Teorijsko proučavanje interakcija glikozilovanih flavonoida sa lipokalinima

AB  - Beta laktoglobulin (BLG) je glavni protein surutke i veoma poznat alergen koji poseduje
više mesta za vezivanje liganda. BLG pokazuje afinitet prema širokom spektru
bioaktivnih jedinjenja, kao što su masne kiseline, vitamini i polifenolna jedinjenja. Bet v
1 je protein koji je u velikoj meri prisutan u polenu stabla bele breze i primarni je uzrok
alergije na polen breze kod ljudi. Bet v 1 vezuje niz fiziološki relevantnih liganda
uključujući masne kiseline, flavonoide i citokinine. Biološka funkcija Bet v 1 nije
poznata. Međutim, pretpostavlja se da neobična karakteristika tercijarne strukture Bet v 1,
unutrašnja šupljina, igra centralnu ulogu u biološkoj funkciji molekula, i verovatno to
čini kroz specifičnu interakciju sa nepoznatim ligandom. Flavonoidi su jedna od grupa
bioaktivnih jedinjenja koja su opsežno proučavana. Bioaktivna jedinjenja imaju veliki
potencijal da budu korišćena u „ojačavanju“ hrane zarad zdravstvenih koristi.
U ovom radu su teorijskim metodama proučavane interakcije i efekti vezivanja
glikozilovanih polifenola, (izoramnetin-3-O-glukozid, izoramnetin 3-O-(malonil 3-O-
glukozid), kvercetin 3-O (malonil 3-O-glukozid) i izoramnetin-3-O-rutinozid)), sa Bet v1 i
BLG u obliku monomera i dimera.
Metodom slepog dokinga su pronađena mesta vezivanja flavonoida za ispitivane proteine i
analizirane interakcije koje ovi flavonoidi grade sa aminokiselinskim ostacima u
okruženju. Pokazano je da na vezivanje glikozilovanih flavonoida za lipokaline najviše
utiče šećerna komponenta koja, sa jedne strane, može uticati na povećan afinitet vezivanja
usled većeg broja hidroksilnih grupa koje poseduje, a sa druge strane, može smanjiti ili čak
onemogućiti pristup unutrašnjoj šupljini proteina, usled velike zapremine.

Uticaj hroma (VI) na aktivnost enzima oksidativnog stresa pekarskog i pivskog kvasca

AB  - Cilj ovog rada je određivanje uticaja hroma na pekarski i pivski kvasac. Uticaj je praćen postavljanjem tečnih fermentacija sa 400 μM Cr i 600 μM Cr u podlozi, a rezultati su poređeni sa kontrolom.
Nakon završene fermentacije određivani su ukupan broj mikroorganizama metodom serijskog razblaženja i količina biomase. Posebna pažnja je posvećena uticaju Cr6+ na enzim oksidativnog stresa pekarskog i pivskog kvasca, superoksid dismutazu. Enzimska aktivnost je određena nakon homogenizacije uzoraka sa ditiotreitolom (DTT) i izražena je kao specifična aktivnost.
Značajne razlike u rezultatima su dobijene za pekarski i pivski kvasac.

AB  - Rod Alnus Mill. (jove), sa više od 30 vrsta, pripada familiji Betulaceae. Alnus vrste koje rastu u Srbiji su: A.glutinosa (crna jova), A.viridis (zelena jova) i A.incana (siva jova).
Jedinjenja izolovana iz jova poseduju širok spektar bioloških aktivnosti, a najvažnije su: citotoksična, antitumorska, antivirusna, antimikrobna i antiparazitska. Vrste roda Alnus su bogat izvor diarilheptanoida. Oni se u biljci prvenstveno nalaze u kori i lišću, kao prosti, glikozidi ili složeni – diarilheptanoid glikozidi esterifikovani različitim fenolnim kiselinama, najčešće cinamičnim. Biljke ovog roda su u tradicionalnoj medicini korišćene u brojne svrhe kao alterativi, purgativi, emetici, hemostatici, za lečenje raznih bolesti, uključujući i tumor.
Cilj ovog rada bilo je izolovanje i određivanje strukturnih izomera alnuzida A i alnuzida B iz kore sive jove.

AB  - The citric acid (CA), dextran (DEX) and (3-aminopropyl)triethoxysilane (APTES) are frequently used ligands for surface modification of magnetite nanoparticles to improve their colloidal stability and biocompatibility for use in biomedicine. Here we report the synthesis, surface modification of magnetite (Fe3O4) nanoparticles and analysis of the influence of coating materials on heating efficiency in magnetic hyperthermia, as well as on electrochemical sensing toward the detection of neurotransmitter dopamine (DOP). Spherical magnetite nanoparticles (~13 nm) were prepared by co-precipitation and surface-modified with CA, APTES and DEX. The hyperthermia tests revealed that Fe3O4@APTES resulted in the highest specific absorption rate (SAR) of 67.2 W/g. The engineered sensor, by surface modification of screen-printed carbon (SPC) electrodes with Fe3O4@APTES, showed linear response with DOP in the concentration range of 1−200 μM and a low detection limit of 34.3 nM. Besides, the sensor offered remarkable selectivity, repeatability and reproducibility. Application of SPC-, Fe3O4-, Fe3O4@CA- and Fe3O4@DEX-modified SPC electrode resulted in lower electrocatalytic activity compared with Fe3O4@APTES/SPC. The obtained results indicate the effects of a surface modifier especially that Fe3O4@APTES, alone and combining with SPC electrodes, can serve as an agent in various types of biomedical applications, like hyperthermia and electrochemical sensing of biologically active compounds such as DOP.

AB  - The goal of this work was to develop green electrode material that unites all advantages of domestic made, synthesized porous carbon powder and ceria dioxide nanoparticles known due to exceptional catalytic properties. Thermal decomposition of Novolac phenol-formaldehyde resin and cerium sulfate resulting in producing a high-performance CeO2 porous carbon material highly sensitive to dopamine (DA) electrooxidation. Morphological and structural characteristics of the material were determined by SEM and XRD measurements, while electrochemical characterization was performed by EIS and CV. The sensitivity of DA determination on the proposed CeO2-doped carbon material was enhanced by adding multi-wall carbon nanotubes to finally prepare a mixture for a specific carbon paste electrode (TPCeO2&MWCNT@CPE). SWV technique was used for quantification of dopamine in Britton-Robinson buffer pH 6 in the concentration range of 0.5-100 μM of DA, with the detection limit of 0.14 μM and quantification limit of 0.44 μM. abstract = "The goal of this work was to develop green electrode material that unites all advantages of domestic made, synthesized porous carbon powder and ceria dioxide nanoparticles known due to exceptional catalytic properties. Thermal decomposition of Novolac phenol-formaldehyde resin and cerium sulfate resulting in producing a high-performance CeO2 porous carbon material highly sensitive to dopamine (DA) electrooxidation. Morphological and structural characteristics of the material were determined by SEM and XRD measurements, while electrochemical characterization was performed by EIS and CV. The sensitivity of DA determination on the proposed CeO2-doped carbon material was enhanced by adding multi-wall carbon nanotubes to finally prepare a mixture for a specific carbon paste electrode (TPCeO2&MWCNT@CPE). SWV technique was used for quantification of dopamine in Britton-Robinson buffer pH 6 in the concentration range of 0.5-100 μM of DA, with the detection limit of 0.14 μM and quantification limit of 0.44 μM. AB  - L-DOPA (L-3,4-dihydroxyphenylalanine), the precursor of dopamine, is widely used in the treatment of Parkinson’s disease, thus determining and monitoring the concentration of L-DOPA is of utmost importance for both medical and scientific purposes. Although many analytical approaches, designed for drug detection and quantification, already exist, there is a constant need for modification of old and tailoring of new, faster, and selective methods. Redox active chemical species, such as L-DOPA, can be measured directly by electrochemical means, whereas electrochemical sensors combine sensitivity and selectivity within a small analytical device. This work demonstrates the development of such electrochemical sensor, based on carboxylated single-wall carbon nanotubes (SWCNT-COOH) decorated with SiO2 coated-Nd2O3 nanoparticles, and further application for the detection of L-DOPA. Developed SWCNT-COOH@Nd2O3-SiO2 sensor shows linear response in the range from 2 µmol L−1 to 52 µmol L−1 analyte concentration, and beside the low detection limit, it is characterized by a fast response time, as well as good life-time, reproducibility and repeatability.

Coupling these two reactions results in a powerful combination that allows for a rapid, stereoselective access to substituted, highly functionalized five- and six-membered N-heterocycles. This minireview illustrates the applicability of this approach as the key feature in syntheses of various iminosugars – a class of polyhydroxylated alkaloids with a range of interesting biological activities and high pharmacological potential.

AB  - Ispitivane su interakcije poznatih lekova (indapamid, olanzapin i atorvastatin) sa strukturnim analozima serum albumina korišćenjem fluorescentne spektroskopije, UV/Vis spektrofotometrije i ravnotežne dijalize. Pokazane su razlike u vezivanjima ove tri supstance: indapamid se ne vezuje za serum albumin, a olanzapin i atorvastatin se vezuju ali sa razlikama u vezivnim mestima. Ove razlike u vezivanju se pripisuju strukturnim razlikama supstanci jer indapamid ima sulfonamidnu grupu, olanzapin tiofensko, a atorvastatin pirolno jezgro. Promene u fluorescenciji serum albumina praćene su na tri temperature (20, 25 i 37 °C) i analizirane su Stern-Volmerovom jednačinom koja je pokazala statički mehanizam vezivanja. Ravnotežnom dijalizom su potvrđene konstante vezivanja olanzapina i atorvastatina za serum albumin.

AB  - Pripremljeni su molekulski obeleženi polimeri na bazi β-ciklodekstrina kod kojih je teofilin služio kao templat, primenom dva sintetička puta. Jedan je podrazumevao upotrebu β ciklodekstrina kao monomera pri čemu je dobijen monolitni polimer, dok je kod drugog korišćen monomer nastao modifikacijom β-ciklodekstrina metakrilnom kiselinom i nastali polimer je bio u obliku sitnih čestica. Pripremljeni su i polimeri obeleženi donepezilom, upotrebom metakrilne kiseline kao funkcionalnog monomera i divinilbenzena i etilenglikol-dimetakrilata kao umreživača. Prepolimerizaciona smeša je ispitana konduktometrijski. Dobijeni polimeri su okarakterisani pomoću FTIR spektroskopije, skenirajuće elektronske mikroskopije (SEM) i adsorpcionih izotermi. Na osnovu ponovnog vezivanja templata za dobijene obeležene i neobeležene polimere, utvrđeno je da je obeležavanje uspešno izvedeno. Jedan od obeleženih polimera je odabran na osnovu prethodnih analiza i testiran u uslovima sličnim fiziološkim kako bi se dobile informacije koliko dugo i koje količine aktivnih farmaceutskih supstanci (templata/donepezila) otpušta.

Zbog prisustva bočnog lanca alkilamina ovi molekuli se ponašaju nalik surfaktantima. Ukoliko je koncentracija supstance u rastvoru iznad određene koncentracije može doći do formiranja agregata (tzv. micela). Ta granična koncentracija iznad koje dolazi do formiranja micela naziva se kritična micelarna koncentracija (kmk). U ovom završnom radu korišćene su konduktometrijske titracije za određivanje kmk ImiHCl i AmiHCl. Specifična provodljivost, merena je na konstantnoj temperaturi od 25±1 °C, u rastvorima NaH2PO4 i NaCl. Takođe, određena je rastvorljivost ImiHCl i AmiHCl u rastvorima sa i bez pufera, u prisustvu fiziološki relevantne koncentracije hlorida, primenom pH-Ramp Shake-Flask metode. Rastvorljivost je određena titracijama polazeći od bazne ka kiseloj sredini, i obrnuto. pH Vrednost je podešena korišćenjem HCl, H3PO4 i/ili NaOH u zasićenim rastvorima fosfatnog pufera. Nakon uspostavljanja stabilnog ravnotežnog stanja (nakon mešanja od 6 h i 18 h sedimentacije), centrifugiranje je korišćeno za odvajanje faza. Koncentracija imipramina i amitriptilina je izmerena pomoću HPLC sa UV/VIS detekcijom.

AB  - Rastvorljivost se izražava kao koncentracija supstance u određenoj zapremini rastvarača na određenoj temperaturi i predstavlja važan fizičko-hemijski parametar u ranoj fazi dizajna i razvoja lekova. Nortriptilin-hidrohlorid spada u grupu tricikličnih antidepresiva koji se koriste u lečenju depresije. pH-Zavisna rastvorljivost nortriptilin-hidrohlorida proučavana je primenom pH-Ramp shake-flask metode. Određivanje rastvorljivosti je od velikog značaja, budući da je u digestivnom traktu zastupljen širok opseg pH vrednosti (od jako kisele sredine u želucu do slabo bazne u crevima), pa samim tim visoka rastvorljivost jonizovanog (kiselog ili baznog) oblika supstance omogućava bolju apsorpciju iz digestivnog trakta u krvotok i veću biodostupnost na mestu dejstva. Primenom softverskog paketa pDISOL-X (in-ADME Research, New York, USA) određene su ravnoteže u heterogenom sistemu koji čini nortriptilin-hidrohlorid i fosfatni pufer.

AB  - Different phosphates and phosphonates have shown excellent coating ability toward magnetic nanoparticles, improving their stability and biocompatibility which enables their biomedical application. The magnetic hyperthermia efficiency of phosphates (IDP and IHP) and phosphonates (MDP and HEDP) coated Fe3O4 magnetic nanoparticles (MNPs) were evaluated in an alternating magnetic field. For a deeper understanding of hyperthermia, the behavior of investigated MNPs in the non-alternating magnetic field was monitored by measuring the transparency of the sample. To investigate their theranostic potential coated Fe3O4-MNPs were radiolabeled with radionuclide 177Lu. Phosphate coated MNPs were radiolabeled in high radiolabeling yield (> 99%) while phosphonate coated MNPs reached maximum radiolabeling yield of 78%. Regardless lower radiolabeling yield both radiolabeled phosphonate MNPs may be further purified reaching radiochemical purity of more than 95%. In vitro stabile radiolabeled nanoparticles in saline and HSA were obtained. The high heating ability of phosphates and phosphonates coated MNPs as sine qua non for efficient in vivo hyperthermia treatment and satisfactory radiolabeling yield justifies their further research in order to develop new theranostic agents. © 2021, The Author(s), under exclusive licence to Springer Science+Business Media, LLC, part of Springer Nature.

AB  - Usled eksponencijalnog rasta populacije ljudi na Zemlji, kao i veće ekološke osvešćenosti sve više se radi na istraživanjima alternativnih izvora energije. Jedan takav izvor predstavljaju mikrobne gorivne ćelije koje koriste električnu energiju mikroorganizama za napajanje malih potrošača. Ove ćelije, usled inherentnih ograničenja u pogledu gubitaka koji tokom njihovog rada nastaju, zahtevaju poboljšanja na nivou elektroda, mikrobnih populacija, konfiguracija više povezanih ćelija i sl. U ovom radu proučavan je uticaj inkubacije elektroda kulturom mikroorganizama u cilju poboljšanja perofrmansi. Došli smo do saznanja da su najadekvatniji materijali za izradu elektroda na bazi ugljenika, kao i da inkubirane elektrode poboljšavaju performanse u kontekstu smanjenja početnog perioda porasta napona (eng. start-up), kao i dužeg vremena u kojem ćelije pružaju relativno visok, koristan, napon.

AB  - We describe herein the synthesis and laccase mediated oxidation of six novel 1,4-dihydropyridine (DHP)-based hexahydroquinolines (DHP1-DHP3) and decahydroacridines (DHP4-DHP6). We employed different laccase enzymes with varying redox potential to convert DHP1-DHP3 and DHP4-DHP6 to the corresponding pyridine-containing tetrahydroquinoline and octahydroacridine derivatives, respectively. Intensively coloured products were detected in all biocatalytic reactions using laccase from Trametes versicolor (TvLacc), possibly due to the presence of conjugated chromophores formed in products after oxidation. The NMR assessment confirmed that the oxidation product of DHP1 was its corresponding pyridine-bearing tetrahydroquinoline derivative. Laccase from Bacillus subtillis (BacillusLacc) was the most efficient enzyme for this group of substrates using HPLC assessment. Overall, it could be concluded that DHP2 and DHP5, bearing catecholic structures, were easily oxidized by all tested laccases, while DHP3 and DHP6 containing electron-withdrawing nitro-groups are not readily oxidized by laccases. DHP4 with decahydroacridine moiety consisting of three condensed six-membered rings that contribute not only to the volume but also to the higher redox potential of the substrate rendered this compound not to be biotransformed with any of the mentioned enzymes. abstract = "We describe herein the synthesis and laccase mediated oxidation of six novel 1,4-dihydropyridine (DHP)-based hexahydroquinolines (DHP1-DHP3) and decahydroacridines (DHP4-DHP6). We employed different laccase enzymes with varying redox potential to convert DHP1-DHP3 and DHP4-DHP6 to the corresponding pyridine-containing tetrahydroquinoline and octahydroacridine derivatives, respectively. Intensively coloured products were detected in all biocatalytic reactions using laccase from Trametes versicolor (TvLacc), possibly due to the presence of conjugated chromophores formed in products after oxidation. The NMR assessment confirmed that the oxidation product of DHP1 was its corresponding pyridine-bearing tetrahydroquinoline derivative. Laccase from Bacillus subtillis (BacillusLacc) was the most efficient enzyme for this group of substrates using HPLC assessment. Overall, it could be concluded that DHP2 and DHP5, bearing catecholic structures, were easily oxidized by all tested laccases, while DHP3 and DHP6 containing electron-withdrawing nitro-groups are not readily oxidized by laccases. DHP4 with decahydroacridine moiety consisting of three condensed six-membered rings that contribute not only to the volume but also to the higher redox potential of the substrate rendered this compound not to be biotransformed with any of the mentioned enzymes. Overall, we showed that multiple analytical approaches are needed in order to assess biocatalytical reactions.",

AB  - Cerium oxide nanoparticles (nCeO2) have widespread applications, but they can be hazardous to the environment. Some reports indicate the toxic effect of nCeO2 on tested animals, but literature data are mainly contradictory. Coating of nCeO2 can improve their suspension stability and change their interaction with the environment, which can consequently decrease their toxic effects. Herein, the exopolysaccharides levan and pullulan, due to their high water solubility, biocompatibility, and ability to form film, were used to coat nCeO2. Additionally, the monosaccharide glucose was used, since it is a common material for nanoparticle coating. This is the first study investigating the impact of carbohydrate-coated nCeO2 in comparison to uncoated nCeO2 using different model organisms. The aim of this study was to test the acute toxicity of carbohydrate-coated nCeO2 on the bacterium Vibrio fischeri NRRL B-11177, the crustacean Daphnia magna, and zebrafish Danio rerio. The second aim was to investigate the effects of nCeO2 on respiration in Daphnia magna which was performed for the first time. Finally, it was important to see the relation between Ce bioaccumulation in Daphnia magna and Danio rerio and other investigated parameters. Our results revealed that the coating decreased the toxicity of nCeO2 on Vibrio fischeri. The coating of nCeO2 did not affect the nanoparticles’ accumulation/adsorption or mortality in Daphnia magna or Danio rerio. Monitoring of respiration in Daphnia magna revealed changes in CO2 production after exposure to coated nCeO2, while the crustacean's O2 consumption was not affected by any of the coated nCeO2. In summary, this study revealed that, at 200 mg L−1, uncoated and carbohydrate-coated nCeO2 are not toxic for the tested organisms, however, the CO2 production in Daphnia magna is different when they are treated with coated and uncoated nCeO2. AB  - In vivo measurements of the optical properties of plant leaves, by spectrophotometric methods, could potentially provide very useful information for the assessment of the plant’s health. This paper describes a novel experimental set-up that enables continuous measurements of the optical reflection and transmission coefficients of broad-leaved plants. The optical spectral characteristics of leaves, in the broad wavelength range, are often a discussed topic in the scientific literature. Nevertheless, to the best of our knowledge, the real-time spectral analyses of the leaf haven’t been reported so far. Monitoring the evolution of the plant activity in real-time has resulted in the graph of the spectral Circadian rhythm as a function of time. The set-up was tested on Phaseolus vulgaris and the results of the measurements are presented and discussed.

AB  - Termin pektin se odnosi na strukturno najsloženiju porodicu polisaharida u prirodi. Nalazi se u ćelijskom zidu biljaka. Jako je značajan po svojoj sposobnosti da poboljša kvalitet hrane. Njegova izvanredna struktura i svojstva su doprinela da se dosta koristi kao stabilizator i zgušnjivač. Prvo pominjanje pektina je u jednom Engleskom članku 1750. godine o pripremi želea od jabuka. Francuski hemičar i farmaceut, Henri Braconnot je 1825. god. izolovao i opisao ovo jedinjenje. Kada govorimo o komercijalnoj proizvodnji, 1930-ih godina Hermann Herbstherith je pisao o mogućoj upotrebi jabukovih komina koje su se odbacivale kao nusproizvod u proizvodnji voćnog soka.
Naziv pektin potiče od grčke reći „pektos“, što znači čvrst i tvrd i ukazuje na sposobnost pektina da formira hidrogel.
Sadržaj pektina varira u zavisnosti od izvora biljnog materijala. Postoji nekoliko nusproizvoda poljoprivredne i prehrambene industrije koje imaju visok sadržaj pektina: komine od jabuka, šećerna repa, suncokret itd.
Pektin je jako bitan u ljudskoj ishrani jer predstavlja prebiotsko i rastvorljivo dijetetsko vlakno, deluje tako što smanjuje koncentracuju glukoze u krvi, koncentraciju LDL holesterola, imunostimulišuće i antikancerogeno.
Cilj ovog završnog rada je bilo ispitivanje antioksidativnih osobina kiselih i alkalih hidrolizata pektina iz kore i pulpe dunje.

AB  - U ovom radu analizirana je poliesterazna i lipazna aktivnosti mikroorganizama izolovanih iz zagađene životne sredine sa ciljem izolovanja sojeva koji se potencijalno mogu primeniti za biodegradaciju složenijih organskih polimernih molekula koji su danas prepoznati kao kontaminanti.
U tom cilju bilo je neophodno testirati 40 izolovanih sojeva mikroorganizama na rast na podlogama sa tributirinom, kokosovim uljem i polikaprolakton diolom kao jedinim izvorom ugljenika i utvrditi nakon dvonedeljne inkubacije koji mikroorganizam pokazuje pozitivan test na sva tri tipa podloge, kako bi se dalje primenili za dalja istraživanja u okviru biorazgradnje plastike.

AB  - BACKGROUND: Pectin is a complex polysaccharide, present in fresh and processed food as an additive, which also serves as an important source of fiber. In this work, the aim was to investigate in-vitro application of apple pectin as a possible antimicrobial, antioxidant and prebiotic agent for growth of bacteria selected from human milk and infant feces, as well as standard strains Lactobacillus rhamnosus and Bifidobacterium bifidum.
METHODS : All the bacteria isolated from the milk of mothers of preterm and term-born infants, bacteria from infant feces and standard strains were tested for fermentation of apple pectin as the main carbohydrate in selective broth media.
RESULTS: An increase in the growth of bacteria Bifidobacteria, Lactobacillus, and a consortium of bacteria selected from human milk and infant feces was noticed after pectin fermentation. It was found that pectin has a positive impact on the increase of the antioxidant capacity in all the tested samples. A high correlation was achieved, comparing electrochemical with spectrophotometric methods. Antimicrobial tests showed that pectin and fermentation products have slight or no influence at all on the growth of standard strains of Escherichia coli, Staphylococcus aureus and Candida albicans.
CONCLUSIONS: The results showed the importance of pectin as an antioxidant and its impact on the development of potential probiotic microbiota in mothers and infants. The study indicated a new potential application of pectin as a supplement in infant food.
