Novel derivatives of aroylacrylic acid phenylamides as inducers of apoptosis through the ROS-mediated pathway in several cancer cell lines

AB  - In the present work, the α,β-double bond of the aroylacrylic acid phenylamides was suitably modified to optimize the toxicity–antiproliferative activity ratio of the resulting compounds 1–5. The phenylamides were modified by Michael's addition of suitably chosen piperidine-containing fragments: 1-amino-N-benzylpiperidine (a1), 4-benzylpiperidine (a2), and N,N-dimethyl-N-[2-(1-piperazinyl)-ethyl]amine (a3). The compounds exerted micromolar activity toward three cancer cell lines, A549, LoVo, and Skov-3, causing apoptotic cell death. It was shown that the nature of the cyclic amine moiety at position C2 of the compounds is probably the primary determinant of anticancer activity toward tested cell lines and the acute toxicity toward brine shrimp (Artemia salina). The majority of compounds revealed the ability to vigorously induce mitochondrial superoxide anion generation in all treated cell lines, which together with cell cycle arrest at the S phase and activation of intrinsic caspase cascade, indicates the possibility that apoptosis was triggered due to irreparable chromosomal damage by acute oxidative stress. Two derivatives also exerted significant antibacterial activity with one order of magnitude higher potency than chloramphenicol in most of the investigated bacterial strains. Also, the drug-like properties for all compounds were estimated by available software tools.

AB  - Benefits and risks of the fish consumption should be assessed to balance the ingestion of healthy omega-3 fatty acids and adverse chemicals. Persistent organic pollutants-POPs (organochlorine pesticides-OCPs and polychlorinated biphenyls-PCB), macro- and micro-elements and fatty acid contents were determined in six fish species from the Adriatic Sea to assess health risks for consumers (worst-case scenario, diseases development risks and benefit-risk). 16 element, 24 POPs and 14 fatty acid contents were determined in six pelagic species which can be used in human diet. Element concentrations were determined by inductively coupled plasma mass spectrometry (ICP-MS), POPs by high-resolution gas chromatography and fatty acid content by gas–liquid chromatography. Diet based on chub mackerel and round sardinella showed lower daily intake (DI) of POPs and highest DI of essential omega-3 fatty acids than other investigated species. Lower ingestion of toxic elements can be observed by consuming anchovy and round sardinella. Based on POP concentrations, there was not observed non-carcinogenic (HI) nor carcinogenic (CR) risks for consumers. Based on element concentrations, there was low HI (0.1 ≥ HI ≥ 1), while the maximum HIs and outlier values (horse mackerel and anchovy samples) implied the presence of HI (HI > 1). The most significant contributor to total non-carcinogenic and carcinogenic risks was inorganic arsenic (inorganic As). Acceptable CR for consumers was assessed, but maximum CR for consumers of horse mackerel and anchovy (CR ≥ 1 × 10–6) showed adverse effects on human health. There were low HIs for developing cardiovascular, nervous, and reproductive diseases, and maximum HIs were higher than 1. Acceptable (1 × 10−4≥CR ≥ 1 × 10−6) risks were observed for developing cancer of nervous system and reproductive organs. Among investigated fish samples, those with higher ƩBR (benefit-risks) and BR for arsenic (As) than median value have a higher risk than benefits in the human diet.

Electrooxidation of 1-propanol on the mixture of nanoparticles of Pt and RuO2

It was found that the increase in the RuO2 content resulted in the increase in the catalytic activity, which reached its maximum value and then decreased. The catalytic effect was caused by the bifunctional mechanism of the catalyst Pt/RuO2. Ru-OH species were formed on Ru atoms of RuO2 nanocrystals at more negative potentials than on Pt. These oxy species oxidized firmly adsorbed intermediates propionyl, COad, C2Hyad, and CHad and, thus, released Pt atoms for the adsorption and dehydrogenation of the subsequent molecules of 1-propanol.

Elemental Composition of Pheochromocytoma Resolved on Solid/Adrenal Tissue and Whole Blood Level

AB  - Pheochromocytoma (PCC) is an unusual benign adrenal tumor with an unexamined status of the elements. This study delivers the first insight into the levels of microelements (Mn, Co, Cu, Zn, As, Se, Th), toxic elements (Cd, Pb, U), and macroelements (Na, K, Mg, Ca) in both adrenal tissue and whole blood samples collected from PCC patients. The results were strengthened by comparing recorded findings with the patients’ healthy adrenal tissue (HAT) and with whole blood samples from the healthy individuals. PCCs had significantly higher levels of Zn, Se, Na, K, and Mg and lower levels of Mn, Co, Pb, and As than that of HATs. Compared to healthy blood samples, the patients’ blood exhibited considerably higher levels of Na, K, and Ca but significantly lower levels of Mn, Cu, Zn, Se, and Mg. Females had significantly higher levels of essential Se and toxic Pb and Cd in their PCC tissue samples compared to males. PCC tissue levels of Mn and Cu were significantly elevated in smokers over levels in nonsmokers and in PCC patients with tumor sizes below 5 cm compared to PCC diameters above 5 cm. The data presented in this study provide a new insight into the pathophysiology of PCC. Thus, recorded elements should be considered as initiators/modifiers of PCC and potential inductors of malignant transformation. abstract = "Pheochromocytoma (PCC) is an unusual benign adrenal tumor with an unexamined status of the elements. This study delivers the first insight into the levels of microelements (Mn, Co, Cu, Zn, As, Se, Th), toxic elements (Cd, Pb, U), and macroelements (Na, K, Mg, Ca) in both adrenal tissue and whole blood samples collected from PCC patients. The results were strengthened by comparing recorded findings with the patients’ healthy adrenal tissue (HAT) and with whole blood samples from the healthy individuals. PCCs had significantly higher levels of Zn, Se, Na, K, and Mg and lower levels of Mn, Co, Pb, and As than that of HATs. Compared to healthy blood samples, the patients’ blood exhibited considerably higher levels of Na, K, and Ca but significantly lower levels of Mn, Cu, Zn, Se, and Mg. Females had significantly higher levels of essential Se and toxic Pb and Cd in their PCC tissue samples compared to males. PCC tissue levels of Mn and Cu were significantly elevated in smokers over levels in nonsmokers and in PCC patients with tumor sizes below 5 cm compared to PCC diameters above 5 cm. The data presented in this study provide a new insight into the pathophysiology of PCC. Thus, recorded elements should be considered as initiators/modifiers of PCC and potential inductors of malignant transformation. Recovery tests indicate excellent accuracy and precision of the method, concluding that the biosensor can offer a fast, accurate, reliable and precise determination of the polyphenolic index. More importantly, our results suggest a great potential for the application in real samples.

AB  - A catalytic coating, composed of a mixture of nanocrystals of Pt and RuO2, used for oxidation of CH3OH, was formed by the thermal procedure. The size of the RuO2 nanocrystals was increasing and of Pt was decreasing with increasing the content of RuO2. The optimal coating composition depended on potential. At more positive potentials, the optimal coatings contained lower amounts of RuO2. The oxidation reaction of CH3OH on the coatings with the RuO2 content higher than optimal, was determined by dehydrogenization of CH3OH. At lower amounts of RuO2, oxidation of CH3OH was determined by the oxidation reaction of intermediates COad with oxy species of ruthenium. The catalytic effect was caused by a bifunctional mechanism. The bifunctional mechanism is based on the fact that oxy species were formed on Ru at more negative potentials than on Pt. These oxy species oxidized COad intermediates, bound to adjacent Pt atoms and thus discharged them for dehydrogenation of the subsequent CH3OH molecules.

AB  - Background Mammalian meat is the most common trigger of the allergic reactions in patients with α-Gal syndrome (AGS). Milk and dairy, although less often, also cause a significant number of allergic manifestations. The aim of this study was to identify α-Gal-containing bovine milk proteins with allergenic properties among AGS patients. Methods Thirty-eight AGS patients with IgE to milk were included in the study. Milk proteins were analyzed for the presence of α-Gal and for binding by patients’ IgE using immunoblot, ImmunoCAP, and inhibition ELISA. Allergenicity of milk and milk proteins was assessed by basophil activation test. Results More than half of the AGS patients reported allergic reactions to milk or dairy products. Bovine γ-globulin (BGG), lactoferrin (LF), and lactoperoxidase (LPO) were identified as α-Gal carrying proteins which were recognized by AGS patients’ IgE. Whey mirrored the anti-α-Gal and IgE reactivity of BGG, LF, and LPO. Eighty-nine percent of the patients displayed IgE to BGG, 91% to LF, and 57% to LPO. Inhibition of α-Gal-specific IgE binding was achieved by BGG, LF, LPO, and whey. These proteins also activated AGS patients’ basophils. Interestingly, at lower concentrations, LF was the most potent inhibitor of IgE binding, and the most potent activator of basophils. Conclusion BGG, LF, and LPO were all found to be relevant milk α-Gal-containing glycoproteins that bound AGS patients’ IgE antibodies and activated their basophils. These proteins are probably involved in the allergic reactions to milk in AGS patients. LPO was for the first time shown to be an allergen.

AB  - Solid-state reaction between BaTiO3and Fe2O3was used to produce a multiferroic heterostructure composite. Commercial BaTiO3and Fe(NO3)3•9H2O were suspended in ethanol for 30 minutes in an ultrasound bath. The prepared mixture was thermally processed at 300°C for 6 h. Sintering at 1300 °C for 1 h resulted in a mixture of different phases, BaTiO3, BaFe12O19and Ba12Ti28Fe15O84, which were confirmed by x-ray powder diffraction. A dense microstructure with a small volume fraction of closed porosity was indicated by the scanning electron microscopy, while a homogeneous distribution of Fe ions over BaTiO3phase was visible from energy dispersive spectroscopy mapping. Doping of BaTiO3with Fe2O3resulted in formation of magnetic hexaferrite phases, as confirmed by dielectric measurements that showed a broadened maximum of the permittivity measured as a function of temperature. © 2021 Authors. AB  - Dithiocarbamates, with their high lipophilic character and good chelating properties, could provide stabile transition metal complexes and enable these metal-based drugs to reach their biological targets. Palladium(II) and gold(III) complexes, due to its structural similarity with platinum(II)compounds, could be competitive candidates for implementing and developing new pharmacological agents. Herein, novel gold(III) complex with triammonium N-carboxyiminodiacetate as ligand has been synthesized and characterized by elemental analysis, molar conductivity measurements, FT-IR and 1H and 13C NMR spectroscopy. The proposed structure was examined and compared with analogue palladium(II) and zinc(II) complexes by density functional theory (B3LYP/def2tzvp). Additionally, DNA-binding studies by UV/Vis-spectrometer as well as ethidium-bromide (EB) and bovine serum albumin (BSA) quenching studies by spectrofluorometer were performed for gold(III), palladium(II) and zinc(II) dithiocarbamate complexes. The investigated complexes showed a good affinity to calf thymus DNA (CT-DNA) and BSA, with a higher affinity to BSA. Palladium(II) complex exhibited 5-fold a stronger affinity to DNA binding in comparison to zinc(II), but no significantly higher than gold(III) complex. Furthermore, palladium(II) and gold(III) complexes demonstrated a similar affinity toward BSA and these interactions are stronger than showed by zinc(II) complex.

AB  - Metabolism of sulfur (sulfur assimilation pathway, SAP) is one of the key pathways for the pathogenesis and survival of persistant bacteria, such as Mycobacterium tuberculosis (Mtb), in the latent period. Adenosine 5'-phosphosulfate reductase (APSR) is an important enzyme involved in the SAP, absent from the human body, so it might represent a valid target for development of new antituberculosis drugs. This work aimed to develop 3D-QSAR model based on the crystal structure of APSR from Pseudomonas aeruginosa, which shows high degree of homology with APSR from Mtb, in complex with its substrate, adenosine 5'-phosphosulfate (APS). 3D-QSAR model was built from a set of 16 nucleotide analogues of APS using alignment-independent descriptors derived from molecular interaction fields (MIF). The model improves the understanding of the key characteristics of molecules necessary for the interaction with target, and enables the rational design of novel small molecule inhibitors of Mtb APSR.

AB  - Ugalj je čvrsto gorivo i veoma je značajan za energetiku, metalurgiju i hemijsku
industriju. Raznolikost ugljeva predstavlja veliki izazov prilikom proučavanja geohemije
ugljeva, kao i procesa koji utiču na hemijski sastav uglja. Određivanje sastava biomarkera u
uzorcima uglja ima veliki značaj u ispitivanju geohemije uglja. Cilj rada je bio da se
primenom savremenih organsko-geohemijskih metoda, utvrdi poreklo i geološka evolucija
uglja (donji eocen) iz Mdeker (MD) regiona u Tunisu. Ispitivano je 5 uzoraka. Nakon
izolovanja rastvorne organske supstance (bitumena) ekstrakcijom po Soxlet-u i taloženja
asfaltena, malteni su primenom hromatografije na mikrokoloni razdvojeni na frakcije
alifatičnih, aromatičnih ugljovodonika i polarnih jedinjenja sa azotom, sumporom i
kiseonikom (NSO-jedinjenja). Alifatična i aromatična frakcija su analizirane
gasnohromatografsko-masenospektrometrijskom tehnikom (GC-MS). U cilju utvrđivanja
porekla i geološke istorije uglja, upotrebljena su biomarkeri alifatične frakcije i specifična
aromatična jedinjenja.

AB  - Poznato je da je elektrostatički potencijal na površini molekula jedan od ključnih faktora za predviđanje osetljivosti eksploziva i da veća pozitivnost potencijala rezultuje osetljivijim eksplozivom. U ovoj studiji su ispitani nitro derivati benzena, naftalena, antracena i tetracena primenom B3LYP metode sa CC-PVDZ baznim setom. Na osnovu dobijenih mapa elektrostatičkog potencijala, utvrđeno je da povećanje broja prstenova u molekulu ima uticaj na smanjenje pozitivnosti potencijala u centru molekula. Takođe, pokazano je da raspored i blizina nitro grupa u molekulu imaju uticaj na vrednosti potencijala u kritičnim tačkama. Ispitan je i uticaj hloro i formil grupe na elektrostatički potencijal u centru nitro derivata antracena i tetracena, pri čemu su dobijene pozitivnije vrednosti potencijala. Ovi rezultati mogu poslužiti za dizajniranje novih, manje osetljivih visokoenergetskih materijala.

AB  - In this manuscript, samples of Kupa River sediments were examined using three different extraction agents. The aim of this study was to evaluate the applicability of single extraction procedures to investigate the bioavailability and mobility of major and trace elements (Al, As, Ba, Cd, Co, Cr, Cu, Fe, K, Li, Mg, Mn, Na, Ni, P, Pb, S, Si, Sr, Ti, V, and Zn) from river sediment. Two forms of studied elements were evaluated: mobile, the most toxic element form (extraction with 1 M CH3COONH4 and 0.01 M CaCl2) and potentially mobilized form (2 M HNO3 extraction). The estimation of the ecological risk, with the application of the probability distribution of RI (potential ecological risk index) values, is yielded with the help of the Monte Carlo simulation (MCS). Ammonium acetate is proved to be a better extraction agent than calcium chloride. A positive correlation between the content of all extracted elements with nitric acid and the total element content indicates that 2 M HNO3 efficiently extracts all studied elements. Results showed anthropogenic sources of cadmium and copper and high barium mobility. The MCS suggests that risk of Cr, Cu, Ni, Pb, and Zn was low; As and Cd posed a lower and median ecological risk in the studied areas.

AB  - Papain is a proteolytic enzyme of great commercial value. It is a cysteine protease highly expressed in Carica papaya fruit latex, but also present in papaya leaves. Purification procedures mostly deal with the latex and include a combination of precipitation and/or chromatographic techniques. Due to its solubility, structure and activity characteristics, the pH and salt content play significant roles in handling papain extracts. Here we report a simple, rapid and easily scalable procedure for papain purification from papaya leaves, which contain different contaminants as compared to papaya latex. Sodium chloride precipitation of contaminants at pH 5 followed by ammonium sulphate precipitation resulted in the removal of other leaf proteins and protein fragments from papain solution and about a 3-fold purification. The procedure also benefits from the suppression of autoproteolysis and preservation of the native structure, as confirmed by FTIR analysis, and the high recovery of activity of over 80%.

Several structural motifs featuring mono-, bi-, and pentadentate binding modes are discussed in a combination with a variety of different substituents with the pyridine unit. In this context, the current review underlines a brief summary of advances on metal complexes with pyridine-based ligands aimed for further development as metallodrug. In line with this, we also highlight their key properties, different synthetic strategies employed in the preparation, and possible structure–property correlations, indicating advantages and limitations of the applied methods used within documented studies.

AB  - This research proposed two pretreatments of viscose fabrics: oxidation with 2,2,6,6-tetramethylpiperidine-1-oxy radical (TEMPO) and coating with TEMPO-oxidized cellulose nanofibrils (TOCN), to introduce functional groups (COOH and CHO) suitable for irreversible binding of chitosan nanoparticles without and with embedded zinc (NCS and NCS + Zn, respectively) and consequently achieving washing durable antibacterial properties of the chitosan nanoparticles functionalized fabrics. The characterizations of pretreated and chitosan nanoparticles functionalized fabrics were performed by FTIR and XPS spectroscopy, elemental analysis, inductively coupled plasma optical emission spectrometry, zeta potential measurements, scanning electron microscopy, determination of COOH and CHO groups content, and antimicrobial activity under dynamic contact conditions. Influence of pretreatments on NCS and NCS + Zn adsorption, chemical, electrokinetic, and antibacterial properties as well as morphology, and washing durability of NCS and NCS + Zn functionalized fabrics were studied and compared. Washing durability was evaluated through changes in the chitosan and zinc content, zeta potential, and antibacterial activity after 1, 3, and 5 washing cycles. Pretreatments improved washing durability of antibacterial properties of chitosan nanoparticles functionalized fabrics. The NCS and NCS + Zn functionalized pretreated fabrics preserved antibacterial activity against S. aureus after five washing cycles, while antibacterial activity against E. coli was preserved only after one washing cycle in the case NCS + Zn functionalized pretreated viscose fabrics. © 2021 by the authors.
