AB  - Lipophilicity and ionization constant of sixteen fentanyl analogues were investigated by reversed-phase thin-layer chromatography (RP-TLC). Fourteen compounds have nitrogen containing groups at C-3 position of thepiperidine ring and two compounds are 3-carbomethoxy derivatives of fentanyl. Among them, five are diaste-reomeric cis/trans couples and six novel trans diastereomers. In addition, the lipophilicity and ionization constantof fentanyl were also determined by the same method, as a reference. The physicochemical property, lip-ophilicity, expressed as retention indices RM0 , b, and C0, as well as PC1, was determined and correlated with insilico values. Ionization constants were determined on the basis of the relationships between analyte’s retentionexpressed as RF and mobile phase pH. Calculated structural descriptors together with the retention indices, weresubjected to the principal component analysis – PCA and hierarchical cluster analysis – HCA, in order to providebasic insights into the similarities among the studied compounds. The blood – brain barrier (BBB) permeationwas investigated in the function of experimentally obtained values of lipophilicity, polar surface area and mo-lecular weight. In general, results of the present research corroborate well with previously determined anti-nociceptive activities of the investigated compounds, pointing out that besides the cis/trans isomerism, anotherset of important parameters should be taken into account when designing new derivatives of C-3 substitutedfentanyl, especially lipophilicity, voluminosity and HBD/A character of the substituents. Accordingly, RP-TLCcan be considered as a valuable asset in the ligand-based drug design

AB  - Considering the health-benefits of berry fruits consumption and increased market demands for food authenticityas one of the most important quality assurances, phenolic profiling by high-performance thin layer chroma-tography and ultra-high-performance liquid chromatography hyphenated with mass spectrometry was combinedwith multivariate analysis for phytochemical characterization and intercultivar discrimination of cultivatedberry seeds. The phenolic profiles of 45 berry seeds from nine genuine Serbian cultivated fruit species (straw-berry, raspberry, blackberry, black currant, blueberry, gooseberry, cape gooseberry, chokeberry, and goji berry)revealed a good differentiation according to botanical origin. In order to determine biomarkers responsible forthe classification, a total of 103 phenolic compounds were identified, including 53 phenolic acids and theirderivatives, 26 flavonoids and 24 glycosides. Biomarkers derived from the phenolic profile of berry seeds provedto be a powerful tool in the authentication of botanical origin, and may be useful in detection of frauds in berry-based seed-containing product.

AB  - Gamma irradiation provides an alternative pathway to conventional gold nanoparticle synthesis because it is simple, fast, and economical. Here, we employed gamma irradiation at low doses (1–20 kGy) to obtain gold nanoparticles (Au NPs) anchored onto graphene oxide (GO) sheets. GO was selected as a suitable platform for the nucleation and growth of Au NPs because of its large surface area and good dispersibility in water due to the presence of polar oxygen-containing functional groups in its structure. Gamma irradiation at all the applied doses led to the reduction of chloroauric acid and the formation of evenly distributed Au NPs at the GO surface, simultaneously causing the reduction of GO and partial restoration of the graphene structure. As-prepared Au NPs have predominately spheric shapes and the smallest nanoparticles were reported for the dose of 1 kGy. The increase in the irradiation dose caused either the growth of larger particles (5 and 10 kGy) or the broad distribution of particles’ sizes (20 kGy). All samples showed a temperature increase upon exposure to 800 nm laser and photothermal efficiency was the highest for the sample prepared at 20 kGy.

AB  - Most of the food allergens sensitized via the gastrointestinal tract resist thermal treatments and digestion, particularly digestion by pepsin. Roasted hazelnuts are more commonly consumed than raw ones. Since no studies have characterized gastric digestion protein fragments of raw and roasted hazelnuts nor their IgE binding properties, we compared these aspects of raw and roasted hazelnuts’ gastric digesta obtained by INFOGEST protocol. Their electrophoretically resolved profiles were probed with hazelnut allergic patients’ sera in 1D and 2D immunoblots. Electrophoretic profiles demonstrated pepsin digestion of all hazelnut allergens to varying extents. While 2D immunoblots indicated that roasting slightly reduced allergenicity, IgE ELISA with the pool of sera showed a slight significant (10%) increase in IgE binding in both gastric digesta. Cor a 9 isolated from the raw and roasted hazelnuts, characterized by far and near CD, remained stable after roasting, with preserved IgE reactivity. Its immunoreactivity contribution by inhibitory ELISA was noticeable in raw and roasted hazelnut digesta; its activity was slightly stronger in the roasted preparations. Roasting has a visible impact on proteins; however, it did not affect overall IgE reactivity. Gastric digestion slightly increases the overall IgE reactivity in raw and roasted hazelnuts, and may therefore impact the profiles of allergens and their fragments available to interact with the immune system in the small intestine. © 2022 by the authors.

AB  - This study aimed to investigate the chemical composition of steam distillate essential oiland corresponding hydrolate obtained from S. officinalis grown in Serbia, as well as the influenceof weather conditions (temperature and precipitations) on their chemical profiles. Furthermore,their antimicrobial activity was investigated in vitro. The main compounds in essential oil were cisthujone, followed by camphor, trans-thujone, and 1,8-cineole, while hydrolate was slightly differentfrom the essential oil, with camphor, cis-thujone, and 1,8-cineole as the main compounds. Amongthe eight respiratory-associated microorganisms, Klebsiella oxytoca was the most sensitive to thetested EOs (minimum inhibitory concentration (MIC)/minimal bactericidal/fungicidal concentration(MBC/MFC) were 14.20 and 28.4 µL mL−1, respectively). MIC and MBC values of other testedbacteria ranged between 28.40 and 227.25 µL mL−1 while for Candida albicans MIC/MFC rangedfrom 28.40/56.81 to 56.81–113.63 µL mL−1. Antibiotic susceptibility patterns for the analyzed eightrespiratory-associated microorganisms showed an intermediate level of resistance to commonly usedantibiotics such as ampicillin, levofloxacin, and ciprofloxacin. As a preliminary approach to theantimicrobial profiling of the tested EO, the obtained results revealed that the tested samples possessremarkable antibacterial activities and could be used to develop pharmaceutical formulations as analternative to conventional antibiotic therapy

Uric acid (UA) has been classified as a damage-associated molecular pattern (DAMP) molecule that serves a physiological purpose inside the cell, while outside the cell it can be an indicator of cell damage. Cell damage or stress can be caused by different health problems or by environmental irritants, such as allergens. We can act and prevent the events that generate stress by determining the extent to which cells are under stress. Amperometric calibration measurements were performed with a carbon paste electrode modified with La(OH)3@MWCNT, at the potential of 0.3 V. The calibration curve was constructed in a linear operating range from 0.67 μM to 121 μM UA. The proposed sensor displayed good reproducibility with an RSD of 3.65% calculated for five subsequent measurements, and a low detection limit of 64.28 nM, determined using the 3 S/m method. Interference studies and the real sample analysis of allergen-treated cell lines proved that the proposed sensing platform possesses excellent sensitivity, reproducibility, and stability. Therefore, it can potentially be used to evaluate stress factors in medical research and clinical practice.

AB  - Winter savory (Satureja montana L.) is a well-known spice and medicinal plant with a wide range of activities and applications. Two subspecies of S. montana, subsp. montana and subsp. variegata, were used for the preparation of seven different extracts: steam distillation (essential oil (EO) and hydrolate (HY)), subcritical water (SWE), ultrasound-assisted (UAE-MeOH and UAE-H2O), and microwave-assisted (MAE-MeOH and MAE-H2O) extraction. The obtained EOs, HYs, and extracts were used for an in vitro evaluation of the antioxidant activity (DPPH, ABTS, reducing power, and superoxide anion methods) and in vitro antimicrobial activity against Bacillus cereus, Staphylococcus aureus, Enterococcus faecalis, Escherichia coli, Salmonella Typhimurium, Saccharomyces cerevisiae, and Candida albicans. The antimicrobial screening was conducted using disk-diffusion assessment, minimal inhibitory concentration, time–kill kinetics modeling, and pharmacodynamic study of the biocide effect. The total phenolic content (TPC) was highest in EO, followed by SWE, MAE, and UAE, and the lowest was in HY. The highest antimicrobial activity shows EO and SWE for both varieties, while different UAE and MAE extracts have not exhibited antimicrobial activity. The natural antimicrobials in the S. montana extract samples obtained by green extraction methods, indicated the possibility of ecologically and economically better solutions for future in vivo application of the selected plant subspecies.

AB  - Allium ursinum and poisonous adulterants Convallaria majalis and Arum maculatum were used as a model for detection of adulterants in edible plant. A. ursinum samples were spiked with C. majalis and A. maculatum to mimic adulteration. Metabolomic fingerprinting of all samples was performed using 1H NMR spectroscopy, and the resulting data sets were subjected to multivariate data analysis. As a result of this analysis, signals of adulterants were extracted from the data, and the structures of biomarkers of adulteration from partially purified samples were elucidated using 2D NMR and LC-MS techniques. Thus, isovitexin and vicenin II, azetidine-2-carboxylic acid, and trigonelline indicated adulteration of A. ursinum samples with C. majalis. Isovitexin was also recognized to be an indicator of adulteration of A. ursinum with A. maculatum. In conclusion, the case study of A. ursinum suggested that plant metabolomics approach could be utilized for identification of low molecular weight biomarkers of adulteration in edible plants. © 2022 by the authors.

AB  - Raspberries are considered valuable fruits due to their high levels of nutrients and phytochemicals, which have many beneficial effects on humans. As many external factors affect the composition of these fruits (the type of cultivation, soil characteristics, ripeness, storage time and post-harvest technologies, cultivar/genotype, and climatic conditions), the goal of this study was to analyze different raspberry cultivars grown in Norway. Considering that Norway is a country with specific climatic conditions, as well as has a limited period of fruit vegetation, another important goal of this study was also to compare raspberries from different Norwegian areas, as well as different grown cultivars. Modern analytical techniques, such as high-performance anion-exchange liquid chromatography with pulsed amperometric detection (HPEAC-PAD), ultra-high-performance liquid chromatography with diode array detector coupled to triple quadrupole mass spectrometry (UHPLC-DAD MS/MS), and inductively coupled plasma–optical emission spectrometry (ICP-OES), provided a detailed examination of the raspberry extract samples. Based on their high levels of minerals (especially N, P, and K), organic acids (predominantly citric and malic acids), sugars (glucose, fructose, sucrose, and galactose), and polyphenols (ellagic acid, syringic acid, quercetin, and rutin), Norwegian raspberries could be considered fruits with increased health-beneficial compounds. AB  - Most of the food allergens sensitized via the gastrointestinal tract resist thermal treatments and digestion, particularly digestion by pepsin. Roasted hazelnuts are more commonly consumed than raw ones. Since no studies have characterized gastric digestion protein fragments of raw and roasted hazelnuts nor their IgE binding properties, we compared these aspects of raw and roasted hazelnuts’ gastric digesta obtained by INFOGEST protocol. Their electrophoretically resolved profiles were probed with hazelnut allergic patients’ sera in 1D and 2D immunoblots. Electrophoretic profiles demonstrated pepsin digestion of all hazelnut allergens to varying extents. While 2D immunoblots indicated that roasting slightly reduced allergenicity, IgE ELISA with the pool of sera showed a slight significant (10%) increase in IgE binding in both gastric digesta. Cor a 9 isolated from the raw and roasted hazelnuts, characterized by far and near CD, remained stable after roasting, with preserved IgE reactivity. Its immunoreactivity contribution by inhibitory ELISA was noticeable in raw and roasted hazelnut digesta; its activity was slightly stronger in the roasted preparations. Roasting has a visible impact on proteins; however, it did not affect overall IgE reactivity. Gastric digestion slightly increases the overall IgE reactivity in raw and roasted hazelnuts, and may therefore impact the profiles of allergens and their fragments available to interact with the immune system in the small intestine. © 2022 by the authors.

AB  - Lipophilicity and ionization constant of sixteen fentanyl analogues were investigated by reversed-phase thin-
layer chromatography (RP-TLC). Fourteen compounds have nitrogen containing groups at C-3 position of the
piperidine ring and two compounds are 3-carbomethoxy derivatives of fentanyl. Among them, five are diaste-
reomeric cis/trans couples and six novel trans diastereomers. In addition, the lipophilicity and ionization constant
of fentanyl were also determined by the same method, as a reference. The physicochemical property, lip-
ophilicity, expressed as retention indices RM
0 , b, and C0, as well as PC1, was determined and correlated with in
silico values. Ionization constants were determined on the basis of the relationships between analyte’s retention
expressed as RF and mobile phase pH. Calculated structural descriptors together with the retention indices, were
subjected to the principal component analysis – PCA and hierarchical cluster analysis – HCA, in order to provide
basic insights into the similarities among the studied compounds. The blood – brain barrier (BBB) permeation
was investigated in the function of experimentally obtained values of lipophilicity, polar surface area and mo-
lecular weight. In general, results of the present research corroborate well with previously determined anti-
nociceptive activities of the investigated compounds, pointing out that besides the cis/trans isomerism, another
set of important parameters should be taken into account when designing new derivatives of C-3 substituted
fentanyl, especially lipophilicity, voluminosity and HBD/A character of the substituents. Accordingly, RP-TLC
can be considered as a valuable asset in the ligand-based drug design

AB  - Strawberries are an important fruit in the European diet because of their unique taste and high content of essential nutrients and bioactive compounds. The anthocyanins are known to be colorful phenolics in strawberries. In 17 samples of six strawberry cultivars produced in Serbia, i.e., the common varieties Alba, Asia, and Clery as well as promising breeding materials (11.29.11, 11.34.6, and 11.39.3), the anthocyanin profile as well as antimicrobial and antioxidative activity profiles were determined. All investigated extracts showed antioxidative and antibacterial activities against Gram-negative Aliivibrio fischeri. The responses were quite similar in number and intensity. The HPTLC-DPPH• scavenging assay and HPTLC-Aliivibrio fischeri bioassay coupled with high-resolution mass spectrometry identified pelargonidin-3-O-glucoside (Pg-3-glc) as the main anthocyanin and prominent antioxidative and antimicrobial compound in strawberries. The density functional theory calculations at the M06-2X/6-31+G(d,p) level showed that Pg-3-glc quenches free radicals via sequential proton loss electron transfer mechanism in water and in pentyl ethanoate, where the 5-OH group is the most reactive site for proton and hydrogen atom transfer. The results were confirmed via spectrophotometry. The highest total phenolic content was found in Clery and 11.39.3, while statistically significant differences between the genotypes regarding the antioxidant activity were not confirmed. Although very similar in the anthocyanin, antioxidative, and antimicrobial profile patterns, the strawberry genotypes were successfully classified using principal component analysis.

AB  - The main objectives of this research were to (i) investigate the concentration; (ii) characterize the distribution;
(iii) determine the sources apportionment; (iv) estimate environmental and health risks of heavy metals in soil
from mountain beech forest. A total of 76 soil samples from 20 pure beech forest stands from Bosnia and Her-
zegovina (BA), Bulgaria (BG), Check Republic (CZ), Germany (DE), Italy (IT), Poland (PL), Romania (RO), Serbia
(RS), Slovakia (SK), Slovenia (SL), and Spain (ES) were collected. The content of major elements was measured
by X-ray fluorescence spectroscopy (XRF). The content of heavy metals was measured by inductively coupled
plasma–optical emission spectrometry (ICP/OES). Heavy metals had a specific concentration range, which fol-
lowed in soil samples from depth 0–40 cm the common order (low to high): Hg < Cd < As < Co < Pb < Ni < Cu
< Cr < Zn, and from depth 40–80 cm: Hg < Cd < As < Pb < Co < Ni < Cu < Cr < Zn. The grouping of the
examined parameters according to rock types, soil types, and localities indicated the separation of carbonate
rocks from other substrates, luvisol, and rendzina from other soil types, and samples from BA, SL, and IT from
other localities. According to sources apportionment As, Pb and Zn are of anthropogenic origin, Cd, Co, Cr, and Ni are of geogenic origin, while the middle position of Cu and Hg indicates a combined contribution of both
sources. Elements Cd and Hg indicated severe to extremely severe enrichment with a mean value of 24.3 and
70.6, respectively. Based on the determined values Ni, Cr, As and Cd do not pose a health risk.

One of them, Ocimum basilicum L., commonly known as basil, has a vital role due to its various medicinal goods. It is best known as a plant with pharmacological activities, but also as an antioxidant, antimicrobial, and larvicidal agent. Although it has been traditionally used in Serbia in traditional medicine for centuries as an insecticidal, antibacterial, and antifungal plant as well as a traditional culinary plant, none of the O. basilicum varieties have been commercialised until today. There are significant numbers of information across the world that oils and by-products are part of the global market, but no references to the essential oil composition of Serbian plants were found. Therefore, the objective of this work was to evaluate the antioxidant and antimicrobial potentials of essential oil and hydrolate of two different varieties: O. basilicum var. genovese and Ocimum. basilicum var. minimum originating from Serbia for further industrial production of antimicrobial- and/or antioxidant-valued products. The results of this study confirm that essential oils of O. basilicum var. genovese and var. minimum represent a significant source of bioactive compounds, especially linalool, with a high rate of biological activities. Similar behaviour is observed for hydrolates, which are the by-product of the essential oil distillation process and can be utilised as bioactive-rich waste in further investigation.

AB  - Strawberries are an important fruit in the European diet because of their unique taste and high content of essential nutrients and bioactive compounds. The anthocyanins are known to be colorful phenolics in strawberries. In 17 samples of six strawberry cultivars produced in Serbia, i.e., the common varieties Alba, Asia, and Clery as well as promising breeding materials (11.29.11, 11.34.6, and 11.39.3), the anthocyanin profile as well as antimicrobial and antioxidative activity profiles were determined. All investigated extracts showed antioxidative and antibacterial activities against Gram-negative Aliivibrio fischeri. The responses were quite similar in number and intensity. The HPTLC-DPPH• scavenging assay and HPTLC-Aliivibrio fischeri bioassay coupled with high-resolution mass spectrometry identified pelargonidin-3-O-glucoside (Pg-3-glc) as the main anthocyanin and prominent antioxidative and antimicrobial compound in strawberries. The density functional theory calculations at the M06-2X/6-31+G(d,p) level showed that Pg-3-glc quenches free radicals via sequential proton loss electron transfer mechanism in water and in pentyl ethanoate, where the 5-OH group is the most reactive site for proton and hydrogen atom transfer. The results were confirmed via spectrophotometry. The highest total phenolic content was found in Clery and 11.39.3, while statistically significant differences between the genotypes regarding the antioxidant activity were not confirmed. Although very similar in the anthocyanin, antioxidative, and antimicrobial profile patterns, the strawberry genotypes were successfully classified using principal component analysis.

Among all nine cloned adrenoceptor subtypes and the subsequent development of animal models, a significant target for various neurological conditions treatment is alpha1-adrenergic receptors. 2-{[4-(2-Methoxyphenyl)piperazin-1-yl]alkyl}-1H-benzo[d]imidazoles, their 5 substituted derivatives, and structurally similar, arylpiperazine based alpha1-adrenergic receptors antagonists (trazodone, naftopidil, and urapidil) have been subjects of comparative analysis. Most of the novel compounds showed alpha1-adrenergic affinity in the range from 22 nM to 250 nM. The in silico docking and molecular dynamics simulations, binding data together with absorption, distribution, metabolism, and excretion (ADME) calculations identified the promising lead compounds. The results brought out the conclusions which allowed us to propose a rationale for the activity of these molecules and to highlight six compounds (2-5, 8, and 12) that exhibited an acceptable pharmacokinetic profile to the advanced investigation as the potential alpha1-adrenergic receptor antagonists.

AB  - Adrenergic receptors are among the most studied G protein-coupled receptors. Activation or blockade of these receptors is a major therapeutic approach for the treatment of numerous disorders such as cardiac hypertrophy, congestive heart failure, hypertension, angina pectoris, cardiac arrhythmias, depression, benign prostate hyperplasia, anaphylaxis, asthma, and hyperthyroidism. Among all nine cloned adrenoceptor subtypes and the subsequent development of animal models, a significant target for various neurological conditions treatment is alpha1-adrenergic receptors. 2-{[4-(2-Methoxyphenyl)piperazin-1-yl]alkyl}-1H-benzo[d]imidazoles, their 5 substituted derivatives, and structurally similar, arylpiperazine based alpha1-adrenergic receptors antagonists (trazodone, naftopidil, and urapidil) have been subjects of comparative analysis. Most of the novel compounds showed alpha1-adrenergic affinity in the range from 22 nM to 250 nM. The in silico docking and molecular dynamics simulations, binding data together with absorption, distribution, metabolism, and excretion (ADME) calculations identified the promising lead compounds. The results brought out the conclusions which allowed us to propose a rationale for the activity of these molecules and to highlight six compounds (2-5, 8, and 12) that exhibited an acceptable pharmacokinetic profile to the advanced investigation as the potential alpha1-adrenergic receptor antagonists.

AB  - Quality control, nutritional value and the monitoring of hazardous residues in honey bee- products have become
major topics for both producers and consumers. Due to its potential role in human health, bee-products rich in
bioactive compounds are becoming increasingly popular. This review aims to provide an overview of thin-layer
chromatography methods used in quality control, authenticity testing and chemical profiling of bee-products in
order to help scientists engaged in the field of bee-products chemistry to utilize the advantages of this technique
in the detection and elimination of fraudulent practices in bee-product manufacturing. Recently, hyphenation of
thin-layer chromatography, image analysis and chemometrics support bee-products analysis by simultaneous determination of analytes with different detection principles, identification of individual
bioactive compounds as well as structure elucidation of compounds. Highlighted opportunities of thin-layer
chromatography could encourage further investigations that would lead to improvements in the detection and
elimination of marketing fraudulent practices.

Fast detection of apricot product frauds by added pumpkin via planar chromatography and chemometrics: Greenness assessment by analytical eco-scale

AB  - The European Commission requires that fruit products distributed on the market meet standards of high quality and authenticity. For the quality assessment of apricot products susceptible to food fraud, an environmentally friendly, simple and cost-effective analytical profiling was developed by high-performance thin-layer chromatography multi-imaging (HPTLC-FLD/Vis). The new phytochemical profiling was applied for analysis of authentic samples (7 apricot and 5 pumpkin samples) and simulated adulterated products (11 mixture samples prepared by addition of 2.5–53% pooled pumpkin to pooled apricot). Based on the analytical eco-scale assessment, the HPTLC-FLD/Vis method was proven as an excellent green analytical method with low energy and solvent consumption. Chemometric data analysis confirmed the difference between apricot and apricot-pumpkin mixtures based on the phytochemical profile. Chemical markers responsible for their differentiation were identified. The results indicated that frauds by adding pumpkin to apricot products can be detected at added contents as low as 2.5%.
