AB  - Rosehip (pseudo-fruit) of dog rose (Rosa canina L.) is highly valued, and owing to nutritional and sensory properties it has a significant place in the food industry. This work represents an innovative report focusing on the evaluation of the phytochemical composition of rosehips (hypanthium and seed) grown in different locations in Serbia, using Raman microspectroscopy combined with multivariate data analysis. Some significant differences arose between the analysed rosehip samples with regard to the chemical profile of both hypanthium parenchyma cells and seed, although no evident discrimination was recorded between the samples of wild and cultivated rosehip. The differences between the hypanthium samples compared were mainly determined by the content of carotenoids, phenolic compounds and polysaccharides, whereas phenolics, polysaccharides (pectin, cellulose and hemicellulose) and lipids (to a lower extent) contributed to the seed sample discrimination. The differences observed between the rosehip samples may be attributed to abiotic factors (growing, ripening and storage conditions), which had a significant impact on the carotenoid and polyphenols biosynthesis.

AB  - Background Mammalian meat is the most common trigger of the allergic reactions in patients with α-Gal syndrome (AGS). Milk and dairy, although less often, also cause a significant number of allergic manifestations. The aim of this study was to identify α-Gal-containing bovine milk proteins with allergenic properties among AGS patients. Methods Thirty-eight AGS patients with IgE to milk were included in the study. Milk proteins were analyzed for the presence of α-Gal and for binding by patients’ IgE using immunoblot, ImmunoCAP, and inhibition ELISA. Allergenicity of milk and milk proteins was assessed by basophil activation test. Results More than half of the AGS patients reported allergic reactions to milk or dairy products. Bovine γ-globulin (BGG), lactoferrin (LF), and lactoperoxidase (LPO) were identified as α-Gal carrying proteins which were recognized by AGS patients’ IgE. Whey mirrored the anti-α-Gal and IgE reactivity of BGG, LF, and LPO. Eighty-nine percent of the patients displayed IgE to BGG, 91% to LF, and 57% to LPO. Inhibition of α-Gal-specific IgE binding was achieved by BGG, LF, LPO, and whey. These proteins also activated AGS patients’ basophils. Interestingly, at lower concentrations, LF was the most potent inhibitor of IgE binding, and the most potent activator of basophils. Conclusion BGG, LF, and LPO were all found to be relevant milk α-Gal-containing glycoproteins that bound AGS patients’ IgE antibodies and activated their basophils. These proteins are probably involved in the allergic reactions to milk in AGS patients. LPO was for the first time shown to be an allergen.

AB  - A focus of research in recent years is the comparison of honey as the final product of bees with pollen and nectar of the plant from which the honey originates, as the main food source for bees. Buckwheat honey is recognized as a nutritionally valuable product, which provides a scientifically proven health benefit and is confirmed as a functional food. The quality of this type of honey is attributed to high levels of phytochemicals in buckwheat. The purpose of this study was the examination of similarity between buckwheat honey and buckwheat nectar and pollen, as well as simultaneous investigation of their chemical profiles and the origin of the honey. The phenolic profile of buckwheat pollen showed a lower number of flavonoids and phenolic acids than those of nectar and honey samples, but confirmed the presence of the most characteristic polyphenols derived from the buckwheat plant. The notable difference was found to be the presence of (epi)catechin units, its galloylated derivatives and procyanidin dimers, which were not present in honey. Honey polyphenols displayed a pronounced correlation with those of nectar, but not with those of pollen. Finally, by comparing the polyphenolic profiles of honey, nectar and pollen sharing the same geographical origin, new data could be provided for a potential assessment of the botanical origin of buckwheat honey.

In vitro ispitivanje prebiotske, antioksidativne i antimikrobne aktivnosti pektina

AB  - Ovaj master rad fokusiran je na ispitivanje prebiotske aktivnosti pektina, njegove antioksidativne i
antimikrobne aktivnosti, kao i ispitivanja digestije pektina enzimima prisutnih u gastrointestinalnom
traktu.
Prebiotska aktivnost pektina ispitana je fermentacijom bakterijama selektovanih iz kolostruma,
prelaznog i zrelog mleka, kao i sojevima bifidobakterija (Bifidobacterium breve i Bifidobacterium
bifidum) i Lactobacillus rhamnosus. Upoređena je antimikrobna i antioksidativna aktivnost pektina pre
i nakon fermentacije. Pored klasične spektrofotometrijske, DPPH metode, kojom je određena
antioksidativna aktivnost, primenjena je i elektrohemijska metoda ciklične voltametrije.
Eksperimentom in vitro digestije pektina, digestovan je 0,5 % vodeni rastvor pektina enzimima
digestivnog trakta, nakon čega je određen uticaj na rast laktobacila, promena pH vrednosti i
antioksidativna aktivnost pre i nakon digestije.
Eksperimentalno dobijeni podaci ukazuju da pektin ima prebiotsku aktivnost i da povoljno utiče na rast
i aktivnost probiotskih bakterija. Takođe su dokazana i njegova antioksidativna i antimikrobna
svojstva, kao i sposobnost enzima gastrointestinalnog trakta da digestuju pektin.

Kako učenici koriste informacije iz Periodnog sistema elemenata

AB  - Predmet ovog završnog rada odnosi se na osposobljenost učenika osmog razreda osnovne škole i različitih razreda srednje škole da koriste informacije iz Periodnog sitema elemenata u rešavanju zadataka iz hemije. Iz tako postavljenog predmeta proistekao je cilj završnog rada. Cilj rada je da se ispita u kojoj meri učenici osmog razreda osnovne škole i srednjoškolci primenjuju informacije koje pruža Periodni sistem elemenata u rešavanju zadataka iz hemije. Uzorak u istraživanju je činilo ukupno 397 učenika osmog razreda osnovne škole, prvog razreda gimnazije, drugog razreda gimnazije, trećeg razreda gimnazije, četvrtog razreda gimnazije i prvog razreda srednje medicinske škole. Za potrebe istraživanja napravljeni su instrumenti: test znanja i upitnik. Donja granica procentata tačnih odgovora za nazive i hemijske simbole elemenata prve tri periode Periodnog sistema elemenata nije ispod 60 % u svih šest grupa ispitanika. U visokom procentu učenici u svim grupama su na osnovu položaja elementa u Periodnom sistemu navodili grupu i periodu u kojima je element, atomski i maseni broj. U odnosu na to, procenat tačnih odgovora je značajno niži na zahtevima da se odredi broj protona, elektrona i neutrona u atomima elemenata na osnovu položaja elementa u Periodnom sistemu, a niži je i za prepoznavanje trendova u Periodnom sistemu. Učenici svih razreda u najvećoj meri iz Periodnog sistema elemenata koriste podatke o atomskom i masenom broju, simbolu i nazivu elementa, kao i položaju elementa u PSE.

Efekti primene onlajn materijala za učenje o protolitičkoj teoriji u prvom razredu gimnazije

AB  - Cilj ovog istraživanja je bilo da se ispitaju efekti primene onlajn materijala za učenje o protolitičkoj teoriji u prvom razredu gimnazije. Istraživanje je sprovedeno na 130 učenika prvog razreda dve gimnazije. Instrumenti korišćeni u istraživanju su ulazni test, kojim se proveravalo prethodno znanje učenika o kiselinama i bazama, izlazni test, kojim se proveravala uspešnost usvajanja gradiva o protolitičkoj teoroji pomoću onlajn materijala i upitnik za ispitivanje učeničkih stavova. Svim učesnicima istraživanja je bio dostavljen onlajn materijal identičnog sadržaja, ali na jeziku na kojem prate nastavu hemije (srpski i slovački), u okviru kojeg su usvajali znanja o protolitičkoj teoriji sopstvenim tempom kroz male porcije gradiva, praćene zadacima za utvrđivanje. Iz inicijalnog testa se uviđa da obe grupe učenika imaju predznanje o kiselinama i bazama. Rezultati izlaznog testa sugerišu se da je grupa Gimnazije Uroš Predić postigla bolje rezultate nakon korišćenja onlajn materijala, nego što je tu slučaj kod ispitanika iz uzorka koji pohađaju Gimnaziju Mihajlo Pupin.

AB  - Ljudska koža je najveći ljudski organ i ima površinu od 1,5 do 2 m2 što čini približno 15% težine celog organizma. Koža se sastoji iz tri sloja epidermisa, dermisa i potkožnog tkiva (hipodermisa). Epidermis, koji ima važnu ulogu u održavanju temperature tela, sadrži keratinocite, melanocite, Merkelove i Langerhans-ove ćelije. U dermisu, lociranom ispod epidermisa, se nalaze brojni nervni završeci, folikuli dlaka, znojne, lojne i apokrine žlezde, krvni i limfni sudovi. Potkožno tkivo se sastoji od areolarnog vezivnog tkiva, adipoznog tkiva, elastina, fibroblasta, makrofaga i adipocita.
Za boju ljudske kože je u najvećoj meri odgovoran pigment melanin koji se sintetiše u melanocitima, u bazalnom sloju epidermisa unutar sitnih organela-melanozoma, procesom koji se naziva melanogeneza.

AB  - Papain is a proteolytic enzyme of great commercial value. It is a cysteine protease highly expressed in Carica papaya fruit latex, but also present in papaya leaves. Purification procedures mostly deal with the latex and include a combination of precipitation and/or chromatographic techniques. Due to its solubility, structure and activity characteristics, the pH and salt content play significant roles in handling papain extracts. Here we report a simple, rapid and easily scalable procedure for papain purification from papaya leaves, which contain different contaminants as compared to papaya latex. Sodium chloride precipitation of contaminants at pH 5 followed by ammonium sulphate precipitation resulted in the removal of other leaf proteins and protein fragments from papain solution and about a 3-fold purification. The procedure also benefits from the suppression of autoproteolysis and preservation of the native structure, as confirmed by FTIR analysis, and the high recovery of activity of over 80%.

AB  - Pollutants such as pharmaceutical products and pesticides are still present in treated water. Several of these compounds are photoactive. The photodegradation of eight organic pollutants was studied using listed advanced oxidation processes: UV/H2O2, UV/persulfate, Fenton, photo-Fenton and UV/TiO2. The results show that photodegradation is most effective in the first 10 min of treatments in simulated urban wastewater collected from the local Nišava River. To identify the correlation between applied degradation processes as well as the relationship among the structure of investigated compounds and efficiency of degradation, Pearson’s correlation coefficient analysis and principal component analysis (PCA) were performed. The established correlations between the applied processes, as well as the determination of the influence of the structure on the efficiency of degradation, enable a more efficient choice of the degradation procedure, especially in the case of interference. Additionally, process electrical energy consumption and treatment costs for simulated urban wastewater with different pharmaceuticals and pesticides were determined.

AB  - Measurement of antithyroglobulin antibodies (TgAb) is an inevitable laboratory tool in the management of thyroid gland diseases. Currently available immunoassays still have limitations underlying the necessity of the introduction of fast, sensitive, and label-free technologies. Our aim was to develop a method for TgAb measurement in human serum based on the quartz crystal microbalance (QCM) technology. We immobilized thyroglobulin on the surface of Attana LNB Carboxyl sensor chip®, prepared standard curve covering the range of 1–50000 kIU/L, and established optimal measurement conditions. The validation included determination of the detection limit (LOD), functional sensitivity, linearity, precision, as well as the comparison with the results of the radioimmunoassay (RIA). The LOD and functional sensitivity were 4.2 kIU/L and 4.7 kIU/L, respectively. The method was linear in the range of 20–10000 kIU/L. The regression equation for comparison with RIA was CQCM = 1.0056 • CRIA – 24.2778, whereby no significant proportional or systematic difference was present. There was a good agreement with RIA in the classification of patients according to the clinical significance of the results. The developed method has advantages over currently available assays in terms of better LOQ, a higher upper limit of linearity, and precision. The characteristics of the developed method unambiguously show that the application of the QCM biosensors offers a highly reliable novel approach for the measurement of TgAb in human serum.

abstract = "Measurement of antithyroglobulin antibodies (TgAb) is an inevitable laboratory tool in the management of thyroid gland diseases. Currently available immunoassays still have limitations underlying the necessity of the introduction of fast, sensitive, and label-free technologies. Our aim was to develop a method for TgAb measurement in human serum based on the quartz crystal microbalance (QCM) technology. We immobilized thyroglobulin on the surface of Attana LNB Carboxyl sensor chip®, prepared standard curve covering the range of 1–50000 kIU/L, and established optimal measurement conditions. The validation included determination of the detection limit (LOD), functional sensitivity, linearity, precision, as well as the comparison with the results of the radioimmunoassay (RIA). The LOD and functional sensitivity were 4.2 kIU/L and 4.7 kIU/L, respectively. The method was linear in the range of 20–10000 kIU/L. The regression equation for comparison with RIA was CQCM = 1.0056 • CRIA – 24.2778, whereby no significant proportional or systematic difference was present. There was a good agreement with RIA in the classification of patients according to the clinical significance of the results. The developed method has advantages over currently available assays in terms of better LOQ, a higher upper limit of linearity, and precision. The characteristics of the developed method unambiguously show that the application of the QCM biosensors offers a highly reliable novel approach for the measurement of TgAb in human serum.",

AB  - Oligomeric intermediates on the pathway of amyloid fibrillation are suspected as the main cytotoxins responsible for amyloid-related pathogenicity. As they appear to be a part of the lag phase of amyloid fibrillation when analyzed using standard methods such as Thioflavin T (ThT) fluorescence, a more sensitive method is needed for their detection. Here we apply Fourier transform infrared spectroscopy (FTIR) in attenuated total reflectance (ATR) mode for fast and cheap analysis of destabilized hen-egg-white lysozyme solution and detection of oligomer intermediates of amyloid fibrillation. Standard methods of protein aggregation analysis— Thioflavin T (ThT) fluorescence, atomic force microscopy (AFM), and 8-anilinonaphthalene-1-sulphonic acid (ANS) fluorescence were applied and compared to FTIR spectroscopy data. Results show the great potential of FTIR for both, qualitative and quantitative monitoring of oligomer formation based on the secondary structure changes. While oligomer intermediates do not induce significant changes in ThT fluorescence, their secondary structure changes were very prominent. Normalization of specific Amide I region peak intensities by using Amide II peak intensity as an internal standard provides an opportunity to use FTIR spectroscopy for both qualitative and quantitative analysis of biological samples and detection of potentially toxic oligomers, as well as for screening of efficiency of fibrillation procedures.

AB  - Od Werner-ove sinteze prvih kompleksa iz kobalt(III)-hlorida i amonijaka pa do danas, dobijen je veliki broj koordinacionih jedinjenja koja sadrže različite centralne metalne jone i ligande. Za eksponencijalni razvoj koordinacione hemije i interesovanje za ovaj vid jedinjenja najviše je zaslužna njihova raznolika primena u medicini, za lečenje različitih oboljenja, kao i u dijagnostici. Osim u terapeutske svrhe kompleksna jedinjenja našla su ogromnu primenu kao katalizatori u sintetičkoj i organskoj hemiji. U prilog ovoj činjenici idu i nekoliko Nobelovih nagrada za hemiju koje su dodeljene za transformacije koje katalizuju kompleksi paladijuma i rutenijuma.
Cilj ovog završnog rada je sinteza i karakterizacija novih kompleksnih jedinjenja na bazi rutenijuma sa 3-amino-2-hlorpiridinom kao ligandom i njihova potencijalna primena u medicinskoj hemiji.

AB  - Different Schiff base complexes have biological activities that make them suitable for drug design. The biological properties of tetradentate Schiff base copper(II) complexed with N,N′-bis(acetylacetone)propylenediimine have been studied. The cytotoxic activity towards Caco-2 cells were determined by MTT, Anexin V and PI apoptosis assays. N,N′-bis(acetylacetone)propylenediimine-copper(II) showed the anti-cancer and anti-proliferative properties by inducing apoptosis in Caco-2 cells. A comparison of the cytotoxic activity of this compound with cisplatin shows that it is more effective on the colorectal cancer cell line Caco-2. The binding capacity and interaction of N,N′-bis(acetylacetone)propylenediimine-copper(II) with HSA were systemically investigated by in vitro fluorescence spectroscopy, CD spectroscopy, and in silico molecular docking study. Furthermore, in vitro and in silico interaction studies indicated that the complex binds to HSA through a static quenching mechanism without changes in protein conformation. The calculated number of binding sites was in line with molecular docking studies. The obtained Ka value suggests that the compound can be released from the protein in target cells. The tetradentate Schiff base copper(II) complex exhibited in vitro biological activities against cancer epithelial cells, which depend on the molecular structure of the complex, causing apoptosis, and the complex can bind to the protein drug carrier in circulation to the target tissue.

AB  - Background: Diazepam belongs to the group of 1,4-benzodiapines. It is used for the treatment of anxiety, convulsions and as a muscle relaxant. The presence of 4,5-azomethine group enables its electrochemical detection.

Introduction: A screen-printed electrode modified with antimony film was used for the determination of diazepam in pharmaceutical preparations

Methods: Electrode modification was done by ex-situ deposition of antimony on commercially available screen-printed electrode. Parameters affecting the electroanalytical response of the sensor, such as deposition potential, deposition time, and antimony concentration, were examined and optimized. The modified electrode showed enhanced electroactivity for diazepam reduction compared to unmodified electrode. Under optimal conditions, linear sweep voltammetry was used for the determination of analyte.

Results: The sensor showed linear dependence in the range from 0.5 to 10 μmol/L, the correlation coefficient was 0.9992. The limit of detection was 0.33 μmol/L, corresponding limit of quantification was 1.08 μmol/L. Modification enabled determination of diazepam in the presence of oxygen. 

Conclusion: The modified electrode was used for the determination of diazepam in tablets. Results confirmed the applicability of the electrochemical sensor.

Introduction: A screen-printed electrode modified with antimony film was used for the determination of diazepam in pharmaceutical preparations

Methods: Electrode modification was done by ex-situ deposition of antimony on commercially available screen-printed electrode.
