AB  - Serotonin receptors modulate numerous behavioral and neuropsychological processes. Therefore, they are the target for the action of many drugs, such as antipsychotics, antidepressants, antiemetics, migraine remedies, and many others. The 5-HT1A receptors have been involved in the pathogenesis and treatment of anxiety and depression and represent a promising target for new drugs with reduced extrapyramidal side effects. In most antidepressants, a piperazine-based structural motif can be identified as a common moiety. Here we describe the synthesis, pharmacological, and in silico characterization of a novel arylpiperazines series with excellent 5-HT1A affinity. The final compounds, 4a, 8a, and 8b, were selected according to predictions of in silico pharmacokinetics, docking analysis, and molecular dynamics in conjunction with physical properties, and metabolic stability. The accentuated molecules could serve as a lead compound for developing 5-HT1A drug-like molecules for depression treatment.

AB  - We present the green, highly atom-economical, solid-state silica gel-catalyzed synthesis of polysubstituted 1,4- and 1,2-dihydropyridines (DHPs) from commercially available materials, amines and ethyl propiolate. The DHP skeleton was assembled by heating the reactants and silica gel in a closed vessel. Aliphatic amines provided 1,4-isomers as the main or only DHP products, but the reactions of aromatic amines yielded a mixture of 1,4- and 1,2-isomers. To the best of our knowledge, this is the first example of the formation of a 1,2-DHP structure by the reaction of an amine with propiolic ester. Addition of 1 mass percent of H2SO4 to silica gel shifted the product distribution to 1,4-DHP as the main or the only isomer obtained. Experimental and theoretical analyses led to the identification of two key intermediates en route to DHPs and the explanation of the observed regioisomeric ratios. 1,2-DHPs show blue-cyan fluorescence in MeOH with the quantum yield Φ = 0.10–0.22 relative to quinine sulfate Φ = 0.58 and 1,4-DHPs show blue-violet fluorescence with Φ = 0.09–0.81.

AB  - Banana allergy is often associated with the pollen and latex allergies, which led us to the  hypothesis that some yet unidentified banana allergen could provide a basis of the latex-pollen-fruit syndrome. S-adenosyl-L-homocysteine hydrolase (SAHH) was recently identified in the literature as a novel plant allergen. This study aimed to assess the allergenic potential of the naturally occurring banana SAHH (nSAHH) and its recombinant homolog produced in E. coli (rSAHH). nSAHH showed IgE reactivity with a serum pool of twelve banana-allergic persons,
while rSAHH displayed IgE reactivity in ten out of the twelve tested patients. Five linear B-cell epitopes were identified on the rSAHH surface, exhibiting ≥ 90 % sequence homology with relevant plant SAHH allergens. Our findings have elucidated SAHH as a novel plant panallergen, underlying the cross-reactivity between plant derived food and respiratory allergens, confirming our initial hypothesis.

AB  - The widespread abuse of traditional antibiotics has led to a global rise in antibiotic-resistant bacteria, which give 
in return unprecedented health risks. Therefore, there is a large and urgent need for the development of new, 
smart antibacterial agents able to efficiently kill or inhibit bacterial growth. In this study, we investigated the 
antibacterial activity of S, N-doped Graphene Quantum Dots (GQDs) as a light-triggered antibacterial agent. 
Gamma irradiation was employed as a tool to achieve one-step modification of GQDs in the presence of L cysteine amino acid as a source of heteroatoms. X-ray Photoelectron Spectroscopy (XPS), nuclear magnetic 
resonance (NMR), and zeta potential measurements provided the necessary data to clarify the structure of 
modified dots and verify the introduction of both S- and N-atoms in GQDs structure, but also severe changes in 
the aromatic, sp2 domains. Namely, γ-irradiation caused a bonding of S atoms in 1.14 at.% mainly as thiol 
groups, and N in 1.81 at.% as amino groups, but sp2 contribution in GQD structure was lowered from 63.00 to 
4.86 at.%, as measured in dots irradiated at a dose of 200 kGy. Fluorescence quenching measurements showed 
that L-cysteine-modified dots are able to bind to human serum albumin. The antibacterial activity of GQDs 
combined with 1 and 6 h of blue light (470 nm) irradiation was tested against 8 bacterial strains. GQD-cys-25 
sample provided the best results, with minimum inhibitory concentration (MIC) as low as 125 μg/mL against 
S. aureus, E. faecalis, and E. coli after only 1 h of blue light exposure.

AB  - Fe(III) and Mn(II) complexes with condensation product of thiosemicarbazide and 2-acetylthiazole (HL1, (E)-2-(1-(thiazol-2-yl)ethylidene)hydrazine-1-carbothioamide) have been synthesized and characterized by single-crystal X-ray diffraction, IR spectroscopy, and elemental analysis. In both complexes, the thiosemicarbazone ligand is coordinated in deprotonated form through the NNS donor set of atoms. However, while Fe(III) complex is in the doublet ground state with distorted octahedral geometry, the coordination environment around Mn(II) is distorted trigonal-prismatic, and the sextet state is found to be the ground state. Antimicrobial activity (against a panel of Gram-negative and Gram-positive bacteria, two yeast, and one fungal strain), brine shrimp assay, and DPPH radical scavenging activity of both complexes were evaluated, and these results relate to the electronic structure of the complexes.

AB  - Two new Zn(II) complexes bearing tridentate hydrazone-based ligands with NNO or NNS donor atoms were synthesised and characterised by elemental analysis, infrared (IR) and nuclear magnetic resonance (NMR) spectroscopies, and single crystal X-ray diffraction methods. These complexes, together with four previously synthesised analogues, having hydrazone ligands with a NNO donor set of atoms, were successfully employed as catalysts in the ketone-amine-alkyne (KA2) coupling reaction, furnishing tetrasubstituted propargylamines, compounds with unique applications in organic chemistry. DFT calculations at the CAM-B3LYP/TZP level of theory were performed to elucidate the electronic structure of the investigated Zn(II) complexes, excellently correlating the structure of the complexes to their catalytic reactivity.

održavanje svežine poljoprivrednih namirnica. Zbog sve veće i nekontrolisane primene,
pesticidi i njihovi derivati predstavljaju velike pretnje za ekosistem. Razvoj novih modernih 
metoda za njihovo praćenje i detekciju je od primarnog značaja. U ovom radu je korišćena 
nemodifikovana dijamantska elektroda dopovana borom. Cilj ovog rada je jednostavno, brzo, 
efikasno izvršiti detekciju mezotriona, opasnog herbicida koji se najviše koristi u kulturi 
kukuruza. Prvi put je korišćena nemodifikovana dijamantska elektroda dopovana borom za 
detekciju mezotriona. Struktura površine same elektrode omogućava uspešnu oksidaciju 
mezotriona pri viskoj vrednosti potencijala (+1.4V). Postignuta je granica detekcije 0,34μM i 
izuzetno dobra selektivnost između interferirajućih molekula. Ovom metodom omogućena je
laka i jednostavna primena u analizi realnih uzoraka – prehrambenih proizvoda kukuruznog 
porekla.

AB  - Backgound/ObjectivesUrolithins (UROs) are the metabolites derived from the gut microbial action on ellagitannins and ellagic acid-rich foods. Following their absorption in the intestine, UROs are transported through the systemic circulation to various tissues where they can express their biological function as antimicrobial, anti-inflammatory, and anticancer agents. In addition to blood plasma, where they can be found as glucuronide and sulfate conjugates, they are also found in urine. Therefore, the interactions of UROs with serum proteins are of great clinical interest.MethodsA powerful technique for examining these urolithin-serum protein interactions is fluorescence spectroscopy. Bovine serum albumin (BSA) is a particularly suitable model protein because it is readily available, affordable, and similar to human serum albumin. This work aimed to study the binding of UROs (urolithin A, UROA and urolithin B, UROB) and their glucuronide conjugates (UROAG and UROBG) to BSA by quenching the intrinsic fluorescence of protein.ResultsThe spectra obtained showed that the binding process is influenced by the polyphenol's structure and the conjugation process with the glucuronide. The calculated Stern Vollmer binding constants (Ksv): UROA and UROB Ksv were 59236   ±   5706 and 69653   ±   14922, respectively, while for UROAG and UROBG, these values were 15179   ±   2770 and 9462   ±   1955, respectively, which showed that the binding affinity decreased with glucuronidation. Molecular docking studies confirmed that all of the studied molecules will bind favorably to BSA. The preferential binding site for both UROs and UROGs is Sudlow I, while UROs will also bind to Sudlow II. URO-Gs can bind to BSA in the cleft region with lower binding scores than for the Sudlow I binding site.ConclusionThe aglycone's higher hydrophobicity increases the binding affinity to BSA, thus reducing its bioavailability in the blood.

The preferential binding site for both UROs and UROGs is Sudlow I, while UROs will also bind to Sudlow II. The purpose of our study is to better understand the chemistry of the pygidial gland secretions of subterranean ground beetles, as well as the function and structure of the glands. We studied both the chemical composition of the pygidial gland secretion and morphology of the glands in adults of the troglophilic ground beetle species Laemostenus (Antisphodrus) cavicola (Schaum, 1858). The chemical composition of its defensive secretion was revealed using gas chromatography-mass spectrometry (GC-MS), while pygidial gland morphology of the beetle was investigated using bright-field microcopy. In total, seven chemical compounds were detected in the secretion mixture. Formic acid was the most dominant compound, followed by dodecyl acetate and undecane. Other chemicals were present in minor amounts. The morphological structure of the pygidial glands of L. (A.) cavicola was compared with the structure of the glands of the related congeneric troglophilic species Laemostenus (Pristonychus) punctatus (Dejean, 1828). Summary data on the semiochemicals that have been recorded so far in subterranean ground beetle species are presented, and the differences in the chemical composition of the secretions between and among troglobitic and troglophilic species are discussed. So far, forty-four compounds have been detected in four subterranean ground beetle species (two troglobites belonging to the tribe Trechini and two troglophiles belonging to the tribe Sphodrini). The results of this study indicate the great diversity of chemicals in the pygidial gland secretions of subterranean ground beetles. © 2023 by the authors.

AB  - The pygidial glands of carabids produce strong-smelling vapours. In this study, we examined the chemical composition of the gland secretions and the structure of the glands in five species of Carabini ground beetles (one species from the subtribe Calosomatina and four species from the subtribe Carabina): Calosoma (Calosoma) maderae (Fabricius, 1775), Carabus (Carabus) granulatus Linnaeus, 1758, C. (Limnocarabus) clathratus Linnaeus, 1760, C. (Carabus) ulrichii Germar, 1823, and C. (Procerus) gigas Creutzer, 1799. Additionally, we tested the antibacterial potential of the pygidial gland secretions of the two latter species against 11 bacterial strains. In order to detect the chemical content of the secretions, we used gas chromatography–mass spectrometry (GC–MS). The secretion extracts were applied against selected strains of medically important bacteria. We used bright-field microscopy to examine the morphology of the glands. We discovered a total of 11 chemical compounds in the pygidial gland extracts of the ground beetles we analysed. Ten of these compounds were identified as seven carboxylic acids, two hydrocarbons, and one aromatic aldehyde, while one chemical remained unidentified. Most of the components were isolated from the secretion of C. (L.) clathratus (nine), while the lowest number of compounds was found in C. (P.) gigas (two). Methacrylic acid was the most dominant compound by percentage in all five species, while angelic acid was also detected in all samples. As expected, salicylaldehyde was exclusively found in the species of the genus Calosoma Weber, 1801. The secretion of C. (P.) gigas was shown to achieve the highest level of antibacterial activity against Pseudomonas aeruginosa, Salmonella enterica, and S. typhimurium (even the same level as the positive control streptomycin), while the secretion of C. (C.) ulrichii achieved the highest antimicrobial potential against Staphylococcus epidermidis, S. aureus, Listeria monocytogenes, and Bacillus cereus. The most noticeable difference in the structure of the glands between the two genera is that the reservoir in Calosoma is more significantly narrowed as it leads into the efferent duct, compared to that of Carabus.

AB  - In the present study, we investigated in vitro the antimicrobial activity of the pygidial gland secretion of the guanophilic ground beetle Laemostenus (Pristonychus) punctatus (Dejean, 1828) and some of its chemicals against resistant and non-resistant bacteria and Candida species, the synergistic and additive potential of combinations of selected chemicals and antimicrobial drugs against resistant bacterial and fungal strains, anti-adhesive and anti-invasive potential of the secretion and formic acid alone and in selected combinations with antimicrobial drugs against methicillin-resistant Staphylococcus aureus (MRSA) toward spontaneously immortalized human keratinocyte cell line (HaCaT cells). In addition, we examined the antiproliferative activity of the secretion and formic acid in vitro. The tested secretion and the standards of formic and oleic acids possessed a significant level of antimicrobial potential against all tested strains (P < 0.05). The isolate from guano Pseudomonas monteilii showed the highest resistance to the secretion and formic acid, while MRSA achieved a significantly high level of susceptibility to all agents tested, particularly to the combinations of formic acid and antibiotics, but at the same time showed a certain level of resistance to the antibiotics tested individually. Candida albicans and C. tropicalis were found to be the most sensitive fungal strains to the secretion. Formic acid (MIC 0.0005 mg/mL) and gentamicin (MIC 0.0010 mg/mL) in the mixture achieved synergistic antibacterial activity against MRSA (FICI = 0.5, P < 0.05). The combination of formic acid, gentamicin and ampicillin accomplished an additive effect against this resistant bacterial strain (FICI = 1.5, P < 0.05). The secretion achieved a better inhibitory effect on the adhesion ability of MRSA toward HaCaT cells compared to formic acid alone, while formic acid showed better results regarding the invasion (P < 0.001). The combinations of gentamicin and ampicillin, as well as of formic acid and gentamicin and ampicillin achieved similar anti-adhesive and anti-invasive effects, with a slight advantage of formic acid and antibiotics in combination (P < 0.001). The secretion and formic acid were found to be non-toxic to HaCaT cells in vitro (IC50 ≥ 401 μg/mL).

AB  - The use of truffles in food is based mainly on the addition of artificial flavors, aiming to achieve an intense aroma in the products. As truffle is a natural product with nutritional and functional properties, it is important to find an optimal method for truffle storage. As the microbiota contribute to truffle aroma, the bacterial and yeast compositions in the rhizosphere and fruiting body of the truffle and the impact of different freezing methods on the volatile profile of the truffle Tuber aestivum during 90 days of the storage were determined. Bacteria and yeasts isolates were identified using 16s rRNA and 18s rRNA. The effect of freezing truffles at -20°C and -80°C with and without previous dipping in liquid N2 on the volatile compounds was observed using GC/MS. The results demonstrated that the isolated bacteria belonged to the phylum Proteobacteria, Firmicutes and Actinobacteria, and the identified species mainly belonged to Firmicutes, genus Bacillus sp. Isolated yeasts were identified as Cryptococcus sp., Debaromyces hanseinii, Candida fermentati and Rhodotorula mucilaginosa. The GC/MS analysis revealed that frozen truffle samples were richer in the compounds 2-butanone, 2-methyl-butanal, methanethiol and 2-butanol after freezing or during storage. The content of DMS, acetaldehyde, 3-octanone, ethanol, and 2-methyl-1-propanol significantly decreased immediately after freezing. Overall, the gained results indicated that freezing of truffles as a preservation method had profound effects on the volatile compounds, while previous dipping in liquid N2 showed no significant impact on the volatile profile of truffle Tuber aestivum. © 2023 University of Belgrade - Faculty of Agriculture. Red-Light Transmittance Changes in Variegated Pelargonium zonale—Diurnal Variation in Chloroplast Movement and Photosystem II Efficiency

AB  - Chloroplast movement rapidly ameliorates the effects of suboptimal light intensity by accumulating along the periclinal cell walls, as well as the effects of excess light by shifting to the anticlinal cell walls. These acclimation responses are triggered by phototropins located at the plasma membrane and chloroplast envelope. Here, we used a recently developed non-invasive system sensitive to very small changes in red light leaf transmittance to perform long-term continuous measurements of dark–light transitions. As a model system, we used variegated Pelargonium zonale leaves containing green sectors (GS) with fully developed chloroplasts and achlorophyllous, white sectors (WS) with undifferentiated plastids, and higher phototropin expression levels. We observed biphasic changes in the red-light transmittance and oscillations triggered by medium intensities of white light, described by a transient peak preceded by a constant decrease in transmittance level. A slight change in red-light transmittance was recorded even in WS. Furthermore, the chloroplast position at lower light intensities affected the rapid light curves, while high light intensity decreased saturated electron transport, maximum quantum efficiency of photosystem II, and increased non-photochemical quenching of chlorophyll fluorescence and epidermal flavonoids. Our results extend the knowledge of light-dependent chloroplast movements and thus contribute to a better understanding of their role in regulating photosynthesis under fluctuating light conditions.

AB  - Chemotaxonomy presents various challenges that need to be overcome in order to obtain valid and reliable results. Individual genetic and environmental variations can give a false picture and lead to wrong conclusions. Applying a holistic approach, based on multivariate data analysis, these challenges can be overcome. Thus, a metabolomics approach has to be optimized depending on the subject of research. We used 1H NMR-based metabolomics as a potential chemotaxonomic tool on the selected Euphorbia species growing wild in Serbia. Principal components analysis (PCA), soft independent modeling by class analogy (SIMCA) and Orthogonal Projections to Latent Structures Discriminant Analysis (OPLS-DA) were used to analyze obtained NMR data in order to reveal chemotaxonomic biomarkers. The standard protocol for plant metabolomics was optimized aiming to extract more specific metabolites, which are characteristic for the Euphorbia genus. The obtained models were validated, which revealed that variables unique for each species were associated with certain classes of molecules according to literature data. In E. salicifolia, acacetin-7-O-glycoside (not found before in the species) was detected, and the structure of the aglycone part was solved based on 2D NMR data. In the presented paper, we have shown that metabolomics can be successfully used in Euphorbia chemotaxonomy.

AB  - This study aimed to show aromatic profile of wines produced from two autochthonous grape cultivars Krstač (K) and Žižak (Z). During the wine production two enzymatic preparations (EP) Lallzyme cuvee blanc (CB) and Lallzyme enzymatic preparation β (EB) and different time of skin contact (4 and 8 h) were applied. Aromatic compounds were detected by GC/FID–MS ana­lysis. Significantly higher content of total detected aromatic compounds com­pared to appropriate controls (168.54 and 161.72 mg L-1) was observed for K EB4h (176.33 mg L-1) and Z CB4h (177.29 mg L-1) wines. Skin contact and usage of EP mostly increased content of 2-phenylethyl and isoamyl alcohols. Wines from both varieties showed higher content of hexanoic and octanoic acids compared to the control. It is interesting to emphasize that content of esters that are responsible for fruity aroma of wine which is important for plea­sant taste (isoamyl acetate – banana, ethyl hexanoate – ripe banana, 2-phe­nyl­ethyl acetate – powerful fruity rose like) were increased in all samples com­pared to the controls. The highest grades, after sensory evaluation, were obtained for K EB 8h (18.0 out of 20.0) and Z CB 8h (18.2 out of 20.0).

AB  - As byproducts of essential oil distillation, hydrolates are used in natural cosmetics/biomedicine due to their beneficial skin effects. However, data on their safety with relevant biological targets, such as human skin cells, are scarce. Therefore, we have tested nine hydrolates from the Lamiaceae family with skin fibroblasts that are responsible for extracellular collagenous matrix builds. Thyme, oregano, and winter savoury hydrolates showed several times higher total phenolics, which correlated strongly with their radical scavenging and antioxidative capacity; there was no correlation between their viability profiles and the reducing sugar levels. No proteins/peptides were detected. All hydrolates appeared safe for prolonged skin exposure except for 10-fold diluted lavender, which showed cytotoxicity (~20%), as well as rosemary and lavandin (~10%) using viability, DNA synthesis, and cell count testing. Clary sage, oregano, lemon balm, and thyme hydrolates (10-fold diluted) increased fibroblast viability and/or proliferation by 10–30% compared with the control, while their viability remained unaffected by Mentha and winter savoury. In line with the STITCH database, increased viability could be attributed to thymol presence in oregano and thyme hydrolates in lemon balm, which is most likely attributable to neral and geranial. The proliferative effect of clary sage could be supported by alpha-terpineol, not linalool. The major volatile organic compounds (VOCs) associated with cytotoxic effects on fibroblasts were borneol, 1,8-cineole, and terpinene-4-ol. Further research with pure compounds is warranted to confirm the roles of VOCs in the observed effects that are relevant to cosmetic and wound healing aspects.

Nevertheless, data on the effects of the endemic species Origanum minutiflorum, also known as wild or Turkish oregano, is scarce. On the other hand, various chronic lung diseases, characterised by persistent inflammation, oxidative stress, and common bacterial infections, do not have effective pharmacological therapy. Hence, the aim of this study is to examine the effects of wild oregano essential oil (WOEO) on human bronchial epithelial cells and lung pathogens.MethodsWe provided a detailed chemical composition of WOEO using GC-MS and GC-FID analysis. Anti-inflammatory effects of WOEO were analysed using the qRT-PCR and ELISA methods, while antioxidative properties were examined by using the dichlorofluorescein assay in BEAS-2B cells. Antibacterial activity was tested on lung pathogens by using the agar diffusion assay.ResultsThe major constituents of WOEO, analysed in this study, were carvacrol, linalool, p-cymene, γ-terpinene, and (E)-caryophyllene. We found that treatment with WOEO attenuated LPS-induced IL8 gene expression and hydrogen peroxide-induced oxidative stress in BEAS-2B cells. Moreover, WOEO showed an inhibitory effect on pathogenic bacteria Acinetobacter baumannii and highly resistant Klebsiella pneumoniae commonly seen in healthcare-associated pneumonia.ConclusionOur work presents new insights into the anti-inflammatory, antioxidative, and antimicrobial properties of WOEO which may be used as a simple and local treatment in various chronic lung diseases.

AB  - AbstractBackground and objectiveCotinus coggygria Scop. is a valuable medicinal plant species with pronounced pharmacological potential due to its numerous biological activities. The herb is characterized by a high content of polyphenols among which is fustin. The anticancer activities of fustin, however, are extremely weakly studied. The aim of the present study was to investigate the in vitro antiproliferative potential of fustin isolated from the heartwood of C. coggygria against cell lines originating from two of the most common cancer types – breast (MDA-MB-231 and MCF7), and colon cancer (Colon 26).Materials and methodsCell growth inhibitory properties of fustin were examined by MTT assay. Subsequently, phase-contrast and fluorescence microscopy analysis as well as colonyforming assay were carried out on the most sensitive to the cytostatic action of the fustin cell line.ResultsThe obtained results showed that fustin reduced the proliferation of all studied cell lines. The highest cytostatic effect was registered towards breast cancer MDA-MB-231 cells with a half maximal inhibitory concentration (IC50) value of 56.02 μg/ml followed by colon cancer cells with an IC50 of 78.07 μg/ml. AB  - U okviru ove doktorske disertacije proučavan je nutritivni i fitonutritivni sadržaj blitve koja je gajena u uslovima različitih režima đubrenja i navodnjavanja, pri čemu su ispitivane i funkcionalne osobine blitve u cilju utvrđivanja njenog potencijala kao funkcionalne hrane.U početnoj fazi eksperimenta određene su nutritivne, antinutritivne i fitonutritivne komponente u blitvi, kao i uticaj primjenjenih agrotehničkih mjera na pomenute komponente. Korišćenjem standardnih metoda za proksimativnu analizu (AOAC metode) utvrđeno je da svježa blitva ima sledeći sastav: voda- 89%, ukupni šećeri- 4,9%, proteini- 2,5%, pepeo- 1,8% i lipidi-0,45%. Savremenim instrumentalnim tehnikama (spektrofotometrija, ICP OES, tečna hromatografija) su kvantifikovane druge nutritivne, antinutritivne i fitonutritivne komponente u blitvi. Nađeno je da su prosječne koncentracije vitamina C, ukupnog hlorofila, karotenoida u 100 g svježe blitve su redom: 26,9 mg, 47,13 mg, 9,85 mg. Blitva ima visok sadržaj makro- i mikroelemenata od kojih su najzastupljeniji sledeći: kalijum (372,49 mg), kalcijum (178,6 mg), mangan (8,35 mg) i gvožđe (1,46 mg) izraženo na 100 g svježe blitve. Analizom ukupnog polifenolnog i flavonoidnog sadržaja pokazano je da je blitva bogata ovim fitonutrijentima, nakon čega su identifikovane i kvantifikovane četiri flavonoidne komponente: viteksin-2"-O-ksilozid, viteksin-6″-O-malonil-2″-O-ksilozid, izoramnetin-3,7-O-diglukozid i viteksin-6″-O-acetil-2″-O-ksilozid. Strukture ovih jedinjenja su potvrđene 1D i 2D NMR spektroskopijom (1H NMR,13 C NMR, COSY, HSQC, HMBC). Osim toga, određen je sadržaj nitrata u blitvi, koji je bio u granicama dozvoljenih vrijednosti.Statističkom analizom uticaja različitih režima navodnjavanja i đubrenja nađeno je da je đubrenje uticalo na prinos, morfološke karakteristike, količinu proteina, sadržaj fosfora, sadržaj izoramnetin-3,7-O-diglukozida, sadržaj hlorofila i vitamina C. Povećanjem količine primjenjene prihrane povećali su se svi pomenuti prametri, osim sadržaj fosfora i sadržaj izoramnetin-3,7-O-diglukozida, koji su bili niži sa povećanjem nivoa đubrenja. Interakcija navodnjavanja i đubrenja je značajno uticala na sadržaj Na i Cu i sadržaj viteksin-2"-O-ksilozida. Navodnjavanje nije imalo značajnog efekta na testirane parametre, osim na sadržaj nitrata, gdje je povećanjem nivoa navodnjavanja sadržaj nitrata u blitvi opadao, dok je povećanjem đubrenja rastao njihov sadržaj.Poslednja faza eksperimenta je obuhvatila ispitivanje antioksidativne, antiproliferativne i antimikrobne aktivnosti ekstrakta blitve. Rezultati su pokazali antioksidativnu aktivnost vodenog ekstrakta blitve u vrijednosti IC50=3,79 mg/ml (mjereno DPPH testom) dok je metanolni ekstrakt imao prosječni antioksidativni kapacitet od 8,0 (mjereno DPPH testom) odnosno 8,57 μmola ekvivalenta trolox-a/g (mjereno ABTS testom). Analizom antiproliferativne aktivnosti ekstrakta blitve je utvrđeno da postoji inhibicija rasta malignih MCF-7 ćelija i to 70% ćelija je inhibirano nakon 48 h inkubacije. Ispitivani ekstrakti blitve nijesu pokazali antimikrobnu aktivnost.
AB  - In this doctoral dissertation, the nutritional and phytonutrient content of Swiss chard grown under different irrigation and fertilization regimes was studied, while the functional properties of Swiss chard were also examined in order to determine its potential as a functional food.In the initial phase of the experiment, nutritional, anti-nutritive and phyto-nutritive components in Swiss chard were determined, as well as the influence of applied agrotechnical measurments on all tested parameters was monitored. Using standard methods for proximate analysis (AOAC methods), it was determined that fresh chard has the following composition: water- 89%, total sugars- 4,9 %, proteins- 2,5 %, ash- 1,8 % and lipids- 0,45%. Using modern instrumental techniques (spectrophotometry, ICP OES, liquid chromatography) other nutritional and phytonutritive components in Swiss chard were quantified. It was found that the average concentrations of vitamin C, total chlorophyll, carotenoids in 100 g of fresh chard are: 26,9 mg, 47,13 mg and 9,85 mg, respectively. Swiss chard has a high content of macro- and microelements, of which the most abundant are the following: potassium (372,49 mg), calcium (178,6 mg), followed by manganese (8,35 mg) and iron (1,46 mg) expressed per 100 g of fresh Swiss chard. The analysis of the total polyphenolic and flavonoid content showed that Swiss chard is rich in these phytonutrients, after which four flavonoid components were identified and quantified: vitexin-2"-O-xyloside, vitexin-6"-O-malonyl-2"-O-xyloside, isorhamnetin-3,7-O-diglucoside and vitexin-6″-O-acetyl-2″-O-xyloside. The structures of these compounds were confirmed by 1D and 2D NMR spectroscopy (1H NMR,13 C NMR, COSY, HSQC, HMBC). In addition, the content of nitrates in Swiss chard was determined, which was within the limits of permitted values.A statistical analysis of the effect of different irrigation and fertilization regimes found that fertilization influenced the yield, morphological characteristics, amount of protein, as well as phosphorus, isorhamnetin-3,7-O-diglucoside, chlorophyll, and vitamin C content. With increasing the amount of fertilization, values for all parameters mentioned above increased, except phosphorus and isorhamnetin-3,7-O-diglucoside content, which were found to be lower with increasing fertilization level. The interaction of irrigation and fertilization significantly affected Na and Cu content, as well as the content of vitexin-2"-O-xyloside. Irrigation had no significant effect on the studied parameters, except for the nitrate content. While the increased level of irrigation led to decreased nitrate content in Swiss chard, with increasing fertilization their content increased.The last phase of the experiment included investigation of the antioxidant, antiproliferative and antimicrobial activity of Swiss chard extract. The results showed the antioxidant activity of the aqueous extract of Swiss chard with an IC50 value of 3,79 mg/ml (measured by the DPPH test), while the methanolic extract had an average antioxidant capacity of 8,0 (measured by the DPPH test), 8,57 μmol of trolox equivalents per gram (measured by ABTS test). Analysis of the antiproliferative activity of Swiss chard extract revealed that there is an inhibition of the growth of malignant MCF-7 cells and that 70% of the cells were inhibited after 48 hours of incubation. The studied Swiss chard extracts did not show antimicrobial activity.

Savremenim instrumentalnim tehnikama (spektrofotometrija, ICP OES, tečna hromatografija) su kvantifikovane druge nutritivne, antinutritivne i fitonutritivne komponente u blitvi. Nađeno je da su prosječne koncentracije vitamina C, ukupnog hlorofila, karotenoida u 100 g svježe blitve su redom: 26,9 mg, 47,13 mg, 9,85 mg. Blitva ima visok sadržaj makro- i mikroelemenata od kojih su najzastupljeniji sledeći: kalijum (372,49 mg), kalcijum (178,6 mg), mangan (8,35 mg) i gvožđe (1,46 mg) izraženo na 100 g svježe blitve. Analizom ukupnog polifenolnog i flavonoidnog sadržaja pokazano je da je blitva bogata ovim fitonutrijentima, nakon čega su identifikovane i kvantifikovane četiri flavonoidne komponente: viteksin-2"-O-ksilozid, viteksin-6″-O-malonil-2″-O-ksilozid, izoramnetin-3,7-O-diglukozid i viteksin-6″-O-acetil-2″-O-ksilozid. Strukture ovih jedinjenja su potvrđene 1D i 2D NMR spektroskopijom (1H NMR,13 C NMR, COSY, HSQC, HMBC). Osim toga, određen je sadržaj nitrata u blitvi, koji je bio u granicama dozvoljenih vrijednosti.Statističkom analizom uticaja različitih režima navodnjavanja i đubrenja nađeno je da je đubrenje uticalo na prinos, morfološke karakteristike, količinu proteina, sadržaj fosfora, sadržaj izoramnetin-3,7-O-diglukozida, sadržaj hlorofila i vitamina C. Povećanjem količine primjenjene prihrane povećali su se svi pomenuti prametri, osim sadržaj fosfora i sadržaj izoramnetin-3,7-O-diglukozida, koji su bili niži sa povećanjem nivoa đubrenja. Interakcija navodnjavanja i đubrenja je značajno uticala na sadržaj Na i Cu i sadržaj viteksin-2"-O-ksilozida. Navodnjavanje nije imalo značajnog efekta na testirane parametre, osim na sadržaj nitrata, gdje je povećanjem nivoa navodnjavanja sadržaj nitrata u blitvi opadao, dok je povećanjem đubrenja rastao njihov sadržaj.Poslednja faza eksperimenta je obuhvatila ispitivanje antioksidativne, antiproliferativne i antimikrobne aktivnosti ekstrakta blitve. Influence of Different Pre-Distillation Steps on Aromatic Profile of Plum Spirits Produced by Traditional and Modified Methods

AB  - The aim of this study was to compare the aromatic profile of plum spirits, obtained from the Požegača and Stanley varieties, as affected by different combinations of pre-distillation steps used in traditional and six modified production methods that are common in Serbian distilleries. Traditional plum spirits (produced from spontaneously fermented mashes of crushed plums with stones distilled after two months of storage) had the highest contents of ethyl acetate, benzaldehyde, and total acids, which resulted in the occurrence of an unpleasant solvent-like and stone-like odour and acidic taste. These sensory defects were overcome by the distillation of fermented mashes immediately after the completion of alcoholic fermentation. Depending on the combination of the pre-distillation steps, plum spirits from mashes distilled immediately after alcoholic fermentation were characterized by different aromatic profiles: closer to traditional (from spontaneously fermented crushed plums with/without stones) or with a more pronounced fruity character (from pulped plums without stones regardless of the way of fermentation). These differences in aroma profiles have arisen mostly because of the significantly different contents and OAVs of ethyl esters and volatile fatty acids. The appropriate combination of the pre-distillation steps, which is adapted to the variety, can significantly improve the quality of the plum spirit compared to the traditionally produced spirit.
