Uspostavljanje metode za dokazivanje prisustva mutacije P28T u genu GALK1 kod Roma sa teritorije Srbije

AB  - Tema ovog rada biće uspostavljanje metode za dokazivanje prisustva mutacije P28T u genu GALK1 kod Roma sa teritorije Srbije. U uvodu će prvo biti reči o istorijatu romske populacije i zastupljenosti genetski nasledivih bolesti u romskoj populaciji. Potom će biti reči o opštim karakteristikama galaktozemije, čiji je osnovni uzrok mutacija P28T. U glavnom delu rada biće opisane metode pomoću kojih je izolovana DNK iz brisa bukalne sluznice ispitivanih osoba, zatim umnožen željeni fragment GALK1 gena i restrikcionom digestijom ispitano da li postoji mutacija u datom genu. U sledećem delu će biti prikazani rezultati. Naposletku, biće reči o verovatnoći nasleđivanja galaktozemije i tretiranju oboljenja.

AB  - Od decembra 2019. godine i samog početka pandemije SARS-CoV-2 pa do danas, nije pronađen efikasan lek protiv kovida-19 izazvanog ovim virusom. U nedostatku vremena, pribegava se analizi lekova sa antivirusnim svojstvima koji su već u upotrebi, a koriste se u terapiji drugih oboljenja. U ovom završnom radu, kao okosnica istraživanja korišćen je hidroksihlorokin, dizajnirana su nova jedinjenja sa idejom retencije imunosupresorskih svojstva hidroksihlorokina, dok bi izmene imale za svrhu poboljšanje potencijalnih antivirusnih svojstava. Afinitet glavne proteaze koronavirusa prema dizajniranim jedinjenjima ispitan je in silico, molekulskim dokingom, u svrhu nalaženja bioizostera hidroksihlorokina sa strukturnim i hemijskim svojstvima, koji bi inhibicijom aktivnog mesta glavne proteaze koronavirusa mogli da zaustave infekciju u ljudskom organizmu.

AB  - Predmet ove disertacije bio je proučavanje uticaja herbicida sa ili bez primene folijarnog đubriva na sadržaj fitohemikalija u listu i zrnu četiri hibrida kukuruza šećerca (ZP504su, ZP355su, ZP553su i ZP515su) tokom dve godine, u cilju definisanja njihove tolerantnosti i karakterističnog odgovora. Sadržaj fitohemikalija određen je primenom visoko-efikasne tečne hromatografije sa ultravioletnim detektorom sa više dioda i fluorescentnim detektorom i visoko-efikasne jonske hromatografije sa pulsno-amperometrijskim detektorom. Sadržaj ukupnih fenolnih jedinjenja, glutationa i antioksidativna aktivnost određeni su primenom UV/VIS spektrofotometrije. Hromatografska razdvajanja su postignuta pri nižim protocima mobilnih faza uz smanjen utrošak organskih rastvarača podržavajući na taj način trendove zelene analitičke hemije. Dobra linearnost, tačnost, niska granica detekcije, direktno HPLC određivanje ekstrakata bez derivatizacije, skraćeno vreme ekstrakcije i trajanja analize omogućili su rutinsko korišćenje primenjenih hromatografskih metoda. Rezultati su pokazali da je uticaj tretmana bio izraženiji u zrnu, dok u listu uglavnom nije bio statistički značajan. Primena oba herbicida dovela je do statistički značajnog povećanja ukupnog sadržaja svih fitohemikalija u ZP553su na osnovu čega se on može definisati kao tolerantan genotip. Primenjeni tretmani izazvali su aktiviranje neenzimskih antioksidanasa kod ZP355su, a kod ZP515su aktiviranje metabolizma ugljenih hidrata. Rezultati su pokazali da se sadržaj karotenoida, vitamina E, fenolnih kiselina i šećera može koristiti kao jedinstven pokazatelj genotipa nakon primenjenih tretmana. Pokazano je da kontrolisana upotreba herbicida može dovesti do poboljšanja fitohemijskog profila i povećanja nutritivnog statusa kukuruza šećerca.
AB  - The subject of this doctoral dissertation was the impact of herbicides with or without foliar fertilizer on phytochemicals content in leaf and grain in four hybrids of sweet maize (ZP504su, ZP355su, ZP553su, and ZP515su) during two years in order to define their tolerance and characteristic response. The content of phytochemicals was determined using high-performance liquid chromatography with an ultraviolet multi-diode detector and a fluorescent detector, and high-performance anion exchange chromatography with pulsed amperometric detection. The content of total phenolic compounds, glutathione and antioxidant activity was determined using UV / VIS spectrophotometry. Chromatographic separations were achieved at lower mobile phase flows with reduced consumption of organic solvents, thus supporting the trends of green analytical chemistry. Good linearity, accuracy, low limit of detection, direct HPLC determination of extracts without derivatization, shortened extraction time, and duration of analysis enabled the routine use of applied chromatographic methods. The results showed that the impact of the treatment was more conspicuous in the grain, while in the leaf it was generally not statistically significant. The application of both herbicides led to a statistically significant increase in the total content of all phytochemicals in ZP553su, based on which it can be defined as a tolerant genotype. The applied treatments caused the activation of non-enzymatic antioxidants in ZP355su, and in ZP515su the activation of carbohydrate metabolism. The results of this study indicated that the content of carotenoids, vitamin E, phenolic acids and sugars can be used as a characteristic phytochemical parameter of the genotype after the applied treatments. It has been shown that the controlled use of herbicides can lead to an improvement in the phytochemical profile and an increase in the nutritional status of sweet maize.

AB  - The citric acid (CA), dextran (DEX) and (3-aminopropyl)triethoxysilane (APTES) are frequently used ligands for surface modification of magnetite nanoparticles to improve their colloidal stability and biocompatibility for use in biomedicine. Here we report the synthesis, surface modification of magnetite (Fe3O4) nanoparticles and analysis of the influence of coating materials on heating efficiency in magnetic hyperthermia, as well as on electrochemical sensing toward the detection of neurotransmitter dopamine (DOP). Spherical magnetite nanoparticles (~13 nm) were prepared by co-precipitation and surface-modified with CA, APTES and DEX. The hyperthermia tests revealed that Fe3O4@APTES resulted in the highest specific absorption rate (SAR) of 67.2 W/g. The engineered sensor, by surface modification of screen-printed carbon (SPC) electrodes with Fe3O4@APTES, showed linear response with DOP in the concentration range of 1−200 μM and a low detection limit of 34.3 nM. Besides, the sensor offered remarkable selectivity, repeatability and reproducibility. Application of SPC-, Fe3O4-, Fe3O4@CA- and Fe3O4@DEX-modified SPC electrode resulted in lower electrocatalytic activity compared with Fe3O4@APTES/SPC. The obtained results indicate the effects of a surface modifier especially that Fe3O4@APTES, alone and combining with SPC electrodes, can serve as an agent in various types of biomedical applications, like hyperthermia and electrochemical sensing of biologically active compounds such as DOP.

BanLec-eGFP Chimera as a Tool for Evaluation of Lectin Binding to High-Mannose Glycans on Microorganisms

BanLec-eGFP, produced as a soluble cytosolic protein of about 42 kDa, revealed β-sheets (41%) as the predominant secondary structures, with the emission peak maximum detected at 509 nm (excitation wavelength 488 nm). More than 65% of the primary structure was confirmed by mass spectrometry. Competitive BanLec-eGFP binding to high mannose glycans of the influenza vaccine (Vaxigrip®) was shown in a fluorescence-linked lectin sorbent assay (FLLSA) with monosaccharides (mannose and glucose) and wild type BanLec and H84T BanLec mutant. BanLec-eGFP exhibited binding to mannose residues on different strains of Salmonella in flow cytometry, with especially pronounced binding to a Salmonella Typhi clinical isolate. BanLec-eGFP can be a useful tool for screening high-mannose glycosylation sites on different microorganisms.

AB  - Synthesis of novel aminoquinoline derivatives has been accomplished and their activity against malaria strains has been examined. The compounds showed moderate in vitro antimalarial activity against two P. falciparum strains, 3D7 (CQ susceptible clone) and Dd2 (CQ resistant clone). Three aminoquinolines were further examined for antimalarial efficacy in a mouse model using a modified Thompson test. In this model, mice were infected with P. berghei-infected red blood cells, and drugs were administered orally. Antimalarial 3 was found toxic at a dose of 320 (mg/kg)/day in 3/6 mice, however, 2/6 mice of the same group survived through day 31, and one of them was cured. © 2021 Serbian Chemical Society. AB  - Derivatives of thiocarbohydrazone studied so far have shown great biological activity such as antioxidant, antimicrobial, and anticancer. Most of these compounds are bis-substituted derivatives, while monothiocarbohydrazones are much less investigated. Еighteen monothiocarbohydrazones were synthesized and subjected to physicochemical characterization in order to facilitate the examination of their potential biological activity and application in future studies. The structure of synthesized derivatives was confirmed with NMR and FT–IR spectroscopy, and with elemental analysis. For one of the compounds, single-crystal X-ray diffraction analysis was performed. Specific and non-specific molecular interactions were interpreted by LSER principles, using Catalan’s model. For additional information about the dominance and influence of the interactions presented, correlations with Hansen’s solubility parameters were calculated. Influence of the type and position of the substituent on absorption maxima was determined with LFER (linear free-energy relationship) principles, using Hammett’s equation. Acidity constants of the synthesized compounds were theoretically calculated and experimentally determined. Moreover, the excitation of a molecule by a photon of UV–Vis light was interpreted by time-dependent density functional theory (TD–DFT) calculations of UV absorption bands, and intramolecular charge transfer (ICT) was quantified by calculations of the charge transfer distances (DCT). © 2021, The Author(s), under exclusive licence to Springer Science+Business Media, LLC part of Springer Nature.

AB  - Sinteza fluorescentnih derivata estradiola i estrona je urađena na 8 načina. Dobijeni derivati
su karakterisani ispitivanjem interakcija sa antitelima za estrogene uz praćenje
fluorescentnom polarizacijom. Estrogeni su grupa ženskih polnih hormona veoma bitnih za
praćenje fizioloških stanja kod jedinki ovog pola. Pored toga ova grupa hormona je od
interesa kada je u pitanju detekcija oplodnje i praćenje toka trudnoće. Fluorescentna
polarizacija kao veoma osetljiva, jednostavna, nedestruktivna, jeftinametoda je veoma
pogodna za veliki broj analiza koje je neophodno obaviti u kratkom roku (npr. kod praćenja
oplodnje na velikim farmama životinja). Takođe pogodnost ovog načina analize je i jeftin
material i hemikalije koje su neophodne za sintezu fluorescentnih derivate od interesa. Način
izvođenja analiza i i kompleti neophodnih reagenasa i opreme su prilagođeni i terenskoj
upotrebi. Pored toga, lako rukovanje uređajem i jednostavnost izvođenja analize
omogućavaju licima koja nisu eksperti u ovoj oblasti da sama izvode analize.

AB  - Rastvorljivost se izražava kao koncentracija supstance u određenoj zapremini rastvarača na određenoj temperaturi i predstavlja važan fizičko-hemijski parametar u ranoj fazi dizajna i razvoja lekova. Nortriptilin-hidrohlorid spada u grupu tricikličnih antidepresiva koji se koriste u lečenju depresije. pH-Zavisna rastvorljivost nortriptilin-hidrohlorida proučavana je primenom pH-Ramp shake-flask metode. Određivanje rastvorljivosti je od velikog značaja, budući da je u digestivnom traktu zastupljen širok opseg pH vrednosti (od jako kisele sredine u želucu do slabo bazne u crevima), pa samim tim visoka rastvorljivost jonizovanog (kiselog ili baznog) oblika supstance omogućava bolju apsorpciju iz digestivnog trakta u krvotok i veću biodostupnost na mestu dejstva. Primenom softverskog paketa pDISOL-X (in-ADME Research, New York, USA) određene su ravnoteže u heterogenom sistemu koji čini nortriptilin-hidrohlorid i fosfatni pufer.

AB  - Imipramin-hidrohlorid (ImiHCl) i amitriptilin-hidrohlorid (AmiHCl) su površinski aktivni, u vodenoj sredini slabo rastvorni lekovi iz grupe tricikličnih antidepresiva, koji mogu formirati submicelarne agregate u slabo kiseloj sredini. Zbog prisustva bočnog lanca alkilamina ovi molekuli se ponašaju nalik surfaktantima. Ukoliko je koncentracija supstance u rastvoru iznad određene koncentracije može doći do formiranja agregata (tzv. micela). Ta granična koncentracija iznad koje dolazi do formiranja micela naziva se kritična micelarna koncentracija (kmk). U ovom završnom radu korišćene su konduktometrijske titracije za određivanje kmk ImiHCl i AmiHCl. Specifična provodljivost, merena je na konstantnoj temperaturi od 25±1 °C, u rastvorima NaH2PO4 i NaCl. Takođe, određena je rastvorljivost ImiHCl i AmiHCl u rastvorima sa i bez pufera, u prisustvu fiziološki relevantne koncentracije hlorida, primenom pH-Ramp Shake-Flask metode. Rastvorljivost je određena titracijama polazeći od bazne ka kiseloj sredini, i obrnuto. pH Vrednost je podešena korišćenjem HCl, H3PO4 i/ili NaOH u zasićenim rastvorima fosfatnog pufera. Nakon uspostavljanja stabilnog ravnotežnog stanja (nakon mešanja od 6 h i 18 h sedimentacije), centrifugiranje je korišćeno za odvajanje faza. Koncentracija imipramina i amitriptilina je izmerena pomoću HPLC sa UV/VIS detekcijom.

AB  - Pripremljeni su molekulski obeleženi polimeri na bazi β-ciklodekstrina kod kojih je teofilin služio kao templat, primenom dva sintetička puta. Jedan je podrazumevao upotrebu β ciklodekstrina kao monomera pri čemu je dobijen monolitni polimer, dok je kod drugog korišćen monomer nastao modifikacijom β-ciklodekstrina metakrilnom kiselinom i nastali polimer je bio u obliku sitnih čestica. Pripremljeni su i polimeri obeleženi donepezilom, upotrebom metakrilne kiseline kao funkcionalnog monomera i divinilbenzena i etilenglikol-dimetakrilata kao umreživača. Prepolimerizaciona smeša je ispitana konduktometrijski. Dobijeni polimeri su okarakterisani pomoću FTIR spektroskopije, skenirajuće elektronske mikroskopije (SEM) i adsorpcionih izotermi. Na osnovu ponovnog vezivanja templata za dobijene obeležene i neobeležene polimere, utvrđeno je da je obeležavanje uspešno izvedeno. Jedan od obeleženih polimera je odabran na osnovu prethodnih analiza i testiran u uslovima sličnim fiziološkim kako bi se dobile informacije koliko dugo i koje količine aktivnih farmaceutskih supstanci (templata/donepezila) otpušta.

AB  - Ispitivane su interakcije poznatih lekova (indapamid, olanzapin i atorvastatin) sa strukturnim analozima serum albumina korišćenjem fluorescentne spektroskopije, UV/Vis spektrofotometrije i ravnotežne dijalize. Pokazane su razlike u vezivanjima ove tri supstance: indapamid se ne vezuje za serum albumin, a olanzapin i atorvastatin se vezuju ali sa razlikama u vezivnim mestima. Ove razlike u vezivanju se pripisuju strukturnim razlikama supstanci jer indapamid ima sulfonamidnu grupu, olanzapin tiofensko, a atorvastatin pirolno jezgro. Promene u fluorescenciji serum albumina praćene su na tri temperature (20, 25 i 37 °C) i analizirane su Stern-Volmerovom jednačinom koja je pokazala statički mehanizam vezivanja. Ravnotežnom dijalizom su potvrđene konstante vezivanja olanzapina i atorvastatina za serum albumin.

AB  - Plastične čestice sa prečnikom manjim od nekoliko 1mm predstavljaju ozbiljan problem za životnu sredinu, sa štetnim uticajem na sve žive organizme. Iako je štetnost ovih čestica dokazana, do sada nisu u potpunosti ispitani i poznati uticaji mikroplastike na vezivanje različitih supstanci kao što su proteini, teški metali, PAHovi, kao i ostale zagađujuće supstance.

AB  - Because it is important to develop new sustainable sources of edible protein, insects have been recommended as a new protein source. This study applied Life Cycle Assessment (LCA) to investigate the environmental impact of small-scale edible insect production unit in South Korea. IMPACT 2002 + was applied as the baseline impact assessment (IA) methodology. The CML-IA baseline, EDIP 2003, EDP 2013, ILCD 2011 Midpoint, and ReCiPe midpoint IA methodologies were also used for LCIA methodology sensitivity analysis. The protein, fat contents, and fatty acid profile of the investigated insect (Protaetia brevitarsis seulensis larvae) were analyzed to determine its potential food application. The results revealed that the studied edible insect production system has beneficial environmental effects on various impact categories (ICs), i.e., land occupation, mineral extraction, aquatic and terrestrial ecotoxicity, due to utilization of bio-waste to feed insects. This food production system can mitigate the negative environmental effects of those ICs, but has negative environmental impact on some other ICs such as global warming potential. By managing the consumption of various inputs, edible insects can become an environmentally efficient food production system for human nutrition.

AB  - Etarska ulja predstavljaju prirodne komponente aromatičnih biljaka. Zahvaljujući lako isparljivim jedinjenjima koja ulaze u njihov sastav, ona doprinose specifičnom mirisu i ukusu kod ove vrste biljaka. Glavna uloga ulja je ipak u odbrani bilja od raznih štetočina. Većina od njih poseduje terapeutska svojstva, a mnoga i antimikrobno dejstvo. Izoluju se na razne načine, ali najčešće je to postupak destilacije vodenom parom.
Cilj ovog rada je da se testira antimikrobna aktivnost 10 različitih vrsta etarskih ulja (Nane, Bosiljka, Žalfije, Lavande, Morača, Miloduha, Mirodjije, Smilja, Anisa i Divljeg origana) na 5 sojeva mikroorganizama: E. coli, B. subtilis, C. albicans, S. cerevisiae i A. niger. U drugom delu rada, ponovljen je test antimikrobne aktivnosti na spomenute sojeve mikroorganizama, ali prethodno su ulja tretirana ozonom. Rezultati su pokazali da je na neka ulja ozon imao stimulativno dejstvo i da je njihova aktivnost samim tim dosta poboljšana na inhibiciju rasta mikroorganizama.

AB  - Serological testing is important method for diagnosis of severe acute respiratory syndrome coronavirus 2 (SARS-CoV-2) infection. Nucleocapsid (N) protein is the most abundant virus derived protein and strong immunogen. We aimed to find its efficient, low-cost production.

SARS-CoV-2 recombinant fragment of nucleocapsid protein (rfNP; 58–419 aa) was expressed in E. coli in soluble form, purified and characterized biochemically and immunologically.

Purified rfNP has secondary structure of full-length recombinant N protein, with high percentage of disordered structure (34.2%) and of β-sheet (40.7%). rfNP was tested in immunoblot using sera of COVID-19 convalescent patients. ELISA was optimized with sera of RT-PCR confirmed positive symptomatic patients and healthy individuals. IgG detection sensitivity was 96% (47/50) and specificity 97% (67/68), while IgM detection was slightly lower (94% and 96.5%, respectively).

Cost-effective approach for soluble recombinant N protein fragment production was developed, with reliable IgG and IgM antibodies detection of SARS-CoV-2 infection.

abstract = "Serological testing is important method for diagnosis of severe acute respiratory syndrome coronavirus 2 (SARS-CoV-2) infection. Nucleocapsid (N) protein is the most abundant virus derived protein and strong immunogen. We aimed to find its efficient, low-cost production.SARS-CoV-2 recombinant fragment of nucleocapsid protein (rfNP; 58–419 aa) was expressed in E. coli in soluble form, purified and characterized biochemically and immunologically.Purified rfNP has secondary structure of full-length recombinant N protein, with high percentage of disordered structure (34.2%) and of β-sheet (40.7%). rfNP was tested in immunoblot using sera of COVID-19 convalescent patients. ELISA was optimized with sera of RT-PCR confirmed positive symptomatic patients and healthy individuals. IgG detection sensitivity was 96% (47/50) and specificity 97% (67/68), while IgM detection was slightly lower (94% and 96.5%, respectively).Cost-effective approach for soluble recombinant N protein fragment production was developed, with reliable IgG and IgM antibodies detection of SARS-CoV-2 infection.",

AB  - Demand for camel milk (CM) is increasing worldwide, due to its high nutritious value and health benefits. In this study, whole CM powders were produced by spray drying (SD) at six inlet temperatures (190 °C–250 °C) and by freeze drying (FD). Physicochemical and functional properties of CM powder proteins were investigated. SD at higher inlet temperatures (230 °C–250 °C) resulted in higher extent of Maillard reaction (MR), in comparison to lower temperatures (190 °C–200 °C) and FD treatment. Both treatments had negative effect on casein solubility, while whey proteins remained soluble and slightly increased its solubility with the extent of MR. The CM powders obtained at higher inlet temperatures demonstrated improved antioxidant activity. Secondary structure of whey proteins did not differ among the samples, while surface hydrophobicity of whey proteins was higher in all SD than in FD samples, suggesting only limited denaturation of camel whey proteins at higher inlet temperatures of drying. Thus, the effects of SD under the conditions applied in our study did not decrease camel whey protein solubility, while drying procedure itself regardless of temperature decreased solubility of camel milk caseins. MR generated during CM processing could be an important means of compensating for the lack of antioxidant protection normally associated with β-lactoglobulin but happens to be absent from this milk.
