Cytotoxicity of some platinum(IV) complexes with ethylenediamine-N,N'-di-3-propionato ligand

AB  - The cytotoxicities of two platinum(IV) complexes of formula [PtX(2)(eddp)] - nH(2)O (eddp = ethylenediamine-N,N'-di-3-propionate, X = chloro [11 or bromo [11], n = I or 1.24) are reported. The complexes have been obtained by direct reaction of potassium hexahaloplatinate(IV) with H(2)eddp.2HCl followed by addition of a base (LiOH). The crystal and molecular structure has confirmed that the complex with bromo ligands, similarly to the complex with chloro ligands previously reported, has trans configuration of the halogens. In both the chloro and bromo complexes there appear to be intramolecular N-H... X interactions which account for a narrowing of the corresponding X-Pt-N angles below 90degrees. The trans isomer (configuration index OC-6-13, two nitrogens and two oxygens of eddp bound in the equatorial plane) is the only one obtained in the reaction of hexahaloplatinate(IV) with the eddp ligand while a similar reaction performed with ethylenediamine-N,N'-diacetate (edda) affords exclusively the symmetrical cis-isomer (configuration index OC-6-33, equatorial nitrogen and axial oxygen atoms of edda). The longer chain of the propionato groups (as compared to the acetato ones) is responsible for such a change in preferred configuration. NMR data have revealed a very large diastereotopic splitting of the propionato methylene protons a to the nitrogens (0.88 ppm). The trans disposition of the halogen ligands in the compounds with eddp leads to deactivation of platinum(IV) complexes in comparison to those with edda having cis disposition of the leaving chlorides (human ovarian cancer cell line A2780, IC(50) [muM] of 92.6 +/- 12 and 30.3 +/- 7.5 for [1] and [11], respectively). AB  - The title compound was obtained by a template synthesis from 2,6-diacetylpyridine, ethylhydrazinoacetate and CuCl2 . 2H(2)O (molar ratio 1 : 2 : 1, in methanol), and its structure has been determined by single-crystal X-ray diffraction: monoclinic space group P2(1)/c, a = 13.906(5), b = 18.199(6), c = 16.641(6) Angstrom, beta = 107.18(3)degrees and z = 4. The ligand was found to be tridentate in a trigonal-bipyramidal coordination geometry with two chloride ligands. There are two independent complex molecules, one of which is hydrogen bonded to the water of crystallization.

Copper(II) complexes of usnic acid condensation products and their antibacterial activities

AB  - The complexes of Cu(II) and condensation products of usnic acid with the acyl hydrazides of alpha-naphthoic, caprylic and oxamic acid, as well as the-derivative of usnic acid with thiosemicarbazide have been synthesized and characterized by elemental analyses, molar conductivity, magnetic moment measurements and IR spectroscopy. It was found that the derivatives act as tridentates occupying three coordination places in the square planar molecules with the fourth coordination site occupied by a solvent molecule. The complexes were tested for in vitro growth inhibitory activity against the bacterial species Escherichia coli and Staphylococcus aureus. The complexes have higher antibacterial activities compared to both the usnic acid derivatives and Cu(II) ions.

One of them was identified as the title complex. The infrared and electronic absorption spectra, as well as the magnetic momentum confirm a binuclear structure and the existence of hydroxo and O, O′-glycinato bridges.

The crystal structure of the complex has also been determined. The cobalt atom is seven coordinated with approximately pentagonal-bipyramidal geometry. The H2O and methanol are axial with the planar pentadentate H(2)L ligand forming the equatorial plane. The possible reaction mechanism has been proposed.
