 with varying morphology. The obtained materials were combined with g-C3N4 for the preparation of the final composite materials, which were then characterized morphologically and electrochemically. After optimizing the degradation method, it was shown that an anode comprising a steel electrode coated with the composite of PbO2 synthesized using CTAB as template and g-C3N4, and using 0.15 M Na2SO4 as the supporting electrolyte, gave the best performance for RB5 dye removal from a 35 mg/L solution. The treatment duration was 60 min, applying a current of 0.17 A (electrode surface 4 cm2, current density of 42.5 mA/cm2), while the initial pH of the testing solution was 2. The reusability and longevity of the electrode surface (which showed no significant change in activity throughout the study) may suggest that this approach is a promising candidate for wastewater treatment and pollutant removal.

AB - There are ten million people in the world who have Parkinson’s disease. The most potent
medicine for Parkinson’s disease is levodopa (L-DOPA). However, long-term consumption of L-DOPA
leads to the appearance of side effects, as a result of which the control and monitoring of its concentrations are of great importance. In this work, we have designed a new electrochemical sensor for
detecting L-DOPA using a carbon paste electrode (CPE) modified with Eu2O3@Cr2O3 composite
nanoparticles. Rare earth elements, including Eu, are increasingly used to design new electrode
nanocomposites with enhanced electrocatalytic properties. Europium has been considered a significant lanthanide element with greater redox reaction behavior. We conducted a hydrothermal
synthesis of Eu2O3@Cr2O3 and, for the first time, the acquired nanoparticles were used to modify
CPE. The proposed Eu2O3@Cr2O3/CPE electrode was investigated in terms of its electrocatalytic
properties and then used to develop an analytical method for detecting and quantifying L-DOPA.
The proposed sensor offers a wide linear range (1–100 µM), high sensitivity (1.38 µA µM−1
cm−2
) and
a low detection limit (0.72 µM). The practical application of the proposed sensor was investigated by
analyzing commercially available pharmaceutical tablets of L-DOPA. The corresponding results indicate
the excellent potential of the Eu2O3@Cr2O3/CPE sensor for application in real-time L-DOPA detection
